Abstract

ABSTRACT Disease-free potato seeds of high quality can be obtained via in vitro culture. The use of liquid medium during in vitro cultures might boost the number of plantlets produced, however, the problem of hyperhydricity in plantlets was often encountered. This study aimed to investigate effects of different sucrose concentrations and application of aeration system on micropropagation of potato cv. Granola using liquid medium. Aseptic nodal explants with 3-4 nodes from established in vitro cultures were subjected to MS liquid medium with a factorial treatment of three sucrose concentrations (0, 7.5, 15 g. L-1) and two culture  (with and without aeration). The results showed that MS medium with 7.5 g L-1 sucrose was the best medium to produce the highest number of shoots and nodes. Furthermore, it was found that application of aeration system in MS liquid culture decreased plantlet hyperhydricity and increased the number of shoots, number nodes, plantlet height, as well as improved plantlet morphology and vigor. Application of the aeration system in liquid medium produced 200-230 new potato plants per bioreactor in the acclimatization stage and an average of 2773.5 G0 mini tubers.   ABSTRAK Benih kentang bebas penyakit berkualitas tinggi dapat diperoleh dari kultur in vitro.  Penggunaan media cair selama kultur in vitro dapat meningkatkan jumlah planlet yang dihasilkan, namun masalah hiperhidrisitas plantlet masih sering dijumpai. Penelitian ini bertujuan mengetahui pengaruh perbedaan konsentrasi sukrosa dan penerapan sistem aerasi terhadap perbanyakan kentang cv. Granola menggunakan media cair. Eksplan dengan 3-4 nodus dari kultur in vitro ditumbuhkan pada media cair MS dengan perlakuan faktorial, faktor pertama: tiga tingkat konsentrasi sukrosa (0, 7,5, 15 g. L-1) dan faktor kedua: dua sistem kultur (dengan dan tanpa aplikasi aerasi).  Hasil menunjukkan bahwa media MS dengan 7,5 g. L-1 sukrosa merupakan media terbaik untuk menghasilkan jumlah tunas dan nodus terbanyak. Selanjutnya ditemukan bahwa penerapan sistem aerasi pada kultur cair MS menurunkan hiperhidrisitas plantlet dan meningkatkan jumlah tunas, jumlah buku, tinggi plantlet, serta memperbaiki morfologi dan vigor plantlet. Penerapan sistem aerasi pada media cair menghasilkan 200-230 tanaman kentang baru per bioreaktor dalam tahap aklimatisasi dan rata-rata 2773,5 G0 umbi mini.

Abstract

Monstera adansonii has wide leaves with a split pattern on its strands (split leaf), smooth and shiny to create a basic visualization (background filler) in flower arrangements. This study aims to obtain genetic changes using chemical mutagens. Chemical mutagens used were 250 ppm EMS, 400 and 800 ppm streptomycin, 400 and 800 ppm GA3. The observations were performed after treatments on the parameter of amount of chlorophyll, stomata and leaf color changes. The results showed that both streptomycin treatments significantly reduced the plant height and the number of leaves. The application of streptomycin in high concentration (800 ppm) decreased the total amount of chlorophyll content in the leaves and changed the color as well as the shape of the Monstera leaves. All chemical mutagen treatments had no effect on stomatal density and stomatal area. The changes in leaf color and shape occurred in the high concentration of 800 ppm streptomycin treatment, could not detected by SSR marker. Monstera adansonii memiliki daun yang lebar dengan pola belahan pada helaian (split leaf), halus dan mengkilap dapat membuat visualisasi dasar (background filler) pada rangkaian bunga. Penelitian ini bertujuan untuk memperoleh perubahan genetik dengan berbagai mutagen kimia. Mutagen kimia digunakan yaitu EMS 250 ppm, streptomisin 400 dan 800 ppm, serta GA3 400 dan 800 ppm. Pengamatan dilakukan setelah perlakuan, yaitu jumlah klorofil, stomata dan perubahan warna daun. Hasil penelitian menunjukkan bahwa perlakuan streptomycin secara nyata mengurangi tinggi tanaman dan jumlah daun. Sedangkan perlakuan streptomycin dengan konsentrasi tinggi (800 ppm) berhasil menurunkan jumlah klorofil total, serta merubah warna dan bentuk daun Monstera. Semua perlakuan mutagen kimia tidak berpengaruh terhadap kerapatan stomata dan luas bukaan stomata. Perubahan warna dan bentuk daun yang terjadi pada perlakuan streptomisin 800 ppm, tidak dapat dideteksi dengan marka SSR.

Abstract

Mangrove endophytic fungi have the potential to produce secondary metabolites with antibacterial properties. This study aimed to obtain endophytic mold isolates associated with mangrove plants and their antimicrobial activity. Seventeen endophytic molds were isolated from 7 mangrove species originating from Berau, East Kalimantan, Indonesia. Three extracts from the mold isolates OJ-B, OJ-Bu, and 6Xg-Bh showed the highest antibacterial activity with an inhibition zone of 9.63–25.4 mm and with MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration) values ??of around 0.625–5 and 2.5–10 mg mL–1, respectively. LC-MS (liquid chromatography-mass spectrometry) analysis identified several compounds with antibacterial potential, namely beauvericin, choline, nicotinic acid, pyridoxine, quinoline, cyclo (phenylalanyl-prolyl), fusarin c, oleamide and borrelidin. The 3 species were molecularly identified as Diaporthe phaseolorum, Fusarium proliferatum and Phomopsis sp. Thus, the endophytic molds from several of the mangrove species produced bioactive compounds as antibacterial.  Kapang endofit mangrove berpotensi menghasilkan metabolit sekunder yang bersifat antibakteri. Penelitian ini bertujuan mendapatkan isolat kapang endofit yang berasosiasi dengan tanaman mangrove beserta aktivitas antimikrobanya. Tujuh belas kapang endofit diisolasi dari 7 spesies mangrove asal Berau, Kalimantan Timur, Indonesia. Tiga ekstrak dari isolat kapang OJ-B, OJ-Bu, dan 6Xg-Bh memiliki aktivitas antibakteri tertinggi dengan zona hambat 9.63–25.4 mm dan dengan nilai MIC dan MBC masing-masing sekitar 0.625-5 dan 2.5–10 mg mL–1. Analisis LC-MS menunjukkan beberapa senyawa dengan potensi antibakteri yaitu beauvericin, choline, nicotinic acid, pyridoxine, quinoline, cyclo (phenylalanyl-prolyl), fusarin c, oleamide dan borrelidin. Uji molekuler menunjukkan 3 spesies terindentifikasi sebagai Diaporthe phaseolorum, Fusarium proliferatum dan Phomopsis sp.  Hasil menunjukkan kapang endofit dari beberapa spesies mangrove tersebut mampu menghasilkan senyawa bioaktif sebagai antibakteri.
