Abstract

Paracetamol is a well known and commonly used analgesic-antipyretical agent. However, it exhibits hepatotoxic side effect if used for the long term or using exessive dose (10-15 g for single dose). A new compound of 1.3 bis (p-hydroxyphenyl)urea (code-name as HP2009) is an analgesic agent, and it is predicted that its hepatotoxic side effect is lower than that of paracetamol. The compound HP2009 was succesfully synthesized. The result showed that by using molar ratios of p-aminophenol and urea 2:9.5, pH 1 for reaction condition, refluxing for about 1 hr and evaporating time was set up for 30 minutes, the yield of HP 2009 will be 99.49%. The crystals obtained was characterized using spectroscopic methods, and showed undoubtedly that the product was 1.3 bis(p-hydroxyphenyl)urea.

Abstract

Effects of ratios of ethanol-water as extracting solvent in obtaining extractable material, phenolic compounds and antioxidant activity from the herb of Phyllanthus niruri Linn. have been investigated. The ratios tested were 100:0, 80:20, 70:30, 60:40 and 50:50. The results revealed that the ratio of ethanol-Water used gave significant effect on extractable material, phenolic compounds and antioxidant activity (p<0.05). Among the ratios tested, the best result was obtained by ethanol-water of 60:40 as extracting solvent for the herb of Phyllanthus niruri Linn. to obtain phenolic compound which has antioxidant activity.

Abstract

An investigation on the quality service at the Pharmacy Department of Cilacap Local Government Hospital (CLGH) and Mohammadiyah Hospital in Karanganyar (MHK) which covered five service dimensions, i.e. reliability, responsiveness, assurance, empathy and tangible factors, has been conducted, The quality of services was analyzes by using the SERVQUAL gap model. The study Was a non-experimental descriptive research. The data were collected using purposive sampling with questionnaires as the research tools. The results showed that in the pharmacy departments, of both CLGH and MHK), the above-mentioned pharmacy service dimensions observed performed below customer expectations. The highest gap score in CLGH is on responsiveness followed by tangible factors, reliability, empathy and assurance. The biggest gap score in MHK is on tangible factors, followed by empathy, assurance, responsiveness and reliability, Loyalty of customer to pharmacy department of CLGH for regular patient is around 50,6%, MHK 39,5%, neutral customer 35,1%, and non-loyal customer 25,4%. The results indicated that both hospitals need to strengthen quality services of their pharmacy departments, particularly on service dimensions which have high score gap.

Abstract

A 6-week longitudinal prospective study was conducted to assess the effectiveness and the safety antibiotic used in chronic kidney disease (CKD) patients in internal medicine ward. We compared white blood count and glomerular fi ltration rate before and after antibiotic used. The CKD patients who admitted in the internal medicine ward and age ≥18 years old were included this study. Patients with incomplete laboratory data and renal replacement therapy were excluded in this study. The 25 patients who enrolled in this study were recruited. The majority gender of CKD was male (64%), the mean of age was 61.52±14.17 years old with length of stay (LOS) was 6.92±4.05 days. The highest number of patients was in CKD stage 3 (n=10, 40%) and was followed by CKD stage 2 (n=6, 24%). Most of them were diagnosed community acquired pneumonia. Tablet azithromycin (n=16, 64%) then Cefotaxime intra venous injection (IV) (n= 6, 24%), and Ceftazidime IV (n=5, 20%), Cloxacillin IV (n=4, 16%) were the most antibiotics prescribed. Generally patients had been prescribed appropriate dose of antibiotic and 88% of them showed improved white blood count. In contrast, the glomerular fi ltration rate of 44% CKD patients was getting worse. In conclusion, this study clearly indicate the CKD patients require close monitoring to maintenance of renal function even the antibiotic had been prescribed appropriately.

Abstract

Xylanase enzyme activity of endophytic fungi from Brucea javanica (L.) Merr has been done. Fruit, leave and stem of the plant collected from Bogor, Cianjur, and Tawangmangu which used in this study. To evaluate the enzyme activity the endophytic fungi isolate were inoculated on PDY medium and fermented at room temperature, shake at 150 rpm for 14 days. The supernatan were used to examine the extracellular enzyme and the biomass for intracellular enzyme. The DNS methods was used to test the enzyme activity. The results showed isolate 1.1.8; 1.2.17; 1.3.16 and 1.3.6 produced intra and extracellular xylanase enzyme. Isolate 1.1.6 produced only extracellular enzyme while isolate 1.2.2 did not produce both intra and extracellular enzyme.

Abstract

The aims of the research are to determine the Significant effect factor among mixmg rate, mixing time and their interaction on the physical properties and stability of green tea emulgel and to determine the optimum composition area which can produce good emulgel. In our previous investigation was determined the optimum composition between emulsifying agent (Tween 80 and Span 80) and gelling agent (Carbopol) This research determined the optimum mixing process, particularly on mixing rate and mixing time usig the factorial design with two factors (mixing rate and mixing time) and two levels (high level and low level). The physical properties and stability parameter investigated were including viscosity, spreading ability and viscosity shift atter storage at predetermined temperature and humidity for a month. The results showed that mixing rate Significantly influence on the viscosity and spreading ability and the mixing time significantly influence on the Viscosity shift. The optimum composttion area was not produced because the factorial design equation for viscosity respond and viscosity shin were not significant so can not be used to predict responds.

Abstract

To design new drugs, physical-chemical characteristics of drug molecules can be predicted by in silico test before drugs are synthesized. Ribonucleotide reductase is the main target or receptor of antineoplastic compounds such as hydroxyurea (HU) and its derivatives like 1-(4-trifluoromethylbenzoyloxy)urea or 4-CF3BOU. This compound forms a complex with crystal structure of ribonucleotide reductase I enzyme, which is 2EUD. The hydrogen bond and bond energy in the form of rerank score from both complexes was calculated with Molegro program. Theoretically, compound activity is indicated by rerank score. The compound whose rerank score is small is predicted to have greater activity. The activity of 4-CF3BOU was found to be greater than HU. The reaction mechanism of synthesis 4-CF3BOU was the substitution of nucleophilic hydroxyl group from HU to carbonyl group of 4–trifluoromethylbenzoyl chloride (4-CF3BCl). Purity test was conducted using TLC and melting point. Structure identification was performed based on the spectra of UV-VIS, FT-IR, H/C-NMR and MS. In this study, 4-CF3BOU was discovered to have antineoplastic activity with the IC50 value of 82.37 μg/mL and was tested towards HeLa cells. On the other hand, HU had the IC50 value of 430.21 μg/mL. The antineoplastic activity of 4-CF3BOU was greater than HU.

Abstract

Secara empiris penggunaan daun senggani mempunyai kemampuan sebagai anti-inflamasi dan analgesik. Penelitian ini bertujuan menguji efek anti-inflamasi dan analgesik dari infusa daun senggani. Pada uji anti-inflamasi menggunakan metode Winter, tikus dibagi menjadi 5 kelompok yaitu kelompok kontrol negatif diberi air suling, kontrol positif diberi natrium diklofenak 1,8 mg/200 g bb, kelompok uji diberi infusa daun senggani 2,8 mL/200 g bb dengan konsentrasi 10%, 20% dan 40%. Setelah 30 menit diinduksi karagenan 1%, 0,2 mL/200 g bb. Pada pengujian analgesik menggunakan metode Sigmund, mencit dibagi menjadi 5 kelompok yaitu kelompok kontrol negatif diberi air suling, kontrol positif diberi natrium diklofenak 0,26 mg/20 g bb, kelompok uji diberi infusa daun senggani 0,4 mL/20 g bb dengan konsentrasi 10%, 20% dan 40%. Setelah 30 menit diinduksi asam asetat 3%, 0,2 mL/20 g bb. Dari hasil ANOVA satu arah infusa daun senggani dengan dosis 2,8 mL/200 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek sebagai anti-inflamasi dan infusa daun senggani dosis 0,4 mL/20 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek sebagai analgesik. Infusa daun senggani dengan dosis 2,8 mL/200 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek anti-inflamasi yang lebih rendah dibandingkan natrium diklofenak dan infusa daun senggani dosis 0,4 mL/20 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek analgesik yang lebih rendah dibandingkan natrium diklofenak. Peningkatan dosis infusa daun senggani tidak memberikan peningkatan efek sebagai anti-inflamasi maupun analgesik.

Abstract

The synergic effects of n-butanolic fraction of secondary metabolite of endophytic fungus 1.3.11 (FB) and doxorubicin (Dox) on cell cycle regulation and the expression of Bcl-2 gene expression were investigated on MCF-7 and T47-D cells by iiow cytometry and immunocytochemical techniques respectively. The results showed that after 12 hours of incubation period with FB at its IC50 dose, MCF-7 cell cycle was inhibited at G1 phase while Dox inhibited the cell cycle at G2/M phase. Similar results were observed in T47-D cells when incubated with Dox and FB individually under the same treatment condition. Further treatment was then performed to these cells where both Dox and FB were combined at their IC50 and lC50 dose and added to incubate with the cells over 12 hours period. Interestingly, the modified treatment combination showed that MCF-7 and T47-D cell cycle regulation were inhibited at G2/M phase. Our immunocytochemical study also showed no significant inhibition suppression of Bel-2 gene expression in both MCF-7 and T47-D cells when compared with their corresponding positive control after treatment with FB and Dox or both combined FB and Dox at 1C50 and IC50 dosage over 15 hours incubation.

Abstract

Listeria monocytogenes is a Gram positive rod-shape bacteria. Other foodborne pathogens, mostly, may cause typical symptoms of gastroenteritis. Unlike these microorganisms, Listeria monocytogenes infection may result in more serious symptoms, usually take in the form of meningitis or septicaemia. People with vulnerable immune system are in a bigger risk of being exposed. Furthermore, the bacterium is also severer than any other bacteria found in the food, with more resistant to heat, drying, or salty environment. This research was purposed to identify and characterize a sample named LM 25722248, which was isolated from fresh food factory in a city of United Kingdom. The isolate was characterized as a Gram positive bacterium, showed tumbling motility, and gave positive result for catalase test and haemolysin assay. Moreover, benefiting from molecular technology, it was possible to reveal that the isolate was Listeria monocytogenes.
