Abstract

Ciguatera Fish Poisoning (CFP) merupakan salah satu tipe keracunan pada manusia yang terjadi akibat pengkonsumsian ikan-ikan karang yang telah terkontaminasi ciguatoksin. Toksin tersebut dihasilkan oleh beberapa jenis dinoflagellata bentik yang umumnya ditemukan menempel pada makroalga. Penelitian mengenai inventarisasi dan kelimpahan dinoflagellata bentik pada makroalga di perairan Pulau Tidung telah dilakukan pada tanggal 21 September 2014, mengingat lokasi tersebut telah mengalami tekanan dari berbagai aktivitas manusia. Penelitian dilakukan dengan mengkoleksi makroalga Sargassum dan Padina dari rataan terumbu Pulau Tidung, untuk kemudian dimasukkan ke dalam wadah plastik berisi air laut. Setelah itu, untuk melepaskan dinoflagellata bentik dari makroalga, dilakukan proses pengocokan dan penyaringan dengan saringan bertingkat (125μm dan 20μm). Sampel yang telah disaring kemudian diamati dengan Sedgewick rafter cells di bawah mikroskop. Dari hasil penelitian diperoleh sebelas jenis dinoflagellata bentik, dimana enam diantaranya berpotensi menyebabkan CFP yaitu Amphidinium sp., Ostreopsis lenticularis, O. siamensis, Prorocentrum concavum, P. lima, dan P. rhathymum. Jenis-jenis dinoflagellata toksik yang ditemukan mengindikasikan perlunya dilakukan monitoring di perairan Pulau Tidung.Kata kunci : Dinoflagellata bentik, Ciguatera Fish Poisoning, makroalga, Pulau Tidung

Abstract

Species composition and abundances of phytoplankton in the shellfish farming area in Jakarta Bay, Kali Baru-Cilincing, were examined in order to identify harmful algae species. Phytoplankton contained in the shellfish was also observed to detect the presence of toxic species, as well as the phycotoxin contained in  visceral of green mussel (Perna viridis) collected from the farming area using brine shrimp lethality test (BSLT). We detected the presence of  Alexandrium spp. (PSP causing species), Dinophysis caudata, D. miles and D. rotundata (DSP causing species), and Pseudo-nitzschia spp. (ASP causing species). The last mentioned species was also detected in thestomach content of green mussel. The toxicity of green mussels tested was recorded LC 50 ≤ 1,000 ppm and indicatedthe presence of an active compound in the green mussel viscera. Meanwhile, preliminary PSP test of shellfish did not showed any toxin.

Abstract

The Ciguatera Fish Poisoning (CFP) causing microorganisms were observed at the reef flat of Penjaliran Barat Islands and Pramuka Islands District, Seribu Island National Park, North Jakarta, Indonesia. Of the samples collected, four potentially toxic benthic dinoflagellates species were found, which are Gambierdiscus toxicus, Prorocentrum concavum, Prorocentrum lima, and Ostreopsis lenticularis. The dinoflagellates were found attached on the brown macroalgae Padina and Sargassum. Most of the benthic dinoflagellates from both locations were found in Pramuka Islands District, the inhabitated islands where human activities which could damage the coral reef areas were relatively high. Beside the macroalgae substrate preference of each dinoflagellate species, several environmental factors were also obtained in this research.

Abstract

Abstract

Penelitian tentang Dinoflagellata epifitik pada lamun Enhalus acoroides telah dilakukan di rataan terumbu Pulau Pari, Kepulauan Seribu pada bulan April 2012. Penelitian dilakukan dengan mengoleksi daun lamun Enhalus acoroides dari rataan terumbu, kemudian dimasukkan ke dalam botol plastik berisi air laut.  Setelah itu, dilakukan pengocokan dan penyaringan dengan saringan bertingkat (125µm dan 20µm). Dinoflagellata epifitik yang ditemukan berjumlah 8 jenis, yaitu Gambierdiscus toxicus Prorocentrum concavum, P. emarginatum, P.lima, P. rhathymum, Ostreopsis lenticularis, O. ovata, dan O. siamensis. Enam dari jenis tersebut termasuk Dinoflagellata epifitik yang berpotensi menyebabkan Ciguatera Fish Poisoning.Kelimpahan Dinoflagellata epifitik tertinggi dimiliki oleh Prorocentrum lima (903 sel/cm2), sedangkan kelimpahan terendah dimiliki oleh Gambierdiscus toxicus (1 sel/cm2).Berdasarkan uji korelasi Spearman, parameter lingkungan perairan yang memengaruhi kelimpahan Dinoflagellata epifitik saat penelitian adalah kecepatan arus.

Abstract

Keberhasilan proses transplantasi karang sangat tergantung pada kelimpahan zooxanthella. Semakin padat jumlah zooxanthella dalam karang, maka akan semakin tinggi efisiensi pertumbuhan karang dalam suatu perairan. Cahaya sangat mempengaruhi kelimpahan zooxanthella pada karang, dimana semakin tinggi intensitas cahaya maka semakin besar pula kelimpahan zooxanthella. Penelitian mengenai perbedaan intensitas cahaya terhadap kelimpahan zooxanthella pada karang bercabang di perairan Pulau Pari, Kepulauan Seribu telah dilakukan pada bulan April 2012. Penelitian dilakukan dengan cara menutup ujung cabang koloni karang dengan plastik terang (intensitas cahaya 58 μE/m2s), plastik setengah gelap (intensitas cahaya 26 μE/m2s), dan plastik gelap (intensitas cahaya 0 μE/m2s) selama 4 hari. Zooxanthella dalam fragmen karang dikeluarkan dengan cara dipanaskan, dan kelimpahannya dihitung di bawah mikroskop. Data hasil penelitian ditabulasi dan dianalisis menggunakan uji Regresi Linier dan uji ANAVA satu arah. Hasil penelitian menunjukkan bahwa kelimpahan zooxanthella mengalami penurunan dibandingkan kontrol (1.302.425 sel/cm2) yaitu pada perlakuan 1, perlakuan 2, dan perlakuan 3 dengan jumlah masing-masing 1.201.644 sel/cm2, 933.944 sel/cm2, dan 507.458 sel/cm2. Hasil analisis statistik juga menunjukkan korelasi positif antara kelimpahan zooxanthella dengan kenaikan intensitas cahaya. Kata kunci : kelimpahan zooxanthella, perbedaan intensitas cahaya, dan pulau pari

Abstract

Telah dilakukan penelitian tentang sebaran spasial spesies HAB di lokasi budidaya kerang hijau (Perna viridis), Kamal Muara, Jakarta Utara pada bulan Mei 2011. Sampel diambil secara vertikal di sembilan stasiun dengan plankton-net.  Spesies HAB yang ditemukan berasal dari kelas Dinophyceae (Ceratium furca, Dinophysis caudata, Gonyaulax polygramma, Gonyaulax spinifera, Gymnodinium catenatum, Gymnodinium sanguineum, Prorocentrum micans, dan Prorocentrum sigmoides); Bacillariophyceae (Nitzschia spp., Chaetoceros spp., Skeletonema costatum, dan Thalassiosira spp.) dan Raphidophyceae (Chattonella spp.).  Berdasarkan peta isoplank diketahui bahwa sebaran terpadat pada bulan Mei 2011 adalah di stasiun dekat muara.   Kata kunci : Harmful Algal Blooms, Kamal Muara, kerang hijau, dan sebaran spasial

Abstract

Ciguatera Fish Poisoning (CFP) yang disebabkan oleh Ciguatoksin, dihasilkan oleh beberapa jenis Dinoflagellata epibentik yang umumnya menempel pada makroalga. Dinoflagellata epibentik juga dapat menempel pada substrat lain, seperti pada sedimen atau pecahan karang. Penelitian mengenai Dinoflagellata epibentik telah dilakukan di Pulau Harapan, Kepulauan Seribu pada bulan Maret 2013, di empat stasiun yang terletak di sisi timur, utara, selatan, dan barat pulau. Penelitian ini bertujuan untuk mengetahui spesifisitas substrat Dinoflagellata epibentik pada berbagai macam tipe substrat.Penelitian dilakukan dengan mengoleksi berbagai substrat di rataan terumbu setiap stasiun yaitu pasir, karang mati, lamun Thallasia, dan makroalga Padina, untuk kemudian dimasukkan ke dalam wadah plastik berisi air laut. Setelah itu, untuk melepaskan Dinoflagellata bentik dari substrat, dilakukan proses pengocokan. Sampel kemudian disaring dengan saringan bertingkat (125µm dan 20µm) dan diamati di bawah mikroskop. Data hasil penelitian dianalisis menggunakan Analisis Faktorial Koresponden (AFK) dan Analisis Komponen Utama (AKU).Berdasarkan hasil penelitian, diperoleh dua belas spesies Dinoflagellata epibentik yang tujuh diantaranya merupakan spesies yang berpotensi toksik. Berdasarkan hasil analisis AFK dan AKU diketahui bahwa spesifisitas substrat dari Gambierdiscus toxicus adalah makroalga; Amphidiniopsis hirsutum, Prorocentrum concavum, Coolia sp., dan Amphidinium sp. adalah substrat pasir; Ostreopsis ovata, Ostreopsis lenticularis, dan Prorocentrum rhatymum adalah substrat lamun, karang, dan pasir; Ostreopsis siamensis, Prorocentrum lima, Prorocentrum emarginatum, dan Sinophysis microcephalus adalah substrat lamun, karang, dan makroalga.

Abstract

Benthic dinoflagellates causing Ciguatera Fish Poisoning (CFP), could be found attached either on macroalgae or sea grasses. Research on density and distribution of benthic dinoflagellates on sea grass leaves was conducted in Pari Island waters, Seribu Islands, in April 2012. Research was carried out by collecting Enhalus acoroides leaves from each site, and put inside the plastic jars containing seawater. In order to separate the dinoflagellates species from the leaves, the plastic jars were shaken vigorously and the seawater filtered through a series of sieves (125μm and 20μm). The resulted residue was than observed using Sedgewick rafter cell under light microscope. Based on samples collected, eight benthic dinoflagellates were found, where five of them were potentially toxic. They were Prorocentrum concavum, P. lima, P. rhathymum, Ostreopsis lenticularis, and O. siamensis. The highest abundance was found in the reef flat on the southern side of the island (652 sel/cm2 seagrass leaf). Research showed that the density and distribution of toxic dinoflagellates on sea grass at Pari Island, Seribu Islands were more influenced by local water currents.Keywords: benthic dinoflagellates, Ciguatera Fish Poisoning, Pari Island, seagrass
