Abstract. Pratasik SB, Lalamentik LTHX, Manoppo L, Budiman J. 2022. Deep sea squid in Sulawesi Sea, North Sulawesi Province, Indonesia. Biodiversitas 23: 1774-1779. This study aimed to identify one of the deep sea squids caught in artisanal fisheries by traditional fishermen in Sulawesi Sea, North Sulawesi Province, Indonesia. Samples were collected using the vertical hand line of traditional fishermen. The specimen was identified using Cytochrome c oxidase subunit 1 (CO1). For morphometric characteristics, all body parts were measured, and each part was compared with mantle size. Results showed that the specimen was identified as purpleback flying squid Sthenoteuthis oualaniensis and had 99.35% similarity to S. oualaniensis (CO1) from China (NCBI - MN101944) with sufficient genetic diversity. Based on the body size, species recorded in the present study belong to the dwarf form, the smallest form of S. oualaniensis groups. The species has the following morphometric characteristics: Head length is 0.28 of mantle length, tentacle length is 1.158 of mantle length, and fin area is 0.5 of mantle length. The fourth arm is the shortest and the second arm is the longest. S. oualaniensis recorded in the present study belonged to dwarf form living in equatorial waters. This finding may contribute to the list of marine resources of fisheries in Indonesian waters.

Carsono N, Desiana N, Nurrizqi FM, Elfakhriano IF, Anas, Sari S, Kusumiyati, Ohsawa R, Shimono A, Ezura H. 2022. Evaluation of agronomic and fruit quality traits of miraculin transgenic tomato. Biodiversitas 23: 2004-2009. Tomato cv. Moneymaker has been inserted with miraculin gene which can modify sour taste becomes sweet in the human tongue and, thus, it has potential as a sweetener. Evaluation of agronomic and fruit quality traits for this special transgenic tomato is a major concern that should be performed as substantial equivalence. The study's objective was to compare agronomic and fruit quality traits of transgenic miraculin tomato with those of its wild-type. The experiment employed a randomized block design, one genotype factor with two levels (transgenic vs. non-transgenic tomato cv. Moneymaker), and each was four replicates. Data were subjected to the Student t-test. The study revealed no significant differences between the transgenic tomato plants cv. Moneymaker and the wild type for important agronomic traits such as plant height, stem diameter, leaf area index, fruit weight, fruit weight per cluster, fruit weight per plant, fruit diameter, number of fruits per cluster, number of fruits per plant, and harvest index of miraculin. Some fruit quality traits such as fruit shelf-life, fruit hardness, total soluble solids, and content of glucose, sucrose, and fructose were equivalent to those of its counterpart, suggesting that miraculin transgene did not affect the expression of the observed traits. The miraculin transgenic tomato traits were more likely identical to those of wild-type except for the existence of the miraculin transgene. This will open the possibility for this special transgenic tomato to undergo further environmental safety assessment in the field test facility as a part of the biosafety assessment of genetically modified organism (GMO).

Visual soil structure quality and its correlation to quantitative soil physical properties of upland rice site in Falcataria moluccana agroforestry system. Biodiversitas 23: 1894-1903. Sengon and upland rice are plants that have the potential to be combined in the agroforestry system. However, its development often results in poor quantitative soil physical properties. Measuring the soil’s physical properties is expensive and complicated to do by some people. This study aims to measure the visual soil structure quality from different land covers and calculate its correlation to the parameters of the soil’s physical properties. The research was conducted in the Cikabayan forest and used the purposive sampling method to take soil samples. The research was conducted by comparing nine land covers in the form of agroforestry and monoculture developed in organic and conventional agriculture with tropical rainforests as natural vegetation. Three samples were taken for each land cover type and analyzed with a completely randomized design. The research showed that the vegetation of tropical rainforest generated the best score on all visual soil structure quality measurement methods (VESS, GrassVESS Sq and GrassVESS Rm). The measurement of soil physical properties at natural vegetation of tropical rainforests generated the highest score on the porosity (57.303%). Meanwhile, monoculture-conventional land generated the highest bulk density (1.373 g.cm-3). The VESS method had the highest correlation with soil physical properties compared to other visual soil structure quality measurement methods. Thus, natural vegetation is the best type of land cover compared to all cultivated land and the VESS method is the most appropriate method for estimating soil physical properties.

Food, medicinal plants, and homemade beverages, used as a response to the pandemic in Ethiopia. Biodiversitas 23: 2146-2155. SARS-CoV-2, which is responsible for an infectious disease called COVID-19, significantly impacted the global community, including Ethiopia. Meanwhile, people responded to the disease in various ways using available household materials. These include food, medicinal plants, and household beverages used for preventing and curing symptoms of COVID-19. Thus, the present study was initiated to assess these foods, medicinal plants, and beverages used in Ethiopia during the COVID-19 pandemic. This study used a web-based survey and phone call to collect data related to food, medicinal plants, and beverages used during COVID-19 by emphasizing medicinal plants. The questionnaire was created through Google Form and circulated through social media such as Facebook, Telegram, and Whatsapp. A total of 680 respondents took part in the survey. Relative citation frequencies (RFC) were calculated to assess popular medicinal plants used to prevent or mitigate the disease. A total of 53 medicinal plants used to prevent and cure COVID-19 distributed to 24 families were recorded. Leaves (30%) were the most frequently utilized plant part. The respondents indicated that they were using medicinal plants for COVID-19 prevention at least once a day, accounting for (41.3%). Most of them recommended using medicinal plants to prevent the disease. Relative frequency of citation (RFC) indicated that Zingiber officinale Roscoe was the most popular medicinal plant used during the pandemic. Apart from medicinal plants, foods and beverages were also frequently used during the pandemic. The majority of respondents (49.6%) mentioned that local communities were their source of information. Respondents also indicated the use of the medicinal plant has increased during the first three months of the COVID-19 pandemic claiming that they have the potential to prevent and cure COVID-19. However, this should not be considered a panacea since the effectiveness of these medicinal plants, foods, and beverages has not been confirmed.

Abstract. Retuerma-Dioneda A, Alejandro GJD. 2022. Inventory of Annonaceae in Abasig-Matogdon-Mananap Natural Biotic Area, Camarines Norte, Bicol, Philippines. Biodiversitas 23: 2213-2224. The Abasig-Matogdon-Mananap Natural Biotic Area (AMMNBA) is a protected area home to endemic plants; however, Annonaceae remains understudied. The study provided a checklist of Annonaceae species in the area and their taxonomic keys, endemicity and conservation status. Field surveys were conducted in January 2020 and July 2021 in San Vicente and Labo, Camarines Norte. The 110 species identified belong to 11 genera- Artabotrys, Drepananthus, Fissistigma, Friesodielsia, Goniothalamus, Meiogyne, Monoon, Phaeanthus, Polyalthia, Popowia, and Uvaria and five (5) tribes: Ambaviodeae, Annoneae, Miliuseae, Uvarieae, and Xylopieae. Four (4) endemic species were recorded: Friesodielsia lanceolata, Polyalthia luzonensis, Polyalthia lanceolata and Goniothalamus elmeri. This study provides baseline information on Annonaceae species in AMMNBA for appropriate conservation policies and proper management of the potential economic benefits.

The growth of Paraserianthes falcataria at three different plant ages and soil thickness classes on reclamation sites of post-coal mining areas in East Kalimantan, Indonesia. Biodiversitas 23: 1930-1937. Sengon (Paraserianthes falcataria (L.) Nielsen syn. Falcataria moluccana (Miq.) Barneby & J.W. Grimes) is a fast-growing species commonly used for reclamation of post-coal mining areas in East Kalimantan. The study aimed to determine the impact of different thicknesses of topsoil applied at some reclamation sites after coal mining activities on the growth of P. falcataria. Three coal mining companies were chosen as study areas, i.e., PT Bukit Baiduri Energi (BBE), PT. Mahakam Sumber Jaya (MSJ), and PT. Kaltim Prima Coal (KPC). The regolith soil layer at the reclamation site had three thickness classes (0-10 cm, 0-30 cm, 0-100 cm), and the P. falcataria growing on reclamation had three ages, i.e.,< 2 yrs old (initial growth phase), 2-5 yrs old (tending phase), and > 5 yrs old (independent growth phase). The number of surviving plants, plant height, and plant stem’s diameter were recorded, and the data were analyzed using ANOVA and the least significant difference at 5% level. The results showed that no significant difference in the number of trees among different ages of P. falcataria plantation was found at MSJ. In contrast, at BBE and KPC the number of surviving trees decreased significantly with the increasing plant age. The growth of the plant (height, diameter) increased significantly along with plant age in all sites. The interaction between the top soiling thickness factor and plant age factor produced no different result in plant growth except for plant height at BBE. The P. falcataria plants had not been able to lower the surface soil density in the land reclamation area.

Constructing and expressing acyl-homoserine lactone lactonase (aiiA) gene for enhancing Solanum tuberosum resistance against soft rot disease. Biodiversitas 23: 1780-1787. Expression of the genes associated with pectinase production in soft rot disease of Dickeya spp. are regulated by quorum sensing mechanism using acyl homoserine lactone(AHL). Degradation of AHL using AHL lactonase can be developed to reduce pectinase production in Dickeyaspp.The aiiA gene encodes AHL lactonase from Bacillus spp. This study aims to construct binary vector of inserted aiiA gene (pCAB10) from Bacillus cereus B10 and express the aiiA gene in potato Medians cultivar genome. The aiiAgene was cloned into binary vector and transformed into Agrobacterium tumefaciens LB4404. The Agrobacterium-mediated genetic transformation was conducted with co-cultivation technique.The transgenic plant was confirmed through PCR andselected through kanamycin and soft rot disease resistance to Dickeya dadantii A3. The result showed that aiiA gene was successfully integrated top CAB10 and introduced to A. tumefaciens LB4404. The percentage of pCAB10 transformation was 57.6%, while there generation efficiency was 11.9% with 50 mg/L acetosyringone addition. Four transgenic potato Medians cultivar exhibited significantly higher resistance against soft rot disease than non-transgenic plants. Mahl1 clone had the highest resistance to D. dadantii with soft rot disease incidence as 7.1% and percentage of soft rot disease suppression as 92.85%.

Half diallel of F1 tomato hybrid and its double cross-compatibility. Biodiversitas 23: 1813-1821. Breeding program for improvement of tomato fruit can be done through diallelic crosses genetic combinations. Therefore, genetic evaluation is needed to meet the breeding objective. This process can be combined with crosses between F1 to produce high diversity. However, evaluating the crosses' compatibility is required to increase the effectiveness of the assembly process. This study aimedto assess the half-diallel crosses of F1 tomatoes in the field and determine the most compatible pair cross combinationsin forming double and three-way F1 populations. This study was carried out in the Experimental Farm of the Faculty of Agriculture, Hasanuddin University. The field evaluation was arranged ina randomized complete block design with ten genotypes, while the compatibility evaluationwas carried out using 6 F1 half diallel as the parents. The results showed that the Mawar and Chung varieties were better parents in the crossings. Furthermore, the results showed that F1 K/M (the best yield component), M/C (the best TDS and Lycopene traits), and BC/K (the genotype has good traits of yield component, TDS and Lycopene) were the best single F1 crosses that could be inherited to the F2 generation. In contrast, F1 K/M and F1 K/C crosses were the best male and female parents, respectively. The recommended double-crosses include K/C//BC/M, and BC/C//K/M, while for the F1 unbalanced three-way cross was K/C//K/M.

Genetic parameters for resin production of Pinus merkusii progeny test collected from three landraces in Banyumas Barat Forest District, Indonesia. Biodiversitas 23: 2010-2016. Pinus merkusii Jungh. et de Vriese is a native pine species in Indonesia that is used in the production of essential and economically valued products, such as wood and resin. The resin productivity can be increased significantly via breeding programs. The objective of this study is to analyze the genetic parameters of resin productivity based on the progeny performance in three landrace seeds sources, namely, Sumedang seedling seed orchards (SSOs), Jember SSO, and East Java seed source. The drilling method was used for tapping trees, and then resin production was measured individually. Additional data on height, diameter, and branch-free height were also collected. Based on family variance component analysis data of resin production, it was noted that Sumedang SSO, Jember SSO, and East Java seed sources have relatively high genetic variations of 4.48%, 15.77%, and 3.80%, respectively. These values were then followed by a moderate family heritability value for all of them (0.451-0.551). A high value of individual heritability estimates was obtained for Jember SSO (0.658), and moderate values were found for Sumedang SSO (0.182) and East Java seed sources (0.158), respectively. The estimated genetic gains were calculated for resin production using selection intensity levels at 10%, 25%, and 50%. The result indicated that the Jember SSO landrace obtained the highest genetic gains at 17.55%, 12.73%, and 7.99%, for the above-mentioned intensity levels, respectively. In contrast, the lowest one was found in the Sumedang SSO landrace.

Diversity report of freshwater gastropods in Buton Island, Indonesia. Biodiversitas 23: 1938-1949. This study was located in Buton Island consisted of 5 administrative districts/cities (Baubau City, Buton District, South Buton District, Muna District and North Buton District) and was conducted for 1 year (August 2019-August 2020). The output is to reveal the potential richness of the biodiversity of freshwater gastropods as a first step to optimize the utilization of freshwater gastropod resources in the Buton Islands. Determination of sampling location for gastropods was carried out by purposive sampling method and the selection of gastropod samples using simple random sampling method at a predetermined location based on the presence of gastropods. Gastropod samples (epifauna/infauna) were collected manually without special fishing gear with gloves, paralone pipes (3.5 inches), and filter (1 mm). The gastropods found in Buton Island consist of 18 genera and 40 species. Freshwater gastropods spread over 8 families, namely Cyclophoridae, Planorbidae, Lymnaeidae and Thiaridae. Among the species, there are several invasive alien species (IAS) such as Tarebia granifera, Melanoides tuberculata and Achatina fulica. These three species always dominate the habitat space where they are found. Most gastropod communities in 5 districts/cities live in fast-flowing rivers with rock, gravel and sand substrates. Only a few of them are found in artificial inland waters such as rice fields, embankments, dams and drainage. This study became a first step to optimize the utilization of gastropod resources and sustainable management, especially related to the conservation of native species from the threat of IAS.
