The land suitability assessment for sandalwood is very important for the successful cultivation and development of sandalwood (Santalum album Linn.) in Sumba Island, East Nusa Tenggara Province, Indonesia. The map of land suitability classification for sandalwood is currently available. The land suitability assessment is determined based on designated indicators and predictors. It is important to identify the most influential measures in a particular area to determine the suitable treatment to be applied to the sandalwood plant, as well as to recognize the land suitability evaluation swiftly and efficiently. This study was aimed to determine the variables of indicators or verifiers that have the strongest relationship with the land suitability class for sandalwood. Regression method was used to develop the relationship model and to determine the level of the relationship through the coefficient of determination. The result shows that P2O5 content was the most influential factor to determine the land suitability class for sandalwood in Sumba Island.

Diversity of bird species in the coffee agroforestry landscape: Case study in the Pangalengan Sub-district, Bandung District, West Java, Indonesia. Biodiversitas 21: 2467-2480. Traditionally, shade-grown coffee agroforestry is home to diverse taxa, including insects and mammals. However, research has shown that shade-grown coffee agroforestry is also an important habitat for various types of birds. A study on the diversity of bird species was performed in the coffee agroforestry landscape area of Perum Perhutani, Pangalengan Sub-district, Bandung District, West Java. Bird diversity data was collected using the point count method, and landscape structure analysis was performed, to quantify the characteristics of the landscape. The results were analyzed using Pearson's correlation analysis to determine the relationships between landscape structures and bird species diversity. The results of the study recorded 60 species of birds, from 29 families, among 1,581 individuals. The most dominant bird species were Pycnonotus aurigaster, Orthotomus sutorius, Orthotomus ruficeps, Streptopelia chinensis, and Brachypteryx leucophrys. The highest species abundance value at the research site was recorded for Pycnonotus aurigaster, with a relative abundance value of 22.02%. The diversity of bird species found at the study location was classified as high (H ' = 3.10). Thus, coffee agroforestry landscapes are associated with bird diversity, and diversity appears to increase when the total area (TA) increases; however the patch edge length (TE), number of spots (NP), the complexity of patch forms (MSI, MPFD) and landscape heterogeneity (SHDI) decrease. The coffee agroforestry landscape may represent an important habitat for many bird species.

Abstract

The whale shark (Rhincodon typus) inhabits Cenderawasih Bay at the Birdhead Seascape of Papua in high frequent sightings, but little is known about its genetic characteristics and connectivity. The study was conducted to characterize of the R. typus COI gene fragment from Cenderawasih Bay, in order to accurately compare with the 27 COI gene fragment sequences from the global whale shark deposited in the GenBank. A total of 28 meat samples of whale shark were collected by biopsy punch on a pole spear. The meat samples were extracted to obtain whole genome then amplified and sequenced to obtained COI nucleotides. The determined COI is a 669 bp, comprising of A = 26.5%, T/U = 30.5%, C = 28.3%, dan G = 14.7%. In total, 41 cutting sites of all the sequences ranged in size from 5-7 bp long. One COI single nucleotide polymorphisms and 2 haplotypes were identified. Single site substitution change from T to C for both haplotypes was observed. Overall, the haplotype diversity (Hd= 0.137) and nucleotide diversity (?=0.0002) were relatively low. A different nucleotide composition, number and arrangement of the sequences with the whale shark COI sequences from Cenderawasih Bay compared to COI gene fragment sequences from GenBank were observed. This study shows the availability of sequence of whale shark will contribute to aid further studies of molecular systematics, phylogeography, genetic differentiation, and conservation genetics.

Biodiversitas 21: 3108-3116. Bambara groundnut (Vigna subterranea L. Verdc.) is legume of African origin overlooked in Indonesia. It has underdeveloped in Indonesia; for example, in East Java, it is only cultivated in Gresik, Lamongan, and Bangkalan of Madura. This plant can potentially be developed in dry lands, such as Madura as it has the ability to grow and develop well in a dry environment with low nutrient level. At present, there are sparse researches on the selection and determination of the drought-tolerant character descriptors of bambara groundnut in Indonesia. The present study used the expected lines of bambara groundnut selected from local lines of various regions in Indonesia using the nested design. The results showed that the drought stress treatment led to stunted growth of 12 bambara groundnut genotypes, including the number of leaves, plant height, canopy diameter, leaf thickness, number of flowers, number of stems or branches, number of internodes, length of leaf stems, root length, root wet weight, canopy wet weight, canopy dry weight, root dry weight, and leaf chlorophyll content. However, drought leads to a slight increase in the width and length of the stomata opening and leaf proline contents. The cluster analysis based on stress index and sensitivity index can classify 3 expected lines originating from Gresik Regency (G1, G2, G3), falling into the drought stress-tolerant category. Accumulated proline contents cannot be used as a descriptor of tolerance to drought stresses in bambara groundnut since the expected lines with an increase in proline contents in leaves during drought stresses based on the cluster analysis do not fall into the genotypic cluster tolerant to drought stresses.

Abstract. Tan WA, Teja HS, Stephane. 2020. Ramularia mali strains isolated from petroleum product-contaminated soil are capable to grow on multiple aromatic compounds. Biodiversitas 21: 3590-3595.  Aromatic compounds are present as a complex mixture in the environment, some of which are toxic and carcinogenic in humans and animals. In this study, we reported the first instance of arthroconidial yeast growth on more than one aromatic compound. Yeasts were isolated from soil surrounding gas stations and screened for growth on phenol, followed by qualitative growth testing on chlorobenzene, aniline, toluene, 2-nitrotoluene, benzoate, and naphthalene. Pure isolates identity and phylogeny were determined based on the ITS region. All isolates grew on phenol and benzoate, but none grew on naphthalene. Aside from the inability to grow on naphthalene, SR8 grew on other tested aromatics, while SR3 showed similar growth ability except on 2-nitrotoluene. Growth on 2-nitrotoluene was observed for PPS3, PB4, SR1, SR6, and SR8, a phenotype that has yet been reported in yeasts thus far. All isolates shared 99.81-100% similarity and were phylogenetically clustered with the arthroconidial yeast Ramularia mali CBS 129581, despite that some of them were morphologically different and had varying capability to grow on tested aromatics. Therefore, ITS sequences cannot be used to differentiate Ramularia mali up to the strain level. Overall, we demonstrated the diverse potential of yeast strains in reducing aromatic compound contamination.

Anatomical profile and genetic variability of sweet potato (Ipomoea batatas) cultivars in Banyumas, Central Java, based on RAPD markers. Biodiversitas 21: 1755-1766. Indonesia has developed a food consumption diversification based on local sources. Sweet potato (Ipomoea batatas) is one of the tubers of carbohydrate sources in addition to wheat, rice, corn, potatoes, and cassava. It has a wide distribution, and high cultivars diversity produces a high variation on its morphology, anatomy, and genetic modification. The objectives of this research were to investigate the anatomical profile and genetic variability of eight sweet potatoes (Ipomoea batatas) cultivars found in Banyumas, Central Java, Indonesia. Anatomical profile was observed by the embedding method and subjected to analysis of variance. DNA amplification using Random Amplified Polymorphic DNA (RAPD) method with 10 primers, namely OPA-1, OPA-2, OPA-3, OPA-4, OPA-9, OPA-11, OPA-13, OPA-15, OPA-16, and OPA-18. The result of the anatomical study showed that eight sweet potato cultivars have significantly different (p<0.05) variation in leaf anatomical profile. RAPD primers produced 52 amplified fragments varying from 100 to 1300 bp in size, and 90.40% of the amplification bands were polymorphic. The genetic similarity level is ranged from 0.37 to 0.93. Two unspecific groups were forming at a coefficient of 55% from the dendrogram. The first group consisted of one cultivar, Cangkuang, and the second group consisted of seven cultivars Antin, Ungu Tua, Borobudur, Sukuh, Sari, Beta, and Kidal. The nearest relationships cultivars were Borobudur, and Ungu Tua had the highest similarity coefficient of 93%, and the lowest similarity found in Cangkuang and Antin with a coefficient of 37%. The range of genetic distance of eight cultivars was from 0.37-0.93. Implementation of the research would particularly useful for the identification and evaluation of the genetic improvement of sweet potato cultivars.

Biodiversitas 21: 1476-1482. Increased production can be done by increasing the planting area, but the available land is sub-optimal land with drought stress. Drought-tolerant rice varieties are needed to utilize this land. To create drought-tolerant rice can use induction mutation and in vitro selection methods. This study aims to obtain tolerant upland rice mutants through mutation and in vitro selection. The parents used were Batutegi and Situpatenggang varieties. This research consisted of several main activities, namely callus induction, determination of LD 50% value ofPEG determination of LC 50% value of gamma-ray irradiation, mutation induction and in vitro selection, bud regeneration and acclimatization. The results of this study are mutant callus formed by gamma-ray irradiation at a dose of 24.68 Gy for Situpatenggang and 22.15 Gy for Batutegi (LD50) and selected on PEG media at a dose of 24.11% for Situpatenggang and 25.18% for Batutegi (LC50). The mutant callus regenerated on MS + BA 3 mg/L + Zeatin 0.1 mg/L produced 83 Situpatenggang shoots and 73 Batutegi shoots and successfully acclimatized were 52 Situpatenggang lines and 49 Batuteg linesBatutegi

Trichoderma virens-Tv4 enhances growth promoter and plant defense-related enzymes of mungbean (Vigna radiata) against soil-borne pathogen Rhizoctonia solani. Biodiversitas 21: 2410-2419. Trichoderma virens has been studied for its ability to control various soil-borne pathogens as well as to induce plant resistance. The ability of T. virens control R. solani and its capability to induce resistance was evaluated in two different genotypes of mungbean (Vigna radiata (L.) R. Wilczek). Plant growth-promoting capability and production of plant defense-related enzymes during plant-pathogen-Trichoderma interaction were investigated. Pathogen infection caused the morphological and biochemical changes as well as increased plant defense enzymes activity such as peroxidase, polyphenol oxidase, PAL, phenolics, and flavonoid compared to control uninoculated plants. T. virens improved mungbean seedling growth in terms of increased total biomass, root weight, and root length as well as improved chlorophyll content and IAA-synthase from leaves and roots. T. virens treatment alone or in the presence of pathogen-induced mungbean defense-related enzymes indicated by the increasing of PO and PPO activity, and higher accumulation of total phenolic and flavonoid content. Although most of plant induced resistance parameters showed low and non-significant in direct single-factor comparison, Pearson’s correlation showed there was a positive correlation between plant growth promoter compounds (IAA and chlorophyll) with plant defense-related enzymes (total phenol, and flavonoid content). T. virens treatment can induce systemic defense response of mungbean seedling directly by increasing the activity of some defense-related enzymes, and indirectly by improving plant health, and promoting plant growth. In conclusion, T. virens-Tv4 has potential to be developed as bio-control agents to control R. solani as well as to induce mungbean resistance.

Species density and shell morphology of gold ring cowry (Monetaria annulus, Linnaeus, 1758) (Mollusca: Gastropoda: Cypraeidae) in the coastal waters of Ambon Island, Indonesia. Biodiversitas 21: 1391-1400. The purpose of this research was to examine environmental factors, species density, and the shell morphology of Monetaria annulus. The samples were collected by using the line transect method at 6 research stations. The data of the environmental factors (temperature, salinity, and pH of seawater) as well as species density of M. annulus were collected with in-situ technique, while the shell morphology of M. annulus was measured at Laboratory of the Deep-Sea Research Center, Indonesian Institute of Sciences (LIPI) Ambon. The results of the measurements of the environmental factors in the coastal waters of Ambon Island showed some fluctuations in the measurement values. The seawater temperature ranged between 29.78-30.05 ºC, the salinity of seawater ranged from 32.12-32.33‰, and the pH of the seawater ranged from 8.34-8.49. The density of M. annulus at the research stations from the highest to the lowest was at Tanjung Tiram station, Suli station, Seri station, Hutumuri station, Halong station, and Latuhalat station (0.27 > 0.24 > 0.23 > 0.19 > 0.14 > 0.12 ind/m2). The results of the measurement of the shell morphology showed many variations in the size and shape of the shell of M. annulus with a total number (N) of 294 individuals. The length, the width, and the height of the shell, the length of the blotches, the width of the blotches, and the total weight of M. annulus from each research station were significantly different (p<0.005). These results indicate that the differences in the shell morphology of M. annulus at each research station were influenced by waves, the strength of the currents sea waves, and the different substrate of water. In addition, the variation of shell morphology can be a reason for characterization in the taxonomy of the Cypraeidae family.

Identification Michelia alba barks extract using Gas Chromatography-Mass Spectrometry (GC-MS) and its antifungal properties to inhibit microbial growth. Biodiversitas 21: 1541-1550. Fungicides are substances that inhibit the growth or kill the pathogenic fungi. A substance can be categorized as a fungicide if it shows antifungal activity. This activity is resulting from bioactive compounds derived from the secondary metabolites. Michelia alba bark’s extract could inhibit the growth of Curvularia verruculosa, the cause of leaf spot disease on rice. The purpose of this study was to analyze chemical compounds of bark extract which inhibit the growth of C. verruculosa. The extraction of bark metabolites of M. alba was done using methanol as the solvent. The inhibitory test of the extract was carried out by the diffusion well method on Potato Dextrose Agar (PDA) medium. Analysis of chemical compounds of the M. alba extract was carried out in Gas chromatography-mass spectroscopy. The results of inhibitory test found that the M alba bark extract inhibits the C. verruculosa with the inhibition zone as wide as 36 mm in diameter. The majority of chemical compounds identified from the M. alba extract consisted of 10 compounds, namely: Hexadecanoic acid, methyl ester; 4H-tomentosine; 4H-tomentosine (compound isomer 2); 3-hydroxy pregn-4-ene-20-one; Tomentosine; 2-hydroxy tomentosine; Tert-butyl-2-aminophenylcarbamol; 2H-cyclohepta [b] furan-2-one, 3,3a, 4,7,8,8a-hexahydro-7-methyl-3-methylene-6-(2-formil-3-oxobutyl)-; Isoxanthanol and Xanthanol. To conclude, this extract is useful to treat C. verruculosa that causes leaf spot disease on rice.
