Abstract

Study on isolation of catechin from gambier (Uncaria gambier (Hunter). Roxb) for pharmaceutical and cosmetic uses has been conducted.Raw materials of gambier were collected from three different areas including Andalas University Medicinal Plant Garden, Lima Puluh Kotaand Siguntur Pesisir Selatan, West Sumatera. Catechin was isolated from gambier using two methods, namely purification and pre nonpurification. Several evalutions on quality of the obtained catechin was done including descriptions of the catechins, solubility, colorreactions, thin layer chromatography, yield, drying shrinkage, melting point and maximum uptake levels of catechins. The best results forpharmaceutical and cosmetic uses are gambier originated from Siguntur with pre purification method which obtained 56.3% yield and 96.17%levels of catechins.

Abstract

The formulas of antiacne gel preparation of ethyl acetate extract of gambir have been formulated by using carbopol 940P as its bases. In thisstudy, five difference gel formulas F1, F2, F3, F4 and F5, were prepared with 0.5; 1; 2; 4 and 6% of concentration of extract, respectively.The result showed that organoleptic of al formulas was found unstable, while the appearance of gel F1 and F2 were homogeneous and stableat room temperature as well as cooling process. In addition, all formulas exhibited stable value in pH evaluation and did not irritate the skin.

Abstract

Glutinous rice contains 99.7% of amilopectin. The amilopectin does not swell in cold water and it could retard the desintegration anddissolution of drug. Physical modification of glutinous rice by pregelatization can enhance the swelling power of starch in cold water. In thisstudy, glutinous rice used as sample was collected from three difference places which are kecamatan Air Tiris, Rumbio Jaya dan Salo kabupatenKampar, Riau province. Sample was subjected to pregelatization process manually as well as by using Spray Dryer. The result showed thatpregelatized glutinous rice starch gave the higher swelling power than glutinous rice. Glutinous rice from Rumbio Jaya showed the highestswelling power of 343.87%. This value indicated that it could be potential candidate as sustained release agent.

Abstract

Optimization studies out on isolation of catechin from gambier and pasta for the pharmaceutical and marker compounds have been carried. Gambier gambier and paste were obtained from the Medicinal Plant Garden Andalas University, Siguntur and Lima Puluh Kota. Isolation method used was non-purification method, pre-purification for gambier and fractination for pasta. The analysis performed included catechin solubility, melting point, maximum absorption, thin-layer chromatography, loss on drying, ash content, yield and determination of levels of catechin. Product which has the highest levels of catechins, the determination was continued by analysis of UV spectra and NMR. The best results for pharmaceuticals derived from Siguntur with pre-purification methods which resulted in yield 56.3% with 96.17% catechin content, whereas for a marker compound was obtained from gambier Siguntur pasta with fractionation method which yielded 12.13% with 97.96% of catechin level.

Abstract

Research on the acute toxicity test of the ethanol extract of kaik-kaik (Uncaria cordata (Lour) Merr) leaves has been carried out on male white mice (Mus musculus L.). This study aims to determine the effects of acute and delayed toxicity of the ethanol extract of kaik- kaik root (Uncaria cordata (Lour) Merr) leaves by determining the LD50 value. The experimental animals were divided into 4 groups, namely the negative control group (NaCMC), and the ethanol extract group of kaik-kaik root leaves with doses of 200, 1000, and 5000 mg / KgBB. The results showed that the LD50 value was> 5000 mg / kg BW and was included in the practical non-toxic category. In the observation, there were no tremors, seizures, salivation, diarrhea, lethargy, weakness, sleep, and coma in each experimental animal.

Abstract

Uncaria cordata is a plant that is used to treat diabetes and has antioxidant properties. Research has been carried out on Acute and Delayed Toxicity Tests of Butanol Fraction of Leaves of Kaik-Kaik Root Plant (Uncaria cordata (Lour) Merr.) on Male White Mice (Mus musculus L. This study aims to determine the effect of acute toxicity of the butanol fraction of the leaves of kaik-kaik root by determining the LD50 value. Observations made in this test were the number of dead animals, toxic symptoms, and observations of the organs of the test animals. Observations were made for 24 hours to determine the LD50 value to 14 days after administration of the test preparation. In the observation of toxic symptoms, there were no tremors, seizures, salivation, diarrhea, lethargy, weakness, sleep, and coma in each experimental animal. In the relative weight parameters of organs consisting of the heart, liver and kidneys, there was no effect of this fraction, and there was no change when compared to the control group. The results showed LD50 values > 5000 mg/kgBB are considered relatively harmless and potentially as standardized herbal medicine

Abstract

Genus Uncaria kaya akan kandungan metabolit sekunder. Pengembangan sebagai agen farmakologi dari isolat murni tumbuhan ini perlu untuk diteliti. Tujuan penelitian ini adalah uji sitotoksik senyawa murni ekstrak metanol daun tumbuhan akar kaik-kaik Uncaria cordata (Lour.) Merr. Terlebih dahulu dilakukan tahap ekstraksi dengan metode maserasi dan dilakukan pemisahan dengan kromatografi kolom. Fraksi F1 yang diperoleh dari kromatografi kolom diuji aktivitas sitotoksik memiliki nilai LC50 sebesar 208,92 µg/mL. Senyawa murni hasil isolasi diberi label UcF1, berupa amorf, berwarna hijau dengan titik leleh 188-190 oC. Berdasarkan analisis data pemeriksaan fitokimia, spektroskopi UV dan IR, senyawa UcF1 diprediksi merupakan senyawa golongan terpenoid. Senyawa UcF1 memiliki nilai LC50 sebesar 69,18 µg/mL dan nilai ini menunjukkan bahwa senyawa ini mempunyai aktivitas sitotoksik yang kuat.

Abstract

Kulit batang Bauhinia semibifida Roxb. telah lama digunakan sebagai obat tradisional oleh masyarakat Melayu Lingga, Kepulauan Riau. Penelitian ini bertujuan untuk mengisolasi dan menguji aktivitas antioksidan senyawa metabolit sekunder dari ekstrak metanol kulit batang B. semibifida Roxb. Tiga isolat murni berupa satu senyawa alkaloid (BR1) dan dua senyawa flavonoid (BR2 dan BR3) telah berhasil diisolasi dengan cara ekstraksi menggunakan metode maserasi dan dilanjutkan fraksinasi dengan metode kromatografi kolom. Uji aktivitas antioksidan dilakukan terhadap isolat dengan metode 2,2 difenil-1-pikrilhidrazil (DPPH). Senyawa isolat BR1 menunjukan IC50 >1000 µg/mL dikategorikan tidak aktif sebagai antioksidan, sedangkan senyawa isolat BR2 dan BR3 menunjukan IC50 2,92 µg/mL dan 4,39 µg/mL dikategorikan sangat kuat sebagai antioksidan jika dibandingkan dengan vitamin C sebagai kontrol. Berdasarkan evaluasi aktivitas antioksidan senyawa BR2 adalah senyawa pilihan yang dilanjutkan identifikasi dengan spektroskopi Fourier-Transform Infrared Spectroscopy (FTIR) dan Proton Nuclear Magnetic Resonance (1H-NMR) sehingga diduga senyawa isolat memiliki struktur dasar senyawa flavonoid.Isolation of Secondary Metabolite Compounds of Methanolic Extract of Steam Bark Bauhinia semibifida Roxb. and Their Antioxidant Activites. Stem bark of Bauhinia semibifida Roxb. has been long consumed as a medicinal herb by Lingga Malaya Ethnic. Therefore, this research aimed to isolate and examines the antioxidant activity of secondary metabolites from methanol extract of stembark of Bauhinia semibifida Roxb. Three pure compounds which are an alkaloid (BR1) and two flavonoids (BR2 and BR3) have been isolated by extraction using maceration method followed by fractionation using column chromatography method. Antioxidant activity assay was carried out by 2,2-Diphenyl-1-Picrylhydrazyl (DPPH) method. Compound BR1 showed IC50 >1000 µg/mL categorized as inactive as an antioxidant, while compounds BR2 and BR3 showed IC50 2.92 µg/mL and 4.394 µg/mL, respectively, categorized as very strong as an antioxidant when compared to vitamin C as a control. Based on the evaluation of the antioxidant activity, the BR2 was selected compound and continued to identification by Fourier-Transform Infrared Spectroscopy (FTIR) and Proton Nuclear Magnetic Resonance (1H-NMR) spectroscopy so that the isolate has the basic structure of flavonoid compound.

Abstract

Tin leaf (Ficus carica L.) is one of the traditional medicines that has potential as an antioxidant. The purpose of this study was to isolate secondary metabolites and to test the antioxidant activity of pure isolates from the n-butanol fraction of tin leaves. Extraction was carried out by the fractionation method using n-hexane, ethyl acetate and n-butanol. Then the n-butanol fraction was separated by column chromatography and antioxidant activity was tested using the DPPH method. Pure isolate compounds labeled HNF-3, in the form of crystals, white with a melting point 189-191 ° C. Based on phytochemical examination data analysis, UV spectroscopy, FT-IR and 1H-NMR it can be concluded that the HNF-3 compound is a flavonoid glycoside group and has a moderate category of antioxidant activity with an IC50 value of 160.613 µg / mL.

Abstract

Telah dilakukan skrining aktivitas sitotoksik dari beberapa jenis spon laut yang diperoleh dari Pulau Mandeh Sumatera Barat. Ekstrak methanol spon yang telah dikeringkan difraksinasi ke dalam fraksi non polar, semi polar dan polar, masing-masing dengan menggunakan heksan, etil asetat, dan butanol. Uji aktifitas sitotoksik dilakukan dengan metode Brine Shrimp Lethality Test (BSLT). Hasil Uji sitotoksik terhadap ekstrak methanol menunjukkan bahwa sampel spon AN 07 mempunyai nilai LC50 yang paling tinggi yaitu 26,1036 µg/ml. Hasil uji sitotoksik terhadap fraksi heksan dan etil asetat menunjukkan bahwa sampel spon AN 01 mempunyai nilai LC50 1,4585 µg/ml dan 29,4289 µg/ml sedangkan fraksi butanol menunjukkan bahwa sampel AN 04 mempunyai LC50 yang paling tinggi yaitu 0,0002 µg/ml.   Kata kunci : Sitotoksik, Spon Laut, Brine Shrimp Lethaliy Test
