Abstract

The medicinal plant-derived bioactive compounds have a potential for many biological activities, including antimicrobial activity. Artemisia cina is a medicinal plant from the Compositae family with the potential of having antitumor, antifungal, and antibacterial activity. This study aimed to determine the antibacterial activity and the flavonoid content of A. Cina’s ethyl acetate extract. Plants samples were extracted by ethyl acetate maceration method. Antibacterial activity was tested against Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and Gram-positive bacteria (Bacillus subtilis and Staphylococcus aureus) by a disk diffusion method using 25, 50, and 100 mg/l extract concentrations. The flavonoid contents (quercetin and kaempferol) were measured using High-Performance Liquid Chromatography. The extracts of diploid and polyploid A. cina displayed some antibacterial activity, with the Gram-negative bacteria being more resistant than the Gram-positive counterpart. However, no significant difference was observed between the diploid and polyploid extracts. As for the flavonoid content, the highest quercetin content (0.5501 mg/ml) was found in the polyploid A. cina (J), while the highest kaempferol content (0.5818 mg/ml) was observed in the diploid A. cina (KJT). Although A. cina is widely grown in Indonesia, compared to other Artemisia species, A. cina has not been widely studied, especially its antibacterial  potential and in related to its flavonoid content and the use of ethyl acetate as the extraction solvent.  This study reveals the potential of A. cina as a natural antibacterial agent.

Abstract

Entomopathogenic fungi have the potential to infect most arthropods including termites which are economically important major insects pest of wood, wood products and building structures. However, the application of this fungus in the field has not shown satisfactory results yet, one of which is constrained in mass production of conidia. The purpose of this study was to evaluate 16 types of biodegradable products and waste as substrates for mass production of conidia using solid state fermentation method and two types of inoculum namely solid and liquid inoculum. Toxicity tests were carried out on subterranean termites (Coptotermes sp.) based on JIS K 1571, 2010. The parameters observed were the number and dry weight of the conidia produced, conidial viability, nutrient content of the substrate, and percentage of termite mortality. The results showed that rice, sorghum and corn were the best media for the growth of entomopathogenic fungi based on the number of conidia and dry weight of the conidia produced. Metarhizium sp. T4.B23 produced the highest number of conidia, 1.12 x 1011 conidia/100 g substrate and yielded 180.9 ± 0.623 g dry conidia/kg of rice; followed by Metarhizium sp. B2.2 grown on sorghum that resulted in 1.11 x 1010 conidia/100 g substrates and 8 ± 0.570 g /kg sorghum; and B. bassiana produced 8.3 x 109 conidia/100 g substrate and 31.24 ± 0.407 gr/ kg sorghum. Metarhizium sp. B2.2 showed the highest toxicity to termites with 100% mortality was observed within the second day of testing. Therefore the conidia of Metahizium sp. B2.2 is potential to be developed as a biopesticide using rice or sorghum substrate as a carrier.

Abstract

Parakeets (Melopsittacus undulates) are classified parrot order Psittaciformes. Parakeets are monomorphic birds whose sex is difficult to be distinguished. Sex identification is very important for breeding efforts in order to increase the parakeet population. External morphology was determined to identify the sex of the birds. This research was conducted to determine the sex of parakeets (male and female). This study used five male and five female parakeets aged 4 months as the study objects. This study used the quantitative and qualitative method. Results of this study was verified by performing surgery to determine the sex of parakeet based on their reproductive organs. The t-test results of morphometric characteristics showed no significant different in the length of body, upper bill, lower bill, wing, tail, femur, tibial-tarsus, tarsometatarsus and digits as well as the body weight between male and female parakeets.  The color of the cere was useful to accurately determine sex in parakeets (blue in males and white in females). This research provided information to the public about the differences between male and female parakeets for the selection of good broodstock in order to increase their population in captivity.

Abstract

AbstrakMikroorganisme tanah seperti Gliocladium sp dapat bertindak sebagai dekomposer dan juga sebagai agen pengendali hayati patogen tanaman hal ini memberikan harapan untuk mengurangi penggunaan pupuk dan fungisida sintetik. Tujuan penelitian untuk menguji potensi biofertililzer Gliocladium sp terhadap pertumbuhan dan produksi buah tomat. Variabel bebas yaitu bioferlizer Gliocladium sp  dengan dosis (g/tanaman) 0,50, 100, 150, dan 200 .Variabel tergantung adalah pertumbuhan dan produksi tanaman tomat. Parameter pertumbuhan yang diukur adalah  tinggi tanaman, kandungan klorofil a dan b. Parameter hasil tanaman yang diukur adalah berat buah setelah panen. Hasil uji Anava satu jalan menunjukkan bahwa pemberian Gliocladium sp perpengaruh terhadap tinggi tanaman, kandungan klorofil a dan klorofil b, berat tomat  . Hasil uji lanjut Duncan menunjukkan bahwa dosis 100, g 150 g dan 200 g tidak berbeda signifikan terhadap tinggi tanaman, dosis 150g dan 200 g tidak berbedan nyata dalam pembentukan klorofil a dan klorofil b. Pemberian Gliocladium dosis 150g paling tinggi terhadap berat buah. Simpulan dalam penelitian ini bahwa pemberian Gliokompos sp berpengaruh terhadap pertumbuhan dan produksi buah tomat. AbstractSoil microorganisms such as Gliocladium sp. can play role in the decomposition and can also become the biological control of pathogenic plants. The animal is potentially reduce the use of fertilizers and synthetic fungidid. The aim of the study was to test the potential effect of biofertililzer Gliocladium sp. on the growth and production of tomatoes. The independent variables was biofertilizer Gliocladium sp. at various dosages (g/plant) of 0, 50, 100, 150, dan 200. The dependent variables were the growth and the production of tomatoes. The growth parameter measured were plant height, as well as a and b chlorophyl contents.  The crop parameter was the fruit weight post-harvest.  A one-way anova showed that Gliocladium sp. affected the plant height, the a and b chlorophyl contents, and the tomato weight.  The Duncan’s post-hoc test showed that there was no significant difference on plant height among 100g, 150g and 200g treatment dosages, and 150g and 200g treatment dosages do not affect the formation of a and b chlorophyl. The Gliocladium dosage of 150g was the strongest in influencing the fruit weight.   It was concluded that the administration of Gliokompos sp. affects the growth and production of tomatoes.

Abstract

Feronia limonia (L.) Swingle better known as wood apple is an identity plant of Rembang Regency, Central Java Province, Indonesia. However, the population is very limited. In order to identify the conservation efforts, this ethnobotany study was aimed to determine traditional knowledge on the existence, economic value, utility, breeding technique and conservation of wood apple. The survey was conducted on a total of 102 local people from 10 sub-districts which were determined by proportionally stratified random sampling. Data were collected by questionnaires and individual interviews and then were analyzed descriptively. The results showed that all of respondents knew the figure of wood apple, but only 79.50% understood its status as an identity plant. Most of respondents (96%) stated that the fruits have low economic value, 80.39% respondents have utilized ripe fruit as ingredients of beverages and stems as firewood. This utilization was only equal to 29.72% compared to the potential usability based on research results. It was concluded that the traditional knowledge about the existence, economic value, and breeding technique were good and on the contrary, the knowledge about the plant’s usability was bad. The results will form a basic knowledge for in-depth studies to provide a government policy of Rembang Regency in order to develop an efficient strategy of management and conservation of wood apple as plant identity.

Abstract

Malnutrition, a major risk factor for a number of infectious diseases, including acute upper respiratory tract infections (AURTI), is common in developing countries. Nutritional status is an important index of the quality of life. Objectives:To analyze the correlation between food intake and health status to nutritional status of 9-11 years old children in Semarang. The study was a correlation study carried among school children in Semarang aged 9-11 years old. Data are presented in the descriptive analyses and Spearman correlation. Overall, food intake (energy and protein) of 9-11 years old children in Semarang is normal with ≥ 90% RDA, health status of them was satisfactory (very low AURTI incidence),and their nutritional status were mostly normal. There was a correlation between energy intake with nutritional status with indicators BMI, and z-score of W/A and H/A, but there was no correlation between protein intake and AURTI with nutritional status. Energy and food intake of the children correlate with all nutritional status being studied. It should be suggested to parents to implement balanced diet, to avoid the development of obesity among elementary school children through nutrition education to prevent malnutrition as well as obesity.How to CiteAli, A. Abstract

Mosquitoes are well known as vectors of hazardous diseases for human. Plant extracts can be used as an alternative for larval control due to they are a rich source of bioactive chemicals and safe for the environment. The present study investigated the larvicidal activity of crude extracts derived from leaf and flower of Brugmansia candida against the second larval instar of Aedes aegypti and Culex quinquefasciatus. The larval mortality was observed at 24 and 48 h exposure of both leaf and flower extracts, at the concentration of 100, 250, 500, and 1000 ppm. The 24 h exposure of both extracts at the concentration of 500 and 1000 ppm resulted in larval mortality rates were significantly lower than those of 48 h exposure. However, the mortality rate was not significantly different at the lower concentrations of crude extracts. The results also suggested that there was no significant difference in the larvicidal effect between leaf and flower extracts at 24 and 48 h exposure for all concentrations. The LC50 values at 48 h exposure for leaf extract were 789 and 791 ppm for Ae. aegypti and C. quinquefasciatus, respectively, whereas for flower extract were 772 and 780 ppm for Ae. aegypti and C. quinquefasciatus, respectively. Overall, B. candida showed larvicidal activity against Ae. aegypti and C. quinquefasciatus. This research contributes to new finding regarding the larvicidal activity of B. candida. This finding also supports the next study to develop B. candida as an alternative source for larval control agent.

Abstract

Cholesterol in quail meat is one of the factors causing consumers to control consumption of quail meat. On the other hand, the antioxidant content in quail meat could attracts the consumers. Cinnamon (Cinnamomum sp.) bark and gotu kola (Centella asiatica) leaves are medicinal plants that contain many polyphenol compounds. The objective of this study was to obtain the most optimum formula made from cinnamon and gotu kola as a feed supplement to produce the quail meat which has low cholesterol but rich in antioxidants. This study used a completely randomized design which consisting of six groups with three replications. The six groups included one control (P0), 5 treatments consisted of 5% cinnamon (P1), 5% gotu kola (P2), combination of cinnamon and gotu kola powder with ratio 5%:5% (P3), 5%:10% (P4) and 10%:5% (P5).  The results showed that food supplements made from cinnamon bark and gotu kola leaves could increase the body weight, antioxidant levels, and reduce cholesterol levels in quail meat. Cinnamon as feed supplement with a concentration of 5% or a combination of cinnamon and gotu kola with a ratio of 5%: 10% provided the best effect on antioxidants and cholesterol level in meat. The use of cinnamon and gotu kola supplementation to produce high antioxidant and low cholesterol of quail met have not studied yet. The result of the study would be beneficial for developing the healthy, safe and good quality of quail meat.

Abstract

Housekeeping gene is a gene expressed with a fixed level and in abundant amounts under various conditions. After validation, the housekeeping gene can be used as an internal control to normalize gene expression data. This study reports the isolation of several housekeeping genes in Durik-durik plant (Syzygium sp). This plant material in form of fresh leaves from Durik-durik plants are taken from Kajuik Lake, Riau Province. The next stage is total DNA isolation, polymerase chain reaction, electrophoresis, sequencing and data analysis using bioinformatic tools. The isolated housekeeping genes included 18S rRNA, actin, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), Beta-tubulin and Ubiquitin with sizes of 400 bp, 679 bp, 1134 bp, 836 bp, 1167 bp and 2155 bp, respectively. In addition to 18S rRNA, the five housekeeping genes are the first reported from the genus Syzygium and referable to isolate housekeeping genes in other species in this genus. The six housekeeping genes can be used as internal controls on Durik-durik plants after validation.

Abstract

Allethrin  is  one  of  synthetic  pyrethroid types which  is  used  in many mosquito repellents and has toxic potential. The aim of this research was to examine the infl uence of vitamin C and E antioxidant activity to the sperm quality of allethrin-induced albino rats. This research used a post-test randomized design. The population of the research were  8  hr/day  allethrin-induced male  albino  rats. The  sample  of  the  research was allethrin-induced male albino rats which were divided into 4 groups, i.e. group I was the control group, group II was the group treated with 1.8 mg vitamin C per day, group II was the group treated with 1.44 mg vitamin E per day, and group IV was treated with a combination of 1.8 mg vitamin C per day and 1.44 mg vitamin E per day. The duration of the treatment was 45 days. The sperm sample was taken in day 46. The data consisted of sperm count or amount, sperm motility, sperm viability, and sperm morphology, and then was analyzed with ANOVA followed by BNT. The result showed that vitamin C and E antioxidants signifi cantly infl uenced the the sperm quality of the 45-day mosquito repellent-induced rats. Overall, the allethrin-induced albino rats treated with vitamin C and E antioxidants were better in maintaining the sperm quality than single treatment of vitamin antioxidants.Keywords: allethrin, vitamin C, vitamin E, antioxidant, sperm  quality.
