Abstract

Mesenchymal stem cells have been known to have the nature of supressing immune response. This cell have been proven to reduce the symptoms of organ transplants rejection. Clinically proofing is also being performed on an autoimmune disease caused by the complex immune such as systemic lupus erythematosus (SLE). This study aimed to know aggregated IgG-stimulating effects of mesenchymal stem cells in relation to IDO secretion, an immunosuppressant agent. Mesenchymal stem cells was isolated from liposuction waste and characterized based on its surface molecule accrording to ISCT criterias. The confirmed cell culture was used to the next culture by the addition of heat aggragated immunoglobulins gamma (HAGG) for six days. IDO secreted on supernatant culture was collected and assayed by elisa method at 450 nm. The results showed that there was elevated levels of IDO (16.73-49.5%) on culture with HAGG stimulation than without stimulation. This study could be a part of the basic

Abstract

Plant of dewa leaves (Gynura pseudochina ( L. ) DC) from family Asteraceae is a plant that has been widely used for some treatments including blood circulation , trauma, bleeding and blood clot. In this research, the isolation and identification of flavonoid compounds in n-butanol fraction of the methanol extract of dewa leaves (Gynura pseudochina ( L. ) DC) has been carried out. The compound in Dewa leaves was isolated using maceration with methanol which was then partitioned by n-hexane solvent, ethyl acetate and n-butanol. The n-butanol phase was then isolated by paper chromatography with BAW (4:1:5) as a mobile phase which resulted in seven bands. Each of the bands was further identified using spectrophotometry UV-vis to show wavelength of maximum absorption that was given from only three bands for flavonoids namely NB-III, NB-V and NB-VI. Moreover, the bands were further eluated with 15% acetic acid as second mobile phase and the spot appeared in the presence of ammonia. The final isolates were identified by UV-vis spectrophotometer with addition of a shift reagent. The resulting spectrum from the isolates NB-III was considered as the one of flavonol compounds with the OH group at theposition of 3.7 and oxygenation at 6 or 8 and o– at OH group in ring B; NB-V isolates, OH groups at positions 3, 5, 7 and o– at OH group in ring B and NB-VI isolates, the OH group at position 5,7 and 8.

Abstract

Investigation on formulation of film coated tablets containing combination of three active substances, i.e. extracts of soursop leaf, mangosteen rind and Ling Zhi mushroom has been carried out, which was expected to have high antioxidant and immunomodulator activities. The research consists of formulation of film coated tablets F1 containing extracts of soursop leaf-mangosteen rind-Ling Zhi mushroom (100:100:125 mg) and F2 (125:75:125 mg) in each 840 mg tablet. Full examination showed that film coated tablet F1 and F2 comply with the 4th Indonesian Pharmacopoeia, except for the dissolution test. Determination of antioxidant activity with DPPH method showed IC50 of F1 tablet was 215.93 μg/mL, IC50 of F2 tablet was 363.86 μg/mL, whereas IC50 of soursop leaf extract 602.64 μg/mL, IC50 of mangosteen rind extract 111.80 μg/mL and IC50 of Ling Zhi mushroom extract 2199.50 μg/mL. Determination of immunomodulatory activity by in vivo carbon clearance method showed a moderate phagocytic index of 1.385 for F1 tablet, 1.469 for F2 tablet, and <1 for Echinacea extract as positive control. Stability test performed based on the α-mangostin content showed that the shelf-life of F1 tablet was 1.865 years, and F2 tablet was 1.842 years. The result showed that film coated tablet F1 and F2 have both synergistic antioxidant and immunomodulatory activities as compared to their respective single extract. The result showed that the antioxidant activity and stability of film coated F1 tablet is better than F2 tablet, but the imunomodulatory activity of F2 tablet is better than F1 tablet.

Abstract

Ofloxacin is an antibiotic and categorized as class 2 in Biopharmaceutics Classification System (BCS). Ofloxacin solubility is dependent on pH of the solvent due to its amphoteric properties. The objectives of this research was to investigate the effects of pH and ionic strength on solubility profile of ofloxacin. Ofloxacin solubilities were measured at various pH ranged from 1.5 to 12.5 and ionic strengths from 0.01 to 0.20 respectively. Solubility experiments were carried out using shaking thermostatic-waterbath for 3 hours at a temperature of 37°C ± 1°C. Results showed that ofloxacin solubility increased at pH’s below the pKa1 value (6.08) and above the pKa2 value (8.25). However, the solubility decreased at pH’s between pKa1 and pKa2. Changing the solution pH from 6.2 to 1.5 or to 12.5 affected the solubility of ofloxacin increased about 9.96 and 8.64 times higher, respectively. Ofloxacin solubility in various ionic strengths showed that stronger ionic strength solution caused an increase in ofloxacin solubility. This mean that there was a salting in phenomena. Least-Square Fitting Analysis using Henderson-Hasselbalch equation for amphoteric compound was insufficient to describe ofloxacin solubility process in a series of pH.

Abstract

Trypsin is an enzyme which is important for research as well as for the pharmaceutical industry, medicine and health, which is especially developed as a raw material for digestive enzymes. This study aimed to characterize PST from Lactobacillus plantarum FNCC 0270 that have not been studied as a producer of trypsin. Fermentation for producing PST by L . plantarum FNCC 0270 was performed in the LKB fermentor with working volume of 3.5 liters, at temperature of 37 oC, pH value of 8, agitation rate of 77 rpm and aeration of 0.5 vvm. The media composition were 3.64% baker’s yeast, 1.21% glucose, 0.13% skim milk. The fementation resulted in enzyme activities of 0.00129 U/mL and protein content of 0.49 mg/mL. Purification of the PST conducted by ultrafiltration Hollow Fiber Catridge 5 kD, presipitated with ammonium sulfate saturated (30-70%), 30 kD membrane polyesthersulfone, ion exchange column chromatography Q-XL and HiTrap affinity column chromatography. All of system increased purity of the crude PST. SDS-PAGE of PST indicated the molecular weights of 47.35; 38.42;21.40 and 12.96 kD. The PST was stable at pH value of 8 and temperature range of 25-35 oC. It is proved that the same half-life was 693.2 minutes. Km and Vmax values of the PST were 0.231 mM and 0.001 U/mL, respectively using N-α-benzoyl-DL-arginine-p-nitroanilide as the substrate. PST was inhibited by EDTA, Ca2+, Zn2+, Mg2+, Mn2+ and specific substrates. Based on the result of characterization of PST from L. plantarum FNCC 0270 can be concluded that the trypsin–like protease (PST) was trypsin.

Abstract

Isolation of phenolic polymers from plants are challenging because the low yield and time consuming. Synthesis of phenolic polymer is urgently needed to overcome the lack of the compounds. Phenolic oxidation with H2O2 by peroxidase, results oxidative coupling reaction and finally produces phenolic polymers. Peroxidase (e.g guaiacol) catalyzes oxidative reaction by hydrogen peroxide with substrate acted as hydrogen donor. In this study, peroxidase from sawi hijau (Brassica juncea) was isolated. The obtained crude peroxidase had specific activity 5.618U/mg protein. Reaction between guiacol, H2O2 and crude peroxsidase produced guaiacol dimer. The new dimer was identified by NMR spectrometry, and the product showed coupling in para-para of 4,4’-biguaiakol, with 17% yield. In summary, crude peroxidase obtained from sawi hijau (Brassica juncea) was able to act as a catalyst for the oxidative coupling reaction of guiacol into guaicol dimer compounds.

Abstract

The knowledge of teachers on asthma and its managements are very important, in order that teachers can provide the assistance required by the students in primary school who suffered an asthma attack while at school. This study aimed to examine the effect of an educational book to improve knowledge among primary school teachers on asthma and its management. This study was descriptive study, using questionnaires and educational book “Asthma: The Role of Teachers in the Management of Asthma in Schools”. The research was conducted in 3 phases: (1) asses the knowledge of teachers on asthma before being given an educational book, (2) administration of an educational book “Asthma: The Role of Teachers in the Management of Asthma in Schools”, (3) re-asses the knowledge of teachers on asthma after administration an educational book. A total of 48 teachers from three primary schools in the Medan Deli District involved in the study. In general, their knowledge of asthma before being given education is fairly low. After administering education the teacher knowledge increased, this can be seen from the increase in the number of teachers who answered correctly on any given question. An increase of almost 30% of teachers who correctly answer to questions about the causes of asthma after booklet administration and about 20% for questions about symptoms, and treatment of asthma. It can be concluded that the educational materials such as educational book has the effect to increase the knowledge of teachers about asthma and its treatment. It can be seen from the existence of increase in the number of teachers who answered correct on every the question given. This study concluded that the educational book entitle “Asthma: The Role of Teachers in the Management of Asthma in Schools “ has effective to improve knowledge among primary school teachers on asthma and its managements.

Abstract

Various natural compounds have been reported to be used as anticancer, however their modes of action have not been clearly defined. One of the plant compounds is acetogenin which can induce apoptosis in in vitro study. Two new acetogenins discovered in Annona muricata are annomuricin E and muricapentocin. Molecular docking is one of in silico model to screen coumponds based on their mechanism to protein. Therefore, the two compounds were docked using PLANTS 1.1 to see the affinity to proliferation protein, Protein Kinase C. The RMSD calculations of the best predicting binding site gave 1.3906 Å. The molecular docking results of native ligand (07u), annomuricin E, and muricapentocin are -71.161; -127.739; and -116.868. It showed that annomuricin E has the best in silico affinity to Protein Kinase C, and has better ability to inhibit proliferation as compared to the native ligand and muricapentocin.

Abstract

8-Hydroxy 9,12-octadecadienoic acid (8-HODE) is an oxidized aliphatic fatty acid belongs to the group of oxylipins, which shows bioactivity. It was reported previously that Curvularia lunata BioMCC FE-00283, an endophytic fungi from Cibotium barometz be able to produce 8-HODE by submerged fermentation. Glucose, monosodium glutamate, corn oil are media components which showed significant effect toward HODE production by C. lunata BioMCC FE-00283. In this study we reported the enhancement of 8-HODE production by adding potato extract to previouse medium. These four medium components, which were potato extract, glucose, monosodium glutamate and corn oil, were optimized for 8-HODE production by Response Surface Metodology (RSM). A full factorial Central Composite Design (CCD) was used to explain the interaction effect between media components. Maximum HODE production predicted by the quadratic model was 16.46 mg/L where media composition consits of 36.50% potato extract, 3.60 g/L glucose, 15.92 g/L monosodium glutamate and 0.96 ml/L corn oil. Experimentally verification using these medium composition production of 8-HODE reached 16.189 ± 0.526 mg/L. This optimization,which was added potato extract as variabel in medium optimization was enhanced 3 times fold of 8-HODE concentration compare to the 3 variabel optimization.

Abstract

Rhodamine B is a colouring agent for paint industry, textile and paper. It causes irritation at respiration systems and has a carcinogenic effect. At high concentration rhodamine B causes liver damages. Nowadays, it has been reported being added into cosmetics and food product such as lipstick, eye shadow, rouge, food paste, and kerupuk. This research was conducted to determine rhodamine B in the products by using HPLC and spectrophotometry UV-Vis. It was done through sample collection, sample preparation, validation of analytical method, and analysis of rhodamine B using HPLC and spectrophotometry UV-Vis. The results of HPLC and spectrophotometry UV-Vis showed that four samples of kerupuk (57,14%) and four samples of food paste (50%) contained rhodamine B. However, rhodamine B was not found in cosmetics. The results showed the urgency of fast methods to identify rhodamine, thus its content in food products is carefully controlled.
