Abstract

ABSTRACT The objective of this research was to detect the pollution level of Tondano River based on temperature, BOD, turbidity and dissolved oxygen parameters. Research was conducted at Ternate Baru Village, Manado City, from January to March 2013. Water sample was taken by purposive sampling at three stations at 08.00 AM, 11.00 AM, and 15.00 PM, each with two sampling points. Water quality was measured in situ for temperature and turbidity while dissolved oxygen and BOD were analysed in laboratory. Research results found temperature ranged from 270C-27,8 0C, turbiduty 37,9 - 150 NTU, dissolved oxygen 6,3 - 7,5 mg/L and BOD 15,5-44,5 mg/L. Based on Water Quality Standard Grade II PP No. 82, 2001, BOD and turbidity at Ternate Baru were high. It was concluded that water quality of Tondano River at Ternate Baru Village was categorized polluted. Keywords: water quality, pollution, Tondano River, Ternate Baru Village

Abstract

"Gemusis" fish cultivation group is one of the aquaculture business groups that exist in Tendeki Village, Matuari District, Bitung City, North Sulawesi Province. The width of  pond owned is around 3 ha with an average production of 2 tons per year. The main problem in this group is the difficulty in obtaining superior seeds, expensive feed prices and often the fish that are kept stricken with disease, resulting in less production and even crop failure. Alternative solutions to problems in partner problems are accompanied by two approaches, namely the classical and individual approaches. The classical approach is carried out when delivering the material. An individual approach is taken at the time of practice. Lectures in class in the form of delivery of how to make tilapia seed with sex reversal methods for the maintenance of monosex, making low-quality quality feed made from local raw materials and producing maggot as natural food, as well as how to increase fish immunity by giving immunostimulants to fish feed. Demonstration and practice with techniques for making cheap feed ingredients made from local raw materials. In order for the fish cultivation activities carried out by the partners according to the activity plan, monitoring and assistance will be carried out after the lecture and practice activities. The results obtained showed the group are able to cultivate fish with monosex system, produce feed using local raw materials, maggot cultivation as a natural feed, and can treat fish diseases using mmunostimulant.

Abstract

The objective of research was to determine the concentration of honey in sperm dilution  for improving hatching rate of egg and survival of nile tilapia larvae (Oreochromis niloticus).The number of fish used was five pairs broodstock. Dilution substance was NaCl and honey.  Observations were conducted on spermatozoa motility, fertility and egg hatching rate. Experimental design used was complete randomized design. Sperm dilution substance was prepare by disolving honey  (0 mL; 0,60 mL; 0,65 mL dan 0,70 mL) in 100 mL; 99,40 mL; 99,35 mL and  99,30 mL NaCl respectively.  Each dilution was homogenized using aerator for 15 minutes. Fertilization was done 12 hours after ovulation.  Egg hatching rate was observed after ovulation. Research results showed the use of honey had significant effet on egg hatching rate, but not on larval survival.  Treatment D (0,70 mL honey in 99,30 mL NaCl) had the highest hatching rate (77.33%). Dilution ratio 1:60 was the best indicated by spermatozoa motility 96.66%, fertility 71.65, hatching rate 70% and larval survival 81.67%   Keywords : honey, egg hatching rate, larval survival, Oroechromnis niloticus

Abstract

The objective of research was to evaluate the length and diameter of coral Acropora  formosa cultivated with different seed size in Talengen Bay, Tabukan District, Sangihe Island Regency.  Data were collected from May to June 2016.  Experimental design used was Complete Randomized Design with three treatments, each with three replications.  Size of seed as treatment consisted of  A: 5 cm, B: 7 cm and C: 10 cm. The experiment was conducted using bench  system measuring 60 x 60 cm2, while coral seed was A.  formosa.  The seed was gathered from the nature around research location. Growth data and water quality parameters were measured every two weeks .  Research results showed that the highest absolute length growth was achieved in treatment A (average: 0.70 cm), followed by treatment C (average 0.46 cm) and the lowest was in treatment B (average 0.45 cm).  The highest growth of coral diameter was observed in treatment A (average 0.05 cm) and followed by treatment C (average 0.03 cm) and treatment B (0.03 cm). Statistical analysis showed that absolute length growth and coral diameter   were not significant.  A. formosa cultured with different seed length in Talengen Bay had good growth in which the growth in length was faster than the growth in diameter. Abstract

This research was conducted to evaluate the efficacy of baker’s yeast in enhancing nonspecific immune response and growth of Nile tilapia. After two weeks of acclimatization in fiber tank, juveniles were put into glass aquarium at the density of 15 fish/ aquarium. Fish were fed pellet supplemented with baker’s yeast as treatment for four weeks at 5% of body weight per day, twice daily (08.00 and 17.00). Five doses of treatment used in this research were A (0 g yeast/kg pellet), B (10 g yeast/kg pellet), C (20 g yeast/kg pellet), D (30 g yeast/kg pellet), E (40 g yeast/kg pellet), Immune parameters included total leucocyte count (TLC) and phagocytosis activity were measured at the end of experiment. Fish growth was weighing at the end of research period. Anova was used to evaluate the effect of baker’s yeast on immune parameters and growth while to evaluate the different effect between treatment, Duncan test was used. Research result showed that after four week of feeding, TLC of fish fed diet supplemented with baker’s yeast significantly different compared to control fish. The highest TLC of fish was achieved in treatment B followed by treatment C. It was also found that phagocytosis activity of fish increased significantly in fish fed treatment diet with the highest PA was observed in treatment C. PA of fish in treatment C was different significantly compared to a treatment A, D and E but between treatment C and B, no significant difference was observed. Application of baker’s yeast in diet also significantly increased growth of fish (p=0,00). Fish fed diet supplemented with 20 g baker’s yeast/kg pellet has the highest weight gain compared to other fish in other treatments. It was concluded that the used of baker’s yeast at 20-30 g/kg pellet for four weeks could enhance nonspecific immune response and growth of Nile tilapia. Abstract

The purpose of this study was to determine the concentration of the use of the best fermented coconut pulp in feed on the growth of tilapia.  The research  used 12  nets  measuring 1x1 m placed in a concrete pond measuring 3.5 x 5 cm. The treatments were A (without fermented coconut pulp), B (10% fermented coconut pulp), C (20% fermented coconut pulp), and D (30% fermented coconut pulp). The experimental method used was a completely randomized design with 4 treatments and 3 repetitions. Data collected were absolute growth, daily growth, relative growth, and feed efficiency. The results of the variance analysis showed an influence in the growth of tilapia size 5-8 cm. The best growth and feed efficiency value occurred in treatment D Thus  the use of  30% fermented coconut pulp in fish food could improve the growth of tilapia.Key words : fermented coconut pulp, tilapia, growth feed efficiency

Abstract

Crustacean does not have adaptive immune system and mostly depends on innate or nonspecific immune response. This system can recognize and destroy foreign materials including pathogen.  The nonspecific immune responses of crustacean consist of cellular and humoral responses.  In cellular response, hemocyte plays an important role in body defenses to pathogen including virus, bacteria, fungi, and parasites. In humoral response, recognition of pathogen is mediated by protein and enzyme such as β-1,3-glucan-binding protein (BGBP),  lipopolysaccharide-binding protein (LPS-BP), peptidoglycan recognition protein, phenoloxydase enzime. Abstract

The goal of mass culture of chydoridae is to find out the density of chydoridae that was cultured with yeast and rice bran.  Chydoridae was cultured on 5 aquaria i.e 3 aquaria measuring 100 x 60 x cm (A, B and C) and 2 aquaria measuring 30 x 30 x 30 cm (D and E).  Water quality parameters measured were temperature and pH.  The result of chydoridae mass culture for 21 days reached the peak on 14th day with the result as follow : for aquarium A with the concentration of yeast and rice bran 0.05 g/500 mL is 31 individue/ml; aquarium B with the concentration of yeast and rice bran 0.03 g/500 mL is 13 individue/ml; aquarium C with the concentration of yeast and rice bran 0.03 g/500 mL is 5 individue/ml; aquarium D with the concentration of yeast and rice bran 0.005 g/500 mL and aquarium E with the concentration of yeast and rice bran 0.003 g/500 mL can not be detected.  Counting the density of chydoridae on 21st day gave the result as follow : aquarium A as much as 4 individue/ml; aquarium B as much as 3 individue/ml; aquarium C as much as 2 individu/ml; aquarium D as much as 1 individue/ml and aquarium E as much as 2 individu/ml.  During culturing, water quality such as temperature and pH were on the range that was not harmful the chydoridae life.   Keywords: zooplankton, life food, chydoridae, fresh water, mass culture

Abstract

This study was aimed to find out the growth rate of Nile Tilapia (Oreochromis niloticus) cultured with biofloc technology, and to understand the dynamic of several water quality parameters during culture period. This experiment was conducted in the Laboratory of Aquaculture Technology, FPIK, UNSRAT.  Several substances were added to stimulate the development of biofloc, which were: 0,3 mL of EM-4 containing probiotic-bacteria Lactobacillus casei dan Saccharomyces cerevisiae.; 15 mL of mollases;  0,5 g of  yeast commonly used to ferment soybean ; 12 g of dolomite.    60 seeds of  nile tilapia,  with an individual weight of 6 g, were stocked in each container.   Fish were fed at a dose of 3% of the total biomass/day at the first week, and the dose was decreased  down to 1% of the total biomass per day afterwards.  Fish were weighed at day-1, day-15 and day-30.   The change of water color and the density of the floc was consistently evaluated using imhoff-cone. The initial weight of the tested individual fish were 6,00 g. After 15 days, the weight was increased to 7,37g, and at the end of the experiment the individual fish weighed 11,47 g. Hence, the absolut growth of the tested fish was 5,47g,  the specific growth rate was 91% and the daily growth rate was 2,11%.    The density of flok reached 9,5 mL/L at the beginning of the second week, which was categorized as very high density.   The density of floc then was decreased to 4 mL/L.   The density of the floc was maintained around 5 mL/L during the experiment. The temperature of the medium was at the range of 26-300C.  The level of ammonia was 0,03mg/L and  the level of  nitrite fluctuated between 0,15-3mg/L, which is normal and safe for biofloc system.  The level of nitrate was 4-55 mg/L, which was quite high for biofloc system but not harmful for fish.   Keywords:  Biofloc, Nile-Tilapia, growth, floc-density

Abstract

The objective of research was to isolate the probiotic bacteria from intestine of carp.  Isolation was performed by weighing 1 g of intestine, crushed and inserted in a reaction tube containing 9 mL of NaCl then centrifuged. Supernatant was taken and spread over the agar medium by scatter method. The bacteria-stocked medium was then incubated in an incubator at 28°C for 24-48 hours. The growing colonies were re-isolated on the agar medium to obtain pure colonies. The bacteria were then identified by morphological observation, resulting pigment and gram staining. Five nile tilapia (sized 8 – 10 cm) were used in pathogenicity test and placed in the aquarium. Fish were injected with 0.2 ml of Lactobacillus solution containing 1x107 CFU/mL via intraperitoneal injection. The results showed that the bacteria Lactobacillus sp had rod-shaped characteristics, white pigment and gram positive and  not pathogenic, which means it can be used as probiotic for aquaculture.Keywords : Lactobacillus sp, carp, probiotic, pathogenicity test
