Abstract

The coastal waters of Simpang Pesak Sub-district, East Belitung Regency considered as a productive region and had a broad range of habitats for bivalvia. However, research on spatial distribution and habitat preference of bivalvia in this region is limited to none. The objectives of this research were to examine the effects of marine biophysical parameters and substrate quality on bivalvia. The research was started from Mei 2013 until June 2013 by using survey method and random systematic sampling approach. The results showed that there were 16 species of bivalves consisted of 14 genus from 7 family and dominated by Gafrarium pectinatum at station I (13.62 ind/m2, C=0.52, H’=0.6, muddy sandy), and  station II (28.4 ind/m2, C=0.51, H’=1.04, sandy), Gafrarium tumidum at station III (59.9 ind/m2, C=0.51, H’=1.03, sandy), Scapharca pilula at Stasiun IV (61,6 ind/m2, C=0.5, H’=0.89, clay sandy). Spatial distribution and density of bivalves were influenced by particle size, C-organik, and other environmental factors such as current, dissolved oxygen, TSS, temperature, and salinity. Abstract

Chronic sediment toxicity tests could be used to evaluate the effects of sediment on critical stages in the life of an organism like invertebrate larvae and microalgae. Microalgae play an important role in aquatic ecosystem due to their role as producer in food chain producing organic matter and oxygen through photosynthesis process. This paper aimed to assess sediment quality of Semarang water using chronic sediment toxicity test. Research was conducted by testing 13 sediment samples from Semarang coastal region in August, 2010. Results showed that sediments exposed for 96 hours were still able to stimulate the growth of C. gracilis eventhough the copper concentration was above standard value provided by the CCME. The conclusion from this research was that sediment in Semarang still in good condition and can support growth of C. gracilis as primary producer. Chronic sediment toxicity test using C. gracilis could be combined with heavy metal measurement for sediment quality asessment. Abstract

The utilization of local food commodities such as corn and cassava with seaweed addition as a dietary fiber source for producing artificial rice through extrusion technology is an  alternative for food diversification. The research was carried out to find out the best composition (rice, corn, cassava, and seaweed) and temperature of extrusion process on making artificial rice and the influence of dietary fibre on sensory properties and physicochemical. The composition of rice, corn, and cassava in proportion  of 1:3:1 with 20% seaweed, Eucheuma cottonii, addition and temperature extruder of 90 °C were selected as the best product for artificial rice. The  sensory evaluation was 8.02±0.21 (people’s preference). In physicochemical properties, dietary fiber significantly affected on low bulk density and starch digestibility. This condition is very good for health especially in maintaining the stability of blood glucose in the body. Keywords: artificial rice, composition, extrusion, seaweed, dietary fibre, temperature

Abstract

In this study, cellulase was produced by endophytic fungus isolated from seagrass. Substrate used for incubating the fungus was the waste of agar industry. The objectives of this study were to optimize cellulase production and to characterize the highest activity of fungal crude cellulase. In our previous study, the EN isolate (isolated from Enhalus sp.) showed the highest cellulolytic index. Therefore, in this research we focused on cellulase activity of the isolate. Cellulase activity was determined based on endoglucanase activity, total cellulase activity, and β-glucosidase activity. The highest activity was then used to determine cellulase activity in enzyme characterization. The fungus was cultured in different concentration of agar extraction algal wastes. The fungal culture was incubated for 3-21 days with 120 rpm orbital shaker. The results showed that endoglucanase activity was 0.019-0.031 U/mL, total cellulase activity was 0.007-0.013 U/mL, and β-glucosidase activity was 0.00012-0.00361 U/mL. The highest endoglucanase and total cellulase activity were obtained from the fungal culture after 9 days incubation, β-glucosidase was obtained from the fungal culture after 15 days incubation with 1.5% of algal waste as substrate. The optimum pH and temperature were determined as 4 and 60 oC, maximum reaction rate (Vmax) and Michaelis-Menten constant (Km) for endoglucanase activity was determined as 0.044 U/mL and 0.103% respectively. Abstract

Cellulolytic fungi are potentially useful in biotechnology. Decomposition of substrates containing cellulose can be applied in bioethanol and enzymes production. Fungi are one group of microorganisms that can decompose cellulase materials.  This study aimed to obtain potential cellulolytic fungi isolated from marine habitat. Screening of the fungi was performed using CBM media with different salinity. Six isolates were tested quantitatively for cellulolytic activity using congo red staining. Cellulolytic index of algicolous and manglicolous fungi were lower than those of EN isolate, Veronea sp. KT19, and sponge associate (SMH). Fungi cultured in freshwater and seawater media possessed higher cellulolytic index in compared to those of cultures in 3% NaCl medium. However, freshwater and seawater media showed no significant effect. Cellulolytic index of EN isolate, Veronaea sp. KT19, and SMH after six days of incubation were 1.357; 1.267, and 1.278, respectively. All six isolates potentially produced cellulase in all medium tested. Abstract

An integrated study of sediment distribution was conducted in Weda Bay, Northern Maluku to provide general information on transportation and deposition process based on sediment grain size distribution. The study was conducted during the Weda Bay Expedition using the “Baruna Jaya VII” research vessel in March 13th –22th 2013. Sieving method (granulometric) was used to analyze the grain size. The results indicated that in general the pattern of sea floor sediment distribution was dominated by clay – sand grain-sized. The current speed influenced the sediment transport, deposition, and distribution.  Larger fractions of sediment were quickly settled on the sea floor due to stronger currents around Southern area (Widi islands), meanwhile the lesser fractions of the transported away into other places with weaker currents conditions. Abstract

Turbulent mixing process can be evaluated from density inversions in CTD profiles, that may reveal the overturning eddies. The quality of overturn regions are then determined by the quality of CTD signals. The study removed noise on CTD signals without losing small perturbation which commonly occurred by traditional filter.   By applying wavelet denoising in pre-processing data of Thorpe method, more Thorpe displacements indicated overturn regions, were able to detect especially in small density gradients. Therefore, the identified overturn region using the Thorpe method can be applied to a larger region of density gradients. Abstract

The secondary entry portal of the Indonesian Throughflow (ITF) from the Pacific to Indian Oceans is considered to be via the Halmahera Sea (HS). However, few ITF studies have been done within the passage. This motivated the Internal Tides and Mixing in the Indonesian Througflow (INDOMIX) program to conduct direct measurements of currents and its variability across the eastern path of the ITF. This study focused on the intra-seasonal variability of near-bottom current in HS (129°E, 0°S), its origin and correlation with surface zonal winds and sea surface height over the equatorial Pacific Ocean. The result showed a strong northwestward mean flow with velocity exceeding 40 cm/s, which represented the current-following topography with the northwest orientation. Meridional current component was much stronger than the zonal component. The energy of power spectral density (PSD) of the current peaked on 14-days and 27-days periods. The first period was presumably related to the tidal oscillation, but the latter may be associated with surface winds perturbation. Furthermore, cross-PSD revealed a significant coherency between the observed currents and the surface zonal winds in the central equatorial Pacific zonal winds (180°E-160°W), which corroborates westward propagation of intra-seasonal sea surface height signals along the 5°S with its mean phase speeds of 50 cm/s, depicting the low-latitude westward Rossby waves on intra-seasonal band. Abstract

ABSTRACT Soft corals contain bioactive compounds that can be used as a marine natural product. The puposes of this study was to determine of the soft corals inhibition potential for antibacterial activity. The methodology in this study included field sampling, extraction (in the solvent of n-hexane, ethyl acetate and methanol), antibacterial bioassay, and briefly describe the type of soft coral that possesed the highest inhibition. In sampling treatment, the biomass among wet samples and dry samples varied.  The highest shrinkage due to drying was found in Nephthea, followed by Lobophytum and Sarcophyton, while the lowest was found in the genus of Sinularia. Extracted from 12 samples of soft coral, the lowest extract weight was found in the semi-polar solvent (EtOAc), while the extract weight of n-Hex and MeOH was vary. Inhibition power from all extract samples was found on soft coral extract in the EtOAc and MeOH solvents. The highest value of inhibition power was found in soft coral of Sinularia polydactyla and S. Abstract

ABSTRACT Bungus district of Kabung Bay is a growing region located at coastal zone of southern city of Padang, west sumatra. As a growing region, the Bungus district brings some impacts on population increase and degradation of environment quality. Therefore, it is needed an effort to identify land use changes and the distribution of land use in this region from the year of 2003 until 2013. This research used landsat 7 imagery in 2003 and landsat 8 imagery in 2013. The data were analysed descriptively using geographical informastion system. The result showd that (1) swamp land cover experienced the smallest land use change between 2003 until 2013 (0.02 ha/yr), meanwhile forest land use had the biggest change of about 224.8 ha/yr. The biggest addition of land cover belong to settlement area about 47.59 hectare, and the other hand occur on bush about -31.68 hectare.
