Abstract

Penelitian ini bertujuan untuk menganalisis senyawa metabolit sekunder golongan triterpenoid dari hasil fraksinasi ekstrak air buah buncis (Phaseolus vulgaris Linn). Ekstrak buah buncis di ekstrak dengan cara maserasi dengan pelarut air (1: 18 w/v) dan dianalisis menggunakan KG-SM. Hasil uji pendahuluan menghasilkan bahwa ekstrak kental air buah buncis positif mengandung triterpenoid, Hasil analisis KG-SM menunjukkan bahwa senyawa triterpenoid terbanyak terdapat pada fraksi ekstrak metanol yaitu dua senyawa tetrasiklik triterpenoid tipe lanostane: 9,19-cyclolanost-24-en-3-ol (cycloartenol) yang memiliki rumus molekul C30H50O (m/z = 426) dan senyawa 9,19-cyclolanost-24-en-3-ol,acetat yang memiliki rumus molekul C32H52O2 (m/z = 468) dan hasil fraksinasi kromatografi kolom menghasilkan 78 fraksi dengan hasil uji pereaksi Lieberman Burchard menunjukkan bahwa positif terbanyak mengandung triterpenoid berada pada fraksi 10, fraksi 11 dengan perbandingan eluen heksana : etil asetat = 7 : 3 v/v dan fraksi 17, fraksi 18 dengan perbandingan eluen heksana : etilasetat = 5: 5 v/v.This study aimed to analyze the class of secondary metabolites triterpenoids from the fractionation of aqueous extract of fruit beans (Phaseolus vulgaris Linn). Fruit extract beans in a solvent by maceration with water (1: 18w / v) and analyzed using GC-MS. Lieberman Burchard reagent test results showed that the water condensed fruit beans extract containing triterpenoids and GC-MS analysis showed that the highest triterpenoid compound present in the methanol extract fraction triterpenoid compounds tetrasiklik types lanostane: 9.19-cyclolanost-24-en-3-ol (cycloartenol) which has the molecular formula C30H50O (m / z = 426) and compound9.19-cyclolanost-24-en-3-ol, acetate which has the molecular formula C32H52O2 (m / z = 468) and the results of the fractionation column chromatography produce 78 fractions with Lieberman Burchard reagent test results showed that the most positive fractions containing triterpenoids are at 10, 11 fractions with a ratio of eluent hexane: ethyl acetate = 7: 3 v / v and fraction 17, fraction 18 with a ratio of eluent hexane: ethyl acetate = 5 : 5v / v.

Abstract

ABSTRACTKamu, V. S, 2011. Utilization of pepaya rind extract in VCO production process.A research was done to determine optimum concentration of papaya rind extract in VCO (Virgin Coconut Oil)production process. Concentration of pepaya rind extracts were 10, 20 and 30% used in VCO productionprocess. VCO yield from the process were determine and purified with activated carbon and zeolite, thus storedfor 0, 1, 2, 3, and 4 weeks. Purified VCO were evaluated its moisture content and free fatty acid. Data wereanalyzed by ANOVA from two replication data by SPSS 15.0.The results show that optimum yield of VCO came from concentration of papaya rind extract 20% was 21,9 %.Purifying oil ((VCO) with adsorbent reduce moisture content and fre fatty acid content significantly, so oil quality(VCO) become better, and the best adsorbent is active carbon.Keywords : virgin coconut oil, papaya, moisture content, adsorbent

Abstract

The objective of this research was to determine singlet oxygen quenching (SOQ) effects of colorantextracts of spinach leave on erythrosine-sensitized photooxidation of linoleic acid, protein and vitamine C.Spinach leave was extracted with water, ethanol 40%, 60% and 80% for 30 minutes. Analyses ofantioxidant phytochemicals were based on total phenolic and flavonoid contents. The extracts of spinachleaf evaluated singlet oxygen quenching effect using linoleic acid, protein and vitamine C as substrates eachcontaining 5 ppm erythrosine was exposed under fluorescent light (4000 lux). Extraction with ethanol 40%showed the highest yield of extract, followed with ethanol 60%, water and ethanol 80%. The result showedthat solvent was significantly have an effect on total phenolic content, which ethanol 60% extract was thehighest, followed with 80 and 40% ethanol extracts and water extract. Contrary, ethanol 40%, extractpossessed highest total flavonoid content than water extract, ethanol 60% and ethanol 80% extract.Futhermore, the result determined SOQ, water extract possessed highest SOQ compared with ethanol40%, 60% and 80% in photooxidation of linoleic acid system. However, ethanol extract 40% and ethanolextract 60% showed that highest activity than ethanol extract 80% and water extract in photoxidation ofprotein system whereas in photooxidation of vitamin C system, ethanol extract 60% showed higest activitycompared with etnanol extract 80, ethanol extract 40% and water. Abstract

ABSTRAK Penelitian ini bertujuan untuk menentukan aktivitas antifotoksidasi mengukur nilai SPF dari ekstrak metanol daun Gedi (Abelmoschus manihot L). Daun gedi diekstraksi dengan cara sokletasi.  Dalam penelitian ini sampel diekstraksi dengan pelarut metanol, difraksinasi dengan pelarut petroleum eter, etil asetat, butanol dan akuades. Dari hasil fraksinasi tersebut kemudian diuji aktivitas antifotooksidasi dan fotoprotektif. Kandungan fenolik menggunakan asam galat sebagai larutan standart. Uji Aktivitas antifotooksidasi ditentukan dengan menghitung nilai diena terkojugasi yang merupakan pembentukan awal oksidasi pada lipid. Semakin rendah nila diena terkonjugasi, maka semakin baik sebagai antifotooksidasi. Untuk penentuan fotoprotektif, dilakukan dengan mneggunakan spektrofotometer UV-Vis pada panjang gelombang 290-320 nm dengan konsentrasi 100 μg/mL. Hasil penelitian ini menunjukkan bahwa fraksi etil asetat memiliki kandungan fenolik, aktivitas antioksidan, antifotooksidasi, dan fotoprotektif yang paling tinggi. Hasil yang diperoleh menunjukkan total fenol paling tinggi pada fraksi eti asetat yaitu 110.204 µg/mL, sedangkan untuk aktivitas antioksidan 57.72%. Hasil pada aktivitas antifotooksidasi menunjukkan bahwa fraksi Etil asetat menunjukkan kemampuan yang paling baik dan untuk fotoprotektik memiliki nilai SPF yang tinggi adalah etil asetat yaitu 9.03.  ABSTRACT This study aimed to determine the antifotoxidation activity measuring the SPF value of the solvent fraction of the leaves of Gedi (Abelmoschus manihot L). Gedi leaves extracted by socletation. In this study the samples were extracted with methanol solvent, fractionated with petroleum ether, ethyl acetate, butanol and aquades. The results of the fractionation were then tested for antifotooxidation and photoprotective activity. Phenolic content uses gallic acid as a standard solution. The antifotoxidation activity test is determined by calculating the conjugated diene value which is the initial formation of oxidation in the lipid. The lower the conjugated diene tilapia, the better it is antifotoxidation. For photoprotective determination, a UV-Vis spectrophotometer was carried out at a wavelength of 290-320 nm with a concentration of 100 μg / mL. The results of this study indicate that the highest ethyl acetate fraction has the highest phenolic content, antioxidant activity, antifotoxidation and photoprotective. The results obtained showed the highest total phenol in the eti acetate fraction of 110,204 µg / mL, while for antioxidant activity 57.72%. The results of the anti photoxidation activity showed that the ethyl acetate fraction showed the best ability and for the photoprotectic the high SPF value was ethyl acetate namely 9.03.

Abstract

The objectives of the present research were evaluated effect of solvent extraction on antiphotooxidative from cloveleave (Eugenia aromatic (L). A certain quantity of powdered sample was extracted with methanol 80%, ethanol 80%and aseton 80%. After evaporation the extracts were dissolved in methanol and assayed for phenolic, flavonoid andcondensed tannin content. Singlet oxygen quenching and free radical activity of each extracts of spices at 500 ppmlevel were evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical. Anti-photooxidative activity test wasconducted using linoleic acid as substrates each containing 5 ppm erythrosine as a photosensitizer. The resultshowed that solvent was significantly having effect on total phenolic, flavonoid and condensed tannin content.Methanol extract possessed highest total flavonoid and condensed tannin content than aseton and etanol. Methanolextract of clove leave was found to have the highest anti-photooxidative effect, followed by aseton and ethanol. Inaddition, ekstrak methanol showed any difference in it free radical scavenger effect than that of ethanol and asetonextracts. This results show that clove methanol extract has the best activity on photooxidation of linoleic acid andscavenger of free radical DPPH. These results suggest that the methanolic extract of clove leave contains somecompounds having singlet oxygen quenching and free radical scavenging activities on linoleic acid and free radicalDPPH.Penelitian ini bertujuan untuk mempelajari pengaruh pelarut ekstraksi terhadap aktivitas antifotooksidan dankomposisi fenolik yang terdapat pada ekstrak daun cengkeh. Serbuk daun cengkeh diekstraksi dengan metanol 80%,etanol 80% dan aseton 80%. Setelah evaporasi, ekstrak daun cengkeh dilakukan pengujian untuk kandungan fenolik,flavonoid dan tannin terkondensasi. Aktivitas penstabilan oksigen singlet dan radikal bebas dari ekstrak dievaluasidengan menggunakan asam linoleat sebagai substrat yang mengandung 5 ppm eritrosin sebagai fotosensitiser danradikal bebas 1,1-diphenyl-2-picrylhydrazyl (DPPH). Hasil penelitian ini menunjukkan bahwa pelarut ekstraksimemiliki efek sangat signifikan terhadap kandungan fenolik, flavonoid dan tannin terkondensasi. Pelarut ekstrakmetanol memiliki kandung flavonoid dan tannin terkondensasi paling tinggi daripada aseton dan etanol. Ekstrakmetanol daun cengkeh ditemukan mempunyai efek antifotooksidatif diikuti ekstrak etanol etanol dan aseton. Selainitu, pelarut methanol juga menunjukkan suatu perbedaan dalam penangkalan radikal bebas DPPH. Berdasarkananalisis komposisi fenolik dalam daun cengkeh dengan HPLC teridentifikasi senyawa eugenol, asam galat, kuersetin,rutin dan kaempferol. Hasil ini menyimpulkan bahwa ekstrak metanol daun cengkeh mengandung beberapasenyawa fenolik yang mempunyai aktivitas sebagai penstabilan oksigen singlet dan penangkalan radikal bebas.

Abstract

Traditional medicine usage can be a breakthrough for alternative medication with regard of the resistancy ofdiseases-agent to common drugs. Lansium domesticum has been reported containing therapeutic-substancestherefore, has the capacity as a medicine. This research aimed to further elaborate the antibacterial activities ofseeds, fruitshield and bark skin of Lansium domesticum. The in vitro experimental research was conducted,initiated by extract maseration and continued by antibacterial test to bacterial isolation: Salmonella typhii,Escherichia coli, Vibrio cholerae, dan Staphylococcus aureus. Antibacterial activities test has been done bycomparing to several antibiotics and statistically test by ANOVA followed by LSD or BNT test. The study resultshowed that the extract of L. domesticum effectively inhibited the bacterial’s activities. Fitochemistry test of theextract should be conducted to detect the active compound as well as in vivo research for treatment againstbacterial.

Abstract

ABSTRAK Pisang, jagung dan sagu merupakan sumber makronutrien, vitamin, mineral, serat pangan dan fitokimia. Tujuan penelitian adalah untuk menentukan aktivitas antioksidan dan fotoprotektif tepung komposit dari pisang goroho, jagung manado kuning dan sagu baruk menggunakan perbedaan pelarut. Tepung komposit diproduksi menggunakan campuran tepung pisang, jagung dan sagu pada perbandingan 60:30:10. Tepung komposit dievaluasi menggunakan kandungan total flavonoid (metode aluminium), aktivitas antioksidan (uji radikal 1,1-diphenyl-2-phycrylhidrazyl) dan aktivitas fotoprotektif (uji SPF menggunakan spektrofotometer). Hasil penelitian ini menunjukkan bahwa kandungan total flavonoid tertinggi ditemukan pada pelarut aseton dibandingkan dengan methanol, ethanol, petroleum eter dan TPTJ (kontrol) pada tepung komposit. Aktivitas penangkal radikal bebas ekstrak aseton tepung komposit menunjukkan aktivitas paling tinggi daripada ekstrak etanol, metanol, petroleum eter dan TPTJ (kontrol). Hasil penelitian ini juga menunjukkan bahwa ekstrak aseton mempunyai aktivitas fotoprotektif paling tinggi daripada ekstrak metanol, etanol, petroleum eter dan TPTJ (kontrol) pada nilai SPF (sun protection factor). Secara keseluruhan, penelitian ini menyarankan bahwa ekstrak aseton tepung komposit mengandung komponen yang memberikan aktivitas antioksidan dan fotoprotekstif serta berpotensi melindungi kulit dari efek radiasi UV-B. Kata kunci: tepung komposit, pelarut, antioksidan dan fotoprotektif ABSTRACT  Banana, corn and sago are source of macronutriens, vitamines, minerals, dietary fiber and phytochemicals. The objective of this study was to determine antioxidant and photoprotective activities of composite flour from goroho banana, yellow Manado corn and baruk sago using different solvent. Composite flour was produced using mixture of banana, corn and sago flour in the ratio of 60:30:10. The composite flour extracted with petroleum ether, acetone, ethanol, and methanol by using a maceration extraction for 24 hours. The composite flour was evaluated using total flavonoid content (aluminium ion method), and free radical scavenging activity (1.1-diphenyl-2-phycrylhidrazyl radical assay). The in vitro photoprotective activity was studied by UV spectrophotometric method in the range of 290-320 nm. This result showed that the highest total flavonoid content was found in acetone solvent compared that of methanol, ethanol, petroleum ether, and TPTJ (control) in composite flour. The radical scavenging activity of acetone extract of flour composite showed the highest activity than ethanol, methanol, petroleum ether, and TPTJ (control). The results also showed that the acetone extract has highest photoprotective activity than methanol, ethanol and petroleum ether in sun protection factor (SPF) value. In overall, the present study suggests that acetone extract of composite flour contains components that give antioxidant and photoprotective activities and potency to protect skin of UV-B radiation effects. Keyword: composite flour, solvents, antioxidant, photoprotective

Abstract

Tujuan penelitian ini yaitu menganalisis Hubungan Kuantitatif Struktur Aktivitas (HKSA) turunan amino pirimidoisokuinolin kuinon yang berkhasiat sebagai antitumor telah dilakukan dengan menggunakan pendekatan regresikomponen utama. Data aktivitas biologis yang digunakan pada penelitian ini merupakan aktivitas senyawamelawan sel tumor AGS (Human Gastric Adenocarcinoma Cell Line) dalam ukuran log IC 50 . Sebagai deskriptordigunakan muatan atom bersih bersih (q) pada kerangka struktur senyawa dari hasil optimasi geometrimenggunakan perhitungan mekanika kuantum semiempirik AM1. Analisis komponen utama dilakukan sebelum proses regresi untuk mendapatkan variabel laten yang merupakan hasil transformasi data deskriptor muatanatom bersih (q). Dengan pendekatan regresi komponen utama diperoleh keterkaitan erat antara log IC 50 dan q yang direpresentasikan dengan persamaan HKSA berikut :log IC 50 = 0,671 - 0,041.x 1 + 0,232.x 2 + 0,270.x 3 - 0,087.x 4 + 0,105.x 5dengan, n = 27, r = 0,762, SE = 0,362, F hitung /F tabel = 2,168.Quantitative Structure Activity Relationship (QSAR) analysis of amino-pyrimido-isoquinolin-quinone analoguesas antitumor compound has been done using principal component regression approaches. Biological activitydata that used in this research were the activity against tumor cell AGS (Human Gastric Adenocarcinoma CellLine) as log IC 50 values. Atomic net charges (q) in the skeleton of the compound have been selected asdescriptors and all data were resulted by geometry optimization using AM1 semiempirical quantum mechanicscalculations. Principle component analysis has been performed before regression process to produce latentvariables by transformation of the descriptors. Based on principle component regression approaches, there isa selected quantitative relationship between log IC 50 to the descriptors that was shown by QSAR equation below:log IC 50 = 0.671 – 0.041.x 1 + 0.232.x 2 + 0.270.x 3 – 0.087.x 4 + 0.105.x 5with, n = 27, r = 0.762, SE = 0.362, F calc. /F table = 2.168.

Abstract

ABSTRAK Tujuan penelitian ini adalah menentukan pengaruh pelarut dan waktu ekstraksi berbantu ultrasonik dari tepung serat pangan tak larut (SPTL) dari tongkol jagung dan aktivitas antioksidannya. Etanol dan aseton digunakan untuk mengektrak komponen fenolik dri tepung IDF tongkol jagung dengan waktu ekstraksi (30, 60, 90, 120, 150 dan 180 menit) pada dievaluasi berdasarkan kandungan fenolik dan aktivitas antioksidan dari ekstrak tanaman. Hasil penelitian menunjukkan bahwa ekstrak etanol memiliki kandungan fenolik tertinggi dibandingkan dengan aseton ekstrak semua tingkat waktu ekstraksi. Hasil ini juga menunjukkan bahwa ekstrak etanol memiliki aktivitas antioksidan yang lebih kuat dalam aktivitas pemulungan radikal dan mengurangi daya antioksidan. Studi ini menyimpulkan bahwa waktu ekstraksi dan jenis pelarut mempengaruhi total kandungan fenolik dan aktivitas antioksidan. Kata kunci:  ABSTRACT The objective of this study was to determine solvents effect and extraction time of ultrasound-assisted extraction of corncob insoluble dietary fiber (IDF) powder and their antioxidant activities. Ethanol and acetone were used to extract phenolic components from corncob IDF powder with extraction time (30, 60, 90, 120, 150 and 180 min) in an ultrasonic bath (Krisbow 42 kHz, 50 watt). The both extracts were evaluated on the phenolic content and antioxidant activity with spectrophotometry method. The results of the phenolics content expressed in μg/mL of gallic acid equivalent showed that ethanol extract has highest phenolic content compared with acetone extract of all the extraction time. The radical scavenging activity of extract ethanol exhibited strong than acetone extract of all extraction time tested. The result also showed that the ethanol extract has highest the total antioxidant capacity compared with acetone extract at all extraction time. This result suggest that corncob IDF powder especially the ethanol extract are very rich in phenolic component which is potential as antioxidant dietary fiber.Keyword: corncob, dietary fiber, ultrasonic, extraction time, phenolic, antioxidant

Abstract

The objectives of this research were study the effect of addition oxalat acid on inhibiting of stainless steelcorrosion in sulfuric acid solution. The stainless steel sample sieved and macerated in sulfuric acidsolution with concentration 0.05 M for 24, 48 and 72 hours. The corrosion rate are evaluated with Asdimof method. The result showed that corrosion rate of stainless steel in sulfuric acid for 72 hour is 0,1309mg/ cm2jam. The corrotion rate rate of stainless steel in sulfuric acid with addition axalat acid inhibitor withconcentracion 1.10-3M , 2.10-3M , 3.10-3M , 4.10-3M for 72 jam were 0,0157 , 0,0188, 0,017 , and 0,0165mg/cm2hour. .
