Abstract

Spons merupakan fauna laut yang diketahui memiliki berbagai senyawa bioaktif. Senyawa tersebut berpotensi sebagai antibakteri, antivirus, dan antikanker. Penelitian ini bertujuan mempelajari aktivitas sitotoksik dan apoptosis ekstrak spons spesies A anggota ordo Astrophorida terhadap sel HeLa. Pada penelitian ini pengujian aktivitas sitotoksik ekstrak etanolik, metanolik, dan kloroform spons spesies A terhadap sel HeLa dilakukan menggunakan MTT assay dan uji apoptosis menggunakan double staining, yaitu etidium bromida-acridine orange. Deteksi golongan senyawa yang terkandung di dalam spons spesies A dilakukan menggunakan Kromatografi Lapis Tipis (KLT). Hasil uji sitotoksisitas menunjukkan bahwa ekstrak etanolik, metanolik, dan kloroform spons spesies A masing-masing memiliki nilai IC50 sebesar 18,25; 27,87; dan 13,87 µg/mL. Ekstrak etanolik, metanolik, dan kloroform spons spesies A pada konsentrasi 31,25 µg/mL dapat menginduksi kematian sel melalui apoptosis masing-masing sebesar 35,3 ± 11,16%; 82,64 ± 16,21%; dan 86,76 ± 9,27%. Berdasarkan uji menggunakan KLT diketahui bahwa spons spesies A menggandung golongan senyawa alkaloid, flavonoid, fenol, dan terpenoid. Oleh karena itu, dapat disimpulkan bahwa ekstrak spons spesies A berpotensi untuk dikembangkan sebagai obat antikanker.Kata kunci: ekstrak spons spesies A, sitotoksik, apoptosis, sel HeLa

Abstract

Effect of ethanolic extract of cashew leaf (Anacardium occidentale L.) in the feed on the weight gain and immunity were investigated on Jawa Super Chicken. Ninety chicken were divided into 6 groups those were basal feed group and 5 different concentration of chasew leaf extract groups. Experiment was conducted for 16 days started by 2 days acclimatization after hatching. Body weight was measured every 3 days starting from 0th day. On 16th day, the chicken were sacrificed, spleen and bursa of Fabricius were collected to quantify some histological parameters such as white pulp and follicle area as well as follicle thickness of those organs. The data were analyzed by regression and one way ANOVA followed by Tuckey test. Histological analysis was performed using optilab and Image Raster software. The results showed that growth and development of immune system of chicken increased significantly as could be seen in body weight, organs weight and index, as well as histological parameters. The regression results showed that spleen and bursa of Fabricius weight influenced increasing of body weight by 64.2% and 38.4% respectively. It could be concluded that biosupplementation of cashew leaf extract in basal feed could increase growth and immunity of Jawa Super Chicken.

Abstract

λ-cyhalothrin is phyrethroid insecticide that can pollute the waters environment and cause sublethal as well as lethal effects to non-target organisms such as tilapia. This research were aimed to study the LC50 96 h value and histopathological effects of λ-cyhalothrin on the gills, liver, and small intestine of tilapia. In this research fish were divided into control group and the treatment groups of λ-cyhalothrin at the concentration of 3, 6, 9, and 12 µg/L. The treatment used a static system for 96 hours. The LC50 96 h value was analyzed by linear regression. Histopathological effects on the organs were analyzed using descriptive comparative. Histological preparation was done by the paraffin method, fixed in Bouin solution, and stained with Hematoxylin-Eosin. The results showed that LC50-96 h values of λ-cyhalothrin on tilapia was 6.519 µg/L. Gills damages were in the form of hyperplasia, necrosis, edema, secondary lamella fusion, removal of lamella epithelial cells, and primary lamella dilatation. Liver damages include hemorrhage, intravascular hemolysis on blood vessels, piknosis, and leukocytes infiltration. The intestine damages were fatty degeneration, edema, leukocyte infiltration, hemorhage, cloudy swelling, vacuolisation, necrosis, and arthropy. The gills, liver, and small intestine damages of tilapia range from slight to severe. It could be concluded that λ-cyhalothrin caused histopathological effects on the gills, liver, and small intestine of tilapia.

Abstract

A bat is the only mamal with true flight ability and have capability to perform echolocation. Miniopterus  and Rhinolopus  are bat species that have some different morphological characteristics but both of them commonly live in the cave. The research were purposed to study the differences and connection of echolocation call type and larynx’s structure of Miniopterus and Rhinolopus  The two bat species were collected from 20th and 23th cave of Japanese Cave Complex, Kaliurang, Sleman, Yogyakarta. Specimens were taken to the laboratory for further analysis. Echolocation signals of both bat species were recorded using ultrasonic microphone and were analyzed with Batscan 9.0 software. Specimens were subjected for anatomical and histological structure analysis of the larynx. Anatomical structure observations were carried out with specimens dissection and they were fixed in 20% formol solution. Some samples of larynx of each species were processed and stained with Alizarin Red’s-Alcian Blue staining for larynx’s bones observation. Histological preparation of larynx was performed using paraffin method and stained with Hematoxylin-Eosin and MAF. The results showed that echolocation signals of Minipterus  have lower frequency (peak frequency between 44.1-51.6kHz) compared to that of Rhinolopus  (peak frequency between 54.4-64.1 kHz). However, Miniopterus  has better ability to modulate the signal frequency than Rhinolopus  Both species are able to produce a long-duration signal but Rhinolopus  can produce longer signal duration. These results can be related to the differences of larynx’s anatomical and histological structure of both species which are equally able to keep the intensity of echolocation signals but have different work mechanisms.

Abstract

During the COVID-19 outbreak, economic sectors slowed down and caused a significant impact on almost any level of society, especially the middle to lower class income. Thus, a strategy needs to be developed to overcome this situation. The Ngoro-Oro Village has potency in the livestock sector, especially in the development of superior hybrid Jawa super chicken. Therefore, the purposes of our community service in the Ngoro-Oro Village were to develop superior hybrid Jawa super chicken and use black soldier fly larvae as an alternative feed for chicken. We distributed 105 Jawa super hens aged ± three months and 21 superior free-range roosters. Furthermore, the hens and rooster were mated at the age of ± six months with a ratio of 5:1. The hybrid chickens were evaluated for their body weight and morphometric measurements until the age of four months. We also monitored the farmer community’s use of black soldier fly larvae as an alternativ e feed for chickens. The hybrid chicken at the age of 4 months old has a body weight of about 971.64 ± 271.06 grams with a height of about 41.73 ± 5.87 cm, head’s length of 5.01 ± 0.44 cm, body length of 22.82 ± 3.14 cm, body width of 10.68 ± 1.49 cm, wing’s length 19.14 ± 2.67 cm, thigh’s length 11.95 ± 1.71 cm, and leg’s length is about 7.91 ± 1.41 cm. Moreover, Ngoro-Oro farmers can improve the community’s economy by selling superior hybrid Jawa super chicken at a price 75,000 – 85,000 rupiahs per head. Through all of this community service program, it could be concluded that superior hybrid Jawa super chicken is potential to be developed as broiler, and the farmer community could develop black soldier fly larvae as an alternative feed for chicken.

Abstract

Kelelawar memiliki berbagai peran penting baik secara ekologis maupun ekonomis yaitu dalam pengendalian hama, siklus nutrien, pollinator, dan penghasil guano. Namun, kelelawar mengalami ancaman kepunahan yang disebabkan karena kerusakan lingkungan maupun perburuan liar, sehingga upaya konservasi sangatlah penting. Upaya ini dapat dilakukan melalui pendekatan studi struktur histologis lidah untuk memahami kesukaan pakan binatang tersebut. Di Yogyakarta, Indonesia, terdapat dua spesies kelelawar, Rhinolophus pusillus dan Miniopterus schreibersii yang hidup di habitat sama. Penelitian ini bertujuan untuk mengkaji struktur histologis lidah R. pusillus dan M. schreibersii. Tiga ekor kelelawar pada masing-masing spesies ditangkap menggunakan sweep net dan mist net, selanjutnya kelelawar dianastesi menggunakan kloroform dan dikorbankan. Lidah dikoleksi dan difiksasi dalam neutral buffered formalin 10%. Preparasi histologis dilakukan menggunakan metode parafin dengan pewarnaan Hematoxylin Eosin dan tebal irisan 6 µm. Data histologis dianalisis secara deskriptif komparatif. Hasil menunjukkan bahwa struktur histologis lidah R. pusillus dan M. schreibersii serupa. Secara histologis, lidah kelelawar tersebut tersusun atas 3 lapis yaitu tunika mukosa, tunika submukosa, dan tunika muskularis. Pada bagian lapisan epitel pipih berlapis berkeratin terdapat tonjolan papila filiformis, fungiformis, dan sirkumvalata. Tunika muskularis tersusun dalam 3 orientasi. Pada lidah kedua spesies tersebut juga terdapat kelenjar ludah dengan sel-sel penghasil kelenjar serosa dan mukosa. Kelenjar ludah R. pusillus terdapat di bagian sublingual dan anterior lidah, sedangkan kelenjar ludah M. schreibersii terdapat di bagian antero-inferior dan posterior lidah. Berdasarkan hasil tersebut dapat disimpulkan bahwa kedua spesies kelelawar memiliki struktur lidah yang serupa sehingga dapat mengembangkan pola adaptasi yang serupa terhadap pakan.

Salah satu cara yang dapat dilakukan adalah menyilangkan ayam betina Jawa super dengan ayam jantan Pelung yang diketahui memiliki pertumbuhan lebih cepat dibandingkan ayam lokal. Hasil persilangan tersebut diharapkan memiliki pertumbuhan cepat dan berpotensi sebagai ayam pedaging dengan karakter lokal. Tujuan penelitian ini adalah mempelajari pertumbuhan, morfometri tubuh, dan fenotipe ayam Super pelung (SUPEL) hasil persilangan ayam betina Jawa super dengan ayam jantan Pelung. Parameter yang diukur adalah berat badan, morfometri tubuh, dan fenotipe ayam Supel. Selain itu dilakukan pengukuran berat, warna, morfometri, dan analisis proksimat telur ayam Jawa super. Hasil menunjukkan rerata berat badan ayam Supel saat Day Old Chicken (DOC) sebesar 33,57 ± 2,44 g dan mencapai 1.164,00 ± 98,20 g pada umur 3 bulan. Fenotipe anak ayam Supel menunjukkan ciri-ciri kombinasi dari kedua indukan. Sementara itu hasil pengukuran berat telur ayam Jawa super diperoleh rerata sebesar 43,15 ± 3,57 g. Berdasarkan hasil yang diperoleh dapat disimpulkan bahwa ayam Supel hasil persilangan memiliki pertumbuhan yang cepat dan memiliki karakter fenotipe seperti ayam lokal.AbstractThe increasing demands of Broiler chicken meat in the market lower farmers' interest in the local chicken breed. One of the solutions is to crossbreed between Jawa super hens and Pelung roosters with faster growth than other local chickens. The crossbreed chicken (Supel chicken) is expected to have faster growth and the potential as Broilers with local characteristics. The purpose of this study is to evaluate growth, body morphometry, and the phenotype of Super pelung (SUPEL) chicken. Parameters measured were body weight, body morphometry, and phenotype of Supel chicken. The measurements of weight, color, morphometry, and proximate analysis were also conducted on Jawa super egg. The results showed that the weight of Supel chicken during Day Old Chicken (DOC) was 33.57 ± 2.44 g and reached 1,164.00 ± 98.20 g in 3 months. The phenotype of Supel chicken showed combination characteristics of the two broods. Meanwhile, the weight of the Jawa super chicken egg average of 43.15 ± 3.57 g. It can be concluded that Supel chicken has a faster growth rate and phenotypic characters resembling Indonesia's local chickens.

Abstract

Ulva lactuca is a functional food ingredient with high antioxidant content. The presence of heavy metal contaminants in natural Ulva lactuca needs to be eliminated. Ulva lactuca was investigated for its potential as a nephroprotector in female Wistar rats. The study assessed Blood Urea Nitrogen (BUN) levels, creatinine levels, and kidney histopathology. The experiment included a Group I (control), Group II (administered 1000 mg/Kg BW/day of natural Ulva lactuca), and Group III (administered 1000 mg/Kg BW/day of heavy metal decontaminated Ulva lactuca using bilimbi juice). After 30 days of oral treatment, no significant differences were observed in BUN levels before and after treatment. However, significant variations were found in Group III compared to the Group I creatinine levels after treatment. Both natural and heavy metal decontaminated Ulva lactuca showed no adverse effects on kidney function. Notably, heavy metal decontaminated Ulva lactuca exhibited improved kidney histopathology, suggesting its potential as a nephroprotector. Further research is needed to explore its broader applications in promoting kidney health.

Abstract

Chlorpyrifos is an organophosphate insecticide, acaricide, and nematicide which is used to control leaf pests and soil pest insects. The insecticide chlorpyrifos works by disrupting the insect's nervous system. The use of insecticides on vegetable crops is generally more intensive than on other food crops so the negative impact on the biotic and abiotic environment becomes greater. The purpose of this study was to determine the negative impact of chlorpyrifos exposure with concentrations of 0.01, 0.1, 1 and 10 ppm on the embryonic development of Wader pari (Rasbora lateristriata). The results of observations at 24 and 48 hours showed that there were abnormalities in the development of the Wader pari’s embryo at the concentration of 0,1, 1 and 10 ppm, indicating that chlorpyrifos causes malformations in the form of defects in the development of the embryo of Wader pari fish.
