Abstract

Abstract

Various treatments for Type 2 Diabetes Mellitus (T2DM) aim to alleviate hyperglycemia. Although natural products have been extensively utilized, their mechanism of action and efficacy as antidiabetic agents, particularly for T2DM, has not been extensively assessed in vivo. In this research, the leaves of the Namnam (Cynometra cauliflora) were extracted and tested for their activity as an antidiabetic agent. Sprague Dawley rats aged 3.5-4 months, weighing 200-250 g, were used as the experimental model, with a total of 30 individuals. The rats were induced with a high-fat diet and 30% sucrose until their blood glucose concentration reached ≥ 120 mg/dL. Subsequently, the rats were divided into four groups (groups 1, 2, 3, and 4). Over 21 days, changes in blood glucose, triacylglycerol, glycogen, and blood plasma insulin levels were assessed. The results demonstrated that the methanol extract of Namnam leaves (NLME) effectively reduced blood glucose levels by 23-34%, decreased plasma triglyceride levels by 13-30%, and increased liver glycogen levels by 68-96% compared to the control group (Diabetes). Among all the parameters assessed, NLME exhibited similar performance to metformin, a commonly prescribed diabetes medication (p<0.05). Furthermore, the study revealed that NLME exerted antidiabetic effects, particularly for T2DM, by promoting liver glycogen formation, enhancing insulin secretion control, facilitating glucose absorption by muscles, and restricting fat metabolism in the blood

Abstract

Background Ehrlichiosis is an infection caused by the bacterium Ehrlichia canis, which attacks monocytes. Ehrlichiosis is characterized by anemia, lethargy, lameness, pancytopenia, hemorrhage, and weight loss. Objective This study aimed to describe the clinical findings and treatment of a dog diagnosed with Ehrlichia canis infection. Case A 5-year-old female Alaskan Malamute dog named Salt was brought to the clinic of GloriaVet Pet Health Solution, Bandung, Indonesia, with lameness and weakness, accompanied by high fever and very pale mucous membranes (pallor). Examination and treatments Dog was examined using a general examination, hematology test, biochemistry test, native smear, and IDExx SNAP© 4dx test kit. The examinations showed a positive result for Ehrlichia canis infection, along with severe pancytopenia, hyperglobulinemia, and thrombocytopenia. Diagnosis heavily relied on serology and clinical manifestation, and further PCR testing was not performed. The treatment consisted of doxycycline 10 mg/kg/day for 28 days, a 0.75% ketamine drip infusion, darbepoetin alfa, and a blood transfusion. Conclusion Dog was infected with chronic ehrlichiosis with several clinical symptoms including severe anemia. Treatment with doxycycline as the treatment of choice for ehrlichiosis infection and a blood transfusion for treating severe anemia showed signs of improvement, but were ultimately ineffective due to the poor prognosis associated with chronic ehrlichiosis and severe pancytopenia.

Abstract

Background Diabetes mellitus (DM) is a chronic metabolic disorder in cats characterized by persistent hyperglycemia resulting from impaired insulin secretion, insulin resistance, or both, and is not always accompanied by pancreatic or hepatic dysfunction. Objective This case study aimed to describe the clinical findings and therapeutic management of a cat diagnosed with diabetes mellitus with suspected hepatic and pancreatic dysfunction. Case An 11-year-old female domestic cat named Miau Miau was presented to the Waras Satwa Veterinary Clinic, Batam, Indonesia, with diarrhea, decreased appetite, polyuria, and polydipsia. Examination The diagnostic approach included anamnesis, physical examination, hematological evaluation, blood biochemistry analysis, and serial blood glucose monitoring using a glucometer. Persistent hyperglycemia was established based on repeated blood glucose values above the normal range (75–120 mg/dL) during the observation period. The findings revealed hyperglycemia (318 mg/dL) with increased levels of amylase, alanine aminotransferase (ALT), and total bilirubin, indicating possible involvement of the pancreas and liver. Diagnosis and treatments The cat was diagnosed with diabetes mellitus with suspected hepatic and pancreatic dysfunction. The treatment included Moringa leaf (Moringa oleifera) as an adjunct therapy in oral capsule form at a dose of ±98 mg/kg body weight/day, combined with pancreatic enzyme supplementation, a hepatoprotective agent, and dietary modification to a high-protein, low-carbohydrate diet. After 14 days of hospitalization, the blood glucose levels gradually decreased to 115 mg/dL. Conclusion This case demonstrates that diabetes mellitus in cats may be associated with pancreatic and hepatic dysfunction, as reflected by increased levels of amylase, ALT, and total bilirubin.

Abstract

ARTICLE HIGLIGHTSBusulfan can damage the liver by triggering oxidative stress Physalis angulata leaves show natural liver protective effects Extract reduced oxidative damage in liver tissue Liver structure improved after extract treatment Early treatment provided stronger liver protection than delayed useABSTRACTLiver disease is the leading cause of death worldwide, with drug-induced hepatotoxicity representing a major contributing factor. Busulfan, an anticancer drug used in chronic myeloid leukemia, is cytotoxic to proliferating cells and can induce liver injury. Physalis angulata L. (ground cherry) contains bioactive compounds with potential hepatoprotective properties. This study aimed to evaluate the protective effects of ethanol extract of Phyasalis angulata L. (EEPA) leaves on busulfan-induced liver damage in female Sprague–Dawley rats. Twenty-five rats were randomly divided into five groups: P1 (normal control), P2 (busulfan 40 mg/kg BW, single dose), P3 (extract 300 mg/kg BW, days 1–28), P4 (busulfan + extract, days 1–28), and P5 (busulfan + extract, days 14–28). Parameters assessed were alanine aminotransferase (ALT) and aspartate aminotransferase (AST), the level of malondialdehyde (MDA) in liver, and liver histology. Data were analyzed using one-way analysis of variance (ANOVA) followed by a post hoc test. The results showed that Physalis angulata L. extract did not cause significant differences in the serum ALT and AST levels but significantly reduced the MDA concentrations and improved the liver histological architecture. Busulfan administration significantly decreased hepatocyte count and increased Kupffer and inflammatory cells, fatty degeneration, and necrosis. Early administration of the extract (P4) demonstrated stronger hepatoprotective effects compared with delayed administration (P5). These findings suggest that EEPA leaves exerts hepatoprotective effects against busulfan-induced liver injury in rats, likely through antioxidant activity, as evidenced by subsequent histological improvement, and highlighting its potential as a natural hepatoprotective agent.
