KRAS and BRAF Mutation in Borderline Epithelial Type Ovarian Tumor Suardi, Dodi

Abstract

Objective: To understand the molecular profile by identifying the mutation of KRAS and BRAF in borderline type ovarian tumor. Method: In the study, we examined paraffin tissue sample from Department of Pathology Anatomy, University of Indonesia/Dr. Cipto Mangunkususmo Hospital, Jakarta, that was diagnosed as borderline epithelial ovarian tumor. Seventeen samples were taken to Sandya Laboratory in Bandung for examination of PCR BRAF exon 15 codon 600, and KRAS in exon 2 codon 12 and 13, as well as exon 3 codon 61. Result: Mutation of KRAS occurred in 94% of subjects (serous borderline 62.5%, mucinous bordeline 37.5%), of which 70.6% mutation happened in exon 2 codon 12 (serous borderline 33.3%, mucinous borderline 66.7%), 52.9% mutation in exon 2 codon 13 (serous borderline 33.3%, mucinous borderline 66.7%), and 76.5% mutation in exon 3 codon 61 (serous borderline 30.8%, mucinous borderline 69.2%). Mutation of BRAF occurred only in 47% of subjects, but the results of Exact Fisher test showed that mutation in BRAF gave significant result, while other variables did not give significant result (p=0.009). Conclusion: Molecular pathology in borderline ovarian tumor related with BRAF mutation is more likely to occur in serous borderline type, while KRAS mutation is more likely to occur in mucinous borderline type. Abstract

Diet Makanan Tinggi Garam (DMTG) menyebabkan: (1) Kenaikan nilai peroksida lipid yang diukur secara kualitatif dengan MDA. (2) Persalinan preterm sampai dengan 40% dan peroksida lipidnya tinggi. (3) Hipertensi pada model SDR yang bunting. (4) Kenaikan nilai malondialdehida (MDA) jaringan jantung yang disertai dengan penurunan Glutation Peroksidase (GPx). (5) GPx jaringan tidak berperan untuk menetralkan MDA pada jaringan jantung. (6) tidak menyebabkan perubahan denyut jantung pada model tikus penelitian. (7) Tidak menyebabkan perubahan komponen darah tetapi menaikkan HCT dan WBC pada SDR bunting kelompok perlakuan. (8) Perubahan yang nyata pada struktur otot jantung baik pada induk maupun embrio SDR bunting kelompok perlakuan. (9) Kelainan endotelium pembuluh kapiler jantung. (10) Kelainan struktur mitokondria pada sel jantung induk dan embrio, dan lebih kecil dari kasus kontrol (tanpa perlakuan).

Abstract

Objective: To evaluate the efficacy of PPH management by conservative surgical management using B-Lynch Method and Surabaya method (Modified B-Lynch compression suture) as an alternative to hysterectomy in PPH due to uterine atony. Method: Eighty-four cases of PPH from 2786 deliveries were recruited in period of July 1, 2007 - August 31, 2008. The study was perfomed in Ob/Gyn Dept. Dr. Soetomo Hospital Surabaya, Indonesia. Its management consisted of 72 cases succeed with medical (uterotonics) and mechanical technique (tamponade technique) and the remain of 12 failed cases was done by conservative surgical technique that preserving uterus. The surgically therapy on these 12 cases were proceeded by B-Lynch technique on 4 cases and another 8 cases with Surabaya Method in which the technique was done by performing brace suturing way with 3 longitudinally stitches using chromic catgut no 2 and round needle. Result: From 12 cases showed that all 8 cases with Surabaya Method technique were success to stop bleeding, but 2 cases were failed from B-Lynch Method and hysterectomy were done. Two out of 8 cases Surabaya Method and 1 B-Lynch technique case were ended with death which possible causes were HELLP syndrome, DIC and multiple organ failure complication in severe preeclampsia, although actually the bleeding had already been stopped. Conclusion: The present study showed that Surabaya Method was effective to stop bleeding in PPH conservative surgical management with uterus preserving. Beside simple, effective and easy to implement, the advantage of Surabaya Method was quick. Abstract

Objective: To determine the difference of HER2/neu expression in benign, borderline and malignant epithelial ovarian tumor. Method: An analytic cross-sectional study was performed on 49 samples of epithelial ovarian cases which were divided into three groups: twelve cases of benign epithelial ovarian tumor, 9 cases of borderline epithelial ovarian tumor and 28 cases of malignant epithelial ovarian tumor. Paraffin embedded tissue samples was cut, stained with HER2/neu imunohistochemistry and examined in Pathology Laboratory at Sanglah Hospital. The other data was collected from medical record samples. The data was analysed using One Way Anova test and Chi-square test with level of confidence α = 0.05. Result: There were 13 cases (46.43%) of the HER2/neu over expression in malignant epithelial ovarian tumors, 2 cases (22.22%) of the HER2/neu over expression in borderline epithelial ovarian tumors and 1 case (9.09%) of the HER2/neu over expression in benign epithelial ovarian tumors. With p = 0.048 (p < 0.05), there are differentiation of HER2/neu expression in benign, borderline and malignant epithelial ovarian tumors. Conclusion: In this study HER2/neu expression was different between benign, borderline and malignant epithelial ovarian tumors. Abstract

Objective: To determine the role of progesterone-induced blocking factor (PIBF) in women with threatened abortion. Methods: This was a cross-sectional study. The blood serum of two groups, the first one was women with normal gestation of  20 weeks, and the second one was those with imminent abortion in Prof. Dr. R.D. Kandou Hospital, and Subcenter Hospital in Manado, was collected. Samples were processed with PIBF ELISA-kit. Results: PIBF serum value of women in normal gestation  20 weeks is (47.15323.830)ng/ml and threatened abortion is (11.540 4.892) ng/ml, with p value = 0.000. Conclusion: PIBF serum value of women with threatened abortion is significantly lower compared to women of normal gestation  20 weeks. This study showed that PIBF has an important role in maintaining pregnancy and can be used as a biologic marker of a pathologic process in pregnancy.Keywords: early pregnancy, pregnancy immunology, progesteroneinducedblocking factor, threatened abortion

Abstract

Objective: To determine the effect of polymorphism of estrogen receptor β to the risk of epithelial ovarian carcinoma at Dr. Mohammad Hoesin Hospital Palembang Methods: This population-based case control study included 40 women with epithelial ovarian carcinoma and 40 controls, from January 2010 until December 2011. Data analysis was performed by Chi Square test. Results: Distribution of estrogen receptor beta genotypes, both GT and TT (wild type-mutant and mutant), among case subjects was significantly higher than control (24 subjects {60%} vs. 5 subjects {12.5%}). The genotype effect was statistically significant for rs1271572 (OR=2.636; p=0.039) with Chi Square analysis. Whereas the allotype effect was also statistically significant (OR=1.949; p= 0.047). Conclusion: Polymorphism of estrogen receptor β may play a role in the risk of epithelial ovarian carcinoma at Dr. Mohammad Hoesin Hospital Palembang. Abstract

Objective: To determine the role of matrix metalloproteinase-2(MMP-2) in serum and peritoneal fluid of endometrioticpatients.Methods: Research’s design using cross-sectional method in Dr.Wahidin Sudirohusodo hospital and several other hospitals inMakassar within May 2015 until May 2016. Subjects were chosenusing consecutive sampling technique. The examination usingthe ELISA method. The data were analysed using Fisher exact,t-independent, Mann-Whitney, and Spearman association.Results: A total of 50 subjects were recruited in this study. Mostlythe value of MMP-2 serum and peritoneal fluid in endometriosisgroup was higher compare to study control. There was significantdifferent between the total of MMP-2 serum and peritoneal fluid.There was also a significant association between the value ofMMP-2 serum and peritoneal fluid with endometriosis.Conclusion: The value of MMP-2 serum and peritoneal fluid werehigher in endometriotic patients compared to those withoutendometriosis.
