Abstract

ABSTRACTEffectivity of Immunostimulant Spirulina to Jambal Catfish (Pangasius djambal). Four kinds of diet with difference doses of Spirulina (0 gr.kg-1 diet; 2 gr.kg-1 diet; 4 gr.kg-1 diet and 6 gr.kg-1 diet) is experimented to jambal catfish (Pangasius djambal Bleeker). The aim of research to observed histology limphoid organs. This research is done during 28 days and challenged by intraperitoneal with Aeromonas hydrophila, one of pathogen bacteria on the 29th. The lymphoid organs of fish as taken every weeks to observed histology until the 42nd. The result showed that difference doses of Spirulina influence the histology of lymphoid organs jambal catfish. The dose of Spirulina 4 gr.kg-1 diet is effective to increase the immunity of jambal catfish bodies.Key Words : Spirulina, immunostimulant, lymphoid organ, Aeromonas hydrophila, jambal catfish

Abstract

Panglima Besar Soedirman waters reservoir and the Serayu River in Banjarnegara Regency, Central Java is one of the habitats of hampala fish . Hampala fish is a member of the Cyprinidae family, which has economic value but is fully captured from the wild. The study on the genetic diversity using approaches of isozyme analysis needed to support conservation and domestication of the fish in this area. This study was aimed at the genetic variation of the hampala fish population in PB. Soedirman water reservoir and the Serayu River in Banjarnegara Regency based on esterase (EST), acid phosphatase (ACP), peroxidase (PER), and aspartate aminotransferase (AAT). Visualization of the isozyme was carried out employing horizontal electrophoretic technique with potato starch. From the results of this study it can be concluded that the hampala fish from the reservoir of PB. Soedirman, Serayu River area before reservoir and after reservoir, all of which are in Banjarnegara Regency, can visualized isozymes EST, ACP, and AAT well, except PER isozyme. This finding can be used as based information on population genetics and finally can be used for conservation of this fish. The results of this study are expected to be utilized to evaluate the potential genetic condition of hampala fish, which is the basis for conservation strategy and domestication.

Abstract

Spirulina platensis is a phytoplankton, the cell wall composed of complex sugars so easily digested by fish. The present study was carried out to investigate stimulation cycle of feed deprivation with feed supplemented S. platensis the best to increase growth, hematological and body composition of gurami (Osphronemus gouramy). Groups of 24 fish, each in triplicate, were exposed to four different treatment for a period of 56 days. Sample measurements of growth done every 14 days, hematological and body composition measurements carried out at the end of the experiment. Growth was significantly different between stimulation cycle of feed deprivation and the control (P<0.05). Conclusions result showed that stimulation cycles of feed deprivation could not improve growth and hematological, but could improve body composition. Feed deprivation is done to reduce the cost of production, high production costs due to high feed prices. During research on feed deprivation is done by giving commercial feed, this study is to provide feed supplementation S. platensis. Thus, the results of this study can be useful for science as S. platensis information can be used as a food supplement and and for the people cultivating gurami should be fed daily supplementation of S. platensis.How to CiteSimanjuntak, S. Abstract

Kidney is very susceptible to damage by toxicological compounds such as carbon tetrachloride (CCl­4). CCl4 produce free radicals, which cause lipid peroxidation and kidney damage and free radical release, which can be prevented by the administration of exogenous antioxidants, such as Chlorella vulgaris.The aim of study was to determine an effect of antioxidant of C. vulgaris on the histopathological features of Wistar rat  kidney which is induced by CCl4. Experimental study with completely randomized design. The variable was histopathology features of the kidneys. The doses of C. vulgaris extract were 3 mg, 4 mg, and 5 mg per 100 grams of rat body weight (BW). The administration of C. vulgaris extract was performed within 30 days, while the CCl4 induction (0.25 ml / 100 g BW) was administered orally on the day 9, 12, 16, 19, 23, and 26. Parameters were histopathology features of renal damage, proportion of tubular cell damage, and Bowmans space diameter. The results showed the administration of C. vulgaris extract was able to reduce the impact of damage caused by CCl4 (p<0.05). This was supported by histologic observations, which was showing a decrease of picnotic and vacuolated cells, normal brush border, and decrease of Bowmans space. In conclusion, 5 mg / 100 g BW of C. vulgaris extract can effectively protect the kidney from damage        caused  by        CCl4. The results of this study strongly support further research on immunostimulant content test C. vulgaris and determine the efficient dose for representative animals mammals also in humans.

Abstract

Chlorella vulgaris is an algae with high nutrition content. Carbon tetrachloride (CCl4) is a hepatotoxic chemical.The aims of this study were to determine the effect of C. vulgaris extract on the physiological response of liver such as MDA, SOD and GPx activity on rat after induced by CCl4 exposure as well as to determine the effective dose of C. vulgaris  extract as antioxidant that can neutralize CCl4 exposure.  This research was conducted experimentally with Completely Randomized Design that consists of 6 treatment and 5 times repetition. The doses of C. vulgaris extract used were 3, 4, and 5 mg per /100 g of rat’sbody weight (BW).The administration of C. vulgaris extract was performed within 30 days, while the CCl4 (0.25 ml/100 g BW) was administered orally on the day 9, 12, 16, 19, 23, and 26. Parameters measured were levels of MDA, SOD and GPx of rat blood serum.The results showed that the administration of C. vulgaris extract can inhibit lipid peroxidation indicated by decrease in MDA activities and oxidative stress by increasing SOD and GPx activity. In conclusion, 5 mg/100 g BW of C.vulgaris extract is an effective dose to be uses as endogenous antioxidant to protect the liver cell from damage caused by CCl4exposure. The benefit of C. vulgaris as a supplement for antihepatotoxin in humans.

Abstract

Osteochilus vittatus is a freshwater fish that needs to be developed because it tastes savory and can be used as a biocleaning agent. Research on the effect of supplementation of Spirulina platensis in nilem fish that is maintained in a biofloc system on digestive enzyme activity has never been done before. The objectives of this study were to determine the effect of S. platensis supplementation on Osteochilus vittatus digestive enzyme activity maintained in the biofloc system and to obtain the most optimum level of S. platensis supplementation in improving the  digestive enzymes activity. The study was conducted experimentally with Completely Randomized Design with four treatments and five replications. Osteochilus vittatus were fed with Spirulina platensis level of 0, 2, 4, and 6 g kg-1 for 56 days. Enzyme activity was measured on days 0 and 56 with three kinds of buffer solutions. Amylase and protease activity was analyzed using ANOVA with a confidence level of 95%. The results showed that the highest amylase activity in the liver was 4.764 ± 1.705 U mg-1 protein and proximal intestine was 2.328 ± 0.838 U mg-1 protein. The highest protease activity was found in the liver and proximal intestine with activity of 6.536 ± 0.911 U mg-1 protein and 6.207 ± 2.195 U mg-1protein. Spirulina platensis supplementation level of 6 g kg-1 feed is the most optimum level (p<0.05). Cultivation of fish with S. platensis supplementation in feed can increase enzyme activity and the fish maintenance in biofloc systems can improve water quality.

Abstract

Spirulina platensis is a microalgae that contains nutrients such us iron and phycocianin higher than other microalgae. Research on suplementation of S. platensis in Osteochilus vittatus cultured in biofloc system has never been done. The aim of this study was to determine the blood hematological and biochemical parameters of O. vittatus fed with S. platensis supplementation and maintained in biofloc system. The study was conducted experimentally with Completely Randomized Design, four treatments and five replications. O. vittatus were fed with S. platensis level 0, 2, 4, and 6 g.kg-1 for 56 days. Blood hematological and biochemical parameters were measured on days 0 and 56. Values of those parameters were then analyzed using ANOVA with a confidence level of 95%. The result showed that the highest red blood cells, white blood cells, and hemoglobin counts were 1.57x106­­­­ cell.mm-3, 2.37x105 cell.mm-3, and 6.77 g.dl-1 respectivley, while the highest hematocrit value was 17.5 %. The highest total protein, albumin, and globulin in blood were 7.96 g.dL-1, 4.31 g.dL-1, and 3.79 g.dL-1 respectively, and the best for ratio A/G was 1.14. S. platensis suplementation level of 4g.kg-1 feed was the most optimum level (P<0.05). Cultivation of fish with S. platensis supplementation in feed can increase fish health indicated by the hematological and biochemical parameters of the blood and the fish maintenance in biofloc systems can improve water quality. The benefit of this research is to make a good condition for cultivation and efficiently used not only water but also feed.

Abstract

Osphronemus gouramy is a potential Indonesian fish that needs to be supported by good nutritional feed and water quality in a culture system. The research on the effect of combining Chlorella vulgaris (C. vulgaris) supplementation with culture systems of biofloc and non-biofloc in gourami fish on amylase activity and blood urea level has not been conducted. Therefore, this study aims to determine the effect of C. vulgaris supplementation with different culture systems on the amylase activity of various digestive organs and blood urea levels. It obtains the best C. vulgaris supplementation feed combination to increase the amylase activity of various digestive organs and reduce blood urea levels. This research was carried out experimentally with a Factorial Completely Randomized Design consisting of two factors. Furthermore, gourami was fed with C. vulgaris levels of 0, 2, 3, 4, and 5 g kg-1 for 28 days and divided into biofloc and non-biofloc groups of culture systems. The amylase activity was measured at three pH buffers, and blood urea levels were analyzed using a urea kit. The results showed that amylase activity increased when supplemented with C. vulgaris 5 g kg-1 feed using the biofloc system, while blood urea levels decreased. C. vulgaris supplementation and biofloc system are alternative solutions to improving nutrition and water quality.

Abstract

Limbah batik mengandung logam berbahaya diantaranya adalah Cr, Cu, Zn, dan Mn. Limbah batik yang dihasilkan dari industri tekstil umumnya merupakan senyawa anorganik non-biodegradable, yang dapat menyebabkan pencemaran terutama lingkungan perairan. Logam yang masuk dalam bentuk ion akan diikat oleh protein darah (Metalotionin), disebut ikatan M+Mt. Ikatan tersebut bersifat stabil dan tidak mudah lepas, sehingga memicu pembentukan radikal bebas. Radikal bebas akan menimbulkan reaksi peroksidasi lipid yang dapat merusak hati, sehingga terjadi peningkatan kadar enzim SGPT yang merupakan parameter gangguan pada fungsi hati. Ikan Nila (Oreochromis niloticus) dianggap sebagai bioindikator untuk studi toksikologi perairan dan monitoring lingkungan, mudah beradaptasi di segala kondisi lingkungan, dan memiliki nilai komersial yang tinggi. Penelitian ini dilakukan secara eksperimental dengan Rancangan Acak Lengkap (RAL), dengan perlakuan konsentrasi limbah batik Naftol 17% v/v, 34% v/v, 51% v/v, dan kontrol yang dipaparkan selama 48 jam. Analisis data menggunakan uji Anova dengan taraf kepercayaaan 95% dan dilanjutkan dengan uji Duncan. Hasil penelitian ini menunjukkan bahwa efek subletal limbah batik terhadap aktivitas enzim SGPT pada serum ikan Nila (O. niloticus) nilainya berbanding lurus. Hasil perhitungan rerata aktivitas enzim SGPT selama 48 jam pada perlakuan kontrol 6,17±0,98 U/L, konsentrasi 17% v/v yaitu 11,67±1,51 U/L, konsentrasi 34% v/v yaitu 17,67±1,51 U/L, dan konsentrasi 51% v/v yaitu 121.83±1,9 U/L. Analisis Anova (α=0,05) didapatkan nilai F hitung (199,552) > F tabel (4,30). Hasil uji Duncan menunjukkan bahwa perlakuan limbah batik dengan konsentrasi 51% v/v selama 48 jam, paling berdampak terhadap penurunan fungsi enzim SGPT.
