Abstract

Abstract

Damage to coral reef ecosystems and changes in water quality are capable of increasing the abundance of benthic dinoflagellates which could cause Ciguatera Fish Poisoning (CFP). Therefore, this research aimed to observe spatial and temporal abundance of those potential benthic dinoflagellates and analyze their relationship with environmental factors. Sampling was carried out on September 21-24, 2023 in Pramuka Island, Seribu Islands district, with different treatments, namely depth, location and time. Research conducted by deploy several series of artificial substrates into the waters, and the benthic dinoflagellates were collected from the artificial substrate placed in 1, 3, and 5 meter of water depths for 24, 48, and 72 hours at Odi, Mazu, and Villa Delima Piers. Cell densities were determined by enumeration using a Sedgewick Rafter Counting Chamber under a light microscope, and the relationship with environmental parameters was analyzed by using Principal Component Analysis (PCA). The results showed that 4 genera of benthic dinoflagellates which had the potential to cause CFP, were attached to the artificial substrate, namely Amphidinium, Coolia, Ostreopsis, and Prorocentrum. Prorocentrum cells were observed in highest number on artificial substrates at every treatment with different depths, locations, and times. Furthermore, the PCA analysis showed the abundance of Prorocentrum was influenced by the most typical environmental parameters at each depth, location, and time, namely temperature, nitrate, DO, and light intensity. The research provided valuable information on the benthic dinoflagellates both spatially and temporally, through international standard methods in order to prevent and anticipate negative impacts caused by CFP.

Abstract

Diatoms are the most common and abundant microalgae group in seawater, with an estimated 200 genera and 200,000 species categorized into three distinct taxonomic classes: Mediophyceae, Bacillariophyceae, and Coscinodiscophyceae. This study describes the morphological characteristics of Lampriscus GM052022, a diatom belonging to the Mediophyceae family, which was collected from Gili Matra, Lombok, Indonesia. Analysis using light microscopy (LM) and electron microscopy (EM) showed that the morphological characteristics of the Lampriscus GM052022 specimen are similar to those of Lampriscus shadboltianus var. crenulatus due to the distinct crenulation lines on the frustule, which differentiate this variation from Lampriscus shadboltianus. However, the presence of distinct morphological features in this diatom specimen from Gili Meno suggests that it might not be L. shadboltianus var. crenulatus. The triangular spines (spinules) observed on the Lampriscus GM052022 valve surface have not been previously documented. However, whether the presence of spinules is a sign of different taxa or a result of polymorphism within the species or variation cannot be confirmed. Due to this characteristic, Lampriscus GM052022 is given the temporary name L. cf. shadboltianus var. crenulatus. The next step will be to conduct genetic analyses to confirm whether these morphological differences support the separation of this Gili Meno Lampriscus specimen into a new species (sp. nov.) or variation (var.).

Abstract

Abstract. Widiarti R, Zamani NP, Bengen DG, Madduppa H. 2022. Molecular characterization of toxic benthic dinoflagellate, Prorocentrum lima in west Indonesian waters using LSU 28S rDNA gene. Biodiversitas 23: 3257-3263. Prorocentrum lima is one of the toxic benthic dinoflagellates known to cause Diarrhetic Shellfish Poisoning (DSP), which is also associated with ciguatoxin-producing species, Gambierdiscus toxicus that causes Ciguatera Fish Poisoning (CFP). P. lima has a wide range of morphological variability and genetic diversity, but such research has never been reported from Indonesian waters yet. This study aimed to determine the molecular characteristics of P. limain west Indonesian waters, namely Bintan Island, Belitung Island, Seribu Islands, and Karimunjawa Islands. Molecular characterization was conducted by amplification on large subunit (LSU)28S rDNA gene. Extraction was conducted using freeze-thaw which was continued with single cell PCR method. Genetic distance values and phylogenetic analysis were analyzed using MEGA software. Based on molecular analysis, P. lima from this research was divided into two subclades, namely subclade A from Seribu Islands and Belitung Island, and subclade B from Karimunjawa Islands and Bintan Island. P. lima from Bintan Island showed a closer relationship with the reference sequence from the Genbank. Observation of molecular characters of P. lima showed that the genetic diversity of P. lima depended on the variation of the island’s morphogenesis type. These findings could support a further study on the distribution of P. lima in Indonesian waters, related to the genetic variation and toxin production, since Indonesia consists of many small islands.
