Abstract

Liberibacter bacteria are one of the pathogenic bacteria in citrus plants. Siam Citrus Plant in Setapok Village, Singkawang City, suspected to be infected with Liberibacter spp. bacteria by showing patterns of symptoms of chlorosis accompanied by patches. This study aims to detect Liberibacter spp. on Siam Citrus leaves with symptomatic chlorosis with patches. Leaf sampling uses the roaming method in one of the citrus plantations in Setapok Village, Singkawang City. Detection of Liberibacter spp. done by PCR (Polymerase Chain Reaction) technique. Detection of bacterial DNA through several stages namely, isolation of bacterial plasmid DNA, 16S rDNA gene amplification with a thermocycler machine, and visualization using a set of electrophoresis devices. Electrophoresis results show the presence of DNA band lines, with a size of around 1100 bp for OI1 and OI2c primers, and DNA bands of 703 bp for A2 and J5 specifics. The results showed that there were Liberibacter spp. in the symptomatic chlorosis of Siam Citrus leaves accompanied by patches.

Abstract

Peniti Mangrove is one of the forests in West Kalimantan which has a high forest productivity rate. This highly supports the existence of cellulolytic bacteria, a cellulase-producing microorganism which can degrade cellulose into glucose. This research aims to measure the total density of bacteria, cellulose degradation power and the genera members of cellulolitic bacteria on Mangrove soils in Peniti, Mempawah. The soil samples were obtained through composite samplings, by setting 20 spots in four different locations: A,B,C dan D. Bacterial isolation was conducted using dilution technique and pour plate method on carboxy methyl cellulose (CMC) agar medium. The count of the density was done through total plate count (TPC) method. The degrading power was counted using cellulolytic index value. From the cellulose degrading test, 9 isolates of cellulolytic bacteria from 16 isolates were obtained. Cellulolytic bacteria isolates were characterised by colony morphology, cell morphology and biochemical tests. Through the identification process, 7 genera were identified: Aeromonas, Bacillus, Alcaligenes, Pseudomonas, Actinobacillus, Listeria and Chromobacterium.

Abstract

Bean (Phaseolus vulgaris L.) is a type of legumes from leguminoceae familly which is beneficial to decrease blood pressure and control body metabolism system. Bean can grow well on rich soil which contains much humus. This research aims at identifying the effect of Nipah banana’s skin waste towards the vegetative growth of bean. The research was conducted for thee months since Febuary to April 2017. It was carried out in a lab of biology deapartment, Faculty of Mathematics and Natural Sciences, Tanjungpura Universty. The research employed completely randomized design ( RAL). It consisted of six level of concentration, such as control (P0) 0 (P1) 12.5 (P2) 25 (P3) 37.5 (P4) 50 (P5) 62.5g. They ware carried out with three repetitions so they could gather 18 experimental units. Providing bean with the banana’s skin waste could give a clear effect towards the plant’s heights, gross weight, and dry weight. Using the banana’s skin waste whice has 50g concentration provides the best resulty for the plant’s height (141.30 cm), the number of leaf (37.67 leaves), and dry weight (12.54 g).

Abstract

Stem rot disease is caused by one of Phytophthora pathogenic fungi that attacks several types of cultivated plants. The alternative control against Phytophthora attack can be done with the use of natural fungicides. One of the herbs that can be untilized is the ethanol extract of lakum stem (Cayratia trifolia (L.) Domin). The aim of this research is to know the effect of ethanol extract of lakum stem and its secondary metabolite in inhibiting the growth of isolate Phytophthora sp. Im5. The research was conducted for three months starting from October to December 2017. This research used solid dilution method and completely randomized design (CRD) wth 8 levels of treatments, i.e. positive control, negative control, ethanol extract concentration of 5, 10, 15, 20, 25 and 30 mg/ml. The test for secondary metabolite compounds was done by phytochemical screening method. The result showed that the concentration of 30 mg/ml has the biggest antifugal activity with the inhibition percentage of 34.98%. The activity of lakum stem ethanol extract against isolate Phytophthora sp. Im5 was included in moderate catagory. The results of phytochemical screening showed that the ethanol extract of lakum stem contained alkaloid, flavonoid and tannin compounds.

Abstract

Control of corn leaf blight caused by the fungus Helminthosporium sp., can be carried out by employing natural antifungal of plant extract. One of the plants that have the potential as an antifungal is Sonneratia alba J.E.Sm. The aim of this study is to determine the ability of S. alba fruit as antifungal and to determine the concentration of S. alba fruit extract, that can inhibit the growth of fungus Helminthosporium sp., at the level of very strong activities. Testing of antifungal activity of the fruit extract of S. alba carried out by employing 7 treatments, which the treatments are negative control, solvent control (DMSO 10%), positive control (Dithane M45 10%), concentration of fruit extracts S.alba (25 %, 50%, 75% and 100%). Based on phytochemical test indicates that fruit S. alba contains class of flavonoid compunds, alkaloids and tannins. The results of antifungal testing at level 100% concentration indicate very strong antifungal activity (90.01%). Those conditions indicate that 100% constitutes concentration of S. alba fruit extract, that can inhibit the growth of Helminthosporium sp. fungus at the level of very strong activities.

Abstract

Overfishing has been being the major problem for Leptobarbus melanopterus population in Danau Sentarum National Park. This activity was feared could affect the declining of L. melanopterus population in the future. This research aims to determine growth pattern of L. melanopterus through length-weight relationship and relative weight analysis. The total of 64 fishes were collected which consisted of 32 males and 32 females. The range of male total length was 146-248 mm with weight was 40-170 gram, whereas total length female was ranging 141-299 mm with weight was 40-350 gram. Based on the regression analysis of length-weight data, the b value of male and female were estimated 2,46 and 2,81 respectively. Moreover, the b value of all samples without distinguishing sex type was 2,69 with relative weight was 101,33. The results of this research confirms that either females or males has negative allometric growth pattern.

Abstract

Mangrove is a community of tropical coastal plants dominated by several species of trees with shrubs that have the ability to grow in coastal areas. The purpose of this research is to know various kind of mangroves which located in Tanjung Bila area. The research were done for 4 month start from February until May 2016 in Tanjung Bila Pemangkat region Sambas regency. The data of mangrove vegetation were collected using the transect line plot method. There were 8 species of mangroves found in Tanjung Bila area and classified into 5 family i.e. Avicennia marina, Avicennia alba (Avicenniaceae), Bruguiera cylindrica, Rhizophora apiculata (Rhizophoraceae), Sonneratia caseolaris (Sonneratiaceae), Acanthus ilicifolius, Acanthus volubilis (Acanthaceae) dan Acrostichum speciosum (Pteridaceae).

Abstract

The infection of Diplodia sp. Fungi on tangerine (Citrus nobilis) causes the surface of main trunk or branches to dry up. Generally, the effort of controlling disease on plants is done by the use of synthethic pesticidies that have negative impact, so that it should be mitigated and the use of botanical pesticides should be promoted. The aim of this study was to determine the antifungal activity of methanol extract of    P. serratifolia leaves toward the growth of Diplodia sp. fungi. The completely randomized design (CRD) was used on this study with 6 treatments which were control, DMSO, the extract concentrations of 25%,50%, 75%, and 100%. The result showed that the methanol extract of Premna leaves significantly affecting the growth of Diplodia sp. fungi. The concentration of 100% was the one to produce the highest inhibition percentage on Diplodia sp. fungi growth, which reached 93,31% with very strong activity.

Abstract

Bacteria belongs to the genus of Staphylococcus is pathogenic bacteria and its existence is an indicator of contaminated environment particularly in the air. The aim of this study is to detect the existence of bacteria belongs to Staphylococcus in the traditional market space of Pontianak city. The study was conducted in January - March 2018. Sampling was administered in 4 traditional markets of Pontianak City using volumetric air sampling along with a vacuum air pump YYP 550-12A1 with an air flow rate of 26 l / min for 6 minutes using Mannitol Salt Agar (MSA). The results showed that air bacteria member of the genus Staphylococcus found in all traditional markets in the city of Pontianak. The lowest number of bacteria belongs to the lowest Staphylococcus genus in the Flamboyan Market was 15 CFU/m3 and the highest was in Pasar Mawar 89 CFU/m3. The data indicates the air in the traditional market of Pontianak City has not fully fulfill the air health standards according to Sedyaningsih (2011), which is free of pathogenic bacteria 0 CFU/m3.

Abstract

Kakap River Estuary is a body of water in a direct contact with the sea and has a water salinity changes that affect the composition of Rotifera, is an estuarine ecosystem. The purpose of this study was to determine the species of Rotifera in the Kakap River Estuary. This study was conducted in May to August 2015. The sampling location was divided into seven stations, each station was divided into three sampling points, and each point of sampling was repeated three times. The research found 73 species of Rotifera belong to 14 genera (Anuraeopsis, Brachionus, Keratella, Beauchampiella, Euchlanis, Cephalodella, Colurella, Lepadella,Habrotrocha, Lecane, Philodina, Polyarthra, Trichocerca, and Testudinella). Lecane was the most common genera species found of all stations with a number of 33 species.
