Abstract

Introduction: Seventy-five percent of menopausal women have menopausal syndrome, one of which is vaginal atrophy. DHEA is the only source of postmenopausal sex steroid hormones that provides estrogen and androgens according to cell needs to function normally. The aim of this study is to prove that DHEA supplementation orally can increase epithelium thickness, smooth muscle tissue, and the number of vaginal blood vessels of adult white mice (Rattus norvegicus) after bilateral ovarectomy. Method: This study used true experimental post-test only control group design using 36 white mice aged 10-12 weeks with a body weight of 180-200 grams after bilateral ovarectomy. The mice were divided into two groups with 18 mice each. The first group (controls) was given glycerin orally at a dose of 1 ml per day while the second group (treatment) was given 7.2 mg DHEA dissolved in 1 ml glycerin orally per day. On day 43, vaginal tissue was taken and histopathological examination was performed. Result: The results showed that the mean of epithelial thickness of treatment group was significantly higher than control group (51.397 µm vs. 29.755 µm; p: < 0.05). The smooth muscle thickness was also significantly higher in treatment group compared to control (74.136 µm vs. 47.428 µm; p: <0.05). Finally, the number of blood vessels was also significantly higher in treatment group (18.39 vs. 9.50; p:<0.05). Conclusion: It can be concluded that DHEA supplementation increases epithelium thickness, smooth muscle tissue thickness, and the number of vaginal blood vessels in adult rats after bilateral ovarectomy.

Abstract

Penyebab kegagalan penyembuhan luka pada diabetes mellitus adalah multifaktorial. Kegagalan dari fungsi leukosit adalah faktor yang utama dari penyembuhan luka diabetes yang sulit sembuh. Thymoquinone, zat aktif utama pada jintan hitam, mempunyai efek anti mikroba, anti inflamasi, immunomodulator, dan antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh minyak jintan hitam (Nigella sativa) yang diberikan secara topikal terhadap peningkatan regenerasi jaringan luka yang dilihat dari neovaksularisasi, jumlah fibroblas, dan epitelisasi pada tikus diabetes melitus.Penelitian ini menggunakan tikus (Rattus novergicus) usia 3-4 bulan sebagai subjek yang secara anatomis memiliki kesamaan dengan manusia dewasa. Penelitian eksperimental ini menggunakan rancangan peneltian Posttest Only Control Group Design . Subjek penelitian dibuat menjadi diabetes dengan pemberian Streptozotocin (STZ) dengan dosis tunggal 50 mg/kgBB, yang disuntikkan secara intraperitoneal. Jumlah sampel dalam penelitian ini 30 ekor tikus diabetes yang dibagi menjadi 3 kelompok. Tikus dilukai kulit punggungnya dengan diambil kulit full thickness diameter 0,5 cm menggunakan pump biopsi. Kelompok pertama adalah kelompok kontrol negatif diberi cream emulgide topikal dengan oral amoksilin, kelompok ke-dua adalah kelompok perlakuan diberi minyak jintan hitam 100% topikal dengan oral amoksilin, dan kelompok ke-tiga adalah kelompok kontrol positif diberi salep mupirocin 2% topikal dengan oral amoksilin untuk masing-masing kelompok selama 14 hari. Tikus dikorbankan pada hari ke 14 dan diambil jaringan lukanya untuk dilakukan pemeriksaan histopatologi terhadap neovaskularisasi, jumlah fibroblas, dan epitelisasi.Berdasarkan hasil analisis sesudah perlakuan didapatkan bahwa rerata neovaskularisasi kelompok kontrol negatif adalah 25,00±1,89, kelompok perlakuan adalah 8,30±2,21, dan kelompok kontrol positif adalah 7,50±2,51; rerata jumlah fibroblas kelompok kontrol negatif adalah 79,10±2,81, kelompok perlakuan adalah 32,80±4,73, dan kelompok kontrol positif adalah 29,90±4,99; rerata epitelisasi kontrol negatif adalah 60,84±2,79, kelompok perlakuan adalah 96,96±9,59, dan kelompok kontrol positif adalah 96,30±1,17. Selanjutnya pada rerata neovaskularisasi dan jumlah fibroblas diuji dengan uji One Way Anova diperoleh hasil berbeda secara bermakna (p<0,05), dan dilanjutan dengan uji Least Significant Difference – test LSD pada neovaskulariasi dan jumlah fibroblas antara ketiga kelompok diperoleh hasil perbedaan bermakna kecuali antara kelompok perlakuan dan kelompok kontrol positif. Sedangkan uji perbandingan antara ketiga kelompok terhadap rerata epitelisasi dengan menggunakan Kruskal-Wallis menunjukkan hasil berbeda secara bermakna (p<0,05) dan dilanjutkan dengan uji Mann-Whitney, diperoleh hasil perbedaan bermakna kecuali antara kelompok perlakuan dan kelompok kontrol positif.Kesimpulan dari penelitian ini adalah pemberian minyak jintan hitam secara topikal dapat meningkatkan regenerasi jaringan luka pada tikus diabetes dengan cara meningkatkan epitelisasi, menurunkan neovaskularisasi dan jumlah fibroblas pada hari ke 14.

Abstract

Introduction: Repeated exposure of the skin to UV-B rays induce reactive oxygen species (ROS) formation that activates Nuclear Factor-kappa Beta (NF-κB) and stimulates pro- inflammatory cytokines expression which leads to premature aging of skin. Deep-sea shark liver oil contains ±80% squalene which could help neutralized ROS. Therefore, the purpose of this study was to assess the effect of topical deep sea shark liver oil toward NF-κB expression in UV-B-exposed on wistar rat (Rattus norvegicus) skin. Methods: A true experimental posttest-only control study was conducted using 36 Wistar rats which divided into 2 groups (n= 18). Control group was exposed to UV-B and treated with topical placebo basis of glycerin while the treatment group was treated with topical deep sea shark liver oil. A total of 840 mJ/cm2 UV-B exposure was given for 4 weeks. NF-κB expression was examined by immunohistochemistry. Results: The mean NF-κB expression was significantly higher in control group at 30.8±3.81 while the treatment group treated with topical deep sea shark liver oil has average expression at 10.4±1.98. The independent t-test showed a t-value of 20.090, and a p-value of 0.033. Conclusion: It can be concluded that topical deep sea shark liver oil inhibits the expression of NF-κB expression in UV-B-exposed Wistar rat (Rattus norvegicus) skin.

Abstract

Introduction: Aging is a complex multifactorial process and leads to functional changes one of which is the decline in reproductive function. Decrease in reproductive function in women in in the form of decreased ovarian reserve can be detected by low levels of Anti_mullerian Hormone (AMH). This study aims to prove the effect of administration of Gallus domesticus boiled chicken white egg increased AMH levels of premenopausal female wistar rats. Methods: This research was experimental animal with Pretest-Posttest Control Group Design. The study conducted on 16 premenopausal female wistar mices, aged 17-18 months, weighing 250-260 grams. The mice were divided into 2 goups: the control group (placebo), which was given gelatin flour by 7.3 gram 3 times daily for 4 weeks and the group treted with Gallus domesticus boiled chicken white egg by 7.3 gram 3 times daily for 4 weeks by oral. Blood AMH level were measured before and after treatment for both group. Results: The result showed that before treatment, mean AMH level both in control group and treatment group were not significantly different (p > 0,05). After treatment, mean AMH level both in control group and treatment group were significantly different (p < 0,05). The administration of Gallus domesticus boiled chicken white egg increased AMH levels by 0.804 ng/ml. Conclusion: It can be concluded that administration of Gallus domesticus boiled chicken white egg increased AMH level. The mean AMH level after treatment had a significant difference between control group (placebo) and treatment group with p = 0.001.

Abstract

Introduction: In aging process, the hair experiences thinning and the hair shaft becomes more fragile due to decreased regeneration of hair. Cinnamon essential oil contains cinnamaldehyde compounds which have a vasodilating effect on blood vessels and stimulating IGF-1 can increase fertility and prevent hair loss. Methods: This study was an experimental study with a post test only control group design carried out in the laboratory animal unit of the pharmacology section of the Udayana University medical faculty. In this study 30 male white rats (Rattus norvegicus) wistar lines were sheared 4cmx4cm which were divided into three groups: negative (aquades) control group (2% minoxidil), treatment group with cinnamon essential oil (concentration 100 %) for 30 days. Results: The average hair length of aquades (P0) was 5.88 ± 0.36 mm, minoxidil group 2% (P1) 7.4 ± 0.20 mm. , cinnamon essential oil group (P2) 8.32 ± 0.18 mm In the diameter size of hair follicles in the three groups showed the mean diameter size of hair follicles was significantly different (P <0.05). The average results of the group hair follicle diameter (P0) were 79.73 ± 30.68 µm, group (P1) 127.07 ± 38.20 µm and group (P2) 130.91 ± 49.96 µm. The LSD test results on hair length measurements between the three groups showed a very significant difference (P <0.01). Group P0 and P1-1,51 mm, group P0 and P2-2,44 mm group P1 and P2 0,92 mm. The results of LSD hair follicle diameter P0 and P1 -47.34 µm (P <0.05), P0 and P2 -51.18 µm (P <0.01), P1 and P2 -3.84 µm (P> 0 , 05) Conclusion: The application of topical cinnamon essential oil increases hair length and diameter size of male wistar rat hair follicles as effectively as topical minoxidil 2%.

Abstract

Introduction: Diabetes mellitus is a complex and progressive disease which often lead to several debilitating complications that partly caused by by free radicals which can be overcome with antioxidants. Bay leaves contain essential oils, tannins, flavonoids and terpenoids which have considerable antioxidant properties. Therefore, this study aims to determine the effect combination treatment of vildagliptin and bay leaf (Syzygium polyanthum) extract toward pancreatic beta cells density and glycated albumin levels in diabetic male Wistar rats (Rattus norvegicus). Methods: An experimental posttest only control group study was conducted using 36 albino male rats, aged 2-3 months, weighing 180-200 grams. The rats were divided into 2 groups (n= 18) with control group treated with 1.8mg/200g body weight vildagliptin and 2cc placebo (aquabidest) while the treatment group received 1.8mg/200g BW vildagliptin and 250 mg/200g BW bay leaves extract. All treatment lasted for 21 days. Results: The results showed that pancreatic beta cell counts in the treatment group was significantly higher than the control group (109.07 ± 20.47 cells/field of view vs 90.87 ± 13.91 cells/field of view; p<0.01). However, the levels of Glycated albumin between two groups were not significantly different (treatment vs control: 17.33 ± 4.51 vs 20.18 ± 4.57; p=0.068). Conclusion: This study suggested that combination treatment of vildagliptin and bay leaf extract increased pancreatic beta cells but did not reduce glycated albumin levels in diabetic male Wistar rats.

Abstract

Introduction: Aging can cause decreased testosterone levels in men which is known as andropause. Some herbs are known to increase the levels Testosterone. One of them is Klabet seed that contain phyto-testosterone. The purpose of this study was to prove that the administration of ethanol extract of Klabet seeds increased testosterone levels in old male Wistar rats. Methods: A pretest-posttest control group design study was conducted using 20 male white wistar rats that were divided into 2 (two) groups: control group that received 2 cc of distilled water and treatment group that received 2 cc ethanol extract of Klabet seeds 250mg/250gBB rats/day by sonde for 14 days. The testosterone level was measured on day 1 and 15. Results: The analysis showed that there was no significant difference in the pre-test and post-test testosterone level P0 (Pre-test:16.4±0.553 ng/ml; posttest: 15.9±1.51ng/ml) while increased level of testosterone could be observed in treatment group (P1) (pre-test: 16.6±0.476ng/ml; post-test: 19.4±0.828ng/ml. Comparative analysis between post-test results (P0 vs P1) showed that the difference was statistically significant (p <0.001). Conclusion: Oral administration of Klabet seeds ethanol extract can significantly increase testosterone level in old male wistar mice.

Abstract

Kondisi hiperglikemia pada diabetes melitus merupakan salah satu faktor risiko disfungsi ereksi sebagai akibat dari disfungsi endotel melalui mekanisme autooksidasi glukosa, glikasi protein, dan aktivasi jalur metabolisme poliol yang selanjutnya mempercepat pembentukan senyawa oksigen reaktif. Kakao kaya kandungan antioksidan flavonoids yang dapat menyebabkan penurunan atau pencegahan terjadinya stres oksidatif dan disfungsi endotel sehingga menghambat penurunan sintesa, pelepasan, dan aktivitas Nitric Oxide (NO). Tujuan dari penelitian ini adalah untuk membuktikan pemberian ekstrak biji kakao (Theobroma cacao L.) dapat menghambat penurunan Nitric Oxide (NO) dan jumlah sel endotel korpus kavernosa tikus wistar jantan diabetes. Penelitian eksperimental, completely randomized post test only control group design menggunakan 36 ekor tikus putih (Rattus norvegicus) jantan, dewasa (berumur 10 minggu) sebagai sampel, yang terbagi menjadi 2 (dua) kelompok masing-masing berjumlah 18 ekor tikus, yaitu kelompok kontrol negatif P0 (aquabides) dan kelompok perlakuan P1 (ekstrak biji kakao dosis 280 mg/200 gram berat badan tikus). Hasil penelitian menunjukkan rerata kadar Nitric Oxide (NO) kelompok yang diberi aquades adalah 0,074±0,004 nmol/nl dan kelompok yang diberi ekstrak biji kakao dosis 280 mg/200 gram berat badan tikus adalah 0,137±0,008 nmol/nl (p<0,01). Rerata jumlah sel endotel kelompok yang diberi aquades adalah 39,00±1,847 sel dan kelompok yang diberi ekstrak biji kakao dosis 280 mg/200 gram berat badan tikus adalah 78,78±7,369 sel (p<0,01). Dapat disimpulkan bahwa pemberian ekstrak biji kakao (Theobroma cacao L.) dapat menghambat penurunan Nitric Oxide (NO) dan jumlah sel endotel korpus kavernosa tikus wistar jantan diabetes.

Abstract

Introduction: Piper crocatum leaves extract has been widely proven in reducing blood glucose levels in rats. However, there is still no study that compared its effectiveness to metformin. Therefore, this study aimed to evaluate the efficacy of Piper crotatum leaves extract compared to metformin in reducing fasting blood glucose and glycated albumin. Methods: Experimental pretest-posttest groups study using 16 adult male Wistar rats, aged 2-3 months, and weighing 200-220 grams was conducted. Diabetes was induced by using Streptozotocin and Nicotinamide intraperitonially. The control group was treated with metformin 9 mg/200gBB/day while the treatment group was treated by Piper crocatum leaves extract at 0.6 g/200gBB/day for 14 days. Venous blood sample was used to assess fasting blood glucose and glycated albumin. Resuts: The mean of fasting blood glucose level decreased significantly in the control group from 182.00+19.38 mg/dl to 117.38+12.51 mg/dl (p<0.001) and the levels of GA was also decreased from 3.04+0.99 ng/ml to 1.38+0.63 ng/ml (p<0.001). In the treatment group, the level of fasting blood glucose decreased from 179.50+24.39 mg/dl to 140.13+15.12 mg/dl (p<0.001) and the levels of glycated albumin was also decreased from 4.01+1.12 ng/ml to 3.58+0.90 ng/ml (p=0.036). There was no difference in the level of fasting blood glucose and GA between the two groups before treatment but both parameters were significantly lower compared to the treatment group (p=0.005 and p<0.001 respectively). Conclusion: Administration of piper crotatum leaves extract reduced fasting blood glucose but has lower efficacy compared to metformin in diabetic Wistar rats.

Abstract

Introduction: Supplements milk is a high protein of whey that consumed by many teenagers to maintain the fitness in accordance with the advertisements in the society, the presence of hormones in milk that are not listed in its composition causes a hormonal balance disorder. The aim of this research was to prove that giving supplements milk with a high protein of whey in increasing estrogen levels and lowers testosterone counts in male Wistar rats (Rattus norvegicus). Methods: This research employed experimental research using a pretest-posttest control group design that used 22 white male Wistar rats (Rattus norvegicus), the samples were divided into 2 groups, namely the control group (P0) and the treatment group (P1) that treated by supplements milk with a high-protein of whey once a day for three weeks. The blood was taken before and after treatment for estrogen levels and testosterone counts. Result: The finding indicated that a significant increase in estrogen levels before and after the administration of milk supplement with high whey protein 11.603 ± 0.502 ng/ ml to 16.141 ± 0.347 ng/ ml (p<0.001). There was no decrease in testosterone counts before and after subjects were administered by milk supplement with high whey protein 1.86 ± 0.325 ng/ ml to 1.69 ± 0.260ng/ ml p = 0.220 (p> 0.05). Conclusion: Based on the results of the analysis it could be concluded that the administration of milk supplement with high whey protein could increase estrogen levels while decreasing testosterone in male Wistar rats (Rattus norvegicus).
