Biodiversitas 21: 636-641. Since earlier times, dye has an important role in human life, especially related to the ritual practice. Mineral material was first used as natural dyes. In order to inventory the uses of plants as a dye and natural color binder by the local community of Pringgasela, ethnobotany research was done. This research had been done by interviewing the "penyisik" of traditional woven fabric craftsmen and direct observation in the field. The results showed 13 plant species used as a dye and nine species as color binder. Parts of less utilized plants were used. It is hoped that this research can prevent synthetic dyes that have been circulating widely, which has a potential hazard and cause environmental pollution.

Abstract. Sabdono A, Trianto A, Radjasa OK, Wijayanti DP. 2019. Antagonistic activity of bacteria isolated from coral Acropora sp of Karimunjawa Islands, Indonesia against Acroporid White Syndrome. Biodiversitas 20: 1350-1355. Acroporid White Syndrome (AWS) disease, causing coral death and mortality, has come out as a serious threat to coral reefs in the Karimunjawa Islands, Java Sea. In the present study, the bacterial community associated with the Acroporid were assessed for their antagonistic activity against bacteria associated with AWS disease. A culture collection of 43 bacteria associated with coral Acroporid was established by plating on Zobell’s 2214E. Isolates were screened using the overlay method. The results showed that five isolates (11.62 %) were able to inhibit the growth of the AWS isolates. These isolates were tested further for their consistency of antagonistic activity by using disc-agar diffused method. Based on their consistency, AU5 isolate was selected for study at advance. Polyphasic and phylogenetic analysis were used to characterized AU5 isolate. The DNA sequence analyses demonstrated that this bacterial isolate affiliated with Firmicutes. The AU5 isolate showed high 16S rDNA sequence identity to Virgibacillus salarius strain muzPC213 (99%). The results demonstrated the existence of bacterial coral symbionts with AWS antipathogenic activity on the coral tissue, possibly performing as biocontrol agent to protect the coral Acroporids against AWS outbreaks.

Growth parameter, mortality, recruitment pattern, and exploitation rate of white shrimp Penaeus indicus on the northern coast of Central Java, Indonesia. Biodiversitas 20: 1318-1324. The aims of this study were to know the growth parameters, first lenght captured (Lc), mortality, recruitment pattern, and the exploitation rate of white shrimp (Penaeus indicus H.Milne Edwards, 1837) in the Western Coastal North Central Java Sea, Indonesia. Samples were collected from May 2016 to July 2017 using survey method and direct observations on fishing unit areas. The results showed that the relationship of the carapace length and weight is allometric. Based on the estimates of von Bertalanffy growth equation to the carapace length, the growth parameters of CL? and K values in male were 37 mm and 0.97, while in female were 38.5 mm and 0.85. While, the rates of total mortality (Z), natural mortality (M), and fishing mortality (F) were 3.5, 1.67, and 1.84 for males, and 5.03, 1.51, and 3.52 for females, respectively. The exploitation rate (E) of male shrimp was 0.52 and of female was 0.7 indicating that female shrimp get higher capture pressure than the males. It also showed that the rate of exploitation has to exceed the optimum sustainable yield (E>0.5). Time recruitment occurred throughout the year, with two peak seasons, i.e., in April-June and September-November. Moreover, the length of the carapace during recruitment was 15-20 mm. While the carapace length of the first caught (CL?50) was 18.3 mm for male and 17.6 mm for female indicating that the P. indicus caught was still in the small size, as well as below the first mature gonad (CLm50 = 22-39 mm). This is because the mesh size of cod-end is 0.75 inches or about 2 cm.

Abstract. Fauzi, Widodo H. 2019. Short Communication: Aphrodisiac plants used by Dayak Ethnic in Central Kalimantan Province, Indonesia. Biodiversitas 20: 1859-1865. Sexual dysfunction in men might render psychological disorders and household harmony. The prevalence increases with age and other risk factors such as systemic diseases and psychological factors. A community with no exception certain ethnic groups uses plants to fulfill various needs, including to treat sexual disorders in men. This paper aims to explore medicinal plants that are often used for aphrodisiacs by Dayak ethnic in Central Kalimantan Province, Indonesia. The method was carried out through reviewing literature, searching for various references and analyzing data of the exploration of ethnomedicine and community-based medicinal plants in Indonesia held in 2012 and 2015. Of the 9 Dayak ethnic groups surveyed, only four ethnic groups (Bawo, Taboyan, Tomum and Siang) posses an aphrodisiac formula. The plants used by ethnic Dayaks for aphrodisiacs are Eurycoma longifolia Jack., Luvunga sarmentosa (Bl.) Kurz., Rennellia elliptica Korth., Mimosa pudica L., Lepisanthes amoena (Hassk) Leenh., Fordia splendidissima (Miq.) Buijsen., Imperata cylindrica (L.) Raeusch., Ficus deltoidea Jack., Koilodepas bantamense Hassk., Eusideroxylon zwageri Teijsm. & Binn., Gnetum sp., Caryota sp. and Calamus sp.

Bacteria associated with tunicate, Polycarpa aurata, from Lease Sea, Maluku, Indonesia exhibiting anti-multidrug resistant bacteria. Biodiversitas 20: 956-964. Tunicate is a rich secondary metabolites producer with various biological activities whether as an original producer or produced by the associated microorganisms. In this study, a total of 11 tunicate specimens were identified as Polycarpa aurata with four color variations based on morphological characteristic and COI gene identification and BLAST analysis. The P. aurata associated-bacteria were isolated and tested for antimicrobial activity against multi-drug resistant (MDR) bacteria. A total of 86 axenic isolates were successfully purified. Furthermore, nine isolates (10.5%) exhibited antibacterial activity on preliminary screening. Nine prospective isolates were fermented in respective medium (Zobell 2216, modified M1 or modified ISP2 media) then extracted using ethyl acetate. The ethyl acetate extracts from liquid fermentation were tested against MDR Escherichia coli, MDR Bacillus cereus, Methicillin-Resistant Staphylococcus aureus (MRSA) and Methicillin-Sensitive and Staphylococcus aureus (MSSA). As a result, seven isolates (8.1%) still retained the activity at the extract concentration 150 µg/disk. Molecular analysis based on 16S rDNA sequencing revealed the most active isolates, TSB 47, TSC 10 and TSB 34 identified as Bacillus tropicus, Vibrio alginolyticus and Virgibacillus massiliensis, with BLAST homology 99%.

Variability of agronomic characters related to resistance to stem canker (Phytophthora palmivora) on duku (Lansium domesticum) along Batanghari River, Sumatra, Indonesia. Biodiversitas 20: 1127-1132. Duku (Lansium domesticum Correa) is a tropical favorite fruit in South East Asia, including Indonesia. However, currently stem canker disease caused by Phytophthora palmivora Butler (Butler) has been a serious problem on duku plant in Indonesia, especially in Jambi Province. Since duku is vegetatively propagated, selecting resistant mother trees is an effective approach to overcome the disease. This research was aimed to identify mother trees and their progenies that resistant to stem canker pathogen. The research was conducted in two stages. The first one was morphological identification of healthy-looked mother trees grown at four locations: three in Batanghari and one in Muaro Jambi District. The second stage was analyzing resistant seedling progeny of mother trees identified at the first stage. At the second stage, 3 month old seedlings were inoculated with P. palmivora, isolated from rhizosphere of trees grown at the researchs location using Completely Randomized Design. The research found 19 healthy mother trees, and showed similarity index of 0.40 based on eleven identified agronomic characters of mother trees. Cluster analysis revealed 2 identified clusters of mother trees with the identified clusters were independent of plant growth locations. Seedling resistant analysis showed that there were only 5 seedlings expressing resistant traits. Correlation analysis showed that the resistant seedlings were originated from mother trees which tend to have longer leaves, more fruits per branch, more yields, and bigger canopies. This information of the characters can possibly become future reference to choose resistant plant as sources of mother trees to develop and maintain duku populations along the Batanghari River.

Abstract. Hartini S. 2019. Orchids Diversity in the Sicikeh-Cikeh Forest, North Sumatra, Indonesia. Biodiversitas 20: 1087-1096. Sicikeh-cikeh forest includes three forest areas, namely Adian Tinjoan Customary Forest, Adian Tinjoan Protection Forest, and Taman Wisata Alam Sicikeh-cikeh. Typical vegetation of this area is mountain forest with large diversity of plant species. Among the species, orchid is one of the potential plants found diversely in this location and has not yet been recorded. The exploration activities were conducted at Sicikeh-cikeh forest. Orchid exploration was conducted to collect living plants for ex situ conservation purpose. Orchid inventory, to record orchid diversity in this area, was based on plant collection by purposive random sampling. The results of the study recorded approximately 102 different species from 30 genera of orchids in this area. Typical epiphytic and terrestrial highland orchids were found and very common in Sumatra. Among the species found are endemic Sumatra, such as Coelogyne brachygyne, Coelogyne salmonicolor, Dendrobium kruiense, Epigeneium pulchellum, Thrixspermum gombakense, Corybas stenotribonos and Paphiopedilum tonsum. Interesting terrestrial orchids include Kuhlhasseltia javanica, Neuwiedia zollingeri var. javanica, Paphiopedilum tonsum, Phaius callosus, Corybas stenotribonos, Calanthe aurantiaca, Calanthe chrysoglossoides, and Calanthe pulchra. While interesting epiphytic orchids include Bulbophyllum absconditum, Bulbophyllum coloriferum, Coelogyne brachygyne, Epigenium pulchellum, Eria bractescens, Eria pachystachya, Pholidota gibbosa, and Thrixspermum amplexicaule.

Differences among clinical isolates of Pseudomonas aeruginosa in their capability of forming biofilms and their susceptibility to antibiotics. Biodiversitas 20: 1450-1456. Pseudomonas aeruginosa is an important nosocomial pathogen capable of causing both acute and chronic infections. The individuals of this bacterium have differences in their capability of forming biofilms.  Biofilm is a collection of bacterial cells attached to the tissue, coated by polysaccharides and extracellular matrix, enabling bacteria to become resistant to antibiotics, so infection is difficult to treat.  The aim of this study was to know the differences among clinical isolates of Pseudomonas aeruginosa in their capability of forming biofilms, and their susceptibility to some antibiotics. The bacteria were isolated from various patient samples at Dr. Moewardi Hospital Surakarta, Indonesia, from August to December 2017. The isolates were purified using single cell colony technique.  The bacterial identification and tests of susceptibility to many antibiotic were conducted using automatic equipment (vitex® 2). Test method of biofilm formation was done using the Tissue Culture Plate. The readings of results were monitored spectrophotometrically at a wavelength of 570 nm and repeated 8 times. DNA isolation was done using Qiagen DNeasy Blood and Tissue Kit, and gene identification (pelD and pslA) was done using PCR (Polymerase Chain Reaction) and visualization through electrophoresis.  The results showed that of the 64 isolates of P. aeruginosa from blood, sputum, urine, ear middle, urine catheter, pleural fluid, pus, stool, aspirate, and cerebrospinal, 22% were low in forming biofilm, 50% moderate, and 28% high. Isolates of P. aeruginosa were resistant to ampicillin, cefazolin, tigecycline, nirofurantoin, and cotrimoxazole, but sensitive to piperacillin, ceftazidime, cefepime, aztreonam, meropenem, amikacin, gentamicin, and ciprofloxacin. The genes of pelD and pslA were present in all P. aeruginosa isolates (low, moderate, dan high).   In conclusion, P. aeruginosa clinical isolates had different capability of forming biofilms and susceptibility to antibiotics. Isolates having high ability to form biofilm were relatively more resistant to many antibiotics. They were most sensitive to amikacin and resistant to ampicillin.  There was no difference in the presence of mop pelD and pslA among all isolates.

Spatial distribution of salinity, mud thickness and slope along mangrove ecosystem of the coast of Kupang District, East Nusa Tenggara, Indonesia. Biodiversitas 20: 1624-1632. Mangrove ecosystem is suffering from continuous damage in terms of quality as well as quantity. Rapid mangrove conversion triggered by the ever-growing human population and activities have affected them adversely and caused mangrove deterioration in the coastal areas of Kupang District, Indonesia. The decline of quality and quantity of mangrove environment has caused highly alarming effects, such as an increase in abrasion, decrease of marine products, and sea-water intrusion. Although various efforts have been deployed for the rehabilitation of mangrove forests, there are no significant results yet. One of the reasons is such efforts are not based on scientific data on suitability of the physico-chemical conditions of the mangrove habitats. The objectives of this study was to analyse the physico-chemical conditions, such as salinity, mud thickness and slope of five selected mangrove locations, namely Tarus, Mata Air, Tanah Merah, Oebelo and Merdeka in Kupang District by systematic sampling method and to prepare spatial distribution maps for each of these parameters. Salinity conditions showed a wide variation in the study locations, starting from 10 ppm to 38.33 ppm. Mud thickness ranged from 20.11 cm to 84.77 cm in the entire study area. The highest mud thickness of 84.77 cm was found in Tanah Merah, followed by Oebelo with the maximum thickness of 74.66 cm, Mata Air with 56.22 cm, Merdeka with 53.66 cm, and the lowest mud thickness was in Tarus with 53.55 cm. Mangrove locations in Kupang District are dominated with 2-3% slope. The study indicated that the habitat conditions in the coastal area of Kupang District are suitable for the growth of mangroves. This study will also provide the basis to plan future mangrove rehabilitation programs in the study area.

Abstract. Hanapiah MFM, Saad S, Ahmad Z, Yusof MH, Khodzori MFA. Assessment of benthic and coral community structure in an inshore reef in Balok, Pahang, Malaysia. Biodiversitas 20: 872-877. Inshore water reef closer to human activities are often neglected since they are not protected by marine park management. Little is known on how this reef responses to challenging environment in terms of abundance, diversity and benthic community composition. This study provides a quantitative assessment on the benthic community composition at 5 reef sites in Balok, Pahang, Malaysia and observed the ecological adaptation in the reef community towards turbid water environment. Balok reef has 39% overall coral cover with very low macroalgae abundance (4%). A total of 28 coral genera from 12 families have been recorded with Porites the most dominant genus in the coral assemblages. Most reef sites in Balok are categorized under conservation class 1 (CC1), which indicated that resilient and survival of the reef rely on stress-tolerators taxa. The data presented here showed that Balok reef is highly resilient towards constant exposure to high sedimentation and wave action.
