Abstract

Cancer therapy using chemotherapeutic agents associated with many adversed side effects, including immunosupressant. The use of immunostimulator together with chemotherapeutic agent (co-chemotherapy) can be the alternative method to solve that problem. Ganoderma lucidum (Ling Zhi) has been reported to have immunostimulatory activity. The aim of this research was to evaluate the immunostimulatory activity of water extract of G. Zucidum by determining the relative CD8+ T lymphocyte cell percentage in rats induced by doxorubicin. Extraction of plant material was carried out by infusion method. Sprague Dawley female rats were divided into six groups, they were doxorubicin as control, commercial product as comparing control, 100 mg/kgBW and 450 mg/kgBW extract treatment, extract control, and without treatment control. Relative CD8+ T lymphocyte cell percentages of blood samples were obtained by flow cytometry by using Multiset program. The data were analyzed statistically using paired sample t test and one way ANOVA continued by Post Hoc test. The result showed that the water extract of G. lucidum increased the relative CD8+ T lymphocyte cell percentage in rats induced by doxorubicin. The water extract of G. lucidum is promising to be developed as co-chemotherapy immunostimulatory agent.

Abstract

Infeksi Ganoderma pada tanaman kelapa sawit merupakan permasalahan utama dalam pengendalian penyakit pada perkebunan kelapa sawit di Indonesia. Sampai saat ini infeksi Ganoderma baru diketahui setelah muncul tubuh buahnya pada batang tanaman kelapa sawit, dan pada saat itu tanaman kelapa sawit sudah tak bisa dipertahankan lagi dan tingkat penyebarannya sudah meluas. Penelitian ini bertujuan untuk mengidentifikasi spesies Ganoderma yang banyak menyerang tanaman kelapa sawit di Indonesia baik dari segi morfologinya maupun dengan pendekatan biologi molekuler, yaitu berdasarkan pada peta genetik (sequence) 18S rRNA dan ITS (Internal Transcribe Spacer). Hasil penelitian menunjukkan bahwa Ganoderma yang banyak menyerang tanaman kelapa sawit di Indonesia adalah Ganoderma boninense. Dari hasil karakterisasi telah didapatkan peta genetik spesifik (specific sequences) ITS dari Ganoderma boninense dibandingkan dengan spesies Ganoderma yang lain, sehingga peta genetik spesifik ini dapat dijadikan dasar untuk mendeteksi secara dini adanya serangan Ganoderma boninense pada areal perkebunan kelapa sawit di Indonesia. Pendeteksian secara dini terhadap serangan Ganoderma ini akan sangat bermanfaat untuk menentukan proses penanganannya dan pada akhirnya diharapkan akan dapat meminimalisasi tingkat kerugian yang terjadi (1).

Abstract

Symptoms of Acquired Immuno Deficiency Syindrome (AIDS) is a militia or disease caused by me infection of Human Immunodeficiency immune deficiency due to Antiretroviral. Due to Virus (HIV). The therapy used is a drug that the use of Antretroviral is the appearance of unwanted reactions or side effects will appear. The objectives of the study were to analyze the side effects of antiretroviral therapy in HIV / AIDS patients in RSPI Sulianti Saroso, North Jakarta, January-June 2016 period. The sample was 95 patients. The research was done by retrospective method, data analysis desknptif data presented in tabular form and presented with pie chart. The result of the research is the combination of TDF + 3TC + Efavirenz Tenofovir-Lamivudin-Efavirenz) as much as 73,68% (0 person). There were 94.73% (90 people had mild adverse reactions and 5.27% (5 people) severe side effects.

Abstract

Aloe vera leaf contains hydroxyanthraquinone derivatives, aloin (barbaloin and the distereoismeric 10-C glucosides of aloe-emodin anthracene) and 7-hydroxyaloin isomer and to a lesser extent aloe emodin, chrysophanol, chromone derivative and aloeresin as major glycoside-anthracene derivative. The aloin content varies in different species of aloe. The main constituent of aloin is barbaloin which is a pale yellow crystalline glycoside, soluble in water. Recent studies confirm that aloe has an antibacterial (against Staphylococcus ssp. and Streptococcus ssp.) and a fungicidal activity, so it can be used as antiseptic agent. The main purpose of this research was to obtain a physically stable liquid soap containing aloe vera. The formulation contained sodium lauryl ether sulphate with various concentrations. The obtained liquid bath soaps were evaluated cencernng their stability i.e.: viscosity, surface tension, and pH for eight weeks. Each formula gave a viscosity in the range of 8100 to 10400 cps and showed a pseudoplastic flow.

Abstract

Previous study reported that Averrhoa bilimbi fruits had a potency as a natural diuretic. This research was conducted to examine the effects of ethanolic extract of Averrhoa bilimbi fruits on diuretic activity, urine sodium and potassium concentrations, and pH of urine male rats Sparague-Dawley. Fifteen rats were divided into 5 groups and 3 replications. Group 1 was given saline (negative control), group 2 was given furosemide 20 mg/kg bw (positive control), group 3 was given extract of Averrhoa bilimbi 0.44 g/kg bw, group 4 was given extract of Averrhoa bilimbi 0.88 g/kg bw, group 5 was given extract of Averhoa bilimbi 1.75 g/kg bw. Averrhoa bilimbi extract, saline, and furosemide were administred orally. The diuretic activity was determined by measuring cumulative urine volume for 10 hours post administration. The result showed that the extract of Averrhoa bilimbi at dosages of 0.88 and 1.75 g/kg bw increased sodium and potassium excretion by 18-36% and 22-27%, respectively. On the other hand, the increased diuretic activity and sodium and potassium excretion dramatically decreased urine pH. It was concluded that extract of Averrhoa bilimbi at dosages of 0.88 and 1.75 g/kg bw increased diuretic activity with enchanced sodium and potassium excretion in urine. The extract of Averrhoa bilimbi cosisted of alkaloid, flavonoid, and saporin that were assumed to have synergic activities as natural diuretics.

Abstract

Merancang obat baru dapat dilakukan melalui modifikasi struktur yaitu dengan mengubah gugus tersubstitusi yang menyebabkan perubahan sifat fisikokimia, farmakokinetik, toksisitas dan aktivitas masing-masing senyawa. Perubahan tersebut dapat diprediksi melalui uji in silico. Penelitian ini bertujuan untuk memprediksi sifat fisikokimia, proses farmakokinetik (ADME), toksisitas dan aktivitas sitotoksik dari 23 senyawa turunan N-benzoil-N’-(4-fluorofenil)tiourea sebagai calon obat anti kanker. Uji in silico dilakukan dengan cara doking senyawa yang akan diprediksi aktivitasnya dengan enzim target SIRT1 kode pdb. 4I5I. Hasil doking berupa energi ikatan yang digambarkan dengan nilai Rerank Score (RS), dengan menggunakan program Molegro Virtual Docker. Senyawa dengan nilai RS kecil diprediksi mempunyai aktivitas yang besar. Hasil uji in silico menggunakan program pkCSM dan Protox online tool dapat disimpulkan bahwa sebagian besar turunan N-benzoil-N’-(4-fluorofenil)tiourea mempunyai sifat farmakokinetik yang baik, menimbulkan toksisitas yang relatif rendah dan mempunyai aktivitas sitotoksik yang lebih besar dari ligan pembanding 4I5_601, dan senyawa N-(4-fenilazobenzoil)-N’-(4-fluorofenil)tiourea merupakan senyawa yang diprediksi mempunyai aktivitas sitotoksik paling besar.

Abstract

Hydroxyl octadecadienoic acid (HODE) is an oxidized aliphatic fatty acid from the oxylipins group, which shows bioactivity and can be produced by Curvularia lunata BioMCC FE-00283 by submerged fermentation. Glucose, monosodium glutamate, and corn oil were media components that showed significant effect toward HODE production by C. lunata BioMCC FE-00283. Composition optimization of these three medium components was carried out by Response surface methodology (RSM). A full factorial central composite design (CCD) was used to explain the interaction effect between media components. Maximum HODE production as predicted by the quadratic model have chromatogram area of 4540000, which was verified experimentally to be 1091338 ± 152489. Eventhough experimental veriiication showed lower result compared to the model prediction, however it was higher compared to the HODE production using basal media with unoptimized composition of carbon, nitrogen, and inducer which only have chromatogram area of 295045 ± 71200. The verification also showed that optimization increased HODE production by 3.7 fold compared to that produced by the unoptimized media.

Abstract

To search for scientific evidence the efficacy of herbal medicine, has conducted clinical trial the efficacy of hypertension herbs compared with hydrochlorotiazide. Efficacy of clinical trials were conducted with the design open-label, randomized clinical trials and parallel design. It was involving 80 subjects who have met the inclusion and exclusion criteria. Subjects randomized study that is divided into two groups, a herbs group and hydrochlorotiazide group. Subjects performed once a week to check the complaint history, the development of clinical symptoms, the possibility of side effects and diagnostic physical examination. Assessment score of quality of life by questionnaires Short Form 36 (SF-36) every four weeks. Results showed that administration of herbal medicine for 56 days hypertension, lower blood pressure (systolic and diastolikic) is equivalent to hydrochlorotiazide (HCT). Lowering blood pressure to normal (normotensive) 62.7% of the subjects. Eliminating the clinical symptoms of hypertension (dizziness / headache, stiff neck / whiny and myalgia) herbs group subjects at almost the same time with the hidrochlorthiazide group. Raising the quality of life scores (SF-36) subjects herbs group equivalent to the increase in quality of life scores (SF-36) hydrochlorotiazide group.

Abstract

Assay for secondary metabolite of endophytic fungi from Cassia siamea Lamk.. (Johar Plant) has been investigated. Endophyitic fungi are microorganisms that live asymtomatically within the living tissue of host plants. Endophytic fungi associate with plant, they can help the metabolism of host plant and produce potensial secondary metabolite. The purpose of this experiment were to investigate endophytic microbial agents. Isolate of endophytic fungi from twig of Cassia siamea Lamk., using direct seed method was carried out. The microbial activity was carried out with dilution method using paper disk and glass cylinder. The results showed 3 isolates of bacteria and 7 isolates of endophtyic fungi. The metabolite from three isolates of bacteria had the anti-microbial activity more poten toward Gram positive than Gram negative and four isolates of fungi had anti-Aspergillus agent.

Abstract

Binahong (Anredera cordi olia (Ten) Steenis) has been used as wound healing in traditional Indonesian medicine. The aim of this study was to examine the wound healing activity and irritation potency of ethanolic extract of Anredera cordifolia (Ten) Steenis. Dry Binahong leaves were ground and screened with mesh no 40. Dry Binahong leaves were macerated with 10 times of volume of 96% ethanol for 90 minutes with constant agitation (200 rpm). It was screened and concentrated until the liquid would be a quarter. Then the extract was incorporated to gel base as 2,5%; 5%; 10% and 20%. Incision wound method was employed to activity test of wound healing. Male mice (strain Swiss), weight between 25 ~ 30 g, was used to this test. The male mice were classified into 7 groups i.e. negative control, control vehicle, positive control and 4 groups of sample. The extract then was assessed to irritation potency using HET-CAM method. Lactic acid was used as the positive control for irritation assessment. The result of this study stated that the highest activity of wound healing was shown by the sample containing 5% of ethanolic extract. The percentage activity of wound healing was 63,3%. Irritation appeared on the 10% and 20% ethanolic extract of binahong leaves.
