Abstract

Insulin leave (Tithonia diversifolia (Hemsl.) A. Gray) empirically used by people as antihyperglycemic drugs. The study was aimed to evaluate antihyperglycemic activity as well as to determine the most optimum dose of T. diversifolia to reduce blood glucose levels. Hyperglycemic condition was induced to male Rattus norvegicus rats by intraperitoneal injection of alloxan at a dose of 150 mg/kg bw (body weight). In this study, completely randomized design was performed with three treatment groups and five times repetition. Group P1 received glibenclamide 10 mg/kg bw, P2 received aqueous extract of T. diversifolia at a dose of 150 mg/kg bw, P3 received aqueous extract of T. diversifolia at a dose of 300 mg/kg bw. Administration of both glibenclamide and aqueous extract of T. diversifolia was conducted orally for 28 days. Data were analyzed using Anova and Duncan’s test with 95% confidence level (α = 0.05). The results showed that the mean percentage of decrease in blood glucose levels, drink intake and body weight of all treatment groups were not significantly different (p0.05). Data of feed intake showed that P1 was significantly different from P2 (p0.05), but P2 was not significantly different from P3 (p0.05). Based on the result of this research, it was found that the low doses of aqueous extract of T. diversifolia has the same ability to decrease blood glucose level compared to glibenclamide. Futhermore, this study provide some information that can be used as preclinical analysis to determine effective doses of aqueous extract of T. diversifolia to decrease blood glucose levels.

Abstract

Penelitian bertujuan mengidentifikasi spesies dan kelimpahan Bactrocera yang menyerang berbagai buah di Kecamtan Demak dan Dempet Kabupaten Demak. Penelitian menggunakan metode purposive sampling. Penelitian dilakukan dengan mengambil 5 macam buah yang terserang (jambu air, belimbing, jambu biji, melinjo dan mangga), pengambilan data faktor klimatik dilakukan pada saat pengambilan sampel. Buah terserang kemudian dilakukan rearing, Bactrocera spp yang didapat dilakukan identifikasi. Bactrocera yang menyerang jambu air yaitu Bactrocera ablistrigta, Bactrocera carambolae menyerang belimbing dan jambu biji, Bactrocera mcgregori menyerang melinjo dan Bactrocera papayae menyerang mangga. Kelimpahan Bactrocera albistrigata sebesar 112 individu/kg jambu air, Bactrocera carambolae pada belimbing sebesar 368 individu/kg belimbing, Bactrocera carambolae pada jambu biji sebesar 86 individu/kg jambu biji, Bactrocera papayae sebesar 48/kg mangga dan Bactrocera mcgregogi sebesar 668 individu/kg melinjo. Kelimpahan tertinggi terdapat pada Bactrocera mcgregogi yaitu 668 individu/kg melinjo. Kelimpahan terendah pada Bactrocera papayae yaitu 48 individu/kg mangga. This study was conduted in Demak and Dempet districts Demak regency. Demak is one of the producer of horticultural crops such as fruits. The objective of this research is identifying the species and abundance of Bactrocera which attacked various fruits in Demak regency. The research used was purposive sampling method. The steps of this research include 5 various infected fruits (water aple (, starfruit, guava, gnetum gnemon and mango climatic factors data collection is done at the time of sampling, developing fruit then carried rearing, Bactrocera spp obtained to identify. Bactrocera that infected the water aple (Bactrocera ablistrigta), Bactrocera carambolae attack starfruit and guava, Bactrocera mcgregori attack melinjo (Gnetum gnemon) and Bactrocera papayae attack mango. Bactrocera albistrigata abundance of 112 individuals/kg water aple, starfruit Bactrocera carambolae on by 368 individual/kg starfruit, guava Bactrocera carambolae on by 172 individual/kg guava, Bactrocera papayae by 48/kg mango and Bactrocera mcgregori of 668 individual/kg Gnetum gnemon.The highest abundance showed by Bactrocera mcgregori that is 668 individual/kg melinjo(Gnetum gnemon). The lowest abundance was (Bactrocera papayae 48 individual/ 0,5 kg mango).

Abstract

Pleurotus ostreatus cultivation is performed using solid medium to harvest fruit body and using liquid medium to harvest mycelia in submerged culture. Modifying nutrients in the medium to increase protein content of the fruitbody and mycelia can be done through addition of nitrogen-containing materials. This study aims to determine: the appropriate composition of the liquid medium for high mycelial growth and protein content; and the exact composition of the solid medium to obtain high fruitbody product and protein content. The method was experimental with completely randomized design (CRD). The treatments were incubation of P. ostreatus on three types of liquid medium and four types of solid medium. The results showed that the optimal liquid medium composition for mycelial  growth was Liquid Fermentation Medium 1 (FC1) with 10% corn fluor, and the highest protein content was in Liquid Fermentation Medium 2 (FC2 = 29.76%). While the optimal solid medium composition for fruitbody production was the medium with 3% corn starch supplement (TJ3), and the highest protein content was obtained from the medium without corn starch supplement (TJ0=24.69%). The increase of mycelial and fruitbody weight from the medium with the addition of corn material indicated a prospective in cultivation process, however effort to increase protein content of the fruit body needs further research. Cultivating P. ostreatus in mycelial phase may take shorter incubation time, may be produced in mass production with less space consuming, and higher protein content than that by producing fruitbody.

Abstract

Pseudomonas aeruginosa is an opportunistic bacterium that causes infections in human. The wall cell of its bacteria contains lipopolysaccharide as virulency factors to protect it from human immunity. Lipopolysaccharide can inhibit phagocytosis in the body. Polysaccharide krestin (PSK) from Coriolus versicolor extract contains β-glucan that can increase phagocytic activity and capacity. This research aimed to identify the effect on polysaccharide krestin from C. versicolor extract on phagocytic activity and capacity of mice exposed by P. aeruginosa. The design of this research was experimental design. There were six treatment groups. The phagocytic activity and capacity were counted on slide smears of mice peritoneal fluid. The data was analyzed by using one way ANOVA. The results of the phagocytic activity and capacity showed that PSK was added before exposure (P1) or after exposure (P2) or before-after exposure (P3) had potential to increase phagocytic activity and capacity.  The conclusion of the research was that adding polysaccharide krestin either before exposure or after exposure or both of them could increase phagocytic activity and capacity. Abstract

Endophytic bacteria are microorganisms that thrive in plant tissue without causing any negative effects to the host plants. The purpose of this study was to apply endophytic bacteria that have the potential to produce IAA hormones as biological agents in rice seeds. This research was conducted by testing IAA hormone using a spectrophotometer, testing HCN production using filter paper, biochemically identification of the isolate, and applicating endophytic bacteria into rice seeds. Endophytic bacteria have different abilities in producing IAA hormone. Based on the results of the study, the highest IAA concentration (57 ppm) was obtained from KSB1 (Potassium Solubilizing Bacteria) isolates and followed by KSB3 isolate with 56 ppm. All the endophytic bacteria isolates were unable to produce HCN. Results of the study showed that KSB1 and KSB3 isolate were able to influence the growth of root length by 5.62 cm and 6.70 respectively as well as the canopy height by 8.56 cm and 8.30 cm respectively. Based on observations of colony morphology, cell morphology and biochemical tests, KSB1 was presumably belong to the genus Staphylococcus. In this study, new information about endophytic bacteria that can increase the growth of Ciherang variety rice can be obtained. This study provides information that the application of environmentally friendly bacteria is important to preserve the environment and agriculture sustainability in Indonesia without reducing the productivity of rice.

Abstract

Pineapple propagation by lateral shoots, suckers or crowns is often confronted with limited number of regenerated seedlings and high diversity in flowering and fruit formation. In order to solve this problem, this study offer an alternative method by using tissue culture techniques. This study aimed to determine the effect of growth regulators on plantlet regeneration from bud slicing of pineapple cv. Tangkit. Four levels of 2.4-D (0.0, 0.001, 0.01 and 0.1 ppm) in combination with BA (0.0, 0.1, 1.0 and 10.0 ppm) were tested on solid MS medium. Cultures were incubated in total darkness for a week followed by transfer to 16-hour photoperiod. Results showed that explants treated with 2,4-D and/or BA succeeded in regenerating adventitious shoots. Average leaf number did not differ significantly among treatments (P = 0.60). Highest leaf number (2.99 ± 0.23) was obtained on medium with 0.01 ppm 2,4-D without BA, followed by 0.1 ppm 2,4-D without BA (2.85 ± 0.33). Meanwhile, roots were only formed on medium with 0.1 ppm 2.4-D without BA (4.2 ± 0.37 per shoot). Thus, complete plantlets were regenerated only on medium supplemented with 0.1 ppm 2,4-D without BA. The growth of plantlets was relatively uniform, and plantlet acclimatization succeeded 100% on Jiffy pots. The finding of optimum concentration of 2.4-D and BA in this study is important to develop standard protocol for in vitro propagation of pineapple cv. Tangkit. Thus, the benefit of producing seeds in large quantities and relatively uniform in growth is made possible through tissue culture technique.

Abstract

AbstrakTujuan penelitian ini adalah mengetahui keanekaragaman fauna khususnya kupu-kupu kekayaan jenis, kelimpahan, indeks keanekaragaman serta indeks kemerataan jenis. Metode yang digunakan adalah point count, line transect dan flight trap. Radius pengamatan pada setiap titik ditentukan berdasarkan tipe vegetasi dan kemampuan pengamat, sementara lamanya waktu pengamatan pada setiap titik hitung adalah 10 menit. Analisis data mengunakan indeks kekayaan jenis Margalef’s (DMg), Indeks keanekaragaman jenis  Shannon-Wiener, dan indeks kemerataan jenis Simpson. Hasil penelitian menunjukkan 55 jenis kupu-kupu ditemukan di kawasan Taman Kehati Universitas Negeri Semarang. Jenis yang ditemukan tergolong dalam 5 (lima) famili yaitu  famili Papilionidae, Pieridae, Lycaenidae , Nymphalidae, dan Hesperidae  (Tabel 1). Hasil penelitian juga menemukan satu jenis termasuk jenis yang dilindungi UU No 5 Tahun 1990, PP No 7 Tahun 1999 dan masuk dalam kategori CITES apendik II, yaitu jenis Troides helena. AbstractThe research tried to examine the diversity of butterflies in terms of species richness, abundance, diversity index, and evenness. The methods employed were point count, line transect and flight trap. The observation radius in each point was determined based on the vegetation type and the observer’s capability, and the interval time for observation in each point was 10 minutes. Data was analyzed using Margalef’s species richness index (DMg), Shannon-Wiener’s species diversity index, and Simpson’s species evenness index. There were 55 butterfly species in the area of Taman Kehati Semarang State University. The butterflies were from five (5) families, i.e. Papilionidae, Pieridae, Lycaenidae, Nymphalidae, and Hesperidae (Table 1). Observation has found one species that is categorized as protected based on Law No. 5 Year 1990, Governmental Regulation No. 7 Year 1999, listed in CITES Appendix II; the species was Troides helena.

Abstract

Biosorption is a new waste treatment technology that can eliminate toxic heavy metals. Biosorption can be considered as an environmentally friendly alternative technology to treat industrial liquid waste that is economically proper to use. One of them is biosorption that utilizes the microorganisms’ absorption ability, especially bacteria that can absorb heavy metals in waters, such as Enterobacter agglomerans. This research aimed to determine the ability of E. agglomerans in reducing heavy metals pollution in local river. The research employed the measurement of the effect of lead (Pb) to E. agglomerans growth using Optical Density (OD) at wavelength 600 nm. The colony numbers were calculated using a standard curve. While the ability of E. agglomerans to reduce heavy metals concentration in liquid media was measured using AAS with a wavelength of 240 nm. The results showed that lead affected the growth of E. agglomerans. The OD value has a negative relationship with the concentration level of Pb. The ODs were decreased from 2.867 to 1.242, using Pb level from 0 ppm to 20 ppm. Therefore, it proved that E. agglomerans could reduce heavy metals concentration in local river in Central Java Province. This research was the first report on E. agglomerans activity on heavy metal in contaminated water. This result can be used as a reference for industrial sites near the river to treat their wastewater before discharging it to the river body to preserve its water purity.

Abstract

Stelechocarpus burahol is one of the medicinal plants that contains flavonoids. The study was carried out to know flavonoid production of cultures in vitro S. burahol from mesocarp explants. Mesocarp explants were cultured on MS medium containing different combination and concentration of plant growth regulators i.e. picloram (5, 7.5 and 10 mg/L) and 2, 4-D (10, 15 and 20 mg/L) under dark condition. Induction of callus formation started on the 20.29th to the 29.86th days. Medium supplemented with Picloram and dark state proved to be the best condition for optimum callus induction from mesocarp explants of S. burahol. Callus grown on medium with the addition of 7.5 mg/l Picloram produces the highest flavonoid. The maximum production of the secondary metabolite was obtained from 8 weeks old callus. However, by the time of callus ageing, its output has declined. It could be concluded that callus cultures from mesocarp S. burahol can be used for flavonoid production. How to CiteHabibah, N.
