Abstract

The high frequency of common cold self medication using amoxycillin should be given serious attention, while the percentage of its use should be known. Investigation was carried out by a cross sectional survey study using questionaire as primary data (containing indication, dosage, and duration of therapy) in several pharmacies in Banyumas and Purbalingga during four months since December 2006 until March 2007. Investigation was carried out using 85 samples, 69 addult (81 %) and 16 children (19 %). Rational use of amoxycillin consisted of 61 samples (71,76 % ) and irrational use 24 samples (28,24 %). Irrational use of amoxycillin was caused by irrational indication, but 100% rational use were observed based on dosage and duration of therapy.

Abstract

Streptococcus mutans is one of the microbes which is responsible for the cause of bad breath. The activity of Kalanchoe pinnata (Lamk.) Pers. leaves ethanol extract is four times stronger than the water extract in inhibiting the growth of Streptococcus mutans. The aim of research is to isolate and characterize an antibacterial compound of ethanol extract of Kalanchoe pinnata (Lamk.) Pers. leaves. The extract was fractionated using column chromatography. The fractions were purified by TLC and the isolate were tested for its purity using HPLC. Isolate was characterized using UV-Vis spectrophotometry and FTIR. Fractionation by column chromatography generated 9 fractions. The fraction VII have the highest diameter of inhibitory against Streptococcus mutans. The yield of isolate was 9.18% of the fraction VII or 0.84% of the ethanol extract with purity of 95%. Isolate was suspected as a group of saponin compound.

Abstract

Ikatan obat dengan protein dalam darah umumnya tidak mempengaruhi efek terapinya. Selama tidak ada pergeseran ikatan, akibat obat lain yang afinitasnya lebih besar terhadap protein atau perubahan kondisi pasien. Terjadinya pergeseran ikatan yang mengakibatkan peningkatan konsentrasi obat tidak terikat juga akan dikompensasi dengan peningkatan volume distribusi sehingga konsentrasi obat tidak terikat kembali normal. Akan tetapi dalam prosedur monitoring obat yang ditetapkan adalah konsentrasi oba total, sehingga salah interprestasi bisa terjadi.

Abstract

Diabetes mellitus (DM) is a chronic disorder metabolism of glucose in the body. Mahogany seeds (Swietenia mahagoni (L.) Jacq.) reportedly contains fl avonoids, alkaloids and saponins which can be used as therapy of diabetes mellitus. This study aimed to determine the eff ects of antihyperglycemic combination of mahogany seed oil-glibenclamide in male rats hyperglycemiced by alloxan induction. The mahogany seed oil liquefaction processed by using hydrolic pressed instrument 100 psi pressure. This study using Wistar strain male rats aged 2-3 months as many as 35 heads which were divided into 7 groups: group I (normal control) with distilled water added, while the II-VII group with alloxan induced intraperitoneal 140 mg/kg body weight two times. Group II as a negative control was added 0.5% CMC, group III was added glibenclamide 0.09 mg/200 g BB, IV groups oiled mahogany seeds 2 mL/200 g BW, group V, VI, and VII were added a combination of mahogany seed oil glibenclamide with each ratio (75%: 25%, 50%: 50%, 75%: 25%). The distribution of the test preparation 1 times a day for 12 days and blood glucose measurement performed on days 3, 6, 9, and 12. Data has been analyzed by oneway ANOVA (signifi cant p <0.05). The results obtained the combination of mahogany seed oil-glibenclamide provides eff ect to reduction in blood glucose levels induced male rats alloxan, the combination group mahoni-glibenclamide seed oil have equal activity in lowering blood glucose levels with a single chemical drug glibenclamide. Mahogany seed oil is potentially antidiabetic.

Abstract

HIV/AIDS has become a pandemic disease Which hits the Whole World including Indonesia. Antiretroviral (ARV) drugs are drugs pursuing HIV replication. A Government Hospital in South Jakarta is among hospitals that accept training on Voluntary Counseling and Testing (VCT) by the Department of Health, Republic of Indonesia, and has been a reference hospital for the medication of HIV/AIDS and its associated opportunistic infection in people With HIV/AIDS. This epidemiologic surveillance was conducted to find out proportion number, Case Fatality Rate (CFR), incidence rate and prevalence rate of all incidents related to HIV/AIDS. Data collection for people with HIV/AIDS was done through drug counseling cards, medical records and the laboratory examination results, and data analysis Was performed using Stata 7.0 software. Antiretroviral diugs often used in the Government Hospital in South Jakarta were Duviral-Neviral, Duviral-Evafir and Hiviral-Stavudin-Evahr. The results of this study show that the goals of the surveilance conducted can be achieved except for the number of incidence. Among 397 patients suspected to have HIV/AIDS, 324 patients had risk factor of HIV/AIDS, 240 patients were diagnosed positive HIV 290 patients Were severed with opportunistic infections, and 104 patients died.

Abstract

The use of antiseptic hand gel with regards to paradigm that is healthy as well as clean practical, as an attempt to avoid the onset of infection. The volatile oil of Alpinia galanga rhizomes are known active as antibacterial. Three formulation of gels containing of 1 mL of galangal oil were made with variations of carbopol 940 from 0.5% - 2 %. Antiseptic test were done using replica methods. Data of viscosity, spread power, and adhesion were analyzed using one way ANAVA. Data of effectiveness was analyzed using one way ANAVA and if there is any differences, it will be continued by using HSD Tukey. The results showed that viscosity of gels were pseudoplastic flows. Spread power of gel preparations increased, adhesion and viscosity decreased during storage. All gel preparation showed the effectiveness of antiseptic gels because be able to reduce the number of bacterial colonies. Effectiveness of antiseptic formula that contains of carbopol 0.5% can reduce the number of colonies 70% and did not differ from the positive control which can reduce the number of colonies up to 79.37%.

Abstract

One mechanism of whitening agent is to inhibit the tyrosinase enzyme in melanogenesis process. However, since the human tyrosinase has not been determinated experimentally, therefore research on human tyrosinase can not be conducted. Homology modeling is one method that can be used to exhibit the three-dimensional structure of human tyrosinase. This model used to predict the interaction of bengkoang (Pachyrrhizus erosus) active compounds with the human tyrosinase enzyme by using a molecular docking approach (pose analysis). In addition, the strength of the bond between the ligand compounds and the site protein target can be predicted by analyzing the resulting score. Homology modeling was carried out using Amber99 method of software MOE 2009.10 and Bacillus megaterium (TyrBm) as a template. The model obtained was evaluated using Ramachandran plot analysis, z-score, and the plot energy ProSA. The best model used for molecular docking using the active site ligand, ALPHA PMI placement method, and affinity DG scoring of MOE 2009.10 software. The best model is the TyrHSM (MOE default alignment) with 96.7% residues are in allowed regions of the Ramachandran plot, has the least positive region, and has a z-score ratio> 1. The isolated compound 2-butoxy-2,5-bis (hydroxymethyl)-tetrahydrofurane-3,4-diol is predicted have the best affinity to human tyrosinase with a score -0.3598 kcal/mol.

Abstract

Four alkaloids, brucine, brucin N-oxide, colobrine N-oxide, and strychine N-oxide were isolated from the bark of Strychnos ligustrida Bl. (Loganiaceae). Their structures were determined by spectroscopic data analysis, in particular by the Nuclear Magnet Resonance ('H and 13C NMR) and 2D NMR ('H-'HCOSY, 13C-'H COSY) and mass spectroscopy (MS).

Abstract

The jackfruit bark extract (A. heterophyllus) could inhibit the activity of tyrosinase enzyme. It was needed to be formulated as whitening cream. Cream formulation need emulsifi er to result physical properties and good stability. Aim of this study was to know the optimum composition of stearic acid and trietanolamine mixture as emulsifi ers in the jackfruit bark extract cream by using Factorial Design method. The fraction of the jackfruit bark extract was formulated in cream. The composition of stearic acid and trietanolamine were F1 (5:0,4), Fa (2,5:0,4), Fb (5:0,2) and Fab (2,5:0,2). The cream’s physical properties (spreadability, viscosity, viscosity shift) were evaluated. Data were analyzed by using Design Expert to obtain emulsifi er optimum composition. The result of study showed cream of jackfruit bark extract with variation of stearic acid and triethanolamine have a the viscosity value : 30628-37227 cps; spreadibility range from 5.20 to 6.80 cm and viscosity shift between 8.34% to 9.95%. The conclusion of this research is the formula that has optimum physical properties obtained by emulsifi er mixture composition of 3.22 gram stearic acid and 0.3 gram trietanolamine.

Abstract

Development and validation of the instrument to assess patient satisfaction to pharmaceutical care is important to be conducted. It will provide a reliable tool so that research to measure patient satisfaction regarding to pharmaceutical care could be further developed. First, content of the questionnaire was developed based on the literature review and focus group discussion of the panel. Then, the draft was tested to 5 respondents as a readability measurement. After received input from readability test’s respondents and made some changes, panel re-tested the questionnaire as content validity analysis. Finally, the questionnaire was distributed to other 30 respondents to get data for factor analysis, construct validity test and reliability test. Thirteen items was found as important factors that inuence patient satisfaction to pharmaceutical care based on literature review and focus group discussion activity. The questionnaire was further tested on 30 respondents. In the factor analysis assessment, categorization of items could not be made. Validation test using Pearson correlation and reliability test using Cronbach’s Alpha toward each item on the questionnaire showed that all items were valid and reliable, except one item in the “hope” section. Further study with greater number of respondents is required to re-validate this questionnaire.
