Abstract

Kokosanolide A (1), from the seeds of Lansium domesticum Corr. cv Kokossan, has been shown strong cytotoxic activities (IC50 = 8.62 μg/mL) against MCF-7 breast cancer cells. The aim of this work was to study the molecular interactions of kokosanolide A and kokosanolide C with the Estrogen Receptor α (ERα) using computer-aided drug design approaches. Molecular docking using Autodock Vina (open-source software PyRx 0.8) was employed to explore the modes of binding of kokosanolide A (1) and kokosanolide C (2) with ERα. Compounds 1 and 2 showed strong bond-free energy (-8.8 kcal/mol and -8.7 kcal/mol) to ERα. These two compounds have a molecular mechanism to inhibit ERα in breast cancer cells.

Abstract

Environmental pollution around the world in the last 10 years has continued to increase with increasing industrial growth. Environmental pollution has reduced the availability of clean water. Clean water is the main thing for all living things on this earth. Therefore, environmental pollution that occurs in the clean water crisis must be handled as soon as possible and reduced by various effective policies. The main focus of this research is to analyze heavy metal pollution on the surface of the water, clean water, and sediments caused by industrial activities. This research is a field study by taking samples directly at the location and then analyzing them in the laboratory. The results of the analysis showed that the highest heavy metal Fe on the water surface was found in Bireuen Regency by 0.28% compared to Lhokseumawe City with 0.0029% and Aceh Utara 0.0029%. Meanwhile, the highest heavy metals contained in clean water were recorded in Bireuen and Lhokseumawe City at 0.007% and 0.0064%, respectively. The results of the heavy metal analysis in the highest sediment were found in North Aceh at 42.93% compared to 8.02% and 18.12% for Bireuen and Lhokseumawe. Overall, the analysis carried out in this study shows that the surface water, clean water, and sediments have been contaminated by heavy metals. Meanwhile, the quality standard for surface water is 0.3 mg/l, net ai is 1.0 mg/l, and sediment is 20bj.

Abstract

Green synthesized nanomaterials have been widely developed because of their less toxicity, low energy process, environmentally friendly, effective, cheap, and pollution-free. Green synthesis of silver doped titanium dioxide nanoparticles (Ag/TiO2 NPs) was carried out using Mirabilis jalapa plant extract. The plant extract was used as a reducing agent. The functional groups, morphology, and crystalline structure of as-synthesized Ag/TiO2 NPs were investigated by FT-IR, FESEM, and XRD. Analysis by FESEM confirmed that the morphology of Ag/TiO2 NPs is spherical with an average size of ~ 400 nm. Crystallite size for the Ag/TiO2 NPs was calculated by the Scherrer formula dan the average size found to be in the range of 15.72 nm. The result of XRD analysis showing the fcc structure for metallic silver and TiO2 particles in the anatase and rutile phases.

Abstract

Ramie (Boehmeria nivea) is one type of plant used to produce high quality cellulose fiber. Its fiber is convertible to cellulose acetate (CA), which is commonly used as membrane’s material. Dense CA membrane is used in gas permeation process and the performance is improvable by mineral blending and annealing. This study aims to determine the characteristics of modified CA membranes and the performance in separation of CO2/CH4 gases. Membrane preparation was carried out by using phase inversion technique, while annealing was carried out at 60oC. Furthermore, NaA zeolite was added with various concentrations, which include 5, 10, 15 and 20%. Gas permeation was carried out at 4 barrer for an hour. The performance of the membrane was measured based on its permeability and selectivity. The results showed that CA membrane with 10% zeolite yielded the best performance with CO2 and CH4 permeability of 140.55 and 22.25 barrer respectively and selectivity of CO2/CH4 6.32.

Abstract

Dadih is naturally fermented buffalo milk in bamboo tubes and known to have antioxidant, antibacterial, and antihypertensive activity. Lactic acid bacteria in dadih can use as a starter to produce yogurt. The study aimed to produce yogurt of cow's milk with dadih as a starter, to determine the antioxidant activity and amino acid composition. Dadih is added with concentration variation of  2.5; 5; 7.5 and 10% (v/v) and it fermented for 48 hours at room temperature. Lactic acid bacteria cell counts of dadih were calculated by the total plate count method. Yogurt was tested by organoleptics with 33 panelists and proximate analysis water, ash, fat and protein contens based on the Association of Official Analytical Chemist (AOAC) in 2005. Antioxidant activities of yogurt were tested by DPPH method (1,1-diphenyl-2-picrylhydrazyl). Amino acid composition of yogurt was analyzed using ultra performance liquid chromatography (UPLC). The research result of amount of LAB on dadih are 1.01 x 1011 CFU/mL, has fulfilled the requirements of SNI 2981: 2009. Yogurt produced with the addition of dadih by 10% (v/v) is most accepted by panelists. The highest antioxidant activity was obtained by yogurt with 2.5% addition of dadih with IC50 value of 78.28 ppm. Yogurt almost contain all essential and non-essensial amino acids including tyrosine and phenylalanine as the antioxidants. Measurement of water, ash, protein and fat content in the sample meets the requirements of SNI 2981: 2009.

Abstract

This paper reports the synthesis of hydroxyapatite from Geloina coaxans shell and sodium dihydrogen phosphate as precursors by facile and simple method. Firstly, Geloina coaxans shell calcined at 1000 oC for 12 hours to produce lime or CaO. The chemical composition of Geloina coaxans shell calcined was analyzed using the XRF technique, and the result shows that the main component of Geloina coaxans shell calcined was calcium. Synthesis of hydroxyapatite has been done with stirring and aging time parameters and molar ratio of precursors was 1.67.  The XRD patterns showed a specific peak of hydroxyapatite that appeared at 2θ=31.8 o and based on the XRD results on the aging time indicated that the rate of growth of HAp crystals was better and faster at aging for 12 hours. The crystal size of HAp obtained was 37 nm. The FTIR spectra of hydroxyapatite showed characteristics of hydroxyl and phosphate bands in sample spectra. The morphology of hydroxyapatite synthesized also was examined by SEM and the results showed spherical morphology.

Abstract

Metode alternatif untuk mengatasi pencemaran logam berat adalah biosorpsi menggunakan biomassa sebagai adsorben. Telah dilakukan penelitian kajian adsorpsi campuran Pb2+ dan Zn2+ pada biomassa Imperata cylindrica sebagai adsorben. Tujuan penelitian adalah mengetahui kemampuan biomassa mengadsorpsi Pb2+ dan Zn2+. Preparasi biomassa dilakukan dengan aktivasi menggunakan asam nitrat dan amonium hidroksida. Adsorpsi dilakukan dengan sistem batch. Parameter yang diukur adalah pH optimum, waktu kontak optimum, kapasitas adsorpsi dan recovery ion logam. Analisis kadar logam dilakukan dengan menggunakan Spektrofotometer Serapan Atom (AAS). Hasil penelitian menunjukkan pH optimum adsorpsi Pb2+ dan Zn2+ masing-masing pada pH 5 dan pH 6. Waktu kontak optimum adsorpsi Pb2+ dan Zn2+ masing masing pada 40 menit dan 30 pertama. Kapasitas adsorpsi Pb2+ dan Zn2+ pada konsentrasi awal 10 ppm masing-masing adalah 90,95% dan 43,60%. Recovery Pb2+ dan Zn2+ masing-masing 84,45% dan 57,13%.

Abstract

Schiff bases of 2,4,5-triphenyl-1H­-imidazole ligand and Cu(II) complex has been synthesized and characterized by 1H and 13C NMR, elemental analyses, molar conductance, thermal analysis (TGA), magnetic moment measurement, IR and UV-Vis. On the basis spectral studied and analytical data, it is evident that the Schiff base acts as monodentat ligand coordinating via nitrogen atom. The result showed that Cu(II) complex has octrahedral geometry. The studied complex was tested for anticancer activity. The anticancer activity of Cu(II) complex is evaluated against liver Carninoma (HEPG2)  cell lines. These compound exhibites a moderate and weak activity against HCT 116 cell line with IC50 19.8 mM. DOI: http://dx.doi.org/10.15408/jkv.v0i0.3328

Abstract

Telah dilakukan penelitian tentang imobilisasi resin bekas pengolah limbah trans-uranium denganresin poliester tak jenuh untuk mengetahui kualitas blok polimer-limbah sebagai fungsi kandunganlimbah. Polimerisasi dilakukan dengan mencampurkan resin poliester tak jenuh dengan katalisdengan perbandingan katalis 1% dari jumlah resin poliester tak jenuh yang digunakan, kemudianditambahkan limbah cair transuranium simulasi. blok polimer-limbah yang terjadi diukur densitas,kuat tekan dengan alat Paul Weber, dan laju pelindihan dengan alat soxhlet pada 100 0C dan 1 atmselama 6 jam. Blok polimer dibuat dengan kandungan limbah 10, 20, 30, 40, dan 50 % berat. Hasilpenelitian menunjukkan bahwa semakin besar kandungan limbah maka kuat tekan blok polimerlimbahsemakin kecil, sedangkan laju pelindihannya semakin besar. Berdasarkan kuat tekan dan lajupelindihan, maka hasil terbaik diperoleh untuk blok-polimer dengan kandungan limbah 20 % dan 30%.

Abstract

Abstrak Penelitian tentang profil protein sarang burung wallet masih terbatas, terutama sarang burung walet dari Indonesia. Oleh karena itu, penelitian ini bertujuan untuk menganalisa profil protein dan asam amino sarang burung walet yang berasal dari daerah Painan, Kabupaten Pesisir Selatan, Sumatera Barat. Analisis protein dilakukan dengan menggunakan SDS-PAGE, sedangkan analisis asam amino dilakukan dengan menggunakan kromatografi cair kinerja tinggi (KCKT). Analisa ekstrak air sarang burung walet dengan SDS-PAGE menunjukan bahwa sarang burung walet terdiri dari 6 protein. Keenam protein tersebut mempunyai bobot molekul masing-masing 147.2 kDa, 142.6 kDa, 133.4 kDa, 73.3 kDa, 66.2 kDa dan 37.7 kDa. Dari analisa asam amino burung walet dengan KCKT didapatkan 16 asam amino yang terkandung dalam sarang burung wallet, yang terdiri dari 7 jenis asam amino esensial yaitu Histidin (2.31%), Leusin (3.84%), Treonin (3.82%), Valin (3.93%), Metionin (0.48%), Isoleusin (1.80%), Fenilalanine (4.49%)  dan 9 asam amino non esensial yaitu Asam Serin (4.56%), Aspartat (4.48%), Arginin (3.93%), Lisin (2.34 %), Prolin (3.64%),  Asam glutamate (3.65%), Glisin (1.87%), Alanin (1.31%), Tirosin (3.92%). Serin merupakan asam amino dengan kadar tertinggi (4.56%), diikuti dengan Fenil alanine (4.49%) dan Asam aspartate (4.48%). Kandungan asam amino ini sedikit berbeda dengan kandungan asam amino sarang burung walet dari daerah dan negara lain. Kata kunci: sarang burung walet, protein, asam amino Abstract Study on protein profile of bird nest is still limited particularly protein profile of bird nest from Indonesia has not been reported. Therefore, this study was aimed to analyze protein profile and amino acid composition of bird nest from Painan, Pesisir Selatan Distric, West Sumatra. Protein analysis was performed by SDS-PAGE, and high performance liquid chromatography (HPLC) was used for amino acid analysis. SDS-PAGE analysis showed  six bands, which molecular weigh of 147.2 kDa, 142.6 kDa, 133.4 kDa, 73.3 kDa, 66.2 kDa and 37.7 kDa, respectively. On the other hand, HPLC analysis demonstrated that bird nest was composed of 16 amino acids. Seven of them were essential amino acids; histidine (2.31%), leucine (3.84%), threonine (3.82%),  valine (3.93%), methionine (0.48%), isoleucine (1.80%), phenylalanine (4.49%), and nine of them were non-essential amino acids; serine (4.56%), aspartic acid (4.48%), arginine (3.93%), lysine (2.34%), proline (3.64%), glutamic acid (3.65%), glycine (1.87%), alanine (1.31%), tyrosine (3.92%). Serine was the highest percentage of amino acid in the bird nest (4.56%), followed by phenylalanine (4.49%) and aspartic acid (4.48%). Composition of amino acid in this bird nest was slightly different with composition of amino acid in bird nest from other area. Keywords : bird nest, protein profile, amino acids
