Abstract

The optimal cellular reservoir for the conservation and propagation of endangered poultry is acknowledged to be primordial germ cells (PGCs). This study aimed to evaluate and compare the efficacy of cryopreserving PGCs derived from purified sources and blood samples. The experimental design encompassed two treatment groups, each iterated six replication. Blood samples that had been filled with 500 µL of phosphate-buffered saline without Ca2+ and Mg2+ (PBS) underwent centrifugation at 1200 rpm for six minutes. The supernatant was discarded and the pellet was mixed with 500 µL 10% FBS-PBS and was then subdivided for cryopreservation and PGCs purification. Cryopreservation included the controlled freezing of blood and purified PGCs samples in Nalgene cryogenic vials with the incorporation of banker cells. Following thawing, statistical analyses revealed no significant variance in the mean PGCs count between purified PGCs and blood samples (p > 0.05). The percentages of PGCs mortality and the recovery rate demonstrated statistical significance (p < 0.05). Further investigations indicated that cryopreserving blood samples significantly increased the recovery rate of PGCs while decreasing post-thaw mortality. Morphology of PGCs from blood cryopreservation showed similar morphology to PGCs in general. The potential application of cell-based cryo-banking will contribute to the preservation of poultry genetic diversity for sustainability and adaptation to future poultry demands.

Abstract

Market organic waste (LOP) is organic residue or residue generated from market trading activities such as residual vegetables, fruits, and other organic foodstuffs. This waste can be a source of environmental problems if not managed properly because it can rot and cause unpleasant odors. Still, if managed properly, it can be used as an alternative feed for livestock. This study aims to evaluate the quality of LOP silage using rejected commercial syrup as silage additives. The manufacture, harvesting, drying, and siege of silage are conducted at the Laboratory of Nutrition and Feed Technology, Faculty of Agriculture and Animal Husbandry, Sultan Syarif Kasim State Islamic University, Riau. The experimental method with a Complete Randomized Design consisting of 5 treatments and five repeats was used in this study. The treatment in question was P1: spinach 33.3% + cabbage 33.3% + kale 33.3% as control, for P2, P3, P4, and P5 plus SKA of 1%, 2, 3, and 4%, respectively, based on dry matter, then enzymatic for 30 days at room temperature. The parameters measured are temperature, humidity, mold growth, and silage pH. The data obtained were analyzed based on variety analysis, and the difference in parameter values between treatments was further tested with a DMRT level of 5%. The results of the variety analysis showed that the use of SKA had a significant effect (P<0.05) on temperature, humidity, fungal growth, and silage pH. The silage temperature at the time of harvesting averages 30±0.39°C; the average humidity is 77±0.25%; mushroom growth averaged 2.87±0.39%; and an average silage pH of 3.67±0.45. The conclusion is that SKA can improve the quality of market organic waste silage by optimizing temperature and humidity during ensilage to minimize pH and mold growth.

Abstract

Abstract This study aims to describe the Planning, Organizing, Implementation, and Supervision of the Package C Equivalence Program Management at PKBM Sanggar Juang, Jatisari District, Karawang Regency. A descriptive qualitative research approach. Data collection techniques, observation, interviews and documentation. The research subjects were one chairperson, one tutor and two residents studying Package C at PKBM Sanggar Juang, Jatisari District, Karawang Regency. The results of the study, in education management there are planning, organizing, implementing and supervising. Planning, in the Package C Equivalence Program at PKBM Sanggar Juang, Jatisari District, Karawang Regency, makes a learning program that is in accordance with the direction of the Karawang Regency education office but in practice it is adjusted again to the needs of the learning community. organizing. PKBM Sanggar Juang cannot be said to be good because the institution still lacks the quality and quantity of its human resources. Implementation of the Package C Equivalence Program at PKBM Sanggar Juang the materials, media, methods, and time used in learning are tailored to the needs of the learning community. The supervision carried out by the PKBM Sanggar Juang creative institution is carried out directly, while the supervision from the education office to the Sanggar Juang PKBM institution is carried out once a year, and there is an evaluation every three months and at the end of each semester. Abstrak Penelitian ini bertujuan untuk mendeskripsikan Perencanaan, Pengorganisasian, Pelaksanaan, dan Pengawasan pada Manajemen Program Kesetaraan Paket C di PKBM Sanggar Juang Kecamatan Jatisari Kabupaten Karawang. Pendekatan penelitian kualitatif yang bersifar deskriptif. Teknik pengumpulan data, observasi, wawancara dan dokumentasi. Subyek penelitiannya yaitu satu orang ketua, satu orang tutor dan dua orang warga belajar Paket C di PKBM Sanggar Juang Kecamatan Jatisari Kabupaten Karawang. Hasil penelitian, dalam manajemen pendidikan ada perencanaan, pengorganisasian, pelaksanaan dan pengawasan. Perencanaan, pada Program Kesetaraan Paket C di PKBM Sanggar Juang Kecamatan Jatisari Kabupaten Karawang membuat pogram pembelajaran yang sesuai dengan arahan dinas pendidikan Kab Karawang tetapi pada praktiknya di sesuaikan lagi dengan kebutuhan warga belajar. Pengorganisasian. PKBM Sanggar Juang belum bisa dikatakan baik karena lembaga tersebut masih kekurangan kualitas serta kuantitas sumberdaya manusianya. Pelaksanaan Program Kesetaraan Paket C di PKBM Sanggar Juang materi, media, metode, dan waktu yang digunakan dalam pembelajaran disesuaikan dengan kebutuhan warga belajar. Pengawasan yang dilakukan oleh kretua lembaga PKBM Sanggar Juang dilakukan ecara langsung sedangkan pengawasan dari dinas pendidikan ke lembaga PKBM Sanggar Juang dilakukan setahun satu kali, serta ada evaluasi tiga bulan sekali dan di setiap akhir semester
