Abstract

INDUCTION MUTATION AND SELECTION OF IN VITRO PLANT OF WHEAT (Triticum aestivum L.)The goal of this research was to produce wheat crop which is tolerant to lowland condition.Six varieties were used, Dewata, Selayar, Alibey, Oasis, Rabe and HP1744. This research consisted of 4 stages: production of the best callus on MS medium containing 3 g/L 2.4-D, induced mutation of embryogenic callus using EMS, in vitro selection of callus at temperature of 27–35°C, and callus regeneration. The best result for callus production was 76% for Dewata and 70% for Selayar varieties. Higher concentration of EMS and longer soaking time decreased the percentage of callus growth. LC50 for Dewata was 0.3% EMS at 30 minutes and that for Selayar was 0.1% EMS at 60 minutes. The higher the temperature was, the lower was the adaptation tolerant of the plants, and callus growth was inhibited. At the highest temperature (35°C) the callus did not grow at all.Keywords: Induced mutation, Triticum aestivum, EMS, in vitro selection, callusABSTRAKTujuan penelitian ini adalah untuk merakit tanaman gandum yang toleran pada dataran rendah. Varietas yang digunakan ada 6 yaitu Dewata, Selayar, Alibey, Oasis, Rabe dan HP-1744. Penelitian terdiri atas empat tahap yaitu induksi pembentukan kalus terbaik menggunakan media MS + 3 g/L 2,4-D (dipilih dua varietas yang terbaik), induksi mutasi kalus embriogenik menggunakan EMS, seleksi kalus in vitro pada suhu 27–35°C, dan regenerasi. Hasil induksi kalus terbaik terdapat pada varietas Dewata sebesar 76% dan Selayar sebesar 70%. Semakin tinggi konsentrasi EMS dan semakin lama waktu perendaman yang digunakan maka semakin menurun persentase pertumbuhan kalus. LC50 varietas Dewata adalah EMS 0,3% waktu 30 menit sedangkan LC50 varietas Selayar adalah EMS 0,1% waktu 60 menit.Semakin tinggi suhunya maka semakin berkurang toleran adaptasi tanaman tersebut, dan pertumbuhan kalus semakin sedikit. Bahkan pada suhu tertinggi yaitu suhu 35°C tidak ada pertumbuhan kalus sama sekali.Kata Kunci: Induksi mutasi, Triticum aestivum, EMS, seleksi in vitro, kalus

Abstract

Process Optimization for Endoglucanase Gene Expression Derived from Bacillus sp. RP1 by Escherichia coli BL21 (DE3)/egcABSTRACTCellulases are one of the most used enzymes in industrial processes. In an effort to increase production, industries have developed strategies such as isolating new cellulase producing strains, genetic engineering and process optimization since the last 50 years. One endoglucanase producing strain, Bacillus sp. RP1 was isolated from hot springs. The ribosome binding site and coding sequence of the endoglucanase gene (egc) from Bacillus sp. RP1 was cloned into pGEM-T Easy. The recombinant plasmid was used to transform E. coli BL21 (DE3). Cloning was followed by process optimization. Medium composition was selected using Plackett-Burman design. The medium components tested were rice hull, molasses, ammonium chloride, urea and fishmeal. Rice hull and molasses were found to be the factors most influencing enzyme activity and dry cell weight, respectively. The next step involved Box-Behnken method and response surface methodology to optimize the responses against molasses concentration, rice hull concentration and fermentation time. The concentration intervals used to test were 1%, 5.5% and 10% while the fermentation time used were 24, 36 and 48 hours. The conditions which optimized both enzyme activity and dry cell weight were 7.45% molasses, 6.45% rice hull and 39.52 hours of fermentation.Keywords: Bacillus sp. Sebagai upaya untuk memenuhi kebutuhan, 50 tahun terakhir dikembangkan beberapa strategi untuk meningkatkan produksi selulase yang mencakup rekayasa genetika dan optimasi proses. Karena itu, dilakukan kloning gen egc dan RBS yang berasal dari Bacillus sp. RP1 yang diisolasi dari sumber air panas ke dalam vektor pGEM-T Easy. E. coli BL21 (DE3) ditransformasikan dengan vektor yang mengandung gen egc tersebut. Setelah kloning, optimasi proses berupa desain medium turut dilakukan untuk mengoptimalkan ekspresi gen egc. Desain medium diawali dengan seleksi komposisi medium menggunakan metode Plackett-Burman. Komponen medium yang diuji adalah kulit beras, molase, amonium klorida, urea dan tepung ikan. Kulit beras dan molase diperoleh sebagai bahan yang paling berpengaruh terhadap aktivitas enzim dan berat kering sel. Tahap selanjutnya melibatkan metode statistik Box-Behnken dan metodologi respons permukaan yang bertujuan mengoptimalkan respons aktivitas enzim dan berat kering sel terhadap konsentrasi molase, konsentrasi kulit beras dan lama fermentasi. Konsentrasi yang diuji adalah 1%, 5,5% dan 10%, sedangkan lama fermentasi yang diuji adalah 24, 36 dan 48 jam. Konsentrasi optimal molase adalah 7,45% dan konsentrasi optimal kulit beras adalah 6,45% dengan lama fermentasi optimal 39,52 jam.Kata Kunci: Bacillus sp. Abstract

Genetic Diversity of 22 Local Rice Accessions from North Toraja Based on Simple Sequence Repeats (SSR) MarkersABSTRACTOne way to explore the potential of local rice is by the characterization that could obtain genetic diversity of that plants. The aim of this study was to obtain the genetic diversity of 22 local rice accession from North Toraja. Twenty-two of local rice accessions from North Toraja were characterized by 30 SSR markers and using NTSYS pc 2.1 program to analyze genetic diversity. The results showed that twenty-six SSR markers that had been analyzed produced some alelles with a size between 106.75-311 bp, the average number of alleles were 3 and the polymorphism rate was 0.53. On coefficient genetic similarity at 0.38, the population formed three clusters. Cluster I and II were dominated by rice that had no hair on the tip of the grain and cluster III were dominated by rice that had hair on the tip of the grain. There were 105 opportunities to crossing between accessions when the genetic distance was above 0.7.Keywords: genetic diversity, local rice, North Toraja, polymorphism rate, SSR markers ABSTRAKSalah satu cara untuk menggali potensi padi lokal adalah dengan karakterisasi. Dengan adanya kegiatan karakterisasi tersebut maka dapat diketahui bagaimana keragaman genetik dari suatu tanaman. Penelitian ini bertujuan untuk mengetahui keragaman genetik dari 22 aksesi padi lokal Toraja Utara. Duapuluh dua aksesi padi lokal Toraja Utara dikarakterisasi menggunakan 30 marka SSR dan dianalisis keragaman genetiknya menggunakan program NTSYS pc 2.1. Hasil penelitian menunjukkan bahwa duapuluh enam marka SSR yang dianalisis memiliki kisaran ukuran alel antara 106.75-311 bp, dengan jumlah alel rata-rata 3 dan tingkat polimorfisme sebesar 0,53. Koefisien kemiripan genetik 0,38 dan terbentuk 3 klaster. Pada klaster I dan klaster II didominasi oleh padi yang tidak memiliki rambut pada ujung gabahnya, dan pada klaster III didominasi oleh padi yang memiliki rambut pada ujung gabahnya. Selain itu, pada jarak genetik diatas 0,7 terdapat 105 peluang persilangan.Kata Kunci: keragaman genetik, marka SSR, padi lokal, tingkat polimorfisme, Toraja Utara

Abstract

To reduce broiler production cost, a study was carried out on utilisation of alternative, less costly feed components, namely kepok banana peel and corn hominy which were mixed and fermented using the fungus Rhizopus oryzae. The fermented substrate was added into commercial feed in order to determine its influence on the poultry’s production performance. This study employed a completely randomized design (CRD) with four treatments: one using 100% pure commercial feed, and the other three commercial feed mixed with the fermented feed component at the levels of 5%, 10% and 15%. All treatments were repeated four times during 25 days growth period. The results showed that supplementation of the fermented ingredient in the broiler ration at the levels of 5%, 10%, or 15% had no significant effect (P>0,05) on the feed consumption, body weight gain, feed conversion ratio, and the final body weight of the animals. Although mixing the fermented feed ingredient up to 15% in the commercial feed promoted the growth of the broilers, the results were not yet comparable to pure commercial feed.Keywords: Corn hominy, broiler, fermentation, kepok banana peel, Rhizopus oryzae AbstrakUntuk mengurangi biaya produksi ayam pedaging, dilakukan pengkajian penggunaan bahan penyusun pakan alternatif yang lebih murah, yaitu kulit pisang kepok dan ampok jagung yang dicampur dan difermentasi menggunakan jamur Rhizopus oryzae. Hasil fermentasi tersebut lalu ditambahkan pada pakan komersial ayam pedaging dengan tujuan mengetahui pengaruhnya terhadap penampilan produksi ayam pedaging. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan empat perlakuan, yaitu satu perlakuan 100% pakan komersial murni, dan tiga perlakuan pakan komersial yang dicampur bahan pakan hasil fermentasi tersebut dengan kadar 5%, 10%, dan 15%. Seluruh perlakukan diulang empat kali selama 25 hari masa pemeliharaan. Hasil penelitian menunjukkan bahwa penambahan bahan pakan terfermentasi ke dalam ransum ayam pedaging pada kadar 5%, 10%, atau 15% tidak memberikan pengaruh nyata (P>0,05) terhadap konsumsi pakan, pertambahan bobot badan, nilai koversi pakan, dan bobot badan akhir hewan. Meskipun penambahan bahan pakan fermentasi tersebut hingga 15% dalam pakan komersial mendukung pertumbuhan ayam pedaging, namun capaian pertumbuhannya belumlah sebaik pakan komersial.Kata kunci: Ampok jagung, ayam pedaging, fermentasi, kulit pisang kepok, Rhizopus oryzae

Abstract

Potency of Plant Bioactive Compounds from the Genus Phyllanthus as Hepatitis B Virus Replication InhibitorIn this research, simulations of molecular docking of Phyllanthus bioactive compounds were performed into the core protein of HBV. This simulation aimed to predict the interaction between compounds with virus core protein causing disruption of capsid formation and inhibiting its replication. The docking simulation was completed by Molegro Virtual Docker 6.0. The 3D stable conformation of molecule structures were docked into HBV core protein downloaded from Protein Data Bank, then the results were analyzed to view the minimum energy and interactions that occurred. The coordinate docking was done at the same coordinate as the previously docked reference ligand position and was validated. From the results it was known that repandusinic acid formed the most stable affinity bond with amino acid residues of viral core proteins. Interaction of B chain forming hydrogen bonds with the amino acid residues of Thr 33, Trp 102, Phe 23, Leu 140, Tyr 118 and Ser 141, and C chain with Thr 128, Val 124 and Glu 117.These compounds can be used as marker for anti HBV.Keyword: Bioactive compounds, core protein, HBV , molecular docking, Phyllanthus ABSTRAKPada penelitian ini dilakukan simulasi penambatan molekul senyawa-senyawa bioaktif Phyllanthus ke dalam protein inti virus hepatitis B. Simulasi ini bertujuan untuk memprediksi interaksi terbentuk antara senyawa dengan protein yang menyebabkan terganggunya pembentukan kapsid virus dan menghambat replikasinya. Simulasi penambatan molekul dilakukan menggunakan program Molegro Virtual Docker 6.0. Sebagai reseptor target digunakan struktur 3D protein inti yang diunduh dari Protein Data Bank. Posisi penambatan dilakukan pada koordinat yang sama dengan posisi ligan referensi yang sudah tertambat sebelumnya dan tervalidasi. Dari hasil simulasi diketahui bahwa asam repandusinat membentuk komplek dengan energi afinitas ikatan yang paling kecil dengan residu asam amino protein inti virus. Interaksi terjadi dengan rantai B yang membentuk ikatan hidrogen dengan asam amino Thr 33, Trp 102, Phe 23, Leu 140, Tyr 118 dan Ser 141, dan rantai C dengan asam amino Thr 128, Val 124 dan Glu 117.
