Abstract

Plant tissue culture is one of the topics in the Biotechnology module of Biology for senior high school class XII. This topic has not been optimally provided in schools, especially in SMA Negeri 7 Yogyakarta due to the limited human resources who are experienced in plant tissue culture and the unavailability of facilities and equipment that support the implementation of the practicum topic. Therefore, this workshop aimed to help students gain knowledge and skills regarding the basic principles, benefits, and stages of plant tissue culture. The workshop was conducted in the Biology Laboratory of SMA Negeri 7 Yogyakarta and participated by 10 class XII students. The workshop consisted of questions and answers before the workshop, materials explanation, hands-on activities, questions and answers after the workshop, and evaluation. The material was delivered using lecturing methods and interactive discussions, followed by practical activities including preparation of culture media, sterilization and inoculation of explants and acclimatization which were carried out by engaging the active participation of each student. The results obtained in this workshop were that students were able to understand and implement plant tissue culture practical activities as well.

Abstract

Perkembangan IPTEK saat ini berkontribusi dalam meningkatnya implementasi bioteknologi dalam kehidupan manusia. Kultur jaringan tumbuhan merupakan salah satu aplikasi bioteknologi di bidang pertanian. Pengenalan kultur jaringan tumbuhan sejak dini bagi siswa SMA menjadi salah satu upaya mempersiapkan SDM yang terampil. Namun demikian, siswa SMA masih mengalami kesulitan dalam proses pembelajaran bioteknologi terutama pada topik ini. Pembelajaran yang berfokus pada teori dan tidak dilengkapi dengan kegiatan praktikum merupakan penyebab siswa sulit memahami pelajaran tersebut. Kendala ini terutama dialami oleh siswa kelas XII SMA BOPKRI I Yogyakarta. Oleh karena itu, kegiatan pengabdian ini bertujuan untuk meningkatkan pengetahuan dan keterampilan dasar siswa pada materi kultur jaringan tumbuhan. Metode kegiatan pengabdian adalah participatory Action Research (PAR) yang diberikan dalam bentuk pelatihan. Pelaksanaan pelatihan dibagi menjadi dua sesi yaitu pemaparan materi pengantar, demonstrasi praktikum dan kegiatan praktikum. Acara praktikum meliputi pembuatan media kultur dan sterilisasi serta inokulasi eksplan. Berdasarkan hasil evaluasi, kegiatan pengabdian ini telah berhasil sebesar 100% dalam meningkatkan pengetahuan berdasarkan hasil tanya jawab selama pelatihan serta keterampilan dalam melaksanakan tahapan kultur jaringan tumbuhan. Kegiatan pengabdian kepada masyarakat ini lebih lanjut dapat diintegrasikan dengan kegiatan pembelajaran biologi di kelas sehingga proses pelatihan dan pendampingan mengenai kultur jaringan tumbuhan dapat berlangsung lebih intensif. Selain itu, project kelompok yang diberikan selama kegiatan pengabdian dapat membantu siswa dalam mencapai tujuan pembelajaran bioteknologi di kelas.

Abstract

Talinum paniculatum Jacq (Gaertn), locally known as Javanese Ginseng, is an essential medicinal plant characterized by high flavonoid content with antioxidant and antimicrobial activities. Compared to other natural sources, a larger number of T. paniculatum leaves is required to obtain high amounts of flavonoid for drug development. In this context, in vitro culture can be used to increase flavonoid production in a controlled condition without reducing plant population. The concentration of Plant Growth Regulators (PGRs) is capable of influencing biomass and flavonoid accumulation in callus culture from several species. Despite these benefits, there is limited information about the optimal concentration of PGRs for flavonoid production from T. paniculatum callus culture. Therefore, this study aimed to investigate the optimum concentration of 2,4-D and kinetin for callus growth, as well as flavonoid production from T. paniculatum. The investigation was carried out using a completely randomized design (CRD), where young leaves explants were cultured on MS media supplemented with various concentrations of 2,4-D (0, 0.5, 1, 2) mg/L and kinetin (0, 0.5, 1, 2) mg/L for callus and flavonoid production. The results showed that the highest callus biomass of 0.105 g was produced from MS medium with 2 mg/L of 2,4-D and 1 mg/L of kinetin. Meanwhile, the highest total flavonoid content of 25.66 mg QE/g DW was produced from media supplemented with 1 mg/Lof 2,4-D and 2 mg/L of kinetin. These results showed that different combinations of 2,4-D and kinetin were required to increase biomass and flavonoid production. In conclusion, this study provided valuable information for the development of bioactive compounds through in vitro culture.
