Abstract

Abstrak: Rutin merupakan senyawa flavonoid yang banyak terdapat pada buah semangka dan kulit dari tanaman jenis jeruk. Rutin memiliki beberapa aktivitas farmakologi, salah satunya sebagai anti artritis reumatoid. Rutin menunjukan absorbsi yang rendah bila diberikan secara peroral karena permeasi yang rendah serta dapat mengalami degradasi di dalam saluran cerna sehingga sebagai alternatif rutin diberikan dalam bentuk transdermal. Transdermal merupakan rute pemberian obat yang mendukung transport obat dari permukaan kulit sampai ke sirkulasi sitemik. Rute pemberian ini dapat mencegah degradasi rutin dalam saluran cerna.Transfersom merupakan salah satu sistem pembawa untuk transdermal yang mampu meningkatkan efektivitas penghantaran obat. Penelitian ini bertujuan untuk melakukan uji penetrasi in-vitro dan aktivitas anti artritis reumatoid dari gel yang mengandung transfersom rutin dan gel rutin non transfersom. Uji penetrasi in-vitro menujukan penetrasi rutin dari sediaan gel transfersom sebesar 14,33% sedangkan untuk gel non transfersom sebesar 9,51%. Aktivitas anti atritis reumatoid diamati selama 28 hari dengan menghitung persen penghambatan udem pada kaki tikus. Hasil menunjukan bahwa persen penghambatan volume udem dari gel transfersom sebesar 57,82% sedangkan gel non transfersom sebesar 45,63%.

Abstract

Green betle leaf (Piper betle L.) and red betle leaf (Piper cf. fragile Benth.) were empirically used for treatment such as anti-microbial, anti-diabetic, antiseptic and resolve nosebleeds and have antioxidant activity. Phytochemical screening and antioxidant activity test have been carried out on (water and ethanol 70%) extracts of both green and red betle leaf. Phytochemical screening showed that both water and ethanol 70% extracts of both green and red betle leaf contain secondary metabolite compounds, flavonoid, alkaloid, tannin, steroid and triterpenoid, the presence of which may produce the antioxidant activity. The DPPH free radical inhibition method showed that antioxidant activity of water extract of both green betle and red betle (IC50) were 36.02 μg/mL and 60.35 μg/mL, whereas the antioxidant activity of 70% ethanol extract (IC50) were 10.59 μg/mL and 28.05 μg/mL, respectively. The highest antioxidant activity was in 70% ethanol extract of green betle (IC50 = 10.59 μg/mL), but was lower than the vitamin C (IC50 = 7.39 μg/mL) or quercetin (IC50 = 2.89 μg/mL) as positive controls. The statistical tests using twowaysanalysis of variance followed by Tukey test showed that there are significant differences between the green and red betle’s, and between water and ethanol 70% extratcs as indicated by the P value < α (α = 0.05).

Abstract

Syzygium campanulatum and Syzygium aromaticum contains antioxidant components suchas flavonoids, phenolic, and terpenoids. May have hepatoprotective properties in reducing SGPT and SGOT activity. This research wants to determine the potency of hepatoprotective of ethanolic extract of Syzygium campanulatum (Korth) and Syzygium aromaticum leaf compared with curcuma tablets. This research uses 24 male Wistar rats divided into 6 groups: I, II, III (as a normal, induction, and compared control), group IV, V, VI were treated 105, 210, and 420 mg/kg BW respectively. The study was conducted for 9 days. After 7 days of treatment, treated groups were exposed by hepatotoxic dose of paracetamol (2000 mg/kg BW). The SGPT and SGOT activity of all groups was measured by enzimatic assay. The result can be concluded that Syzygium campanulatum extract was found to be active as hepatoprotective agent with 210 mg/kg BW dosage (SGPT 21.76 ± 3.98 U/L and SGOT 7.32±6.74U/L) as eff ective as with the curcuma tablets (SGPT 23.91 ± 4.41 U/L and SGOT 14.12±5.37 U/L) and the hepatoprotective activity of Syzygium campanulatum extract at a dosage 420 mg/kg BW better than curcuma tablets (SGPT 12.43 ± 6.51 U/L and SGOT 6.64 ± 5.88 U/L). While the hepatoprotec Syzygium campanulatum and Syzygium aromaticum contains antioxidant components such as flavonoids, phenolic, and terpenoids.May have hepatoprotective properties in reducing SGPT and SGOT activity. Abstract

Catechin is one of bioactive compounds in gambier. Result of preliminary studies showed that catechin is active as an antioxidant and antibacterial, thus it was potential as an oral nutraceutical for prevention of dental plaque. In present study, preparation of the oral nutraceutical in the form of mouthwash was performed by variating content of catechin in the extract as formulation parameter. The contents of catechin in the mouthwash solution are 0.01-0.1% (w/v). The mouthwash products were analyzed for pH, antioxidant and antibacterial activities, also particle size. The results showed that mouthwash with catechin content of 0.01-0.08% (w/v) produced the nanoscale of the particle size of mouthwash.

Abstract

Off-label is a term used in which a drug prescribed outside the official information of the marketing authorization. Off-label prescribing may occur as the result of several factors including lack of clinical trials data involving pediatrics and suitable formulations for medicines commonly prescribed to this fragile population. Objective: This study aimed to estimate the nature and prevalence of off-label prescribing in pediatric inpatients with pneumonia. Material and Methods: a retrospective study was conducted in a study hospital using medical records from pediatric inpatients with pneumonia during the period of January-December 2015. Patient and prescribing data were collected, and drugs were classified as on-label or off-label based on the Indonesia National Drug Information (IONI) and British National Formulary for Children (BNFC). Thereafter, off-label drugs were categorized with a hierarchical system of age, indication, route of administration and dosage. Results: There were 1141 drugs with 77 different types of drug were administered to 207 patient during the study period. The data uncovered that 405 (35,5%) of the drug prescriptions were used off-label based on IONI, and 319 (28%) of the drug were used off-label based on BNFC. Based on IONI and BNFC, most off-label drugs were from anti infection drugs. Conclusion: The prevalence of off-label use in pediatric inpatients with pneumonia is not high. The off-label prescribing may not be necessarily be considered irrational, yet this fact reveals that the use of drugs does not comply with the drug label. Clinical trials for pediatric drugs are essential to provide complete product information for pediatric use.

Abstract

Chemotherapy is one of the breast cancer treatments. However, there are several adverse effects with mild to high severity related to this treatment. This study was made to evaluate adverse effects which occurred in breast cancer patients who are still in their chemotherapy cycle. The evaluation consists of assessment of adverse effects associated with chemotherapy regimens and its dosage, causality assessment, and preventability assessment. This study was done from December 2012 until March 2013 concurrently. The data was collected directly from the patients by doing the interview. The data was also collected from the patient’s medical record and nurse’s daily documentation. Clinically, FAC caused more adverse effects than other chemotherapy regimens. However, in the 6% of excess dosage, there were no different acute adverse effects. The most occurred adverse effect was alopecia with 81% percentage. Causality assessment with probability scale Naranjo and WHO criteria showed that 97.3% and 99.2% were ‘probable’. The preventability assessment which based on Thornton- Schumock criteria showed that only nausea and vomit which were ‘definitely preventable’.

Abstract

Resistance to malarial infection is dependent on the development of an immune response by the host and to a varying extend, a certain innate characteristics possessing protective value against infection. Innate resistance is not always absolute and genetically determined factors, but instead can act to reduce the severity of infection and improve survive. Mechanism of action resistance to malarial infection will be discussed in this article.

Abstract

Gamma irradiation has been used for preservation of herbal medicines, one of which is temu putih Curcuma zedoaria (Berg) Rosc. to cure cervical cancer. This research aimed to study the gamma irradiation effects on microbial contamination, cytotoxic activity against leukemia L1210 cell lines and chromatogram profile of the temu putih active fraction. Gamma irradiation was performed with a 60Co source at 5, 7.5, 10 and 15 kGy. After irradiation at > 7.5 kGy bacteria and mould were not present on herbs. Control and irradiated samples were consecutively macerated with n-hexane, ethyl acetate and ethanol. Ethyl acetate extract was the most cytotoxic gainst L1210 leukemia cells with an IC50 value of 10.60 μg/mL as compared to n-hexane or ethanol extract. Ethyl acetate extract was fractionated with column chromatography producing six fractions. Fraction 2 was the most cytotoxic against L1210 leukemia cells with an IC50 value of 2.44 μg/mL. The thin-layer chromatographic analysis results of ethyl acetate extract and fraction 2 of control and irradiated samples showed the presence of curcumin and chromatographic pattern similar to the control. Curcumin contents in fraction 2 were between 35 to 51%, which were not significantly changed between samples, although was irradiated up to 15 kGy. The maximum dose for irradiation of temu putih herbal preparation is 15kGy.

Abstract

Pada penelitian ini, kelopak rosela dikeringkan kemudian dimaserasi dengan etanol 70% - HCl (99:1) kemudian difraksinasi berturut-turut menggunakan n-heksana, etil asetat, n-butanol dan air. Semua fraksi diuji aktivitas antibakteri terhadap S. aureus dan E.coli dengan menggunakan metode difusi, aktivitas antibiofilm dan aktivitas antikuorum sensing terhadap Pseudomonas aeruginosa. Fraksi etil asetat adalah fraksi yang paling aktif terhadap S. aureus menghasilkan daerah hambatan pertumbuhan pada konsentrasi 10% dan 20% sebesar 15,89 ± 0,37 mm dan 15,93 ± 0,72 mm, sedangkan terhadap E. coli sebesar 17,25 ± 0,86 mm dan 17,35 ± 0,48 mm pada konsentrasi 10% dan 20%. Pada uji aktivitas antibiofilm didapat % penghambatan pada S. aureus sebesar 62,05% ± 17,83 dan pada E.coli sebesar 11,11% ± 23,13, uji aktivitas antikuorum sensing memberikan daerah hambatan sebesar 19,23 ± 1,52 mm dan 20,89 ± 2,35 mm pada konsentrasi 10% dan 20%. Fraksi etil asetat dilakukan isolasi terhadap satu senyawa aktif dengan menggunakan metode VLC dan KLT preparatif kemudian diidentifikasi strukturnya dengan menggunakan spektrofotometer 1H-NMR, 13C-NMR, COSY-NMR, HMBC dan LCMS.
