Abstract

When ammonia in waste water is lost inappropriately, it  will raise an adverse environmental effect for the aquatic cycle. Anammox, anaerobic ammonia oxidation, is a novel process in which nitrite is used as an electron acceptor in the conversion of ammonium to nitrogen gas. The anammox process removes ammonium in the autrotrophic system by leaving little biomass. This study aims to analyze the effect of salinity on the growth of anammox bacteria. The samples used were from the brackish water sediments of the East Flood Canal River of Semarang. The isolation was done by gram staining and the bacteria were inoculated on media with different salinity concentration and the growth was measured using spectrophotometer. The results showed that anammox bacteria had a higher growth rate of 3% (control) when it was grown on a medium with a concentration of 9%. Anammox bacteria grown on anammox selective media showed that the bacteria were able to adapt to environments with different salinity concentrations of 2% and 9%. Key words: anammox, ammonium, nitrogen, anammox bacteria.

Abstract

The determination of antimicrobial and antioxidant activities of Chlorophyll pigment of Caulerpa racemosa (Forsskal) J.Agardh is described in this study. The aim of the study was to find out the effectiveness of chlorophyll extract in inhibiting growth of Staphylococcus aureus and Escherichia coli. Paper disc method was used to study the hampering the ability of chlorophyll toward both bacteria, and the DPPH (1,1-difenil-2-dipikrilhidrazil) method was used to find out the antioxidant activity. The result showed that extract chlorophyll can inhibit the growth of both bacteria at the concentration of 100% with the inhibition diameter zone 2.170 cm and 2.100 cm respectively. Whereas the IC50 of the chlorophyll is 2350.3 ppm can be served as an antioxidants.   Key words: antibacterial, antioxidants, chlorophyll, Caulerparacemosa (Forsskål) J.Agardh.

Abstract

Community structure of bivalvia and gastropods nearby water vegetation in Sentani Lake, Papua. Line’s transect and plot methods were used to determining the structure of mollusks and vegetation by using index, diversity, and dominance as parameters. Intentional measurement were carried out on several biological parameters of the waters at 10 sampling points. From the observation and measurments, we obtained seven types of mollusk with 5644 individuals, where six types are belong to gastropods and one type is a member of bivalves. The highest level of mollusk diversity was in Kwadeware, the highest at the inlet of the lake, and were dominated by Melanoides tuberculata and Pomacea canaliculata. In addition, vegetation found in the area observation were water hyacinth (Eichhornia crassipes), water spinach (Ipomoea aquatica), apu wood (Pistia stratiotes), genjer (Limnocharis flava), Hydrilla verticillata, seagrass (Potomogeton malainus), Nymphaea alba, Ceratophyllum demersum, and Nymphoides indica.Key words: gastropods; molluscs; vegetation; Sentani lake

Abstract

Fungi has several role for humankind, one of them is as food. The potency of wild edible mushrooms as a source of nutrition still need to be explored to support national food security. The aim of this study was to determine the composition of nutrient content of wild edible mushroom called Tambir that commonly be consumed as alternative food by local people in Jayapura, Papua. Samples were collected from the traditional market nearby Uncen campus in Waena, Jayapura, Papua. Micro Kjeldahl method was used to analyze the crude protein content, hexane-gravimetry for lipid, spectrophotometry for carbohydrate, and high performance liquid chromatography (HPLC) for amino acid composition. The results showed that this mushroom has 15.74 % crude protein, 2.59 % lipid, 50.17 % carbohydrate and 8 essential amino acids. The chemical value of those eight essential amino acids were more than 100 that means this mushroom has no amino acid limitation. Therefore, Tambir could be a good alternative protein source for local people. Key words: fungi, proteins, local knowledge, Tambir, Papua.

Abstract

Allupurinol is a medicine to treat gout especially to minimize the quantity of uric acid inside human blood. This study aims to understand the quality of gout drugs particularly the standard of Allupurinol between generic 300 mg and others Allupurinol tablet in retails at Jayapura Regency. Survey method has been used and samples were collected from Sentani district, Jayapura Regency. Spectrophotometer was used to determine the rate of Allupurinol. The results proved that in Allupurinol generic (A) the rate was 100.28% and generic (B) was 99.044%. For others tablet non generic with naming (a) the rate of Allupurinol as 99.46% while (b) was 100.77%. It means the averages of Allupurinol contain in generic was 99.6% and non-generic was 100.12%. According to Indonesian Farmakope Manual 4th Edition, the rate of Allupurinol contains in a drug must be 93.0% for minimum and can’t be more than 107.0%. Our study shown that Allupurinol generic with (A) label and non-generic with (b) label are based on the Indonesian Farmakope Standard 4th Edition.  Key words: Tablet Allopurinol, Penetapan generik, Jayapura.

Abstract

The aims of the study are to evaluate bioactive compounds, antibacterial and cytotoxic potential of D. lasianthera. This orchid grows well all over New Guinea Island as an ornamental plant because of their beautiful flowers. Orchids also known rich of its phytochemical compounds which already used as a traditional medicine in many countries around the world. However, research in pharmacological fields is still limited. In this study, leaves and stem of D. lasianthera were powdered and extracted with ethanol followed by fractionated using n-hexane, ethyl acetate, and ethanol solvent. Extract as well as fraction were tested for phytochemical screening and determined antibacterial activity against Escherichia coli and Staphylococcus aureus using Disc Diffusion Method. Brine Shrimp Lethality Test (BSLT) was used to observe cytotoxic potential of leaves and stem extract and fraction at 10, 50, 100, 250, 500, 750, and 1000 ppm. The results showed that the leaves and stem extract contained tannin and alkaloids, separately. The ethanol extract of D. lasianthera showed 7.35 mm (leaves) and 7.43 mm (stem) inhibition zone against S. aureus Furthermore, the maximum inhibition zone of ethanol fraction of leaves and n-hexane fraction of stem were 8.42 mm and 8.10 mm separately. The LC50 of stem extract and fractions in these study were 699.3 ppm (ethanol extract), 602.1 ppm (ethyl acetate), 329.6 ppm (n-hexane fraction) and 676 ppm (ethanol fraction), whereas for leaves, only ethyl acetate fraction has toxict activity with an average LC50 833.2 ppm. Key words: D. lasianthera; phytochemical screening; antibacterial; cytotoxicity; Papua.

Abstract

Study on the water quality of Sentani Lake has been carried out during October–November 2012 at 3 stations of observation. Observation and retrieval of macrobenthos have been done by using combination between line transect method and square plot. While for physical and chemical  characteristics of water is measured in site and at laboratory. Index of variance Shannon-Wiener, Evenness and Density as well as Pollution Index were used to analyse the physical and chemical  characteristics of water. Result of this study showed that Telaga Maya  area (station 3) has  the highest variation of macrobenthos with H’ value= 2.12 while the lowest variation (H’= 1.66) was found around Ekspo River (station 2). Based on population density of macrobenthos, Pilsbryoconcha exilis which collected in the area of Jembatan Dua River (station 1) was found as  the highest population with density 9.26 ind/m2, while the lowest population density was Melanoides tuberculata with density 0.93 ind/m2. It can be concluded that based on the value of variance index and Pollution Index, the water quality of Sentani Lake belong to low-medium polluted category.Key words: water quality, macrobenthos, sentani lake and Jayapura.

Abstract

Matoa (Pometia pinnata) is parts of Sapindaceae family widely spread in tropical areas, especially in Papua. Several studies mentioned the use of its parts as antioxidants and contain phenolic compounds and flavonoids. This study aim(s) to conduct a phytochemical screening test as well as a cytotoxic test for Pometia pinnata leaves fraction using the BSLT method. The research series began with the extraction process using 96% ethanol, then fractionated using three solvent variations: n-hexane, ethyl acetate, and ethanol. Further, the phytochemical screening test was carried out for cytotoxic testing in order to see the mortality rate of shrimp larvae Artemia salina Leach. The result of the phytochemical showed that the secondary metabolite compounds on the ethanol and ethyl acetate fraction were alkaloids, flavonoids, saponins, tannins, and terpenoids compounds meanwhile the n-hexane fraction contained terpenoids. The results of cytotoxic testing BSLT showed the LC50 value in the n-hexane was 7029.10 ppm, 406.07 ppm for ethyl acetate and 614.47 ppm for ethanol. Thus, it could be said that ethyl acetate and ethanol fractions were classified as a toxic category, while n-hexane was classified as a non-toxic category.Key words: matoa; fraction; phytochemical screening; cytotoxic.

Abstract

Endomycorrhiza palys important role in nutrient uptake of agricultural plant. The purpose of this study was to investigate the presence of endomycorrhiza associated with non-legume plants, i.e.:Zea mays L., Solanum lycopersicum L., Capsicum frutescens L., Brassica oleracea L. and Brassica juncea (L.) Czern,  in agricultural area in Koya Barat, Jayapura. Survey was done before collecting root and soil samples. The root of those non-legume plants were cleaned and stained using method developed by Kormanic and Mc.Graw. Wet seaving method was done to analyze soil samples. The result showed that those non-legume plants were infected by endomycorrhiza. The highest infection percentage was on corn (Zea mays L.), while the lowest was on cabbage (Brassica oleracea L.). Furthermore, from spore identification, 14 species of endomycorrhiza were found on the rhizosphere of those plants which were grouped into genus Glomus (5 species), Gigaspora (2 species), Acaulospora (2 species) and Scutelospora (1), however 4 species were remain unidentified yet. Therefore further study should be done to elucidate this problem.Key words: endomycorhiza, exploration, non-legum, Koya Barat, Jayapura.
