Abstract

Infectious diseases are a public health problem for both developed and developing countries. The cause of an infectious disease such as in Shigella sonnei bacteria which occurs in dysentery is characterized by feces mixed with blood and mucus usually accompanied by fever. One solution to overcome dysentery is by utilizing plants that can be processed into medicine. One of them is mangrove (Avicennia alba Blume) which contains phytochemical compounds as antibacterial activity. This study is an experimental study that aims to determine the minimum inhibitory concentration (MIC) of mangrove leaf ethanol extract (Avicennia alba Blume) against Shigella sonnei bacteria. The minimum inhibitory concentration (MIC) value of the extract was determined using the liquid dilution method and continued with solid dilution, with concentrations of 11%, 12%, 13%, 14%, and 15%. Observations were made to see whether bacterial growth in the media. The test results showed that mangrove leaf ethanol extract (Avicennia alba Blume) had a minimum inhibitory concentration at a concentration of 15% in inhibiting the growth of Shigella sonnei bacteria with clear around the media.

Abstract

Penyakit infeksi merupakan salah satu jenis penyakit yang paling umum diderita oleh penduduk di negara berkembang, termasuk Indonesia. Salah satu penyebab bakteri yang menyebabkan infeksi adalah Klebsiella pneumoniae yang menyebabkan infeksi di berbagai tempat, seperti paru-paru, saluran kemih, aliran darah, luka atau tempat pembedahan, pneumonia, meningitis, abses hati, dan endoftalmitis. Bunga telang merupakan tanaman obat yang memiliki segudang manfaat. Ekstraksi tanaman ini bermanfaat untuk mengobati berbagai penyakit dan dapat menghambat pertumbuhan mikroba, seperti Klebsiella pneumoniae. Penelitian ini dilakukan dengan cara eksperimental, menggunakan metode dilusi cair dan dilusi padat. Pengenceran ekstrak etanol bunga telang ditentukan dengan konsentrasi pengujian 1 (4%, 5%, 6%, 7%, 8%, dan 9%), pengujian 2 (11%, 12%, 13%, 14%, dan 15%). Hasil uji konsentrasi hambat minimum (KHM) pada metode dilusi padat terdapat adanya penghambatan pertumbuhan bakteri pada konsentrasi 14%. Hal ini ditunjukkan oleh media yang jernih di sebagian besar permukaan media. Kemudian pada konsentrasi 15% juga menunjukkan adanya penghambatan pertumbuhan koloni bakteri Klebsiella pneumonia.  konsentrasi hambat minimum, dilusi, ekstrak etanol bunga telang (Clitoria ternatea L.), Klebsiella pneumoniae.

Abstract

Uncaria guianensis is a medicinal plant from the Rubiaceae family, known for its wide range of pharmacological activities, particularly as an anti-inflammatory and immunomodulator. The leaves are rich in secondary metabolites such as alkaloids, flavonoids, tannins, and triterpenoids, whose potential is strongly influenced by the extraction method employed. This study aimed to compare the yield and characteristics of 70% ethanol extracts of Uncaria guianensis leaves obtained through four extraction methods: maceration, percolation, soxhlet extraction, and sonication. Leaf samples were collected from Kampar Regency, Riau, processed into powdered simplicia, and extracted using 70% ethanol. The results of the study showed that the soxhletation method produced the highest yield, namely 65.16%, followed by the maceration, percolation, and sonication methods with yields of 46.42%; 38.20%; and 35.82%, respectively. Phytochemical screening confirmed that all extracts tested positive for the presence of flavonoids, alkaloids, tannins, saponins, steroids, and triterpenoids. In terms of physicochemical characteristics, the soxhlet extract exhibited the best quality, with the lowest ash content (1.00%) and moisture loss (0.53%). In conclusion, soxhlet extraction is the most effective method for producing 70% ethanol extracts of Uncaria guianensis leaves, both in terms of quantity (yield) and quality (ash content and moisture loss).
