Abstract

The success of transgenic fish production has been achieved through eggs fertilization using electroporated sperms carrying exogenous DNA. This study was conducted in order to obtain the optimal electroporation condition for stripped catfish sperm. A plasmid containing green fluorescent protein (GFP) gene driven by carp β-actin promoter was transferred into sperm using electrophoresis method towards transgenic stripped catfish (Pangasionodon hypophthalmus) production. Electroporation was carried out using square wave shock with pulse length of 30 ms and pulse interval of 0.1 sec. Treatments are combination between voltage (50 V, 75 V, and 100 V) and pulse number (1 and 3). Exogenous DNA concentration used was 10 μg/mL of Tris-EDTA. Results showed that increasing the voltage from 50 to 100 decreased sperm motility, while pulse number did not affect sperm motility. Voltage of 50 gave the best motility of sperm, although sperm viability relatively similar between treatments and control except at 100 V with 3 pulses number. Further, electroporation-treated sperms were able to fertilize eggs. Higher hatching rate of eggs was obtained in electroporation treatment at 50 V with pulse number of 1 and 3. The persistence of transferred GFP was detected in electroporated and incubated sperms (control). However, GFP was only detected in larvae from eggs that were fertilized by electroporated sperm. Thus, electroporation could be applied to produce transgenic stripped catfish.

Abstract

In fish culture, optimal growth could be influenced by various culture methods. Aim of the study was to evaluate the productivity of Barbonymus balleroides, lalawak in floating net cages, concrete ponds, and earthen ponds. Cultivation was designed with the circulation water system. Experiment was conducted using completely randomized design with three treatments and three replications for each treatment. The experimental fish, sized of 4.20 ± 0.64 cm SL and weight of 2.14 ± 0.99 g, were obtained from induced breeding. The stocking density used was 20 individuals/m3. Fish were fed 3% of total weight two times every day using commercial pellet with 35% protein content for 90 day. The result showed that lalawak reared in earthen pond was no significant difference on length, weight, and biomass compared with that one in concrete pond (P>0.05), but significantly different (P<0.05) with floating net cages. There were no different (P>0.05) among the three different culture systems for survival rate and FCR. Lalawak reared on earthen pond system supported with optimal water quality could increase productivity value.

Abstract

African catfish, Clarias gariepinus, is one of economically important farmed species in Indonesia. To support the development of aquaculture industry, high genetic quality of both broodstock and seeds is required and breeding program is considered as viable option. Information on genetic variation of the populations being considered to form a base population may give insight toward the appropriate strategy to be implemented in breeding program. This study was aimed to assess genetic variation in farmed populations of catfish in Indonesia using microsatellite markers with special emphasis on their use to develop breeding program. Three populations of farmed catfish, namely Dumbo, Paiton, and Sangkuriang were collected. Fifteen individuals representing each population were screened for microsatellite variability using seven primer sets (cga01, cga02, cga03, cga05, cga06, cga09, cga10). Results found that with exception of two loci (cga01 and cg02) which had a slight increase, the other four loci showed reduction in the number of alleles ranging from 35% to 80% depending on loci. Farmed populations also showed heterozygote deficient and inbreeding level, being the highest was found in Sangkuriang and the least was observed in Dumbo population. Individuals within populations contributed most (95%) while interpopulation variation accounted for only 5% of the total genetic variation. Populations of Dumbo and Sangkuriang were genetically similar while populations of Paiton were genetically different from both Dumbo and Sangkuriang. Viewed from genetic perspective, by combining all information emerging from this study, the best possible strategy to establish a base population with broad genetic base and less inbreeding would be to combine all the populations into a synthetic base population.

Abstract

Inbreeding depression has often been considered to be responsible for the deterioration of performance in aquaculture species. Despite a crucial impact that may result from inbreeding depression, comprehensive information reviewing this subject is limited. This study was aimed to gain information on the effect of inbreeding on the early performance of freshwater prawn. The study was performed by comparing performance of inbred and outbred populations. Inbred population was established by brother-sister mating (inbreeding rate of 25%) while the outbred population was formed by mating unrelated individuals. Several fitness and productivity related traits including survival, the rate of larval development, stage dispersion and growth of larvae were evaluated. Results suggest that inbred families performed poorer than that of the outbred in survival. However, inbreeding depression did not seem to occur in other traits including the rate of larval development, larval stage dispersion and growth. This study implies that to maintain genetic quality of farmed prawn stocks, inbreeding rate in farmed population must be controlled not to exceed that level. Implications that these findings may have on aquaculture practices and possible alternatives for the solutions are discussed.

Abstract

Milky hemolymph disease of spiny lobster (MHD-SL) is categorized as the most destructive disease in farming spiny lobster. Therefore, it is required to investigate the routes of milky disease infection in spiny lobster as a basic knowledge in order to prevent milky disease transmission. The aim of the present study was to perform an experimental infection of milky disease in spiny lobster Panulirus homarus. Experimental infection of milky disease was carried out by several modes of infection which were injection, immersion and per os exposure. Injection of each 0.2 mL undiluted and diluted hemolymph from the diseased lobster resulted in a cumulative mortality of 100% at 15 days post-infection (dpi), and 75% at 16 dpi, respectively. Experimental infection through water immersion caused in a cumulative mortality of 50% at 7 dpi. In contrast, no mortality was observed in per os exposure as well as in control groups. Results of this experimental study provided evidence for horizontal transmission of MHD-SL among P. homarus. Histopathological analysis exhibited that there were masses of Rickettsia-like bacteria (RLB) in the connective tissues of the gill, hepatopancreas, gonad, midgut, and muscle tissues of the affected lobsters. Mass of RLB was not only found in the moribund lobsters but also in the surviving lobsters with milky hemolymph appearance.

Abstract

The role of tiger shrimp defense against invading pathogen on molecular level such antiviral gene expression is limited to be reported. Gene expression is a process which codes information of genes that is converted to the protein as a phenotype. Distribution of PmAV antivirus gene, that has been reported as an important gene on non-specific response immune, is needed to be observed to several organs/tissues and size of tiger shrimp. The aim of this study is to determine the distribution of gene antiviral expression at several organ/tissue and size of shrimp. The organs/tissues observed in this study were: gill, hepatopancres, muscle tissue, eyes, heart, stomach, gonad, and intestine. While the size of shrimp consisted of three groups, those are: (A) 10-20 g/ind., (B) 30-40 g/ind., and (C) 60-70 g/ind. Analysis of antiviral gene expression was performed by RNA extraction, followed by the cDNA syntesis, and amplification of gene expression by semi-quantitative PCR. The result of PCR optimation showed the optimal concentration of cDNA and primer was 1 μL and 50 mol, respectively for PCR final volume of 25 μL. Antiviral gene was expressed on the hepatopancreas and stomach in percentage of 50.0% and 16.7%, respectively. While the highest percentage of individual expressing the antiviral gene was observed in the shrimp size of C (66.7%), followed by B (50.0%) and A (16.7%). The result of study implied that the hepatopancreas has importantly involed in tiger shrimp defense mechanism on viral infection.

Abstract

Common carp and koi (Cyprinus carpio) are the main consumption fish commodity and ornamental fish in Indonesia. Diseases due to koiherpes virus (KHV) infection had caused a huge loss in both common carp and koi culture industries world-wide. This study was generated to select the most potential candidate from out of three koiherpes virus isolates based on their serological performances for vaccine development to control Koiherpes virus disease in koi. Virus collection, isolation, and propagation in cell culture have been conducted from common and koi samples originated from Banjarmasin, South Kalimantan; Takalar, South Sulawesi; Cirata Lake-Cianjur, West Java; Depok, West Java; Ciseeng, Bogor, West Java; and Purwokerto, Central Java. Determination of the causative agent of the disease was done by PCR (Polymerase Chain Reaction) assay and inoculation of filtrate homogenates from infected fish into cell culture. There are three koiherpes virus isolates (BJMN-1, BJMN-2, and SKBM) which are being successfully isolated, cultured and propagated in Koi Fin (KF-1) cell line. Calculation of tissue culture infection dose from each isolates were 104.55 TCID50/mL, 104.72 TCID50/mL, and 103.28 TCID50/mL. Immunoassay of the three isolates was conducted by Indirect-ELISA method. Polyclonal antibody was made by injecting each isolate to the experiment animals (rat/Rattus norwegicus strain Sprague Dawley). It was shown that all of three isolates have high level of immunogenicity, as seen from the absorbance values at 490 nm wavelength. BJMN-1 isolates have a higher potential for it’s ability to cross-react with the other isolates and provided the highest absorbance values. In future, all isolates can be used as KHV vaccine candidates to prevent KHV infection.

Abstract

The successful effect of phages in phage therapy has been proven by an increase of the presence of phages in survivors. Seven previously isolated phages were used to examine the effects of storage temperature and types of media on their survival rate. Phages PSaT-18, PSaT-19, PSaT-20 and PSaT-21 isolated from a Streptococcosis-infected Tilapia (Oreochromis niloticus) were used in this study. Phages PSaG-1, PSaG-2 and PSaG-3 were isolated from a diseased Gouramy (Osphronemus gouramy). All of the phages were stored in refrigerator at temperature of 8oC and kept in two types of media, Brain Heart Infusion Broth (BHIB) and BHIB enriched with glycerol. To determine the lytic activity of the phages, 20 μL of each phage suspension was poured onto BHIA plate and incubated at 25oC for 24 to 48 hours. The result showed that phages PSaT-18, PSaT-19, PSaT-20 and PSaT-21 have lytic activity against S. agalactiae and S. iniae than phages PSaG-1, PSaG-2, and PSaG-3. Phages PSaT-18, PSaT-19, PSaT-20, and PSaT-21 always survived up to three weeks and phage PSaT-18 in BHIB have the highest potential as a candidate of phage therapy for Streptococcosis.

Abstract

Two experiments were conducted to evaluate the hydrolysis of fiber content in palm kernel meal (PKM) by sheep rumen liquor enzyme and to know the apparent digestibility coefficient of hydrolyzed PKM for catfish Pangasius hypophthalmus. The first trial examined effectivity of sheep rumen liquor enzyme to decrease crude fiber content of PKM. The added volume of sheep rumen liquor enzyme was 0, 20, 40, 60, 80, and 100 mL/kg PKM and then it was incubated for 0, 12, and 24 hours. A factorial completely randomized experimental design consisted of 2 variables and triplicates were selected. The second trial was conducted to evaluate the apparent digestibility coefficients of hydrolized PKM for catfish. Apparent digestibility coefficients were determined using chromic oxide indicator added to both reference and test diets. The feed ingredients used in the trial were hydrolyzed PKM (PKMe) and unhydrolyzed PKM (PKM). Ten fishes with weighing around 20 g were used in the trial and held in 80 l tanks. Feces were collected from three replicate groups of fish using a fecal collection column attached to fish rearing tank. PKM hydrolyzed with 100 mL/kg and incubated for 24 hour showed the lowest crude fiber content (6.99%) among the treatments (P<0.05). Apparent digestibility coefficient of hydrolyzed PKM was 57.57% compared with unhydrolyzed PKM 15.31%. Based on the evaluation in those parameters it was concluded that sheep rumen liquor enzyme added to PKM was effective to decrease crude fiber content of PKM and improve apparent digestibility coefficient of PKM for catfish.
