Biodiversitas 21: 3242-3248. Ehrlichia canis is Gram-negative intracellular obligate bacteria that cause ehrlichiosis, a companion vector-borne disease is a potentially fatal disease that attacks dogs. The purpose of this study was to molecular characterize and determine the features of Ehrlichia-infected blood based on the amplification of the gltA gene in Ehrlichia infected dogs from Yogyakarta, Indonesia. Blood samples were collected from 51 dog patients from the Prof. Dr. Soeparwi Animal Hospital, animal clinics, and pet shops based on the anamnesis, clinical sign, and physical examination, followed by microscopic examination, routine hematology, PCR amplification, and DNA sequencing were carried out on the blood samples. Based on positive PCR amplification and blood hematology profile examination ehrlichiosis-positive in dogs showed that thrombocytopenia case was 82.3%, anemia was 70.5%, eosinopenia was 70.5%, neutropenia was 29.4%, monocytopenia was 23%, leukopenia was 17% and lymphopenia was 11.7%. Morulae of Ehrlichia sp.was not found in microscopic examination. Molecularly, detected of E. canis using the gltA gene showed that 34% of samples were positive results. Then 5 of positive Ehrlichia samples were DNA sequenced, they showed a high homology of 100% with Hat Yai isolates (KU765199.1). There was no genetic diversity between E. canis samples in Yogyakarta.

Abstract. Addi A, Soromessa T, Bareke T. 2020. Plant diversity and community analysis of Gesha and Sayilem Forest in Kaffa Zone, southwestern Ethiopia. Biodiversitas 21: 2878-2888. The study was conducted at Gesha and Sayilem districts of the Kaffa Zone with the objective of identifying the floristic compositions, plant community types, and associated environmental factors of the forest. Stratified random sampling technique was used. A total of 90 plots were used to collect vegetation data. The plant community classification was performed using agglomerative Hierarchical cluster analysis Ward’s linkage method was applied in R-software. Species diversity and evenness were evaluated using the Shannon diversity and evenness indices respectively. The study revealed that the study area composed of 300 species that belong to 239 genera in 96 families. Asteraceae was the most abundant family followed by Fabaceae, Acanthaceae, Poaceae, Rubiaceae, and Euphorbiaceae accounting 37%, 15%, 14%, 13%, 12%, and 9% respectively. Five plant community types were identified and these were Ilex mitis-Syzygium guineense, Pouteria adolfi-friedericii-Schefflera abyssinica, Millettia ferruginea-Sapium ellipticum, Arundinaria alpina and Schefflera volkensii-Masea-lanceolata community types. Among the community types, Pouteria adolfi-friedericii-Syzygium guineense community was the most diverse whereas Arundinaria alpina community was the least diverse community. Canonical Correspondence of vegetation data analysis indicated that altitude, disturbance, slope, phosphorus, and the electrical conductivity were the environmental factors that significantly influence the plant communities. The high dependency of local communities on the forest resources is affecting the plant biodiversity. Thus, conservation of the forest through the introduction of sustainable forest management interventions including participatory forest management seems an appropriate action.

Genetic diversity and selection within natural dura oil palm accessions collected in Ghana for oil palm productivity improvement. Biodiversitas 21: 3534-3538. The genetic base of oil palm breeding stock of the Ghanaian Oil Palm Research Institute is considered narrow after two selection cycles. Therefore, 79 accessions of natural dura oil palm collected in Ghana and conserved at Oil Palm Research Institute (OPRI) were field evaluated for 18 agro-morphological traits. ANOVA revealed differences among the 79 dura oil palm accessions for 14 traits. The most heritable traits were single fruit weight (h2 = 0.85), leaf area index (h2 = 0.63), bunch weight (h2 = 0.52), total leaf area (h2 = 0.51), and fresh fruit bunch (h2 = 0.46). Genetic coefficients of variation ranged from 3.30% for frond dry weight to 68.74% for total bunch weight denoting possibility of selection of desirable traits for improvement. Five high fresh fruit bunch yielding accessions (FFB ? 10.81 t/ha) with good heritable bunch characteristics were selected for introduction in the breeding stocks. In fact, their bunch number, average bunch weight, and mesocarp to fruit were higher, in absolute terms, than the controls and trial mean obtained in the study. Although, the height increment was close to those of the controls, short progenies are expected among descendants from their selfings or crosses.

Abstract. Pitopang R, Umrah, Harso W, Nurainas, Zubair MS. 2020. Some botanical aspects and antifungal activity of Etlingera flexuosa (Zingiberaceae) from Central Sulawesi, Indonesia. Biodiversitas 21: 3547-3553. Etlingera flexuosa is a species of Etlingera endemic to the island of Sulawesi and its natural distribution is restricted to Central and South Sulawesi, where it has been utilized for different purposes. Despite the limited knowledge in the aspects of botany and its antimicrobial activities, the use of plants as a source of alternative solutions to health problems is on the rise. The objectives of this study were to obtain some information about the several botanical aspects of E. flexuosa and its antifungal activity. This research was conducted from March to December 2019 and samples were collected from the montane forest of Lore Lindu National Park (LLNP) near Sedoa Village, Lore Utara Sub-district, Poso District, Indonesia. The identification of plant specimens and extractions was carried out at the Laboratory of Plant Biosystematics, Tadulako University, Palu, Indonesia. The antifungal activity was tested using agar diffusion methods. The results showed that E. flexuosa is a perennial herb naturally distributed in Sulawesi island. It usually grows in the pristine submontane and montane forests, sometimes in light and open condition or on the slope of natural disturbed forest at the altitude of 1500 -1700 m a.s.l. The forest is dominated by Fagaceae family, of which E. flexuosa has been utilized extensively by local community of Topo Baria ethnic for various purposes such as flavor enhancer in food, vegetable, traditional medicine, and as roofing materials. It contains some secondary metabolites such as flavonoids, tannins, saponins, terpenoids, alkaloids and steroids, and also shows antioxidant activity. Conclusively, the extract of E. flexuosa could be used as inhibiting agent for the growth of Candida albicans yeast.

Abstract. Sutomo, van Etten E, Iryadi R. 2021. Short communication: Savanna-forest boundary on Mount Rinjani, Lombok Island, West Nusa Tenggara, Indonesia. Biodiversitas 22: 726-731. Seasonally dry tropical forests tend to be bordered by or are mixed with savanna ecosystems. This research investigates the location and nature of forest-savanna boundary on Mt. Rinjani and hypothesizes on potential causes of such boundary formation. The field survey locations were based on MODIS burnt area data. We made 30 plots (50 x 50 m) established along transects to obtain vegetation and environment data across boundaries. For data analysis, we use community correspondence index (CCI), vegetation composition using Importance Value Index (IVI), and Analysis of Similarity (ANOSIM) to detect differences in floristic and environmental characteristics across boundaries. Species composition in the transition zone (based on highest IVI results) comprises Ficus septica, Macaranga tanarius, Lindera sp., Engelhardia spicata, Saurauria sp., Rytidosperma penicillatum, and Athyrium sp. The Non-Metric Multi-Dimensional Scaling (NMDS) based on environmental data showed clear separation between savanna and forest, although boundaries were floristically similar to forest. Micro- and macro-environmental factors, as well as, fire disturbances, are also important features of the forest-savanna boundary on Mt. Rinjani. We present evidence of boundary dynamics in the form of forest advance on the Mt. Rinjani south-west slope.

Abstract. Djaenuddin N, Syafruddin, Patandjengi B, Kuswinanti T. 2020. Short Communication: Potential tests of plant growth bacteria for the control of Peronosclerospora philipinensis in corn. Biodiversitas 21: 3886-3892. The study was conducted at the Laboratory and Screen House of the Indonesian Cereals Research Institute (ICERI). The stages of the study were (i) potential test of bacterial isolates that have the ability to control downy mildew disease in vivo in corn and (ii) molecular identification of the selected bacterial isolates. Experiments were arranged in a Completely Randomized Design (CRD) with treatment of 24 bacteria which suspected to be growth-promoting bacteria. The parameters observed were disease intensity, percent disease suppression, plant height, chlorophyll content, and crop wet weight. The result showed that only five bacterial isolates namely, Bacillus albus strain MCCC 1A02146, Bacillus cereus strain IAM 12605, Bacillus paramycoides strain MCCC 1A04098, Pseudomonas stutzeri strain CCUG 11256, Serratia marcescens subsp. sakuensis strain KRED, have the ability to induce resistance to downy mildew disease caused by Peronosclerospora philipinensis.

The potency of actinomycetes extracts isolated from Pramuka Island, Jakarta, Indonesia as antimicrobial agents. Biodiversitas 22: 1104-1111. Actinomycetes are one of the Gram-positive bacteria which are widely distributed and produce many secondary metabolites including those known as antibiotics, antifungals, anticancer, and antimalarials agents. The secondary metabolites of actinomycetes are abundant, which include many active compounds that have been identified because of the large diversity in the actinomycetes phylum. This study aimed to identify and screen collected by Indonesian Culture Collection (InaCC) from Bojong Gede and Pramuka Island, Jakarta, Indonesia as antibacterial and antifungal agents. Primary screening was done on 16 actinomycetes isolates by well-agar diffusion method. Antimicrobial activity was tested by using micro broth dilution methods to determine minimum inhibitory concentration (MIC). Molecular identification into level genera and species was determined by 16S rRNA gene sequencing. Out of 16 actinomycetes isolates used, 4 isolates have activity against Candida albicans ATCC 10231, Staphylococcus aureus ATCC 6538, Bacillus subtilis BTCC B-612, and Escherichia coli BTCC B-614, specifically InaCC A758, InaCC A759, InaCC A760 and InaCC A765 isolates. InaCC A758 have highest antimicrobial activity against mentioned microbial with MIC value at 50 ?g/mL, 6.25 ?g/mL, 31.25 ?g/mL and 3.125 ?g/mL, respectively. Three genera were found from the samples: i.e. Streptomyces (80%), Microbispora (13%) and Nocardia (6%). Based on 16S rRNA gene identification, the active isolates of actinomycetes InaCC A758, InaCC A759, InaCC A760 and InaCC A765 were similar to Streptomyces badius, Streptomyces olivaceus, Streptomyces sanyensis, and Nocardia otitidiscaviarum, respectively. The secondary metabolites of actinomycetes extracts from Pramuka Island can be potentially developed as antifungal and antibacterial agents.

Diversity, biomass, and carbon stock of understorey plants in the rubber agroforestry and rubber monoculture systems in Central Tapanuli District, North Sumatra, Indonesia. Biodiversitas 21: 3508-3518. Understorey vegetation plays an important role in carbon stock in forest. The objectives of the research were to identify the species of understorey, to find out the amount of carbon stored in the understorey, and to compare the understorey carbon stocks in rubber agroforestry and rubber monoculture. The research was done in Sijungkang Village, Andam Dewi Sub-district, District of Central Tapanuli and Forest Products Chemistry Laboratory, Faculty of Forestry, IPB University on August-October 2016. The method used was the plant vegetation analysis of a stand and understorey. Destructive sampling method was used to harvest of understorey plant in 1 m × 1 m sample plot. The carbon of understorey was analyzed by independent sample t-test. The results showed that there were 23 species in rubber agroforestry and 15 species in rubber monoculture. The understorey species dominant of rubber agroforestry was Stachytarpheta jamaicensis and rubber monoculture was Asystasia gangetica with important value index 30.64% and 33.01%, respectively. The average understorey plant biomass in rubber monoculture was 0.94 tons/ha and in rubber agroforestry were 0.84 tons/ha, respectively. The carbon stocks of understorey in rubber agroforestry were 0.21 tons/ha and in rubber monoculture were 0.25 tons/ha, respectively. The result of t-test showed that there is not significant difference between carbon stock of understorey in rubber agroforestry and rubber monoculture.

Abstract. Hartati S, Muliawati ES. 2020. Short Communication: Genetic variation of Coelogyne pandurata, C. rumphii and their hybrids based on RAPD markers. Biodiversitas 21: 4709-4713. One effort to increase the genetic diversity of orchids is by crossing. This research aims to assess the genetic variation of a hybrid orchid obtained by crossing Coelogyne pandurata and C. rumphii and their hybrids based on RAPD markers. In this research, both parents were analyzed in three replications, while the hybrid was done in 10 replications. The study was conducted by analyzing DNA bands using RAPD markers with six primers, i.e. OPA 02, OPA 07, OPA 13, OPB 12, OPB 17, and OPD 08. Identification of the parents and their F1 hybrids showed 95.83% polymorphic bands with 43 bands measuring 200-2100 bp. The parents of C. rumphii, C. pandurata, and their hybrids showed similarity range of 0.16-1.00. The crossing of C. rumphii and C. pandurata resulted in a similarity of 0.5, shown in two large clusters. The first cluster consisted only of C. pandurata males and the second cluster consisted of C. rumphii females, together with all hybrid individuals. This study succeeded in creating new hybrids of orchids that have different characters from their parents, having a genetic variation of 23%.

Prevalence naturally infected gastrointestinal parasites and complete blood count condition on Wonosobo sheep at Wonosobo District, Central Java, Indonesia. Biodiversitas 21: 3057-3061. The aim of this study was to determine complete blood count of Wonosobo sheep infected with internal parasites on sheep farms in Wonosobo District, Central Java, Indonesia. Fecal and blood samples were taken from 130 female Wonosobo sheep. Fecal samples were individual and the numbers of worm eggs per gram of feces (eggs per gram, EPG) were counted. Complete blood count analyzed with a hematology analyzer machine and total protein were measured with a refractometer. The total average prevalence of gastrointestinal (GI) parasite infections in the Wonosobo sheep are 66.92%. The prevalences of low EPG and high EPG GI parasite infections in female Wonosobo sheep were 19.23% and 47.69%. The gastrointestinal worm species found infecting female Wonosobo sheep in this study were Haemonchus spp., Ostertagia spp., Trichostrongylus spp., Trichuris spp., Bunostomum spp., and Moniezia spp. The most prevalent GI parasites infections in  Wonosobo sheep were caused by Haemonchus spp., and the last prevalent ones were caused by Trichuris spp. Mean Corpuscular Volume (MCV), Mean Corpuscular Hb (MCH), Mean Corpuscular Hb Concentration (MCHC), White Blood Cell (WBC), neutrophil, eosinophil, and lymphocyte counts in female Wonosobo sheep with naturally occurring gastrointestinal worms that had EPG > 600 were significantly higher (p < 0.001) than those in sheep that had EPG <600. Hb, Hematocrit (HCT), Red Blood Cell (RBC), and Total Protein (TP) were significantly lower (p < 0.001) in sheep with EPG values >600 than in sheep with EPG <600. Monocyte and basophil values were not significantly different (p > 0.05) between infected sheep with EPG <600 and those with EPG >600. This study shows that gastrointestinal worm infections naturally affect the hematology and blood chemistry of female Wonosobo sheep in Wonosobo district, Central Java, Indonesia.
