Abstract

This research aimed to diagnose bacterial diseases on nile tilapia cultured in stick net cage in Lake Tondano. Fish samples were taken from three locations of net cages in Lake Tondano namely Paleloan, Eris and Remboken villages. Three samples of fish were taken from each location/village. The samples taken showed clinical symptoms of bacterial infection such as hemorrhagic on the abdomen, the base of the fin, anal and chest, irregular swimming fish, hemorrhagic on the gills, fish refuse food, remain silent on the outskirts of net cage. Bacteria were isolated by using a sterile loop from head kidney (anterior kidney), which is located on the front, below the spine near the head. Bacterial cultures were performed on TSA media by making a zigzag scratches. The petri dish containing bacterial isolates were subsequently incubated in an incubator at a temperature of 35oC for 24 hours. The water quality parameter data measured consisted of temperature, Dissolved Oxygen, pH, ammonia, brightness. The results of the study showed that farmed tilapia in Lake Tondano has been infected with Aeromonas hydrophila that causes MAS (Motile Aeromonad Septicemia). The water quality, both chemical and physics around the net cages in the Lake Tondano was still in good condition and suitable for fish cultivation.   Keywords: Oreocrhomis niloticus, Bacterial disease, septicemia aeromonad motile, fixed net cage, Lake Tondano

Abstract

The research intended to determine the optimal concentration of honey addition in sperm dilution to improve sperm motility, fertilization and eggs hatchability of goldfish (Cyprinus carpio). Experimental design used was a Complete Randomized Design (CRD) with four treatments each with three replications. The concentration of honey addition used for treatment were A (0 ml of honey in 100 mL of physiological NaCl), B (0,2 mL of honey in 99.8 mL of physiological NaCl), C (0.4 mL of honey in 99.6 ml of physiological NaCl), D (0.6 mL of honey in 99.4 mL of physiological NaCl). Sperm dilution containers used was 12 plastic cups and the container fertilization and hatching eggs container used was 12 brass plates.  Spermatozoa motility observation performed immediately after dilution process. Fertilization observation was performed after 12 hours of fertilization process, and Egg Hatchability observation was performed after 72 hours of fertilization.  The observation results showed that the addition of honey in sperm dilution contributed significant effect on spermatozoa motility, fertilization and eggs hatchability of goldfish (Cyprinus carpio). This observation showed that the addition of 0.6 ml of honey in a sperm dilution was the best treatment indicated by the average percentage of spermatozoa motility (80,00%), fertilization (84,00%) and eggs hatchability (82,33%).   Keywords: Honey, spermatozoa motility, fertilization, eggs hatchability, Cyprinus carpio L

Abstract

Absolute Growth, hemolymp  osmolality, oxygen consumption, and basal energy of  Litopenaeus vannamei postlarvae were investigated. After post larval stage, PL-20, the postlarvae were acclimatized from sea water (25 ppt) to low salinity  water (2 ppt) over 96 h. Treatments consisted of different concentration of K+added to distilled water. Four different levels of K+ (0, 30, 60, and 90 ppm) were utilized. Ten shrimp were placed in triplicate 40-L glasses aquarium.  Results from the 42-day K+ growth trial indicated significant differences (P< 0.05) in absolute growth, hemolymp osmolality,  oxygen comsumption, and  basal energy. The values in treatment A was significantly higher than than those under the other four treatments. The results suggest treatment C were the optimum K+ level for culture  vaname postlarvae in low salinity water. Keywords :  Litopenaeus vannamei, oxygen comsumption, hemolymp

Abstract

This study was carried out in the present farming sites of Kappaphycus alvarezii in Minahasa penninsula, Jayakarsa village of North Minahasa, Sondaken of South Minahasa, Buku Tengah of  Southeast Minahasa, and Tumpaan of Minahasa Regencies. It was aimed to know the farming system and financially analyze whether the farming business is feasible or not.  Data were collected through observations, questioneers and interviews.  Literature reviews and financial analyses were also done to know the revenue, the feasibility of  seaweed, K. alvarezii, farming business and its development. The farming system in Minahasa penninsula generally used longline method.  It is used because of being resilient to strong wind, cheap, and easily implemented and long lived so that the line could be repeatedly used up to  4-5 years.  Financial analyses found that among 4 study sites, the highest B/C ratio was recorded in North Minahasa, 6.06, followed by South Minahasa, 2.75, then Southeast Minahasa and Minahasa, 1.67, and BEP values were also different among regencies.  Furthermore, NPV, IRR, ROI and PP of all study sites belonged to feasible criteria for development.  Different conditions among regencies resulted from different geographic conditions affected the planting system.
