Abstract

Cabe jawa (Piper retrofractum Vahl.) merupakan tanaman herbal yang mengandung alkaloid utama piperin. Prospek cabe jawa sebagai bahan obat belum didukung oleh ketersediaan bahan baku yang cukup, karena rendahnya produktivitas tanaman. Salah satu alternatif untuk mengatasi permasalahan tersebut adalah dengan menerapkan kultur kalus. Kalus dapat memproduksi metabolit sekunder relatif cepat dan berkelanjutan. Efektivitas produksi metabolit sekunder seperti piperin pada kalus dapat dilakukan dengan meningkatkan konsentrasi sukrosa dalam medium kultur. Penelitian ini dilakukan untuk mengetahui pengaruh konsentrasi sukrosa pada morfologi kalus, pertumbuhan kalus, dan produksi piperin pada kalus cabe jawa. Medium Murashige and Skoog (MS), dan zat pengatur tumbuh naphtalene asetic acid (NAA) dan benzyl aminopurin (BAP) 1:2 digunakan untuk menginduksi kalus dari daun cabe jawa. Kalus yang terbentuk berwarna hijau muda dan kompak. Kalus disubkultur selama 35 hari pada medium MS dengan konsentrasi sukrosa 30 g/L, 40 g/L, 50 g/L, dan 60 g/L. Setelah inkubasi warna kalus berubah dengan tekstur tetap kompak. Berat segar dan berat kering dari kalus menurun dengan bertambahnya konsentrasi sukrosa. Piperin pada kalus diekstraksi dengan etanol 96% dan diukur kadarnya dengan Kromatografi Lapis Tipis (KLT)-Densitometri. Peningkatan konsentrasi sukrosa pada medium kulur tidak berpengaruh pada kadar piperin dari kalus cabe jawa.

Abstract

Obesity increases the risk of various diseases. Black rice, renowned for its high anthocyanin content, is considered a potential functional food for preventing metabolic disorders. The current study investigated the effects of black rice crunch (BRC) on body weight and haematological profiles in obese rats. Rats were fed with high-fat diet to induce obesity and supplemented with different concentrations of BRC for 4 and 8 weeks. The results showed that high-fat diet effectively induced obesity, as evidenced by significant increase in body weight. Importantly, 75% BRC supplementation resulted in significant weight reduction in obese rats. Further analysis revealed an increase in erythrocyte numbers in obese groups supplemented with 75% BRC, but no significant changes in haemoglobin concentration or haematocrit percentage. Further investigation showed that 75% BRC led to a decrease in mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), and mean corpuscular volume (MCV), potentially affecting the size and concentration of haemoglobin within erythrocytes. The total leucocytes count increased with the high-fat diet, while BRC supplementation alone did not have significant impact. Lymphocyte percentage remained stable across the groups, indicating minimal influence of the dietary interventions. Neutrophil percentage varied initially but was not specific to BRC or the high-fat diet. Platelet count and distribution width were not significantly influenced, but mean platelet volume (MPV) increased after 8 weeks of BRC treatment, suggesting larger platelet sizes associated with obesity. Overall, the study provides important insights into the effects of BRC supplementation on body weight and haematological parameters related to obesity.

Abstract

Gyrinops versteegii (Gilg.) Domke is one of the Thymelaeaceous family species with high economic value. The leaf synthesizes secondary metabolites (SMs) supporting herbal medicine, including a class of SMs that serve as sunscreen. However, the SMs are not equally distributed in the plant, depending on the organs, tissues or cell type. This research aimed to determine the spatial distribution of secondary metabolites in the leaves, especially in the epidermis and mesophyll tissues. The epidermis tissues were separated from the mesophyll using the carborundum abrasion (CA) technique, followed by the extraction of SMs using three different polarity solvents, namely methanol, hexane and chloroform. The photoprotective potential was evaluated using the SPF value with the spectrophotometry approach. Subsequently, the phytochemical analysis was done for the total tannin, flavonoid, and phenols content. The results showed that the highest SPF value was observed in the methanol epidermis extract with SPF value of 8.61, followed by methanol mesophyll extract (4.52), chloroform epidermis extract (3.97), chloroform mesophyll extract (2.52) and hexane epidermis extract (0.60) and hexane mesophyll extract (0.16). Total phenolic content (flavonoid, tannin and phenol) in epidermis extract with methanol solvent was higher (12.347 ± 0.652 mg GAE/g DW), followed by extract with chloroform and hexane solvents. The total phenolic content (flavonoid, tannin and phenol) of mesophyll extract with methanol solvent showed the highest content, followed by chloroform mesophyll and hexane extracts. The results of the histochemical analysis showed that photoprotective compounds accumulate in the epidermis and mesophyll tissue of the leaf of G. versteegii.
