Abstract

The Mount Biru protect forest is still maintained the biodiversity so that it is able to support a variety of animals in it, one of which is a habitat for the release of the javan langurs group (T. auratus). This study aims to determine the daily behavior of the javan langur after rehabilitation and release to evaluate the development of population behavior and barriers in the same group since 2014. The study used a focal animal sampling method combined with scan sampling to observe daily behavior (eating, social, resting, and moving) one female javan langur released in 2014. It was observed that post-release individuals had higher resting activities (38.91%) and the lowest allo-grooming social activities (5.08%). The results of the different test calculations using Chi Square can be concluded that there are significant differences in the daily behavior of the Javan langur after release in 2015, 2019 and 2021.

Abstract

Indonesia is one of the countries that has a high diversity of birds, one of which is the White-winged Duck (Asarcornis scutulata) which is a protected bird. One of habitat white-winged duck habitat in Sumatra is Way Kambas National Park (WKNP). Declain of population the white-winged Duck requires efforts and community participation in its conservation. Community participation is influenced by the understanding of the community. The aims of research to analysis the community's understanding about conservation of white-winged duck in WKNP. Data collection used a interview method using a questionnaire with purposive sampling technique. Data analysis using a quantitative technique with a Likert scale and the indicators used are questions in the two answer categories, namely yes/know with a score of 2 and no/do not know with a score of 1. The result of community knowledge in buffer villages about conservation of white-winged duck have a value of 50.8 including good category. Community experience has a value of 53 including the medium category, and community knowledge of the benefits of white-winged duck is in the medium category with a value of 54.3. The communities of knowledge not the same about white-winged duck, but dominant shown interest in participating in conservation efforts. There is a need for activities that educate communities on white-winged duck conservation. The existence of good knowledge and experience of white-winged duck owned by the community, can influence in realizing white-winged duck conservation efforts in the natural habitat.

Abstract

One of the local souvenirs from Jambi is accessories made from the seed bark of the sebalik sumpah tree. The sebalik sumpah tree is currently quite rare. Through DNA isolation, sebalik sumpah is one of the efforts of genetic conservation and as a first step in cultivating sebalik sumpah trees. DNA isolation is the initial stage in molecular analysis. The purpose of DNA isolation is to obtain genomic DNA from a sample. Plant DNA isolation generally uses fresh plant samples, with young and soft plant parts. This is recommended to obtain good quality genomic DNA. However, with the limitations of fresh samples, a method is needed that is able to extract DNA from plants that have changed shape and have undergone processing. Each plant requires a different method to obtain DNA. The CTAB (Cethyl Trimethyl Ammonium Bromide) buffer method is a method commonly used in plants because it is able to extract DNA with high polysaccharide and phenol content, so many have modified the method. This study aims to determine the success of the CTAB method without nitrogen in extracting processed materials and the length of incubation time at the precipitation stage. Extraction was carried out by liquid nitrogenless CTAB method with the addition of PVP (Polyvinylpolypyrrolidone). The results show that the CTAB method without liquid nitrogen is able to extract plant samples that have undergone processing and the length of incubation time at the precipitation stage affects the DNA produced.
