Abstract

Bakteri asam laktat (BAL) adalah bakteri yang digunakan dalam produk fermentasi. Keberadaan BAL bermanfaat bagi kesehatan. Tujuan pengabdian ini adalah untuk meningkatkan pengetahuan siswa-siswi sekolah menengah atas di Sidoarjo terkait produk bioteknologi yoghurt. Bentuk kegiatan pengabdian adalah sosialiasi melalui edukasi cara pembuatan yoghurt, bakteri yang digunakan dalam pembuatan yoghurt dan manfaat mengkonsumsi yoghurt untuk kesehatan. Hasil pengabdian yaitu, beberapa siswa-siswi yang belum mengkonsumsi yoghurt, telah mencoba sampel yoghurt. Siswa-siswi mengetahui cara pembuatan yoghurt secara sederhana, mengetahui bakteri untuk pembuatan yoghurt serta manfaat yoghurt untuk kesehatan terutama kesehatan saluran pencernaan. Melalui edukasi ini, diharapkan siswa-siswi bisa membuat yoghurt secara mandiri di rumah dan mulai mengkonsumsi yoghurt untuk kesehatan.

Abstract

Aedes aegypti is a type of mosquito that causes dengeu hemorrhagic fever as the main carrier (primary vector) of dengeu virus. This disease is very common in Indonesia, efforts to eradicate mosquito larvae are relatively cheap, simple by means of public health counseling and eradication of mosquito nests using insecticides. The purpose of this study was to determine the effectiveness of the combination of beluntas leaf extract (Pluchea indica linn) and garlic (Allium satisum) in killing Aedes aegypti mosquito larvae at concentrations of 10%, 20%, 50%, and 100% for 24 hours. This type of study used experimental research with a research sample of 400 Aedes aegypti mosquito larvae. The results showed that the combination of beluntas leaf extract (Pluche indica linn) and garlic (Allium satisum) has effectiveness as a larvicide of Aedes aegypti instar III. The effective concentration on the combination of beluntas leaf extract (Pluchea indica linn) and garlic (Allium sativum) is 50% within 24 hours.

Abstract

Soil is a mineral material that has several unconsolidated forms which are located on the surface of the earth and have undergone several treatments. Soil-Transmitted Helminth (STH) is an intestinal nematode that in its life cycle requires a soil medium for the infective form to mature. worms are moist soil, shady, and protected from sunlight. This study aims to determine whether there is contamination of "Soil-Transmitted Helminth" eggs and what species are present in the rice fields using the Native method. This examination uses a 2% eosin solution. The sample uses paddy fields in Sukodono Village. Sampling at 3 sampling points with 3 types of samples, the total sample is 9 samples of paddy soil. The research was conducted on April 26-28 2021 at the Parasitology Laboratory, Faculty of Health Sciences, Maarif Hasyim Latif University, Sidoarjo, on 9 samples of paddy soil, the results obtained were 3 types of samples, or 33% of the soil contaminated with worm eggs and 6 types of samples or 66% not contaminated with worm eggs. Keywords: Paddy fields, Soil Transmitted Helminth, Native Method

Abstract

The aim of this research is to isolate and characterize lactic acid bacteria from mare's milk which have the potential to act as probiotics. Bacterial identification includes gram staining, antimicrobial activity test, hemolytic activity test, pH and bile salt tolerance test. The research results showed that C5 isolates isolated from horse milk samples had potential as probiotics. Characterization includes the form of bacillary bacteria, antimicrobial activity against the pathogenic bacteria Escherichia coli ATCC of 0.40 mm, gamma hemolysis, tolerance to pH 2, 3, 5 and tolerance to bile salts of 0.4-1%.

Abstract

Lactobacillus plantarum BP102 isolated from garlic bulb tissue has probiotic properties, including producing bacteriocin called plantaricin. This study aimed to detect the gene encoding bacteriocin produced by Lactobacillus plantarum BP102, and to evaluate the bacteriocin activity at each stage of partial purification. After the end of the log phase of L. plantarum BP102 was determined, and the bacteriocin-encoding genes were checked by PCR technique. Partial purification of bacteriocin was elucidated including pH-neutralized cell-free-supernatant (CFS), precipitation using 80% of ammonium sulfate, and dialysis (cut-off 10 kDa), then the bacteriocin activity in every partial purification stage was evaluated. The molecular weight of plantaricin was estimated using SDS-PAGE analysis. Lactobacillus plantarum BP102 harbored the gene encoding plantaricin (pln) biosynthesis, namely plnEF and plnK genes. The activity of crude bacteriocin was inactivated by the presence of proteinase-K enzyme. The protein concentration was gradually decreased along with the purification process. The bacteriocin activity was demonstrated at each step of the purification process (CFS, precipitation, and dialysis) against Bacillus cereus by 9.23 ± 0.20 mm, 7.86 ± 0.15 mm, and 7.6 ± 0.10 mm, respectively; while, Escherichia coli by 10.3 ± 0.55 mm, 7.4 ± 0.1 mm, and 6.86 ± 0.45, respectively. The molecular weight of partially purified bacteriocin BP102 was found to be approximately 15.9 kDa. The overlaid part of the gel showed a slight inhibition against E. coli due to a low protein concentration. This bacteriocin purification process should be further optimized to improve the bacteriocin activity that could be useful for food preservation.

Abstract

Collagen milk is a novel substrate that has rarely been investigated as a source of probiotic bacteria, despite its great potential as a functional food. This study aimed to isolate and characterize lactic acid bacteria (LAB) from collagen milk and to evaluate their tolerance to acidic conditions. The isolation and identification of LAB were conducted based on Gram staining, catalase testing, and cell morphology. Acid tolerance was assessed at pH 1, 2, and 3 to simulate gastric conditions. The results showed that a total of 21 LAB isolates were successfully obtained, most of which were rod-shaped and Gram-positive. Among these isolates, three superior strains SKF-174, SKF-106, and SKF-204 exhibited high tolerance to acidic environments, indicating their ability to survive under gastrointestinal conditions. These findings suggest that fermented collagen milk has the potential to serve as a promising source of probiotic lactic acid bacteria with strong acid resistance and desirable functional properties.

Abstract

Papaya fruit has many benefits for body health. Lactic acid bacteria can be used as a starter culture to produce more diverse foods (food diversification) and increase product shelf life. This study was conducted to obtain LAB isolates from California papaya that have potential as starter cultures. The methods in this study include the LAB isolation stage followed by the gram staining process, endospore staining, catalase test, hemolytic test, and antibacterial activity test using S. aurius and E. coli as indicator bacteria. Data were analyzed statistically using one-way ANOVA and Tukey’s post hoc test at 5% of significance level. The total count (TPC) of LAB in California papaya samples was 3.6x105 CFU/mL. The results of LAB isolation from California papaya obtained 14 LAB isolates, after going through the testing and screening stages obtained 8 LAB isolates that could potentially be used as starter culture candidates, namely isolates DPC1, DPC5, DPC6, DPC7, DPC10, DPC11, DPC12, DPC14. Isolate DPC6 is an isolate with the highest antibacterial activity in inhibiting the growth of S. aureus and E. coli bacteria, so it is a potential bacterial candidate that can be used as a starter culture.
