Abstract

Some diseases, such as cancer, hereditary and genetic diseases, as well as viral infectious diseases, have been treated unsatisfied by the conventional therapy so far, and even more, by the gene therapy. Together with the pharmaceutical industries, researchers put their best effort to hunt some molecules that can be more favorable for such kind of therapy. After a pivotal study reported in May, 2001, it is certain that Ribonucleic acid (RNA) could effectively silence gene expression in mammalian cell line, so it was then proposed in 2004 the term RNA therapeutics. Antisense RNA therapy which came into the stage earlier seemed to be the one that can answer all the problems in knocking out the unwanted messenger in gene expression. RNA interference (RNAi) concept, which came later in around 2000, began to look like a possible contender. It was reported in some studies that RNAi seems to have some more advantages over both stronger gene-silencing effects and greater ease of use. However, the main obstacle of all kind of gene therapy is, undoubtedly, on the delivery of this molecule to enter the target cell, and mostly, to where it is needed most inside the body. Some studies on genetic material delivery system have been reported, and their progress has been discussed.

Abstract

One important aspect of district public health service is the management and budgeting of drugs. Up to this time no indicators can be used yet as guidelines for evaluating drug management and budgeting effectiveness in a district in Indonesia. The objective of this study is to determine the effect of training managing drug staff on the evaluation capability increase and their knowledge. Training has been conducted twice in 2 provinces, 4 districts ex HP IV each, covering Dinas Kesehatan, GFK, 5 PHCs per district and 300 outpatients. There are 7 groups of indicator which include a total of 29 indicators. Training involved computation of indicators of drug management and budgeting in a district. Tests were carried out before training and after the first and second training. Data were analyzed using paired t-test for the effect of training on the increase of knowledge and capability of managing drug staff in indicator computation for the evaluation of drug management and budgeting as well as linear regression for the relation between education and working experience of the staff and unpaired t-test for the difference in the increase in knowledge of pharmacy assistant and non pharmacy assistant. Results from the study on 54 managing drug staff in 8 district shows that knowledge increase significantly after the first and second training, whilst the latter increase is greater than the former one and that the increase in knowledge of pharmacy assistant exceeds that of non pharmacy assistant.

Abstract

Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that are suspected having an estrogen-like effect or as phytoestrogen. The objective of this study was to evaluate the estrogen-like effect of biji klabet alcohol extracts. This research used immature Wistar female rats, age of 19 days, which were represent the climacteric/menopause phase. The testing animals were divided into five groups, namely normal group, estradiol control group and three level dose groups of fenugreek extract (30 mg/200 gBW; 60 mg/200 gBW and 120 mg/200 gBW). The result showed that on 120 mg/200 gBW fenugreek extract significantly (a=0.05) increased estradiol and FSH level on immature rat. Analysis of vaginal smear showed vaginal lubrication effect, surprisingly on 120 mg/200 gBW could induce estrous cycle.

Abstract

World Health Organization (WHO) state that breast cancer is second cause of death among other cancer illness. Up to now, what become a major factor that caused the breast cancer has known yet, but it was believed that the relation of some factors can improve the risk of breast cancer. Breast cancer is believed happened because of a complicated interaction between many factors like genetic, environment and hormonal, which one of them is abundant estrogen hormone rate in womenÂ’s body. This research is aim to know the risk level that caused by the use of combination contraception pills, which is combination of estrogen and progesterone with the breast cancer risk. The research use case control method in hospital based with period of September-Desember 2004 at Perjan RS Dr. Cipto Mangunkusumo. The research results show that the user of combination contraception pills has the risk to have breast cancer 1,864 times bigger compare with them who doesnÂ’t take that pills. However, the combination contraception pills is not the major risk factor but itÂ’s only a light factor to increase the risk of breast cancer.

Abstract

The effect of carnauba wax consentration to pharmacotechnical characteristics on the granules and tablets that made by hot melt granulation method have been studied. Fomula I? V with composition : 10, 20, 30, 40, and 50% w/w carnauba wax, propranolol hydrochloride 60 mg/tablet, and lactose to the 340 mg was mixed and melted, and than sieved to the mesh 16 siever. The granules were evaluated and lubricated by 1% magnesium stearat and 2% talcum. Then it were compressed into tablet which each weight was 350 mg and the tablets were evaluated. The results showed that hot melt granulatin form granules and tablets that acceptable pharmacotechnical properties.

Abstract

The red alga, Eucheuma spinosum from South Bangka Waters was shown to contain antioxidant activity. Natural antioxidants from algae are known to play an important role against various diseases and aging processes. The study objective was to investigate phytochemical screenings and antioxidant activity of ethanol fractions Euchema spinosum from south Bangka waters. Antioxidant activities of fresh algae (FA), dry algae (DA), and algae product (AP) were determined with DPPH (1,1-diphenyl-2-picrylhydrazyl) assay. Extraction was conducted using 70% ethanol in acidic environment for 24 hours. Then, the extract was evaporated using rotary evaporator. Dry extract was fractionated with ethanol - n - hexane ( 2 : 3 ) and the ethanol fraction was dried. The phytochemical screenings showed that all fractions had flavonoid, alkaloid and triterpenoid compounds. The most potent antioxidant activity was found in the ethanol fraction of AP with IC50 value 333.66 μg/mL, FA 418.32 μg/mL and DA 472.14 μg/mL. Based on the results, it can be concluded that the ethanol fraction of algae product can be used as a antioxidant source.

Abstract

Green chemistry aspect is the chemical approach that has been studied in the past two decade. One of the principles is the development of green synthesis process that is friendly for the environment. This research showed that papain can be used as catalyst for Knoevenagel reaction with 3 kinds of substituted-benzaldehyde and malononitrile as substrates in aqueous medium. The best reaction condition with 80% yield was reached by utilizing of 25 mg papain/mmol substrate. Reaction was conducted at ambient temperature and pressure for 30 min. Products were as yellowish to yellow needle crystals and successfully characterized by melting point, UV-Vis, IR, mass spectra, and 13C & 1H-NMR, named as 2-(4-hydroxybenzylidene)-malononitrile); 2-(3-hydroxybenzylidene)-malononitrile); and 2-(4-hydroxy-3-methoxybenzylidene)-malononitrile).

Abstract

Guava leaves are used as a cytotoxic. Previous research has demonstrated that guava leaves contained 61.71% of quersetin and has potential as an cytotoxic. The aim of this study was to determine the cytotoxic activity of 70% ethanol extract of guava leaves against cancer cells T47D. The method used direct counting (viable cell count) to obtained LC50 value. This study used a test solution with six concentrations that were 130.62; 67.98; 35.98; 18.43; 9.56 and 5 µg/ml. The LC50 value from the 70% ethanol extract of guava leaves was 27.54 µg/ml. The results suggested the extract of guava leaves has cytotoxic activity against T47D cells

Abstract

Self medications which are ofted applied to children are including coughs, colds, flu, and nasal congestion (17.2%), fever (15%), headache (14%), diarrhea and abdominal pain (9%). The results from previous studies showed only 0.4% caregiver of 4-6 years old children in Banyumas Regency did correctly in children’s fever self medication. Aim of this study is to determine the influence of Community Based Interactive Approach (CBIA) and Focus Group Discussion (FGD) methods for improving knowledge and correctness of caregivers in children’s fever self medication with type of research of quasi experimental design with a control group of non random pretest posttest. Results of Wilcoxon test showed that there were significant differences of knowledge statistically before and after treatment using the FGD and CBIA methods with significancy values in FGD group of 0.000 (p < 0.05) and in CBIA group of 0.002 (p < 0.05), while for correctness before and after treatment with FGD and CBIA methods, there were no statistically significant difference in significancy values of correctness for FGD group of 1,000 (p> 0.05) and for the CBIA group of 1,000 (p> 0.05). Results of Mann-Whitney test showed that the obtained significancy value was 0.012 (p <0.05), it can be concluded that there were significant differences between FGD with CBIA groups. The difference of Mean scores in FGD group (2.45) were greater than the CBIA group (0.96), so that the FGD method was better in improving the knowledge compared with CBIA method even though they both gave some improvements.. Fisher’s Exact test results showed that the ​obtained significancy value was 1,000. According to these statistical analysis results, it can be concluded that based on statistical calculatios, there is no significant difference between FGD and CBIA methods in improving the correctness of children’s fever self medication.

Abstract

The acute toxicity test and antiinflammatory effect of methanolic and n-hexane extracts Carica papaya L leaf have been studied. The results of phytochemical screening show that papaya leaf contains alkaloid, steroid, and flavonoid compound. Flavonoid and alkaloid extracted from methanolic extract of papaya leaf and steroid extracted from n-hexane extract of papaya leaf. The acute toxicity test of methanolic and n-hexane extracts Carica papaya L leaf have been studied on rat with giving a single dose of samples. The aim of this work was to investigate the behavioral responses (pharmacological profile), the development of body weight and mortality for 14 days. The results of acute toxicity test after giving dose of 250 mg/kgbw, 500 mg/kgbw and 1000 mg/kgbw of methanolic and n-hexane extracts of leaves of Carica papaya L to males rat showed no animal died and significant toxic effect. In the study antiinflammatory effects in white rat with carragenan induced 1%, and to determine an effective dose of extract papaya to decrease the edema volume of rat foot. The test was done using rat hind paw edema or esthablished an artificial inflammation in left foot of white male rats.The treatments were carried out on six groups, the positive control group was administered with Indomethacin 10 mg/kgbw per oral, the negative control group was administered with CMC 1%, and the extract groups were administered with 200 mg/kgbw and 400 mg/kgbw of methanolic and n-hexane extracts Carica papaya L leaf. Edema volume were measured every half hour for 5 hours using a pletismometer digital. Obtained data were analyzed by Kruskal-Wallis test followed Mann-Whitney test with confident level was 95%. The result of this study indicate that papaya leaf extract can reduce edema volume of rat foot.
