Abstract

Human Chorionic Gonadotropin (hCG) and Pregnant Mare Serum Gonadotropin (PMSG) hormone is a product that contains the hormone Folicle Stimulating Hormone (FSH) and Litiunizing hormone (LH) that play a role in regulating the development gonad process of fish. This research determine to effect hCG and PMSG hormone induction and find out the best treatment in the induction on gonad maturation of Silver pompano fish. This research used three treatments: control (NaCl), hormone hCG 20 IU.kg-1 and PMSG 20 IU.kg-1 fish body weight. Containers used in the maintenance of fish in the form of Floating Net Cage (KJA) with size 3x3x3 m. The best research result is 20 IU PMSG treatment with GSI value equal to 0.6%, HSI equal to 1.25%, maturity level of gonad reaches TKG III and histologically progress reach secondary oocyte and primary oocyte. The conclusion of this research is hCG and PMSG hormone induction effect on gonad maturation Silver pompano fish for 4 weeks compared to control treatment. The best hormone treatment is PMSG hormone treatment with a dose of 20 IU. Kg-1 fish body weight.

Abstract

One of the preservative agents that was claimed having bactericidal effect is glyroxyl. This study was aimed to observe the effect of glyroxyl on carp’s (Cyprinus carpio) shelf life. Factorial design was used in this experiment with 9 treatments in triplicates. The fish was immersed in glyroxyl solution at concentrations of 0 (control), 0.1, 0.3 and 0.5% (w/v) for 5 min. Following immersion, fish was drained and kept in plastic bucket at ambient temperature. Sampling was done at 0, 12 and 24 h storage and analysis was carried out organoleptically, chemically (TVB) and microbiologically (TPC, Enterobacter). Results showed that organoleptically, chemically and microbiologically, there were no significant different on spoilage inhibition of glyroxyl compared to control. Similar to control, glyroxyl was able to prevent spoilage of carp fish only up to 12 h. The fish was rejected by panelists after 12 h of storage. The TVB value and bacterial counts, including Enterobacter, also increased sharply after that time.

Abstract

One of the ATC quality is influenced by the length time of harvesting and temperature of extraction.The aimof the research was to determine the influenced of different time of harvesting K.alvarezii and the degree temperatureextraction process to the quality of the ATC product (yield, gel strength and viscosity). The material used wastaken from the Bandengan coastal area, Jepara. The treatments used was the difference temperature of extractionprocess which are 70oC and 80oC and the difference of harvesting time 35, 40, and 45 days. K.alvarezii extracted using6% KOH solution for 4 hours, dried and then is milled. The yield test showed that the used extraction temperatureof 80oC produces a higher yield than the temperature of 70oC, the percentage yield of ATC powder was increasein along with harvesting time of K.alvarezii. Abstract

Fish nugget is a product that made from mixture of minced fish meat, tapioca powder, and condiments. This product can be frozen or saved in cold storage before fried. The objectives of this research were to utilize Black Marlin chunk meat residue in making fish nugget and to know the influences of tapioca powder addition to fish nugget’s characteristics and acceptability. This experiment used Complete Block Randomized Design with six treatments of tapioca powder addition, that consist of a1 (3%), a2 (6%), a3 (9%), a4 (12%), a5 (15%), and a6 (18%), with three repetition as block. Protein content, elasticity, and organoleptic test (color, flavor, taste, and texture) of final product were analyzed. The addition of 3%, 6%, 9%, 12%, 15% and 18% tapioca powder resulting to the reduction of protein content as much as 0%, 4.92%, 10.96%, 11.69%, 13.76% and 15.03%, respectively, and increase the elasticity to 0.51, 0.57, 0.60, 0.68, 0.76 and 0.86 N/mm, respectively. The highest acceptance value of fish nugget color, flavor, taste, and texture were 3.75 (a2), 3.63 (a5), 4.11 (a1), and 3.9 (a5), respectively, whereas the lowest were 3.49 (a6), 3.5 (a6), 3.05 (a6), and 2.98 (a1), respectively. The favorite fish nugget chosen by panelists was a2 treatment nugget with the highest total score of acceptance of 4.00.

Abstract

Removing eggs from red nila female during spawning period were observed to know an appropriate time of removal and its effect on inter-spawning period of female broodstock. Spawning experiment was conducted with three replicates in the hapa. The female that cared fertilized eggs were cathed on 7th day, 21th, 28th day, and 35th day after stocking. Other spawning experiment with three relications were cared out to test the effect of removing eggs. On 7th day after spawning produced the highest number of spawned female reduced yhe inter-spawning interval, but there was no effect of male stocking on re-spawning rate of female. The low number of eggs per spawner together with asynchronus spawning would be problems faced on mass production of red nila seed.

Abstract

Kulit ikan bisa digunakan sebagai alternatif bahan baku untuk industri penyamakan kulit mengingat keterbatasan bahan baku kulit mentah hewan darat. Tujuan dari penelitian ini adalah untuk mengetahuipengaruh penggunaan Natrium Sulfida pada proses pengapuran yang berbeda konsentrasinya pada hasil uji fisik kulit ikan buntal, yang meliputi uji kuat tarik, kemuluran dan kuat sobek. Penelitian inimenggunakan kulit ikan buntal awet garaman sejumlah 9 lembar yang disamak menggunakan bahan penyamak nabati (mimosa), sampai menjadi kulit crust ikan buntal. Perlakuan penggunan variasikonsentrasi Natrium Sulfida yang digunakan saat proses pengapuran adalah 0,5%, 1% dan 1,5%. Masingmasing perlakuan tersebut diulang sebanyak tiga kali, kemudian dilakukan uji fisik menggunakan alat tensile strength. Penggunaan konsentrasi Na2S 1% menunjukkan nilai Kekuatan Tarik yang paling tinggi yaitu sebesar 1394.172 N/cm2, dan nilai kemuluran yang paling rendah yaitu sebesar 57.707 %, dan nilai kekuatan sobek yang paling tinggi ditunjukan oleh penggunan konsentrasi Na2S 0,5% yaitu sebesar181.978 N/cm. Penggunaan variasi konsentrasi Natrium Sulfida memberikan pengaruh berbeda nyata terhadap kekuatan tarik, namun tidak memberikan pengaruh yang berbeda nyata terhadap kemulurandan kekuatan sobeknya. Abstract

The aim of this research was to know the reproductive biology of Ompok hypophthalmus included gonadal development stages and spawning season.The study was conducted from June to December 2006 in floodplain of Kampar Kiri River. Samplings were carried out monthly with purposive sampling method. Fish samples were cathed by gillnet, trapnet, hand line and long line. A total of 474 individuals were captured ranged 80-310 mm in total length and 2-143 g in weight. The female and male reached sexual maturity at 115 mm and 214 mm in total length respectively. The spawning season for this species ranges from June to December where a peak season found in October which examined based on variations in gonado somatic index (GSI) and the existence of mature male and females. The fecundity varied from 688-15180 eggs. The correlation coefficient between fecundity with total length and fecundity with weight were very weak. It lead to suggested that the coefficient cannot be used to predict O. hypophthalmus fecundity.

Abstract

The main objectives of this research were to isolate, purify and determine the immunogenicity of the outer membrane protein of V. alginolyticus. The outer membrane protein was isolated by sonication, sodium dodecyl sulphate-polyacrylamide agarose gel electrophoresis (SDS-PAGE) and purified by electroelusion method. Four outer membrane proteins were obtained: namely 32.0; 37.83; 64.13 and 73.43 kDa. Its immunogenicity in grouper Epinephelus fuscoguttatus was compared to that of bacterin of the same isolate. The immunogenicity test was conducted by intra peritoneal injection method. Each protein was dissolved in sterile Phospate Buffer Saline (PBS) and Freund’s Complete Adjuvant (FCA) (1:1) and injected 5 µg/ fish (fish weight 10-15 g). Bacterin (106, 107 and 108 cells/ml) was prepared in the same manner and injected at dosage of 0,2 ml/fish. Control fish were injected with 0.2 ml sterile PBS (pH 7.2). Booster was done a week later by injecting protein or bacterin with the Freund’s Incomplete Adjuvant (FIA). The agglutination test of antibody produced recognized cell surface protein of the whole cell of V. alginolyticus. Outer membrane protein 73.43 kDa was more immunogenic than the rest of proteins and bacterin, based on agglutinating antibodi titer.

Abstract

The success of induced breeding relies on the availability of viable sperm and eggs. Meanwhile information on viability of post-stripped sperm of the nilem is still limited.  This experimental study was conducted to find out the duration of sperm storage that maintains sperm viability.  The nilem’s milt was diluted in Ringer solution at 0, 10, 100 and 1,000 dilution and kept in the refrigerator (4°C)  for 3, 6 and 9 days.  The sperm viability was assessed by its motility, progressivity and ability to fertilize fresh eggs.  The results showed that the sperm were immotile in either undiluted or diluted in Ringer solution.  Upon activation by diluting in water, the sperm of the control groups as well as the tested groups showed a high motility (>70%) and highly progressive (>3).  The sperm were capable to fertilize eggs as indicated by a high fertilization rate of more than 90%.  The highest hatching rate (HR) was produced by the control groups while HR of milt stored for 3, 6 and 9 days were 60%, 1% and 0%, respectively. Low HR produced by milt stored for 6 and 9 days was resulted from a high proportion of abnormal embryos which led to their mortality around gastrula stage.  It could be concluded that the viability of the nilem’s sperm could be diluted in Ringer solution and kept in the refrigerator as long as for 3 days.

Abstract

Level of technical efficiency in fish culture reflecting the fish farmers achievement correlated to their managerial competency to manage the principle factors affecting its productivity.  Objectives of this research were to evaluate technical efficiency in grouper growth out as well as its policy implication. This research was conducted in Lampung Province during September to December 2002. The area was selected by purposive sampling in Padang Cermin Sub district, South Lampung, and the respondent of farmers were selected by census method. The results showed that the average of technical efficiency level achieved by grouper farmers in floating net cages should be classified in middle to high categories with relatively homogen distribution. Principle factors affecting the technical efficiency was level of income from farming fish culture. As the technical efficiency level closed to the frontier, policy of culture development should be conducted using area-based extensive culture approach and should applicate imperative and innovative technologies.
