Abstract

Introduction: Protein is an essential nutrient that serves as an energy source and plays a crucial role in tissue formation. Protein deficiency can lead to various health disorders, ranging from hair loss to serious conditions such as death. High-protein milk serves as an ideal protein source as it contains essential amino acids and complete nutrients, though its consumption should be adjusted to individual needs to avoid side effects. Research Objectives: This study aims to: (1) analyze protein content in high-protein milk (Whey, Casein, and Gainer) using the Kjeldahl and Visible Spectrophotometry methods, and (2) verify the consistency between analysis results and protein content claims on product packaging. Research Methods: Analysis was conducted using two methods: the Kjeldahl method as a high-precision standard method and Visible Spectrophotometry using Biuret reagent which is more sensitive. Each sample was analyzed with six repetitions to ensure data accuracy. Samples were selected purposively based on best-selling products. Results: The results showed the highest protein content in Whey (525.87 mg/g with Kjeldahl; 873.75 mg/g with Spectrophotometry), followed by Casein (316.87 mg/g; 720.83 mg/g), and the lowest in Gainer (227.5 mg/g; 570.5 mg/g). This consistent pattern indicates conformity with packaging claims, although there were value differences between the two methods. Conclusion: The findings emphasize the importance of verifying protein content in supplement products to ensure nutritional information accuracy for consumers. The differences between the two analysis methods are presumably due to their different analytical principles. This study contributes to developing accurate protein analysis methods for high-protein milk products.

Abstract

Background: Gelatin, as a product of partial collagen hydrolysis, is typically derived from pig bones or skin, creating halal compliance issues for Indonesia's Muslim population. Objective: This study aimed to isolate halal gelatin from gulamah fish bones (Johnius trachycephalus) and apply it as a gelling agent in jelly candy production. Methods: The gelatin isolation process involved: (1) degreasing through immersion in boiling water, (2) demineralization using 5% acetic acid for 48 hours, (3) extraction at 60-75°C for 12 hours, and (4) oven drying at 50°C. Gelatin was characterized using FTIR and tested for quality parameters (yield, moisture content, ash content, pH) according to SNI standards. Jelly candies were formulated with two gelatin concentrations (F1=5g; F2=10g) and evaluated for physical properties and hedonic quality. Results: The study achieved a gelatin yield of 7.32% with SNI 06-3735 compliant characteristics: 10.2% moisture content, 1.24% ash content, pH 6.28, and FTIR identification of characteristic functional groups (-OH, C=O, N-H, C-H, and C-N). Formula F2 (10g gelatin) received the highest preference scores for texture (2.93), aroma (2.93), and taste (4.63). Conclusion: Gulamah fish bones show potential as a halal gelatin source meeting quality standards, with optimal application at a 10g concentration in jelly candy formulations

Abstract

Background: Spider lily leaves (Crinum asiaticum L.) contain bioactive compounds, such as flavonoids, which are known for their anti-inflammatory properties. Developing a convenient topical formulation like a stick balm could provide an effective and practical alternative for treating inflammation. Objective: This study aimed to formulate a stick balm containing ethanol extract of spider lily leaves (EESL) and evaluate its physical quality and anti-inflammatory activity. Methods: The extract was obtained by macerating the leaves in 96% ethanol. Stick balm formulations were prepared with varying concentrations of EESL: F0 (base), F1 (1%), F2 (3%), and F3 (5%). The physical quality of the formulations (homogeneity, pH, spreadability, melting point) was evaluated. Anti-inflammatory activity was tested on 24 male white rats induced with carrageenan. The rats were divided into six groups: negative control (induced only), positive control (commercial Geliga balm), F0, F1, F2, and F3. Paw edema volume was measured every hour for 6 hours. Results: All stick balm formulations met the physical quality standards. The anti-inflammatory test showed that the F3 formulation (5% EESL) exhibited the highest activity, with an edema reduction of 0.007 mm, which was equivalent to the positive control group (0.007 mm). The anti-inflammatory effect was concentration-dependent, with lower efficacy observed in the F2 and F1 groups. Conclusion: The ethanol extract of Crinum asiaticum L. leaves can be successfully formulated into a stable stick balm. The 5% EESL stick balm demonstrated significant anti-inflammatory activity that was comparable to a commercial product, making it a promising natural alternative for topical anti-inflammatory therapy.
