Abstract

Merapi volcano as one of national park is an ecotourism site which is very potential to visit. Information about species diversity for a national park is very important especially if its area fluctuated susceptibly. Two years after 2010 Merapi eruption, there are no research about species diversity and evenness of Anuran in that place. The research is carried out during June-November 2012 which is dry season. The objective of this research is to study the species diversity and evenness of Anuran in southern slope of Mount Merapi. The research is carried out in 6 locations; those are Kali Kuning, Telogo Muncar, Telogo Nirmolo, Petak Pitu, Bukit Turgo, and Bukit Plawangan. We used Visual Encounter Survey (VES) method combined with transect in Kali Kuning, Telogo Muncar, and Petak Pitu. VES method combined with time search in Telogo Nirmolo and Bukit Turgo. VES method combined with track exploration in Bukit Plawangan. Species diversity is analyzed with Shanon-Wiener diversity index. Species evenness is analyzed with Pielou evenness index. Total individuals each species is counted to know species abundance. Species diversity of anuran in southern slope of Mount Merapi is consisted of 12 species’ which is distributed in 6 locations. The highest diversity and evenness of anuran is at Bukit Turgo (H’=1.31; E=0.94). The lowest diversity and evenness of anuran is at Petak Pitu (H’=0.49; E=0.3). Species diversity of anuran in southern slope of Mount Merapi is low (Bukit Turgo and Kali Kuning) and very low (Bukit Plawangan, Telogo Muncar, Telogo Nirmolo, and Petak Pitu). Species evenness of anuran in southern slope of Mount Merapi is stable (Telogo Nirmolo and Bukit Turgo), still labil (Bukit Plawangan, Kali Kuning, and Telogo Muncar), and oppressed (Petak Pitu).

Abstract

Sabang seawater have high diversity and its geographical condition allow hot springs to form, therefore, current research on this seawaters become important. The purpose of this study was to discover the quality and diversity of the seawaters based on microbiological aspects specifically the finding of coliform, heterotrophic, halotolerant and sulfur bacteria from several samples from the water and sediment sampling. Analysis and isolation process of the bacteria were using coliform and aerobic count plate petrifilm and specific media. The results indicated that there were coliform bacteria, heterotrophic and halotolerant bacteria and several of hot spring sources under the sea surface with extremely high density of sulfur bacteria as geothermal microbe.

Abstract

The objective of the present study was to obtain good populations of Oreochromis niloticus based on their growth performances. Four populations, i.e GIFT-6, Tempe, Local, and GIFT-3 were used in this study. Sixteen 16 cages laid in four earthen ponds in the Cijeruk Research Installation, Bogor, were used as replication. The results showed that there were no significant difference among four populations for biomass, but a significant difference among populations was observed on length parameter (F < 0.01),in which the local population was the best.

Abstract

The red chick barb [Puntius orphoides (Valenciennes 1842)] is a member of Cyprinidae with high economic value. However, this species is still uncultured. The high market demand is fulfilled by direct collection  from their natural habitat.  In order to avoid overexploitation, domestication effort is needed.  The success of this effort needs some biological information e.g. reproductive characters.  This research aims to know some reproductive characters of red cheek barb collected at Klawing River Purbalingga Central Java.  A survey method was used with performing simple random sampling technique. The measured parameters were gonad maturity, fecundity, and gonad somatic indices (GSI) related to total length. The fish were collected monthly from June until November 2009. Data analysis includes length distribution, and correlation among gonad maturity, fecundity, and gonad somatic indices with body length. The result showed that red cheek barb from Klawing River has variable body length, most of the female individuals (34.37%) had body length ranges from 14.3-16.2 cm, whereas the majority of the male individuals (44.95%) had length distribution from 12.1-14.6 cm. The variation on  gonad maturity was observed during the research both on female and male individuals. However, most of the populations were on the highest level of gonad maturity (level IV) only in September and October. A positive correlation between gonad maturity and body length was observed on the male individuals (r=0.0424) as well as on the female individuals (r=0.4339). The fecundity was ranges from 4.097 up to 32.794 eggs. A positive correlation was also observed between fecundity and body length (r=0.6403). A monthly GSI variation was found both on the female and the male individuals with the highest peak of GSI was observed in September and October. A positive correlation between GSI and body length was found on female individuals (r=7.07 x 10-3) as well as on male individuals (r=0.376).

Abstract

Melon (Cucumis melo L.) has economic potential to be cultivated because the fruit contains protein, fat, carbohydrate, calcium, phosphor, fiber, iron, vitamin A, vitamin B1, vitamin B2, vitamin C, and niacin. Fusarium wilt caused by Fusarium oxysporum will decrease melon crop production. One of controlling method to Fusarium wilt diseases on melon plants which safe for environtmental by using biological control. One of microorganisms which can be biological control agent is Vesicular Arbuscular Mycorrhiza (VAM). This research use experimental method with a Completely Randomized Design (CRD). The experimental treatment consists of two types of treatment which combine 5 doses of VAM mixture ( 0 g/plant, 10 g/plant, 12,5 g/plant, 15 g/plant, 17,5 g/plant) and two inoculation method VAM is inoculated when seeds are planted and inoculation when the seedlings are replanted. Each treatment was repeated 3 times and each unit consist of three plant, so there are 30 units of experiments or 90 plants. The main variabels are observed consist of the incubation periode of the disease and the intensity of fusarium wilt and the supporting variabels consist of pH, temperature, humidity, and the scale of infection. The mixed MVA 15 g/plant dosage inoculated when seeds are planted and 15 g/plant dosage inoculated when the seedlings are replanted is the most effective to suppress incubation period of Fusarium wilt disease.

Abstract

Bukit Barisan Selatan National Parks has been given a named by World Heritage Committee as The Tropical Rain Forest of Sumatra (The World Heritage site) in July 2004 in Shozou China. However, many of Bukit Barisan Selatan National Park’s forest have already been wrecked by irresponsible people. Illegal logging is a big problem in Bukit Barisan Selatan National Parks. Indonesian’s forests are home to megabiodiversity in the world, but deforestation has caused a lengthening list of the inhabitant species to be classified as endangered. For example, the Sumatran tiger has a category to be an appendix I species by Convention on International Trade in Endangered Species (CITES).The decline of a species is very often a strong indicator of degradation in the local habitat, which can also negatively impact the biodiversity of that area. Perhaps the greatest threat to the survival of the tiger is destruction of its habitat. With the expansion of human populations, the logging of forests, the elimination of natural prey, and the spread of agriculture, there is continuation of destruction toward Sumatran tiger population. Monitoring the Sumatran tiger and its prey using camera trap can provide a data base on the continuity of the Sumatran tiger population management.

Abstract

Hormonal manipulation has been widely used in aquaculture to promote fish reproduction.  However some concern has been raised in accordance to it effect of human health as well as environmental issues.  Therefore alternative approaches in promoting fish reproduction need to be continuously studied.  Photoperiod manipulation offers a favorable alternative since it is one of the most crucial environmental cues necessary for fish reproduction.  An experimental study has been conducted to evaluate the effect of various photoperiods on testicular activities in the hard-lipped barb (Osteochilus hasselti C.V.).  Four groups of fish were reared under different photoperiod i.e. 14L:10D, 16L:8D, 18L:6D and the control group were reared under natural photoperiod for eight weeks period. Forth nightly three fish of each group were stripped to aspirate milt for measurement of milt volume and sperm concentration, they were decapitated for gonado-somatic index calculation and spermatogenic activities.  The results showed that the gonado somatic index (GSI) of the control group, 14L:10D and 18L:6D increased throughout the experimental period, meanwhile GSI of the fish reared under 16L:8D increased up to 4th week then decreased until the end of the study.  The fluctuation of the GSI values was followed by the fluctuation of milt volumes and sperm concentration.  This can be concluded that rising daily photoperiod up to 16 hours could improve reproduction in male hard-lipped barb.

Abstract

Genomic library of soybean cv. Lumut had been constructed in l phage vector. Partial digestion of total DNA by 0.014 units Sau3AI per mg DNA at 37oC for 30 minutes gave long fragment DNA. Ligation between insert DNA and l vector with molarity ratio of insert DNA to l vector = 2.9 : 1 followed by phage packaging, resulted in the titer of 2.8x104 to 3.6x105 pfu for 0.5 mg insert plant DNA. Among them, 60 to 91% were recombinants depending on packaging extract protein. By using 1 mg insert plant DNA and packaging extract protein from Stratagene, we obtained 6.6x105 pfu recombinant. Since the length of insert DNAs in l recombinant phages were 15 kb in average and the genome size of diploid soybean was 2.23x106 kb and 90% of the genom was present in 2 copies, then the genomic library constructed had contained all genome of soybean cv. Lumut.

Abstract

Freshwater fish is one of the biodiversity that build river ecosystem structure. Many human activities are carried out in the river as well as in the surrounding areas, such as removal of household waste, excessive fish exploitation and land use activities. This might changes river characteristics and lead to population and biodiversity decline.This research aim to 1) analyzethe diversity and species composition of fishin Luk Ulo River of Kebumen Regency,and 2) analyze the interaction among environmental factors with fish abundance in Luk Ulo River of Kebumen Regency. This research used survey method with cluster random sampling. The river was divided into three different region; upper-stream, middle-stream, and down-stream. Fish collected from 9 stations, i.e. 3 stations in the upper-stream, 3 stations in the middle-stream, and 3 stations in the down-stream. Morphology data were analyzed descriptively in order to make species description. The influences of environmental factor to species abundance were analyzed using Principal Component Analysis (PCA).The results showed that the number of fish obtained in this study were 72 individuals classified into 7 families and 13 species. The diversity and abundance of fish are influenced by physical and chemical factors ie temperature, TSS, pH, BOD5, COD, turbidity, DO, conductivity, salinity, current and substrate. BOD5 concentration was correlated positively with fish abundance with a correlation value of 0.540. In contrast, the effect of temperature and pH has a negative correlation to fish abundance with correlation values of -0.731 and -0.024. Dissolved oxygen (DO) and conductivity no effect on fish abundance.

Abstract

The study of plant diversity and investigation of home garden potential as conservation site for sandalwood at Teun and Lokomea villages had been carried out at East Nusa Tenggara Province. The result showed that home garden in Teun village have 24 species of plant and in Lokomea village have 31 species of plant. Diversity index in both locations are 1.163 and 1.151 with similarity index is 0.80. Based on purpose of flora at home gardens in both villages are divided in to 3 groups are fruit, commercial (estate crops) and vegetable crops. Majority of the local people in both villages have planted mango, coconut, banana, tamarind and sandalwood at their home gardens.  Result of investigation of home garden potency showed that home garden in both villages have 3 points of strength are ecological, economical and social culture potency so that home garden in both villages can be developed as conservation site for sandalwood.
