Abstract

Broiler chicken has high fat content, especially abdominal and subcutan fat which reduced carcass quality and efficiency of feed energy. Genetic approach could be potentially applied to reduce high abdominal and intramuscular fat in broiler chicken through the selection program at gene level related to fat metabolism. This paper describes the metabolism and biosynthesis of body fat and the role of its controlled genes. Fat synthesis is controlled by feed quality and metabolism and biosynthesis process occurred in liver. These processes are controlled by many family genes, but certain genes have dominant role in the process; those are triglyceride lipase genes, fatty acid synthase genes and fatty acid binding protein genes. Expression of fatty acid synthase genes has positive correlation with fat content in liver and intramuscular. Expression of fatty acid binding protein genes was related to the increased abdominal fat deposit. These genes are polymorphic, so that they can be used as a genetic marker in selection to optimize feed efficiency, to minimize abdominal fat and to increase economic value of broiler chicken. Abstract

Gen Toll-like Receptor-4 (TLR4) adalah salah satu gen yang mengontrol ketahanan ayam terhadap Salmonella sp. melalui respons imun non spesifik. Gen tersebut dapat dijadikan sebagai marka genetik pada ayam apabila diketahui keragamannya. Oleh sebab itu tujuan penelitian ini adalah untuk mengidentifikasi keragaman gen TLR4 pada ayam Tolaki yang dibandingkan dengan ayam ras yang ada di Indonesia. Total sampel ada 126 terdiri dari ayam Tolaki (82 ekor), ayam petelur (20 ekor) dan ayam broiler (24 ekor). Tahapan identifikasi meliputi ekstraksi DNA genom, amplifikasi PCR gen TLR4 (220 pb, di exon 2), dan metode RFLP, menggunakan enzim restriksi MscI. Data yang dianalisis meliputi frekuensi alel dan frekuensi genotipe, nilai heterozigositas, dan Polymorphic Informative Content (PIC). Hasil yang diperoleh : gen TLR4|MscI pada ayam Tolaki, petelur dan broiler bersifat polimorfik, dengan ditemukan dua alel yaitu alel A dan G. Frekuensi alel G (pembawa sifat resisten) pada ayam Tolaki, broiler dan ayam petelur adalah 0.77, 0.87 dan 0.20. Dari uji x2 diketahui bahwa genotipe gen TLR4|MscI ayam Tolaki berada dalam keseimbangan Hardy-Weinberg, dengan nilai Ho dan He sebesar 0.31 dan 0.29. Nilai PIC gen TLR4 ayam Tolaki (0.25) termasuk kategori sedang. Berdasarkan hasil tersebut gen TLR4|MscI dapat digunakan sebagai penciri genetik untuk sifat ketahanan ayam terhadap infeksi Salmonella sp. pada ayam Tolaki.Kata kunci : keragaman genetik, gen TLR4, Salmonella sp, respons imun non spesifik

Abstract

Effect of zeolites-spread litters on broiler performances under different floor spaces had been performed in five weeks. As many as 324 chicks of a day-old Hubbard strain were fed rations contained 3,000 – 3,100 Kcal/Kg ME and 21% crude protein during starter period and fed the ratio contained 20% crude protein with similar energy during finisher period. The chicks were randomly allocated to three different floor spaces (10, 12, and 14 heads/m2) and three different zeolites spread litters (0.0, 2.5, and 5.0 kg zeolites/m2) under completely randomized design with factor 3 x 3 and 3 replications. Parameters concerned of present research including feed intake, live weight gain, and mortality rate were analyzed with analysis of variance and least significant different. Results of the present research showed that there was no interaction effect between floor spaces and zeolites spread litters on all parameters concerned. Abstract

The objectives of this study were to identify polymorphism of transforming growth factor β2 (TGF-β2) gene associated with Salmonella pullorum resistance in Indonesian chickens. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assays were used to genotype Indonesian chickens. Analysis of polymorphism was conducted by using PopGen 3.2 software. The effect of genotype on S. pullorum resistance was analyzed using the SAS General Linear Model (GLM) procedure. Genotyping was performed on 278 chickens from 7 Indonesian chicken populations (Sentul, Merawang, Pelung, Kampung, Parent Cobb broiler, The F1 crossbreed of Kampung x Parent Cobb broiler (KB) and F2 KB x KB. The product of amplification was 284 bp. The TGF-β2| RsaI locus was polymorphic in all populations, producing two alleles (T and C) and three genotypes (TT, CT, and CC). The result from the analysis of the allele and genotype frequency showed that the T allele had a higher frequency than the C allele in all populations. The χ2 analysis showed that the 6 chicken populations were deviated from Hardy-Weinberg equilibrium, with the exception of the Parent Cobb broiler chicken. The association result showed that TT genotype was significantly associated with S. pullorum resistance in Sentul chicken. Although the leukocyte concentration, leukocyte differentiation and H/L ratio in Sentul chicken with three of TGF-β2 genotypes (TT, TC, and CC) were not statistically different. In conclusion, polymorphism in the TGF-β2 chicken gene can be used as a candidate marker to increase S. pullorum immune response.

Abstract

ABSTRACT Tenebrio molitor is an insect larvae that has the potential as a protein source for poultry feed. However, no exploratory research has been found on the quality of poultry meat reared with feed containing these insect larvae. The purpose of this study was to evaluate the hedonic and hedonic quality characteristics of fresh and boiled chicken meat in terms of color, texture, aroma and taste of broiler chicken fed with Tenebrio molitor meal. This research method is experimental using a completely randomized design (CRD). As a treatment, there were two types of feed, namely feed containing 5% MBM as control (R0) and feed containing 5% Tenebrio molitor meal (R1). The treatment was repeated five times, ten chickens for each replication. The results of hedonic and hedonic quality observations were analyzed descriptively. Based on hedonic quality characteristics; the color of the raw meat is redder, the aroma is less fishy, and the taste of cooked meat is more savory. And, in hedonic; broiler chicken that is fed a feed containing Tenebrio molitor meal is preferred by the panelists. So it can be concluded that broiler chicken meat that is fed with Tenebrio molitor meal is better than chicken meat that consumes MBM. Key Words: Chicken meat, Hedonic,Tenebrio molitor

Abstract

Ayam IPB-D1 merupakan jenis ayam lokal dari hasil persilangan antara jantan F1 PS (Pelung x Sentul) dengan betina F1 (Kampung x Ras pedaging paren stock Cobb). Penelitian ini bertujuan untuk mengevaluasi performa, kualitas semen, dan freezability spermatozoa ayam IPB-D1 pada konsentrasi Immunoglobulin Yolk (IgY) berbeda. Materi yang digunakan yaitu ayam IPB-D1 jantan sebanyak 20 ekor, terdiri atas 10 ekor ayam dengan konsentrasi IgY rendah dan 10 ekor IgY tinggi dengan ayam umur 8 bulan. Variabel yang diamati yaitu performa yakni konsumsi pakan, pertambahan berat badan, morbiditas dan mortalitas, kualitas semen segar dan kualitas semen beku (freezability). Data dianalis menggunakan uji-t menggunakan program SAS dengan membandingkan antara konsentrasi IgY tinggi dan IgY rendah. Hasil analisis performa ayam IPB-D1 tidak berbeda (P>0.05) terhadap IgY yang berbeda. Hasil kualitas semen menunjukkan perbedaan yang signifikan terhadap konsistensi semen dan motilitas, viabilitas, konsentrasi serta konsentrasi spermatozoa per ejakulat. Ayam IPB D-1 pada dua konsentrasi IgY tidak berbeda nyata (P>0.05) terhadap motilitas semen segar dan recovary rate, namun Ayam IPB D-1 dengan konsentrasi IgY tinggi memiliki motilitas spermatozoa pasca thawing lebih baik daripada (P<0.05) Ayam dengan konsentrasi IgY yang rendah.

Abstract

Stres panas menyebabkan puyuh mengalami stres oksidatif. Stres oksidatif menyebabkan puyuh rentan terhadap infeksi bakteri dan juga akan berpengaruh pada performanya. Stres oksidatif dapat diatasi dengan penambahan antioksidan. Salah satu sumber antioksidan alami adalah bunga kasumba turate. Tujuan penelitian ini adalah untuk menganalisis ketahanan dan performan puyuh yang diberi jus bunga kasumba turate. Materi penelitian ini terdiri dari sari bunga kasumba turate dan puyuh. Penelitian ini terdiri dari 4 perlakuan dan 4 ulangan dengan P0: 100 mL air minum tanpa JBKT (Kontrol), P1: JBKT 0,25 mg/mL dalam 100 mL air minum, P2: JBKT 0,75 mg/mL dalam 100 mL air minum, P3: JBKT 1,25 mg/mL dalam 100 mL air minum. Hasil penelitian tingkat stres menunjukkan bahwa rasio H/L P1, P2 dan P3 lebih rendah dari P0. Hasil uji clearance menunjukkan bahwa mortalitas bakteri Salmonella pullorum mencapai 67,7%. Pemberian JBKT tidak berpengaruh signifikan terhadap konsumsi pakan dan pertambahan bobot badan. Nilai konversi pakan pada perlakuan P1, P2 dan P3 lebih baik dari pada P0. Berdasarkan hasil penelitian dapat disimpulkan bahwa Pemberian jus bunga kasumba turate pada puyuh mampu memperbaiki ketahanan tubuh terhadap panas lingkungan, terhadap infeksi bakteri dan performa pertumbuhan. Konsentrasi 0,25 mg/100 mL JBKT mampu membunuh bakteri S.pullorum tertinggi dan menghasilkan keefisienan pakan terbaik. Kata kunci : Bunga Kasumba Turate, Ketahanan, Performa ABSTRACTHeat stress causes quail to experience oxidative stress. Oxidative stress can make quail susceptible to bacterial infections which will also affect their performance. Oxidative stress can be overcome with the addition of antioxidants. One of the natural sources of antioxidants is the kasumba turate flower. The purpose of this study was to analyze the resistance and performance of quails that were given the flower juice of kasumba turate L). The material of this study consisted of kasumba turate flower juice and quail. This study consisted of 4 treatments and 4 replications with P0: 100 mL of drinking water without JBKT (Control), P1: JBKT 0,25 mg/mL in 100 mL of drinking water, P2: JBKT 0,75 mg/mL in 100 mL of drinking water, P3: JBKT 1,25 mg/mL in 100 mL drinking water. The results of the research on stress levels showed that the H/L ratio P1, P2 and P3 were lower than P0. The clearance test showed that the mortality rate for Salmonella pullorum bacteria reached 67,7%. The provision of JBKT did not significantly affect feed consumption and body weight gain. The feed conversion values of treatment P1, P2 and P3 were better than P0. Based on the results of the study, it can be concluded that the application of kasumba turate flower juice to quails can improve the body's resistance to environmental heat, against bacterial infections and growth performance. The concentration of 0,25 mg/100 mL JBKT was able to kill the highest S.pullorum bacteria and produce the best feed efficiency.Keywords: Kasumba Turate Flower, Performance, Resistance

Spreading zeolites on litter floor of broiler house at rate of 5.0 kg/m2 increased broiler carcass percentages up to 5.41 percents. Effect of zeolites-spread litters on broiler carcass and non-carcaSs components under different floor spaces had been performed in five weeks. As many as 324 chicks of a day-old Hubbard strain were fed with rations contained 3,000 -3,100 Kcal/I<g ME and 21 %crude protein during starter period and fed with the ration contained 20% crude protein duringfinisher period. The chicks were allocated to three different floor spaces (10, 12, and 14 heads/m2) and three different zeolites-spread litters (0.0, 2.5, and 5.0 kg zeolites/m2) under completely randomized design with factor 3 x 3 and 3 replications. Parameters concerned of present research including percentages of both carcasses and non-carcass components were analyzed with analysis of variance (ANOV A) and least .significant different.Results of the preseqt research showed that there was no interaction effect between floor spaces and zeolites-spread litters on all parameters concerned. Floor spaces affected the percentages of gizzard, kidney, and spleen significantly (p<O.05) but neither carcass nor abdominal fat percentage was af.fectedby the floor space. The zeolites-spread litters only affected significantly (P<O.05) on carcass percentage. Spreading zeolites at rates of5.0 kg/m2 on litter floor resulted in the highest carcass percentage (p<O.05) while rates of 25 kg/m2 resulted the same carcass pen:entage as the rates of 0.0 kg/m2. It was concluded that spreading zeolites at rates of 5.0 kg/m2 on litter floor under different floor spaces increased can:ass percentages up to 5.41 percents.

Abstract

The prolactin gene (PRL) is a gene that controls the incubation and egg production in laying chickens. The nature of incubation will reduce egg production and disrupt the reproductive system in local chickens. The purpose of this study was to identify the polymorphism of prolactin genes in IPB-D1 chickens using the direct sequencing method. The polymorphism of the exon 5 prolactin gene was carried out on 46 samples of IPB-D1 chicken DNA which was a collection of the Division of Animal Genetics and Breeding, Faculty of Animal Science IPB. DNA sequences as a reference for designing exon 5 primers were obtained from the National Center for Biotechnology Information (NCBI) with the GenBank access code: AF288765.2. DNA extraction was carried out using the phenol-chloroform technique. DNA amplification resulted in a PCR product with a size of 557 bp. In this study, the genotype frequency, allele frequency, heterozygosity value and Hardy-Weinberg equilibrium were calculated. The results of the study found 5 SNPs in exon 5, namely g.7823A>G, g.7835A>G, g.7886T>A, g.8052T>C, and g.8069T>C. All SNPs are polymorphic and in Hardy-Weinberg equilibrium except g.8052T>C. The g.7823A>G, g.7835A>G, g.8052T>C SNPs are synonymous mutations that do not change amino acids, while the g.7886T>A and g.8069T>C SNPs are non-synonymous that change amino acids. Both g.7886T>A and g.8069T>C SNPs are potential as a marker assisted selection for the characteristics of egg production in IPB-D1 chickens.
