Abstract

Dry extract of sarang semut (Myrmecodia pendens Merr & Perry) water/n-butanol fraction is native plant from Papua is proven to be active against cancer cells. This research aims to improve the quality of extracts of sarang semut become the raw material that meets the standards of medicinal plant extracts in the fulfillment of the quality of the extracts into anticancer Standardized Herbal Medicine / Obat Herbal Terstandar (OHT). Extraction with ultrasonic methods (1:10 with methanol) and fractionation with water:butanol (1:1) as much as 1:20. Standardization and data analysis based on parameters of the Medicinal Plant Extracts (Kepmenkes RI No: 55/Menkes/SK/I/2000) and BPOM RI Regulation No. 12, Year 2014 about quality requirements of traditional medicine. Dry extract of sarang semut (water/n-butanol fraction) met the standards of raw material preparation for OHT with organoleptics shaped dried extracts, dark brown color, aromatic smell, and has a sour-bitter taste. Moisture content was 9,259 ± 0.001 (≤ 10%). Total Plate Count was 5 x 103 colonies/g (< 104 colonies/g). Yeast and Mold Count was 0,5 x 101 colonies/g (< 103 colonies/g). Pathogenic bacteria test shows that MPN Coliform value was 0 colonies/g and negatively to the presence of Escherichia coli, Salmonella sp, Staphylococcus aureus, Pseudomonas aeruginosa, and Shiggella sp. The extract was not detected the presence of aflatoxin total (aflatoxin B1, B2, G1 and G2). The content of heavy metal Cd was 0,0052 ± 0.0005 ppm (≤ 0.3 ppm) and heavy metals Pb was 0,9429 ± 0,0087 ppm (≤ 10 ppm).

Abstract

Marine microorganisms, especially actinomycetes bacteria and fungi produce a variety of secondary metabolites that biologically active and have a unique structure. This study aims to isolate some strains of marine fungi that grow on marine algae as a potential source of antimicrobial agent. Marine fungal strains isolated from the marine algae with antimicrobial activity screening is done by using the disc diffusion method. In this study, it was obtained 18 strains of fungal isolates. The screening results that give best antimicrobial activity shown by the activity of the extract liquid culture of fungal isolate strain AKT.C6 are 10.0 mm against the bacteria Escherichia coli and 10.8 mm against Bacillus subtilis. Mycelium extract of fungal isolates strain AKT.C4 is 16.9 mm against the bacteria Escherichia coli and 17.6 mm against Bacillus subtilis, while the liquid culture extract and mycelium extract did not show any activity against Candida albicans. The isolation of fungi from Kappaphycus alvarezii algaee has potential as an antibacterial agent.

Abstract

Evaluation of teratogenic effects of purified extract isolated fromethyl acetate fractionation of surian leaves (Toona sureni (Blume) Merr.) onwhite female fetus mice has been carried out. The extract was given to miceorally in solution at doses of 5, 10, and 20 mg/kg BW on 6th until 15th day ofpregnancy. On 18th day of pregnancy, laparatomy was performed, then two– third of fetuses were immersed in alizarin red solution and the remainingin Bouin’s solution. The analyzed parameters were the weight of pregnantmice, the number of fetuses, the weight of fetuses, skeletal malformation andmorphology. The result showed that malformations were not shown in dose of 5mg/kg BW, but in dose of 10 mg/kg BW from one mice found one fetus causedlate growth and death. While on dose 20 mg/kg BW, one mice was not death.

Abstract

Pomegranate (Punica granatum L.) is known to have estrogenic activity. Pomegranate peel proven to increase the number of osteoblasts and increases bone density. This study aims to determine the potential of the water fraction of the pomegranate peel as natural Selective Estrogen Receptor Modulators (SERMs) through histological mammary in ovariectomized rats. Ovariectomy method used as a model representing menopause, conducted on 42 rats female Sprague Dawley strain aged 50 days were divided into 7 groups: Sham, OVX, and a group of OVX, each getting estradiol, tamoxifen and fractions water pomegranate peel dose of 50, 100, and 200 mg/kg BW. The results obtained showed water fraction of pomegranate peel did not show any increase in volume and number of mammary ducts.

Abstract

Greenbean (Vigna radiata (L.) R. Wilczek) contained phytoestrogen suspected has a potential effect as an preventive and alternative treatment of osteoporosis. This study is proposed to determine the effect of extracts of greenbean juice (Vigna radiata (L.) R. Wilczek) on bone density by measuring calcium levels using Atomic Absorption Spectrofotometry (AAS) from femur and the number of osteoclasts collected from histological object which made from ovariectomized rat tibia. Ovariectomy method conducted to represent the osteoporosis condition on 36 female Sprague Dawley rats were divided into 7 groups. Group 1 noticed as Sham was the rat control with surgery without ovariectomy and distilled water, group 2 as negative control is given distilled water and ovariectomized rat control, group 3 as positive control was given estradiol and ovariectomized rat control, group 4 as positive control was given vitamin E and ovariectomized rat control. Group 5, 6, and 7 were given 50 g/100 ml the greenbean juice with volume variation 5 ml/200 gBW, 2.5 ml/200 gBW, and 1.25 ml /200 gBW. Six groups had ovariectomized except sham group and rested for 20 days. Greenbean juice was given orally once daily started from day 21 until day 50. Calcium concentration and osteoclast measurement process taken on day 51. The results of bone calcium levels and osteoclast number showed that greenbean juice did not give significant differences among ovariectomized rats.

Abstract

Pecut Kuda leave (Stachytarpheta jamaicensis L.) was one of the many medicinal plants found in Indonesia and it has been widely used by the people in Indonesian to cure infection of tractus urinarius, pharingitis, hepatitis. Although, it has been very popular traditional medicine, very little scientific data were collected until now. This study focused on preliminary studies of antibacterial and antioxidant activity of Pecut Kuda leaves. Extracts were obtained using n-hexane, ethyl acetate and methanol. The antibacterial activity test was performed using the agar diffusion method against Staphylococcus aureus and Escherichia coli. While antioxidant activity was performed by the colorimetric method using DPPH radical reduction. The results showed that n-hexane extracts did not have either antibacterial or antioxidant activity. The ethyl acetate extract had zone inhibition (DDH) between 7 to 11.67 mm against S. aureus bacteria, whereas against E. coli with DDH between 7.67 to 10.83 mm, and had IC50 value of 182.97%. The methanol extract had DDH between 8.33 to 13.67 mm against S.aureus, whereas against E. coli bacteria with DDH between 9 to 14.67 mm, and IC50 value of 14.28%. In conclusion, ethyl acetate and methanol extract of Pecut kuda leaves have antioxidant and antibacterial activity against S. aureus and E. coli bacteria.

Abstract

Diabetes mellitus is a chronic disease with hyperglycemic and oxidative stress component that can lead to various complications. The stem bark of faloak have been used traditionally to treat diabetes by local people in East Nusa Tenggara. Therefore, this research aimed to study antidiabetic activities in alloxan-induced rats of faloak ethanolic extract. The all rats were divide at random into group I : normal control; groups II : diabetic control; group III : standart (glibenclamide); group IV – VI : administrations group of faloak stem bark ethanol extract at a dose of 65 mg / kg, 130 mg / kg and 260 mg / kg for 14 days. Blood samples were collected from rats in duration 0 – 14 days for T0, T4, T7 and T14 after alloxan treatments. The results showed that ethanol extract of faloak’s bark dose of 260 mg/ kg on 14 days had decreased blood glucose levels 46,07%, no different glibenclamide group. The dose of 65 mg/kg and 130 mg/kg reduced glucose level respectively 26,32 % and 38,09 %.

Abstract

Binahong (Anredera cordifolia (Ten.) v. Steenis) leaves empirically have been used to lower cholesterol levels. In this study, preventive activity of ethanol extracts will be tested on the animals fed with a high-cholesterol chow. Hypercholesterolemic animal model established by orally administering high-cholesterol chow, pure cholesterol, cholic acid, and propiltiourasil. The test animals were divided into 5 groups, including the positive control group, ethanol extract of binahong leaves at doses of 50 mg/kg bw, 100 mg/kg bw, and 200 mg/kg bw; and simvastatin at dose of 36 mg/kg bw. Extract was given at the same time as hypercholesterol induction. Lipid profiles were measured at the time before the induction, 15 days and 29 days after induction. Index of atherogenic was also calculated. The results showed that level of cholesterol and LDL of extract binahong at dose of 50 mg/kg (109.17±26.60 mg/dL and 55.78±23.92 mg/dL, respectively) had significant difference compared to a positive control group (p < 0.05). Furthermore, binahong extract at dose of 50 mg/kg bb had the lowest atherogenic index (0,68±0,27) and aorta thickening (150.60±41.72 μm) compared to other extracts at day 15 of induction. In conclusion, extract binahong at dose of 50 mg/kg bw rat gives the best effect to prevent the increase of cholesterol and LDL level on wistar male rats fed with a high-cholesterol chow.

Abstract

Ethanol extract Kencur (Kaempferia galanga L.) has been proven to have antiinflammatory effect as well as to decrease the side effect of NSAIDs drug use, to develop the formulation of ethanol extract of kencur, then made in the dosage form of topical patch. Topical patch preparations are expected to avoid effects on the gastrointestinal tract, first- pass metabolism, and facilitate the use of the patient thereby increasing patient compliance. In the patch formulation, the most important things to note is the penetration enhancer/ enhancer, and enhancer used in this study is sodium lauryl sulphate. This study aims to determine the effectiveness of sodium lauryl sulphate in the preparation of a topical patch of kencur extract as anti inflammatory on the number of neutrophils in the blood. The research method uses canrrageenan as an inflammation inductor and neutrophil count observation was conducted using blood smears, then was observed on the first, third and seventh. Group of treatment which was given sodium diclofenac gel as positive control, group of treatment which was without enhancer as the 1st group, groups of treatment which were givenenhancer 1%, 3%, and 5% as the 2nd, 3rd and 4th group. The result was analyzed by one way ANOVA, followed by Post Hoc Duncan. The experimental results prove that there was a significant difference between the number of neutrophils and macrophages in the negative control group who were not given enhancer and the treatment group were given enhancer sodium lauryl sulphate values ( p < 0.05 ). As well as the group treated with the addition of enhancer sodium lauryl sulfate showed a greater amount of decline compared to the treatment group that contains only ethanol extract kencur.

Abstract

Centella asiatica L., or known as pegagan leaf, empirically can be used as a tonic for organs such as liver, kidneys and brain. It also can increase memory. In our society, Centella asiatica L., hasn’t been optimally processed as atonic. Commonly, it is processed by boiling or grating the leaf. Raw consumption of this leaf inflicts bad taste. The aim of this research is to find out tonic effect and determine the effective dose of herbal syrup Centella asiatica L. The effect of tonic can be determined using natatory exhaustion method, by observing stimulant effect of a certain drug to an animal.Methods : This research used male mice were divided into 5 groups of treatment. The first group: negative control without herbal syrup Centella asiatica L., substance, positive control 50 mg/kg body weight, first dose group 120 mg/kg mice’s body weight, second dose group 150 mg/kg mice’s body weight and third dose group 180 mg/kg mice’s body weight. The effect of tonic was observed through the increasing duration of the mice’s self-defense ability when it was swum.Result: This research resulted that the use of herbal syrup Centella asiatica L., gave tonic effect on dose 120 mg/kg mice’s body weight, 150 mg/kg mice’s body weight, and 180 mg/kg mice’s body weight. The most effective dose of the use of herbal syrup Centella asiatica L., is on dose 180 mg/kg mice’s body weight.
