Short communication: Endophytic actinobacteria isolated from ginger (Zingiber officinale) and its potential as a pancreatic lipase inhibitor and its toxicity. Biodiversitas 20: 1312-1317.  Endophytic actinobacteria from ginger (Zingiber officinale Rosc.) is a bacterium that is capable of producing secondary metabolites that are the same as their hosts. This study aims to look at the potential of endophytic actinobacteria from ginger as a pancreatic lipase inhibitor and its toxicity. Endophytic actinobacteria were isolated, purified, then tested for pancreatic lipase inhibitors and their toxicity using the BSLT method (Brine Shrimp Lethality Test) and phytochemical tested on ethanol extract of selected isolates. Seven endophytic actinobacterial isolates were isolated from the ginger rhizome. The isolates had different morphological diversity based on colony and microscopic observations and 5 isolates had pancreatic lipase inhibitor activity. The highest inhibitors were found in AJ4 isolates (89.9%), compared with pancreatic lipase inhibitors crude extracts of ginger (68.9%) and orlistat (88.1%) as positive controls. The LC50 value of AJ4 isolates was 653,381 ppm and the value of LT50 was 17,569 hours. AJ4 isolates contain terpenoids, phenols, tannins, flavonoids, alkaloids, and saponins. This research data is considered as new information about the potential of endophytic actinobacteria from ginger as pancreatic lipase inhibitors and their toxicity.

Biodiversitas 20: 1839-1845. Wildlife products are valuable commodities, and wild meat is considered as premium value because it has a high value per unit weight compared with other forest products Wildlife products are commonly used for consumption purpose, generating household income and cultural reasons. Because cultural values differ among ethnic groups, it is important to understand how wildlife contributes to local livelihoods in different areas of the Bird’s Head Peninsula. Data was obtained opportunistically during fieldwork to different parts of West Papua Province from 2010 to 2018. Study sites were not intentionally selected, but they were considered during the involvement of authors in research and survey across the Bird’s Head Peninsula of West Papua Province Data was collected through indirect observation and semi-structural interviews. A literature review was also completed to supplement the primary data collected from the fieldwork. The results indicated many people rely on wild meat as an alternative source of food for the household. Wildlife also offered forms of income generation such as sale of bushmeat products. Acquisition of animal parts as cultural artifacts, for personal adornment or for trophies was still a widespread practice throughout the areas. Religious and cultural aspects also contributed significantly to the use of wild meat for traditional or spiritual aspects as well. Finally, some reasons have been identified that derive peoples’ attitudes in using wildlife across the Bird’s Head Peninsula, and they are for consumption, sale and the relationship between wild animal species and cultural values.

(Tunicata, Ascidiacea) colonies growing over corals in Raja Ampat Islands, Indonesia. Biodiversitas 20: 636-642. Indonesia is at the center of the Coral Triangle, the region with the world’s highest marine biodiversity. The Raja Ampat archipelago in east Indonesia has one of the oldest networks of marine protected areas in the country and is a top priority area for marine conservation. The area is however under anthropogenic pressure from growing tourism, developments and exploration of natural resources. The most likely associated introduction of non-native species is however largely unexplored. Colonial ascidians or ‘sea squirts’ comprise a high number of species, many of them reported as introduced or invasive worldwide. In this study, we investigate the presence of white colonial ascidian colonies noticed to overgrow sections of the coral reefs in central Raja Ampat. We use DNA barcoding to address the colonies’ species identification and explore haplotype diversity to determine the species native or introduced status. We produced 22 DNA barcodes belonging to four potential cryptic Didemnum sp. species present in the Raja Ampat archipelago, Indonesia. Overall, the high number of haplotypes found in the area suggest these to most likely to be native species. The present work represents, as far as the authors are aware, the first time that such species were investigated in Raja Ampat. We hope with this work to create awareness for the topic of introduced and invasive species in the area and motivate further studies in Indonesia.

Multi-dimensional rapid appraisal technique for evaluating the sustainability of energy plantation forests in East Lombok District, Indonesia. Biodiversitas 20: 1027-1033. The increased use of woody biomass as a renewable energy source should be supported by development of sustainable energy plantation forest. This study aims to evaluate the sustainability status of energy plantation forest in East Lombok District and to determine the leverage attributes affecting sustainability. The evaluation was carried out using rapid appraisal approach, consisting of multi-dimensional scaling (MDS) analysis to provide sustainability indices for ecological, economic and social dimensions, followed by Monte Carlo simulation to define the validity. Dominant attributes affecting the sustainability were assessed by sensitivity analysis. All analyses were executed using the R software. The evaluation of eight locations generally results in a quite sustainable status for ecological, social, and economic dimensions with the validity index values of 72.73%, 61.07%, and 58.49%, respectively. To maintain or enhance sustainability, some leverage attributes need to be prioritized for policy intervention. In the ecological dimension, increasing species diversity using some potential energy woody species should be prioritized. In the social dimension, optimizing the role and capability of forest farmer groups and their members is needed, while for the economic dimension, government should support the expanding area of energy plantation forest managed by each farmer.

Abstract. Nasution Y, Rasyidin A, Yulnafatmawita, Saidi A. 2019. Evaluation of Salacca sumatrana as soil conservation crop in South Tapanuli, North Sumatra, Indonesia. Biodiversitas 20: 664-670. Salacca sumatrana Becc (snake fruit; or ‘Salak' in local name) is a profitable commodity. Morphologically, it is characterized by low, wide canopy that is potential intercept rainfall, while its roots are able to withstand soil erosion. This study is aimed to evaluate one variety of S. sumatrana named ‘salak sidimpuan" as commodity crop soil conservation based on its capability to intercept rain and trap soil sediment in the catchment area of S. sumatrana field. The research was conducted on mineral soils at West Angkola Sub-district, South Tapanuli District, North Sumatra Province, Indonesia with an altitude of 350-880 m above sea level. The study was carried out from October 2016 until March 2017 using survey method to calculate on rainfall interception, stemflow and throughfall while measurement of sediment used evaporation method. Statistical analysis was used to observe the difference of sediment trapped on the catchment area of S. sumatrana agroforest and the catchment area of mixed forest using Mann-Whitney Test. The results show that rainfall interception on S. sumatrana agroforest is 74.33% of the average rainfall while the Mann-Whitney Test Sig (2-tailed) is 0.000, indicating significant difference in trapping soil sediment between S. sumatrana agroforest and mixed forest. The amount of soil erosion on the catchment of S. sumatrana agroforest is 15.12 ton/ha/year while the erosion on mixed forest is 194.85 ton/ha/year. Thus S. sumatrana plant is capable to withstand rain interception with the result that soil erosion can be held in the catchment area of S. sumatrana agroforest. Instead of the calculation rain interception and soil erosion, then S sumatrana is classified as a soil conservation crop.

Biodiversitas 20: 2119-2127. Air Telang Protected Forest (ATPF) is one of the protected forests in the coastal area of South Sumatra, Indonesia which is around 12,660.9 hectares. This area is strategic because it borders directly with the Bangka Strait and adjacent to Sembilang National Park making it easy to access by the community. Various anthropogenic activities in the region such as residential, farming, agriculture, aquaculture, port and timber harvesting have led to degradation and loss of primary mangrove forest. This study aims to analyze the species composition and community structure of mangrove vegetation in the ATPF. The data were collected through observations at several point-centered samplings which were used to analyze the species composition and community structure of the vegetation and the physicochemical conditions of the environment. The species composition of this area consists of 20 species belonging to 14 families. The dominant species of tree, sapling and seedling stages are Nypa fruticans, Rhizophora apiculata and Acrostichum aureum, respectively. Overall species diversity index in this region is classified as very low, ranging from 0.00 to 0.73. Cluster analysis showed three types of vegetation structures in this region, namely Cyperus-Acrostichum type (shrub vegetation), Acrostichum-Rhizophora type (secondary forests) and Nypa-Avicennia (primary forests).

Abstract. Siregar M, Helmanto H, Rakhmawati SU. 2019. Vegetation analysis of tree communities at some forest patches in North Sulawesi, Indonesia. Biodiversitas 20: 643-655. Deforestation has caused a decline in forest area in Indonesia. Now natural forests is left in the conservation areas while those outside conservation areas are narrow and fragmented. This study aims to analyze forest vegetation and conservation status of the species of trees in some forest patches in North Sulawesi. Diversity, structure and type of tree communities at five forest patches, namely in the villages of Bukaka, Garini and Lambak in East Bolaang Mongondow District, and Soyowan and Basaan villages in Southeast Minahasa District, North Sulawesi Province were studied using a point center quarter method. The total number of tree species (dbh ? 10 cm) across five patches is 84 species. The highest number of species is in Garini (37 species), followed by Lambak (25 species), Soyowan (19 species), Bukaka (10 species) and Basaan (7 species). The species diversity index also shows the same pattern with such species richness, but the dominance index demonstrates opposite trend. The highest evenness index (E) is found in Bukaka (E = 0.9524), followed by Soyowan (E = 0.9061), Garini (E = 0.7873), Basaan (E = 0.7809) and Lambak (E = 0.7099). The most important tree species based on the Species Important Value (SIV) in Bukaka is Ficus sp. (SIV = 52.17), in Garini is Octomeles sumatrana (SIV = 25.73), in Lambak is Boehmeria cylindrica (SIV = 65.03), both in Basaan and Soyowan is Dracontomelon dao with SIV of 96.87 and 46.46 respectively. The beta diversity analyzed based on species similarity index of Jaccard and Whittaker 's index shows a relatively large change in species composition from one location to another. The family of Moraceae, Malvaceae, Anacardiaceae, Annonaceae and Lauraceae are listed as the widest family with a relatively high population. The highest tree density is found in Bukaka (721 trees ha-1), followed by Lambak (482 trees ha-1), Basaan (439 trees ha-1), Garini (292 trees ha-1) and Soyowan (164 trees ha-1). The highest basal area is also found in Garini (53.59 m2 ha-1), followed by Basaan (51.62 m2 ha-1), Lambak (30.74 m2 ha-1), Soyowan (25.13 m2 ha-1) and Bukaka (20.98 m2 ha-1). The results of Cluster analysis using data of species abundance indicate that the five research locations tend to form two types of tree communities. The first community consists of Garini, Soyowan, Lambak and Bukaka, while the second community is represented only by Basaan.

Abstract. Chaiphongpachara T. 2019. Short Communication: A checklist of the mosquito species (Diptera: Culicidae) in the Suan Phueng District, Ratchaburi Province, Thailand. Biodiversitas 20: 468-473. Mosquito is a medically important insect, which is a vector to transmit pathogens to humans. There are several methods to reduce the numbers mosquitoes, which requires entomological knowledge. It is critical to know the species of mosquitoes in the area to choose the most suitable method to provide vector control, specifically, to target mosquito species. In this study, we investigated the species of mosquito vectors in the Huay Nam Nak Village in the Suan Phueng District, Ratchaburi Province, Thailand, which is an endemic area of mosquito-borne diseases, especially malaria and dengue fever. Mosquito species were collected by the Mosquito Magnet® Independence trap from the Huay Nam Nak Village. A total of 1,002 mosquitoes, divided into eight species in four genera, including Anopheles barbirostris s.l. van der Wulp (118 individuals), An. subpictus s.l. Grassi (127 individuals), Culex quinquefasciatus Say (186 individuals), Cx. vishnui Theobald (204 individuals), Cx. whitmorei Giles (198 individuals), Aedes aegypti Linnaeus (45 individuals), Ae. albopictus Skuse (78 individuals), and Armigeres subalbatus Coquillett (46 individuals). This checklist of the mosquito species is very important information to set mosquito control measures that suit each area.

Isolation and characterization of endophytic bacteria from roots of Piper nigrum and their activities against Fusarium oxysporum and Meloidogyne incognita. Biodiversitas 20: 682-687. The endophytic bacteria from spices plants have potential as biocontrol agents. Nevertheless, their potential has not been explored. Pepper plant (Piper nigrum L.) is linked with promising endophytic bacteria as biocontrol agents. This study aimed to obtain isolates of endophytic bacteria from root tissues of pepper plant. The isolation of endophytic bacteria was done using surface-sterilization method and using Tryptone Soya Agar (TSA) medium. The result showed that 10 endophytic bacteria were successfully isolated. We found that a total of 9 bacteria were safe for plants and mammals and used for subsequent steps. The selected bacteria showed inhibition activity of Fusarium oxysporum (18.6 to 43.7%). Furthermore, the secondary metabolites of the endophytic bacteria also promoted lethal effects on Meloidogyne incognita (16.6 to 65.8%). The physiological activity also showed that seven isolates were able to produce chitinase, and four (4) isolates were able to produce protease. Additionally, 55.6% of isolates were also able to dissolve phosphorus and fix nitrogen. This study provided fundamental information related to the biocontrol properties of endophytic bacteria isolated from pepper plant roots.

Isolation and identification of caffeine-degrading bacteria from soil, coffee pulp waste and excreted coffee bean in Luwak feces. Biodiversitas 20: 1580-1587. The present study deals with the isolation and identification of caffeine-degrading bacteria obtained from the caffeine contaminated environment or caffeinated wastes. These bacteria are useful for various biotechnological applications especially in increasing the potential utilization of caffeinated wastes and producing the high-value chemicals. The suspected caffeine-degrading bacteria have been isolated from the soil of coffee plantation area, coffee pulp waste, and the excreted coffee bean in fresh feces of Luwak (Paradoxurus hermaphroditus or Asian Palm Civet) by growing them on the caffeinated agar medium (CAM) containing basal salt medium (M9) and caffeine as a sole source of carbon and nitrogen. CAM-supplemented with 1.5 to 10 g L-1 of caffeine has been used for screening of the potential bacteria which able to grow in high caffeine concentration. Molecular identification based on 16S rRNA gene sequence was performed to identify the selected bacteria. The result revealed that there were 11 and 3 strains of 12 selected bacteria which could grow on the CAM-supplemented with caffeine up to 7 and 10 g L-1, respectively. Based on 16S rRNA gene sequence and phylogenetic analysis, those bacteria were from 5 Gram-negative species, namely Pseudomonas japonica (4/12), Methylobacterium populi (5/12), Raoultella ornithinolytica (1/12), Klebsiella quasipneumoniae (1/12), and Stenotrophomonas chelatiphaga (1/12). Further investigations to determine their metabolic pathway, enzyme, and growth kinetics in the caffeinated medium may provide insights into its possible utilization for scientific or other applications.
