Abstract

Halogenated compound such as 2,2-dichloropropionic acid is known for its toxicity and polluted many areas especially with agricultural activities. This study focused on the isolation and characterization of the bacterium that can utilise 2,2-dichloropropionic acid from palm oil plantation in Lenga, Johor and in silico analysis of putative dehalogenase obtained from NCBI database of the same genus and species. The bacterium was isolated using an enrichment culture media supplemented with 20 mM 2,2-dicholoropropionic acid as a carbon source.  The cells were grown at 30˚C with cells doubling time of 2.00±0.005 hours with the maximum growth at A680nm of 1.047 overnight. The partial biochemical tests and morphological examination concluded that the bacterium belongs to the genus Staphylococcus sp.. This is the first reported studies of  Staphylococcus sp. with the ability to grow on 2,2-dichloropropionic acid. The genomic DNA from NCBI database of the same species was analysed assuming the same genus and has identical genomic sequence.  The full genome of Staphylococcus sp. was screened for dehalogenase gene and  haloacid dehalogenase gene was detected in the mobile genetic element of the species revealed that the dehalogenase sequence has little identities to the previously reported dehalogenases.The main outcome of the studies suggesting an in situ bioremediation can be regarded as a natural process to detoxify the contaminated sites provided that the microorganisms contained a specialised gene sequence within its genome that served the nature for many long years. Whether microorganisms will be successful in destroying man-made contaminants entirely rely on what types of organisms play a role in in situ bioremediation and which contaminants are most susceptible to bioremediation.

Abstract

The detailed causal factors of flood have not been established, whether because of the damage of upper watershed, sedimentation resulting in shallow river, forest conversion, the decrease in abundance and structure or high rainfall. The study aimed at finding out the species diversity and structure of the limited production forest (LPF) from the seedling to the tree stage of Arui watershed in Manokwari district. The species diversity was determined based on Shannon-Wiener index, while vegetation structure was determined based on Importance Value Index. There were totally 92 plots of samples drawn using systematic sampling. The results of vegetation analysis showed that there were 174 species of 43 families with the species diversity of 1.5-1.8 that was categorized as moderate. The four dominant species that were always found in seedling, sapling, pole and tree stages included: Pometia pinnata, Teijsmanniodendron bogoriense, Chisocheton ceramicus, and Horsfieldia irya. The conversion of the LPF into non-forestry sector for the purpose of accelerating development must be reviewed by considering hydrology, land, biodiversity, and social and regional aspects in order to prevent and to reduce flood in the coming days.

Abstract

One of the successful factors of the curriculum 2013 implementation is the availability of appropriate and relevant teaching materials. Teaching materials can be textbooks, interactive videos , virtual labs, maket and so on. Unfortunately, there are still found many materials that are less in accordance with the curriculum of 2013. Therefore it should be developed a more appropriate teaching materials, one of them is a natural sciences module with reflective learning journal. The development of natural sciences modulewas done with research that aims to develop and analyze the effectiveness of natural sciences modulewith reflective learning journals. This research was an RD research using development research procedure according to Thiagarajanthat known as 4D (define, design, develop and dessiminate). The data collection was done by test and non-test technique. Instrument that used in non test technique are interview guides, product validation sheet, students’ response sheet to the readability of science nature modul with reflective learning journal., meanwhile the instrumen that used in test technique is essay question with reporting-interpretative skills’s indicators. The results showed that the leligibility of the natural sciences module with reflective learning journal was categorized as approriate and effective to improve student’s reporting-interpretative skills.

Abstract

Avian Influenza (AI) atau yang lebih dikenal dengan flu burung disebabkan oleh virus influenza yang bermutasi menjadi patogen. Penelitian tentang isolasi dan identifikasi virus AI subtipe H5N1 perlu dilakukan untuk mengetahui keberadaan virus tersebut khususnya di kecamatan Gunungpati. Desain penelitian adalah eks ploratif dengan pengumpulan sampel usap kloaka secara acak di lima kelurahan di kecamatan Gunungpati. Sampel usap kloaka ditumbuhkan pada telur ayam berembrio SPF, kemudian diisolasi RNA-nya dilanjutkan dengan identifikasi subtipe virus AI menggunakan Reverse Transcriptase-Polymerase Chain Reaction (RT–PCR) dengan primer pendeteksi gen H5 dan N1. Hasil positif apabila visualisasi hasil elektroforesis dari produk PCR menunjukkan pita-pita spesifik panjang 219 bp untuk H5 dan 131 bp untuk gen N1-nya. Limapuluh sampel usap kloaka yang diisolasi dari lima kelurahan di Gunungpati, delapan isolat positif VAI dan enam diantaranya positif H5N1 dengan angka prevalensi 12%. Isolat positif berasal dari 2 spesies itik (16,67%), 2 dari entok (11,76%) dan 2 dari angsa (18,18%). Dari lima kelurahan yang diambil sampelnya, tiga kelurahan ditemukan positif virus H5N1 masing-masing kelurahan Sekaran (6,67%), Kalisegoro (16,67%) dan Pakintelan (15,78%). Unggas-unggas air di peternakan unggas tradisional berpotensi sebagai penularan virus AI, khususnya subtipe H5N1.Avian Influenza (AI) or better known as bird flu is caused by influenza viruses that mutate into a pathogen. Research on the isolation and the identification of H5N1 subtype needed to be carried out to determine the presence of the virus, particularly in the subdistrict of Gunungpati. The study design was explorative by collecting cloacal swab samples randomly from five villages in Gunungpati. The cloacal swab samples were cultured in embryonated SPF chicken eggs, then the RNA was isolated and followed by the identification of AI virus subtype using Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) with H5 and N1 gene detecting primers. A positive result is obtained if the visualization of the electrophoresis of PCR products showed bands with specific length of 219 bp for H5 gene and 131 bp for N1 gene. Fifty cloacal swab samples were isolated from five villages in Gunungpati, and eight of them were positive isolates and six of them were H5N1 positive with the prevalence rate of 12%. The positive isolates were derived from two species of duck (16,67%), 2 from wild duck (11,76%) and 2 from geese (18,18%). Of the five sampled villages, three villages were found to be H5N1 positive, i.e. Sekaran village (6,67%), Kalisegoro village (16,67%) and Pakintelan village (15,78%). Water birds in traditional poultry farming were considered potential as the transmitters of AI virus, particularly of H5N1 subtype.

Abstract

The entomopathogenic fungus, Metarhizium anisopliae (Metschnikoff) Sorokin (Ascomycota: Hypocreales)would effectively infect the target host on the appropriate medium water content. The aim of this study was to analyze the influence of water content of medium on the effectiveness of M. anisopliae fungus infection on O. rhinoceros larvae in the laboratory. Fifty healthy third instar larvae of O. rhinoceros were  obtained from field. The M. anisopliae obtained from Estate Crop Protection Board in Salatiga. The conidia density and viability of M. anisopliae were examined before used. The medium for maintaining the larva was the sawdust that had been sterilized. A total of 50 plastic cups were prepared to place 50 larvae (1 larva/cup). Each cup was filled with 100 g medium  of sawdust plus 2 g of M. anisopliae which was then stirred until mixed, with different water content: P1 (20%), P2 (40%), P3 (60%), P4 (80% ) and P5 (98%). The result indicated that  the water content of the medium affected the effectiveness of M. anisopliae fungus infection on O. rhinoceros larvae. The water content influenced the duration of larval mortality at each treatment. An important finding in this study is that controlling O. rhineceros larvae  with M. anisopliae can be done by manipulating the water content of medium. The benefit of this study may be used for the recommendation of O. rhinoceros pest control using M. anisopliae  with an effective water media content.

Abstract

Indonesia as a rich biodiversity country has many superior fruit plant germplasms such as sweet star fruit or carambola (Averrhoa carambola L.). Some varieties of carambola which collected at the Germplasm Garden of Research Center for Biotechnology-LIPI have been used for parent trees of fruit plant production. Therefore, they have to be characterized both phenotypically and genetically. The objective of the study was to analyze the relationship between eight varieties of carambola i.e. Malaysia, Penang, Rawasari, Bangkok, Sembiring, Dewabaru, Demak and Dewimurni at the germplasm garden based on phenotypic and genetic characters. Phenotypic characters were observed directly in the field, whereas genetic characters were observed with RAPD markers using 10 primers. Phylogenetic analysis was done using NT-SYS software showed that there were three clusters of carambola varieties. Meanwhile, Malaysia and Penang varieties have closed relationships (96%) compared with the other varieties. The result of the study would be dedicated to updating and completing the existing fruit plant collection database of Plants Germplasm Garden.

Abstract

Palm sugar solution diets that were offered to the Lorikeets contain  high energy, low protein and rich in amino acids similar to nectar. The major components of palm sugar are sucrose, which has a sweet taste. The Lorikeets  can detect energy-rich from the sweet taste of the feed. The aim of the study was to determine the influence of sweetness level and amino acid composition in palm sugar solution diets on feed intake of T. haematodus. The research method was completely randomized design arranged in a factorial with two factors. The first factor consisted of 4 types of palm sugars, i.e.   aren, coconut, siwalan/palmyra, and nipah sugar with a different degree Brix in 13.5º, 14.0º, 14.5º, and 15.0º Brix.While the second factor consisted of two treatments, with and without the addition of the commercial baby biscuit. The results showed that the feed intake of Siwalan sugar was significantly higher than the other palm sugar solution diets. Siwalan sugar solution diet has high sweetness level, sucrose content and essential amino acid Lysine. This study provides the new information regarding the sweetness level of palm sugar and amino acid composition in the diet that can affect feed intake of T. haematodus. Therefore, this information will help to make a formulation of appropriate diet that meets the nutrient requirements especially amino acid for the birds.

Abstract

Selection of bacteria in yoghurt fermentation is important to produce yoghurt with good quality. Tempoyak lactic acid bacteria is potential to be yoghurt starter culture becouse tempoyak fermentation has similarities in producing lactic acid such as yoghurt. This study aimed to isolate and identify the lactic acid bacteria (LAB) from tempoyak which will be used as a yoghurt starter culture. The methods used in this study included isolation and selection of acid-producing bacteria, lactase and protease activity test, identification of morphology and biochemistry as well as testing the quality of the yoghurt. The results of the study obtained 32 isolates of the LAB with the same characteristic colony, include the round shape, cream-coloured with convex elevation and, smooth surface and entire edge. Selection of acid-producing bacteria obtained 12 isolates with the ability to produce clear zones on MRSA + CaCO3 media ≥ 0.7 cm. Selection of lactase-producing LAB obtained six strains and the protease test obtained two superior strains. Two superior strains namely Tp 12 and Tp 28 have characteristics of coccus, gram-positive, negative catalase, non-endospore and non-motile forms. The organoleptic and several quality tests showed yoghurt using Tp 12 as starter has higher acceptability, the highest levels of lactic acid and lactose levels with values respectively 4.25, 0.84% and 24.53%. This study obtained the LAB strain which can be used as yoghurt starter culture. Tp 12 strain can be used to improve the quality of yoghurt and become a commercial starter that can be applied to various fermented products.

Abstract

Free radicals in cigarette smoke may interfere the balanced of body molecules and cause the damage of hepatic cells. This issues may be prevented by using red fruit oil (Pandanus conoideus L) due to its antioxidant content. This study aimed to evaluate the structure and physiological conditions of liver when the red fruit oil given to the rat that exposed by cigarette smoke. This study carried out for 35 days with female white rats as the sample. The samples were 20 female white rats divided into four groups: P0 (negative control), P1 (positive control), P2 (exposed by cigarette smoke with administration of red fruit oil 0,1 ml/BB), P3 (exposed by cigarette smoke with administration of red fruit oil 0,2 ml/weight). The data of ALT, AST, cell diameter, and hepatic weight were analyzed by using analysis of variants (ANOVA). The results showed that administration of red fruit oil containing antioxidants can reduce levels of ALT, AST, hepatocyte diameter, and increase liver weight, although the difference is not significant compared to the control group and does not affect the eating and drinking behavior so as not to affect the body's metabolism. The conclusion of red fruit oil can reduce ALT, AST, hepatocyte diameter and increase hepatic weight due to exposure to cigarette smoke, although the difference is not significant to the control group, and does not affect the eating and drinking behavior, so good for consumption because it gives a beneficial effect on health liver organ.

Abstract

Cell suspension cultures of Elaeocarpus grandiflorus produce flavonoids and various secondary metabolites. Flavonoid profile and antioxidant activity of the suspension culture extract of E. grandiflorus cells have not been studied. Therefore, this research aimed to analyze the antioxidant activity and flavonoid profile of the suspension culture extract of E. grandiflorus cells. Cell suspension cultures of E. grandiflorus were produced from leaf stalk callus that grew on WPM medium with a growth regulator 2.5 ppm 2,4-D. Cells were harvested at 30 days old and then extracted for profile analysis of bioactive compounds using the LC-MS method. The antioxidant activity analysis was conceded out using the DPPH method. The results showed that there were 32 types of flavonoids, of which 11 compounds had a concentration of more than 1% of the total bioactive compounds and had the potential to have antioxidant activity. The analysis results also showed that flavonoids type, composition, and antioxidant activity were not significantly different between the ages of E. grandiflorus cell suspension cultures. In addition, there was no correlation between flavonoid concentration and antioxidant activity. This study enriches information about secondary metabolites production in the E. grandiflorus cell culture which is still very rarely studied because of its rarity. The study also provides potential antioxidant as a scientific basis evident for E. grandifloras cell culture application in medical purpose.
