Abstract

Abstrak Lebah tanpa sengat atau stinglessbee akhir-akhir ini seringkali dipromosikan sebagai hewan budidaya yang sangat menjanjikan. Akan tetapi masih sangat sedikit literatur yang dapat digunakan dalam pembudidayaan lebah ini. Penelitian ini bertujuan untuk mengetahui karakteristik Beebread yang dikumpulkan oleh 7 spesies stinglessbee. penelitian ini dilaksanakan pada bulan Februari – Mei 2022, sampel beebread diambil langsung dari sarang lebah di PT Suhita Lebah Indonesia kota Bandarlampung dan dari pembudidaya lebah yang ada di desa Harapan Jaya kecamatan Wayratai kabupaten Pesawaran. Sampel dianalisis dengan metode asetolisis dan diamati menggunakan mikroskop serta dikelompokkan berdasarkan ketentuan Wingenroth (2001). Dari pengamatan yang dilakukan diketahui bahwa Spesies Tetrigona apikalis, Tetrigona vidua, dan Tetragonula reepeni memiliki karakteristik beebread monoflora dengan pollen didominasi oleh tumbuhan Cocos nucifera. beebread Tetragonula testaceitarsis dan Tetragonula levicep berkarakter biflora, serta beebread Heterotrigona itama dan Geniotrigona thoracica berkarakter multiflora. Dari ketujuh spesies lebah yang diamati mengumpulkan pollen dari tumbuhan C.nucifera dan memiliki persentase yang tinggi. Pollen C.nucifera dalam jumlah yang signifikan disebabkan oleh preferensi Stinglessbee untuk mencari makan pada bunga dengan konsentrasi gula tinggi yang tersedia sepanjang tahun. Kata Kunci: Beebread, Geniotrigona, Heterotrigona, Stinglessbee, Tetragonula, Tetrigona. Abstract The stingless bee or stinglessbee has recently been promoted as a very promising farm animal. However, there is still very little literature that can be used in the cultivation of these bees. . This study aims to determine the characteristics of beebread collected by 7 species of stinglessbee. This research was conducted in February – May 2022, the beebread samples were taken directly from bee hives at PT Suhita Lebah Indonesia, Bandarlampung city and from bee cultivators in Harapan Jaya village, Wayratai district, Pesawaran district. Samples were analyzed by acetolysis method and observed using a microscope and grouped according to the provisions of Wingenroth (2001). From the observations made, it is known that the species Tetrigona apikalis, Tetrigona vidua, and Tetragonula reepeni have the characteristics of beebread monoflora with pollen dominated by Cocos nucifera plants. Beebreads Tetragonula testaceitarsis and Tetragonula levicep have biflora characteristics, and Heterotrigona itama and Geniotrigona thoracica beebreads have multiflora characteristics. Of the seven species of bees observed, they collected pollen from the C. nucifera plant and had a high percentage. The significant amount of C. nucifera pollen was due to the Stinglessbee's preference for foraging on flowers with high sugar concentrations available throughout the year. Abstract

Pesawaran is one of the areas that has the potential to cultivate stingless bees as producers of honey and propolis. This study was aimed at qualitatively analysing partial amplicons of the 16S rRNA gene from individuals found at the research site. The research was performed from April to September 2023 in an explorative approach. Based on the exploration that has been done in Harapan Jaya Village, Way Ratai District, Pesawaran Regency, individual stingless bees were obtained from 5 different colonies. Individual samples of stingless bees were further extracted and amplified by commercial kits. Each individual from 5 different colonies has successfully obtained its DNA amplicons. Based on qualitative analysis using 1% agarose gel, the size of the partial gene ranged from 400bp to 500bp. This result is in accordance with the data contained in the genbank, where the 16S rRNA gene size is more than 500bp. Therefore, it has been concluded that the size of the partial amplicons of the 16S rRNA gene of individual stingless bees from the 5 colonies that were obtained is actually in range of the size of the 16S rRNA gene of stingless bees that have been recorded in the genbank.
