Abstract

Tuberculosis (TB) is an infectious disease directly transmitted through the air. This disease is caused by the bacterium Mycobacterium tuberculosis. Tuberculosis requires an incubation period of 3-6 months from initial exposure to the bacteria. Patients identified with tuberculosis will be given anti-tuberculosis drugs intensively for 6 months. Long treatment time resulting in drug accumulation can affect the kidneys. Microscopic examination or urine sediment aims to detect kidney and urinary tract abnormalities and monitor the results of treatment. This study used an experimental research type, then the primary data were analyzed descriptively. Identification of the results of macroscopic and microscopic urine in tuberculosis patients in the hospital. Lungs Surabaya, showed the largest percentage of 75% macroscopic results of clear yellow urine. Microscopic results showed normal values compared to normal macroscopic and microscopic ranges of urine. Keywords :Tuberkulosis, Mikroskopis Urin, Makroskopis Urin

Abstract

The COVID-19 pandemic has occurred in Indonesia since March 2020. An increase in cases requires the involvement of various medical elements from the beginning of the patient's condition to the diagnosis decision. Preparation of pre-analysis, analysis, to good post-analysis in the laboratory greatly supports the results of the diagnosis needed. The purpose of this study was to find out various factors that influence invalid results in a real-time examination – polymerase chain reaction (RT-PCR) in patients at Dr. Wahidin Sudirohusodo Mojokerto general hospital. Data collection from the hospital's biomolecular laboratory in a cross-sectional descriptive observational manner was taken from September to November 2020. A total of 40 data processed with descriptive statistics percentage showed a tendency to invalidity results by 30%. Pre-analytic errors play a big role in the RT-PCR method inspection process. Sample misidentification, poor sample quality, non-standard sample transportation conditions,  and sample storage are some of the things that support the invalidity of the results of the COVID-19 RT-PCR examination method. Evaluation during the work process at various stages as well as the use of controls can minimize laboratory results that are not appropriate. Keywords : COVID-19, RT-PCR, Invalid

Abstract

Since the WHO announced the global pandemic caused by COVID-19 in February 2020, various steps have been taken to deal with it, one of which is increasing laboratory tests to support the diagnosis. This study aimed to describe the suitability of the COVID-19 swab results using the TCM (Molecular Rapid Test) tool with RT-PCR as the gold standard for diagnostic COVID-19 laboratory examinations at Dr. Wahidin Sudiro Husodo Hospital, Mojokerto City, reaching 100% or not.  This research is descriptive with a retrospective approach, taking data from the examination results in August 2020, which were carried out at the Clinical Pathology Laboratory and PCR Biomolecular Laboratory, Dr. Wahidin Sudiro Husodo Hospital Mojokerto City.  The results of the 20 samples examined using the TCM tool obtained postpositive results. In contrast,e results in as many as seven samples and 13 samples with negative results. In comparison, with the RT-PCR tool, 6 samples had positive results and 14 samples with negative results, so that 1 (5%) did not come according to the results, namely in samples with code C the results with the TCM tool showed p those with the RT-PCR tool showed negative results. Then the research results using the TCM if the percentage according to RT-PCR is 95%.  Many factors could cause this difference in results. If viewed from pre-analytic factors, it can be caused by improper sampling process, patient cooperation, poor quality of the specimen or containing only very few samples, inaccurate sample delivery and storage conditions, sample contamination, and pipetting errors during sample preparation. At the analytical stage, the presence of cross-contamination, virus mutation, or PCR reaction barriers, the quality of the reagent kit, and the skills of the analyst working on it also affect the results because the more skilled you are, the more precise and correct standard procedures can be performed.  Keywords: COVID-19, TCM (Molecular Rapid Test), RT-PCR

Abstract

Urinary Tract Infection (UTI) is an infection due to the presence of microorganisms in the urine and has the potential to invade the tissues of the urinary tract. Urinary Tract Infection (UTI) depends on many factors such as age, gender, prevalence of bacteriuria and predisposing factors that cause changes in the structure of the urinary tract including the kidneys. Antibiotics are chemical substances produced by microorganisms that have the ability in aqueous solutions to inhibit the growth or kill microorganisms, for example penicillins, cephalosporins, chloramphenicol, tetracycline, and others. Creatinine is the end product of muscle creatine and creatine phosphate metabolism, synthesized in the liver, found in skeletal muscle, blood, and excreted in the urine. Creatinine levels are determined by the amount of muscle mass (the rate of protein catabolism). The type of research used is pre-experimental research with pre-test and post-test group design. In this design, observations were made twice, namely before the experiment was called the pre-test and after the experiment was called the post-test. A total of 30 patient samples, namely the serum creatinine value in the medium category has a value range of 4.9 - 6.9 mg/dl as many as 13 people with a percentage of 43.3%, and the serum creatinine value in the high category has a value range of 7.0 - 10 0.0 mg/dl as many as 17 people with a percentage of 56.7% it can be seen that the serum creatinine value in patients before antibiotic therapy has a range of values in the medium and high categories and after antibiotic treatment is given in the post test table, the serum creatinine value with the medium category has a value range of 4.9 – 6.9 mg/dl as many as 2 people with a percentage of 6.6%, and serum creatinine values in the low category have a value range of 1.1 – 4.8 mg/dl as many as 28 people with a percentage 93.4%. Keywords : Antibiotics, Creatinine

Abstract

Aspartic aminotransferase (AST) is a mitochondrial enzyme that catalyzes the back and forth transfer of amino groups from aspartic acid to ?-oxaloacetic acid to form glutamic acid and oxaloacetic acid. AST is contained in erythrocytes so that if there is hemolysis, a ruptured erythrocyte will cause an increase in the activity of the AST enzyme that comes out of the erythrocyte. This study aims to determine the effect of hemolysis in serum on AST enzyme activity. This research method is experimental by comparing normal serum with hemolytic serum. The research sample involved 30 employees of the Manding Sumenep Public Health Center who would be examined for the AST enzyme activity. The results showed that the AST enzyme activity in normal serum had an average of 24.367 IU / L, while the average hemolytic serum was 39.3 IU / L. Based on the results of these data, it shows that there is an increase in AST enzyme activity in hemolytic serum compared to normal serum.

Abstract

Reticulocytes are red blood cells that are young or immature, do not have a nucleus and derived from the normoblast maturation process in the bone marrow. Hematocrit is a measurement result that states the ratio of red blood cells to blood volume in percent. Objectives of this research are to determine the difference in reticulocyte and hematocrit results using K?EDTA venous blood and 3.2% Sodium Citrate. Examination using manual method (blood smear) on reticulocyte parameters and semi automatic hematology analyzer for hematocrit. The research subjects were 20 students of Universitas Maarif Hasyim Latif Sidoarjo. The results of the Paired T-test statistic for reticulocytes showed that the p value = 0.163 (?? 0.05) there was no difference in the results of K?EDTA blood and 3.2% Sodium Citrate blood with Wesgard QC there were 55% in the Wesgard range and 45% out of range Westgard QC. While the results of the hematocrit with the Wilcoxon statistical test showed a p value = 0.006 (?? 0.05), namely there was a difference in the hematocrit value of K?EDTA blood and moderate sodium citrate with Wesgard QC, the majority of NaCitrate blood HCT values were decreased, namely there were 18 respondents 90% below the standard K?EDTA value.

Abstract

Menstrual disorders are important indicators that indicate impaired reproductive system function that can be associated with an increased risk of various diseases such as uterine cancer, breast cancer, infertility and diabetes mellitus (DM). This is due to the presence of estrogen receptors on pancreatic cells which will increase insulin release and will be influenced by hormonal changes in the menstrual phase. Blood glucose or blood sugar is the main nutrient used for cell metabolism and energy supply in the body, and regulates and maintains glucose within normal limits. This study aims to determine the differences in fasting blood glucose examination in the follicular phase and the luteal phase in menstruating women. This type of research is descriptive with quantitative laboratory analysis techniques, namely calculating fasting blood glucose levels. The material used for this test was the blood of patients as many as 10 samples for the follicular phase and the luteal phase. The results obtained in the follicular phase with results that were not much different from normal values, while in the luteal phase the results slightly increased from the normal value of 70-110 mg/dL and the results obtained a difference in percentage with the results of 13%.
