Abstract

Piroxicam belongs to BCS class II and has low solubility. Self-nanoemulsifying drug delivery systems (SNEDDS) are considered a potential approach for increasing the solubility and release of piroxicam. This study aimed to select the components and component ratios of piroxicam SNEDDS using fractional factorial design 26-2 (FFD). The variables used in the DFT development of piroxicam SNEDDS are the type and concentration of oil (triacetin and oleic acid), surfactant (kolliphor EL and Tween 60), and co-surfactants (Transcutol and PEG 400). The FFD results showed 16 runs with different proportions of the piroxicam SNEDDS components, which were then characterized by critical parameters including emulsification time, %transmittance, droplet size, and drug loading. The components and component ratios of the PKM SNEDDS were determined using single-factor plot analysis. The results showed that triacetin (oil), kolliphor EL (surfactant), Transcutol (co-surfactant) had the greatest contribution to the formation of piroxicam SNEDDS with an oil ratio range of 11.11–28.57%, surfactant 44.44–77.78%, co-surfactant 11.11–44.44 %.

Abstract

Latar belakang: Piroksikam salah satu obat anti inflamasi termasuk dalam golongan BCS kelas II yang memiliki kelarutan yang rendah pada air. Kelarutan piroksikam dapat ditingkatkan dalam bentuk Self Nano Emulsifying Drug Delivery System (SNEDDS). Pada SNEDDS tersusun dari beberapa komponen seperti minyak, surfaktan, dan kosurfaktan yang berpotensi menggangu hasil analisa seperti pada analisa kadar piroksikam.. Tujuan: Pada penelitian ini bertjuan melakukan validasi metode analisa piroksikam dalam formulasi SNEDDS menggunakan spektrofotometer UV-Vis. Metode: parameter International Conference on Harmonization (ICH) yaitu spesifisitas, linearitas, akurasi, presisi, LOD, dan LOQ. Hasil: parameter validasi memenuhi kriteria antara lain spesifitas yang baik, nilai akurasi pada rentang 90-107 %, nilai presisi %RSD <2%, Nilai koefisien korelasi pada linieritas >0,997, dan LOD sebesar 0,256 µg/ml dan LOQ sebesar 0,778 µg/ml.. Simpulan: Analisa piroksikam dalam SNEDDS dengan menggunakan metode spektrofotometri UV-Vis tervalidasi berdasar dari parameter spesifisitas, linearitas, akurasi, presisi, LOD, dan LOQ.

Abstract

Aktivitas fisik yang dilakukan dalam olahraga selain berdampak pada kebugaran tubuh juga beresiko menimbulkan cidera. Pharmaceutical care di bidang olahraga berkontribusi dalam perkembangan olahraga modern. Besarnya manfaat tersebut juga dibarengi masalah penyalahgunaan dan penggunasalahan obat karena terbatasnya pemahaman. Pengabdian ini bertujuan meningkatkan pengetahuan peserta tentang penggunaan obat, suplemen dan herbal pada olahragawan. Kegiatan pengabdian dilakukan dengan pendekatan penyuluhan kelompok dengan sasaran anggota komunitas olahraga. Kegiatan meliputi pengisian pre test, penyampaian materi, diskusi tanya jawab dan pengisian post test. Pre dan Post test berisi pertanyaan tentang pengetahuan DAGUSIBU peserta. Kegiatan pengabdian masyarakat ini memberi dampak pada peningkatan pengetahuan penggunaan obat, supplemen, dan herbal pada olahragawan. Peningkatan pengetahuan tertinggi terjadi pada domain cara mendapatkan dan menggunakan obat.

Abstract

Introdiction: Piroxicam is Non-Steroidal Anti-Inflammatory Drug that belongs to the BCS Class 2, characterized by low solubility. The SNEDDS (Self-Nanoemulsifying Drug Delivery System) approach for piroxicam is believed to enhance its solubility and accelerate drug release. Aims: This study aims to demonstrate that SNEDDS piroxicam has a faster drug release rate compared to piroxicam powder and commercially available piroxicam capsules. Methods: SNEDDS formulation equivalent to 20 mg of piroxicam, containing a combination of oleic acid, Kolliphor EL, and Transcutol in a ratio of 2:7:4, was tested for drug release using a dissolution method in simulated gastric fluid without enzymes at pH 1.2. The concentration of dissolved drug was measured using a validated spectrophotometric method. Result: At the 45th minute, the dissolution rate of piroxicam SNEDDS reached 101.525%, significantly higher than 47.550% achieved by piroxicam powder and 87,081 % commercially piroxicam capsules at the same time. Additionally, the dissolution efficiency of piroxicam SNEDDS is superior, with a rate of 85.539%, compared to 34.510% for piroxicam powder and 66.17% for commercially available piroxicam capsules. Conclusion: The development of a Self-Nanoemulsifying Drug Delivery System (SNEDDS) for piroxicam has shown promising potential to improve solubility and drug release, as demonstrated by superior in-vitro release rates compared to piroxicam powder and commercial piroxicam capsules
