Abstract. Pratiwi NTM, Imran Z, Ayu IP, Iswantari A, Wulandari DY. 2020. The phosphorus load and the variation of the trophic states of Cirata Reservoir (West Java, Indonesia) from 1988 to 2017. Biodiversitas 21: 4176-4183. Cirata Reservoir was constructed in 1987 between Saguling and Jatiluhur Reservoirs; these are the three man-made cascading lakes in West Java, Indonesia. Cirata Reservoir has been used for floating cage aquaculture activities. The high organic content from the remains of the fish pellets raised the phosphorus (P) load, resulting in the eutrophication of the reservoirs. This research was aimed to assess the P load and the variation of the trophic states of the reservoir from year 1988 to 2017. The trophic states and the nutrients’ conditions of the water were determined using Myxophycean Index. Four groups of phytoplankton were found in the reservoir during observation: Cyanophyceae, Chlorophyceae, Bacillariophyceae, and Dinophyceae. In addition, one group was found in a smaller number of observations: Eugolenophyceae. Although the community structure of the phytoplankton has changed, the Cyanophyceae was always dominant in response against the condition of nutrients, especially P. The P load is closely related to the number of floating cages in the reservoir. This evidence showed that Cirata Reservoir tended to be relatively eutrophic. In general, despite that the P load tended to fluctuate, the trophic states of Cirata Reservoir remained dynamic in relatively high conditions; showed by the oligo-mesotrophic state during its first year of operation, then increased to eutrophic and hypertrophic by the next decade. Although the P load tended to decrease, it remains in the eutrophic condition in the recent time.

Abstract

Abstract. Mahulette F, Astuti DI. 2020. The optimization of the Ambonese arrack fermentation using co-culture Pichia Polymorpha and Kloeckera javanica. Biodiversitas 21: xxxx. The fermentation process of the Ambonese arrack is still uncontrollable. As various microbes are involved in the fermentation, which causes inconsistent production of ethanol content. This research aimed to optimize the production of the Ambonese arrack using Pichia polymorpha and Kloeckera javanica in inoculum ratios of 1:1, 1:2, and 1:3. The number of yeast cells and chemical characteristics was observed every three hrs up to 24 hrs. The ethanol content of the fermented coconut sap and the arrack was measured using the titration and Gas Chromatography-Mass Spectrophotometry (GC-MS) methods, respectively. The numbers of P. polymorpha and K. javanica cells at 15 hrs of fermentation were 9.9 log CFU/mL and 10.7 log CFU/mL, respectively. The reducing sugar content decreased from 525 mg/L to 296 mg/L, while the pH from 6.46 to 4.82. The highest ethanol production rate was 1.4 mg/L.3hrs. (ratio 1:1), observed at 12 hrs of the fermentation, while the highest ethanol contents in the fermented coconut sap and the arracks were 11.3 mg/L and 300.400 mg/L, respectively. Different inoculum ratios affect the sensory characteristics of Ambonese arrack. The highest average value of sensory testing was 4.6 (ratio 1:2). Thus, the inoculum ratios of P. polymorpha and K. javanica 1:2 and 1:3 has the best quality in controlled Ambonese arrack processing.

Abstract. Siregar M, Purnomo DW, Siregar HM, Witono JR. 2020. Vegetation and ecoregion analysis at Sipirok Botanic Gardens, South Tapanuli, North Sumatra, Indonesia. Biodiversitas 21: 2526-2535. Botanic Gardens is an ex-situ plant conservation area. Enrichment of plant collections of Botanic Gardens in Indonesia is based on ecoregion types. To find out the type of ecoregion, the existing vegetation, main native species should be known. The research aimed to analyze the existing vegetation and ecoregion type at Sipirok Botanic Gardens. Existing vegetation has been carried out using a separate plot method 20x20 m which is placed on purposively in nine locations considered to have different vegetation types, namely remnant forest, young secondary forests and shrubs, rubber plantation, and grasslands. Around 66 species belonged to 45 genera and 27 families were found in vascular plants (dbh ? 10 cm). There were 66 species of sapling belonged to 54 genera and 35 families, and 110 seedlings belonged to 87 genera and 50 families. The tree species that have the highest Importance Value Index are Ficus sumatrana (PU-1), Myristica fatua (PU-3), Hevea brasiliensis (PU-4 and PU-7), Artocarpus elasticus (PU-8), and Knema cinerea (PU-9). No trees were found in young secondary forest/shrub plots and grasslands. Unlike the species dominance index, the species diversity index and species equitability index are higher in natural forest plots. Beta diversity based on Jaccard similarity index and Whittaker's index shows a relatively different species composition among plots. Cluster analysis shows the tendency of grouping in 2 types of communities, namely: a) remnant forest communities, and b) secondary communities. The natural forest community is further divided into two communities consisting of remnant forest tree species and industrial/plantation plant species also secondary tree species. Secondary communities are also further divided into secondary forest communities and grasslands. Based on ecoregion analysis using previous publications, altitude, and diversity of plant species in the study site, Sipirok Botanic Gardens is a transitional zone of the Sumatran lowland rainforest and mountain rainforest.

LAB from Sumbawa horse milk has good potential antimicrobial and probiotic agents. It is known, the study on LAB diversity based on its phenotypic characters is difficult to be distinguished. However, the development of molecular characterization based on the genotypic characteristic could be done for LAB diversity analysis. The aim of this study is to obtain the genetic diversity of LAB from Sumbawa horse milk collected from Penyaring Village and Lennanguar Village, Sumbawa, West Nusa Tenggara Indonesia. LAB strains were identified based on their genotypic characteristics, including their randomly amplified polymorphic DNA (RAPD) primers profiles and 16S ribosomal RNA (rRNA) sequences. The result of RAPD-PCR analysis showed 5 clusters of dendrograms resulted from GTG5 and LB2 primer amplification. Based on 16 rRNA sequences result, the phylogenetic tree was constructed and revealed 7 species of LAB i.e: SK 1.5, SKP K.3, SKP K.5, SKP K.9/SKP K.7/M.SKP K.3, SKL K.4, M.SKL K.1/ M.SKL K.5, and SKP K.4 belonging to the species of Enterococcus faecium, Weissella confusa, Lactococcus garvieae, Enterococcus thailandicus, Lactobacillus fermentum, Enterococcus faecalis, and Lactococcus petauri. In this study, the bacteria from Enterococcus sp., Lactococcus garvieae, and Lactococcus petauri existed as a novel of bacteria which means they have not been isolated and identified in Sumbawa horse milk compared to the previous findings.

Abstract. Ridwan, Witjaksono. 2020. Induction of autotetraploid Moringa plant (Moringa oleifera) using oryzalin. Biodiversitas 21: 4086-4093. Moringa (Moringa oleifera Lam.) is a plant with high nutritional content and has been widely used as a vegetable and health food ingredient, but its utilization and productivity have not been optimal. Increasing plant productivity can be done by increasing biomass which can be obtained through increasing plant ploidy. This study aims at obtaining tetraploid Moringa by induction using oryzalin compound. Moringa germinating seeds were soaked in oryzalin water solutions for 1, 3, and 5 days with concentrations of 0 µM (control), 15 µM, 30 µM, 60 µM, and 120 µM with 10 seeds for each treatment. We have recovered two tetraploid Moringa plants and two chimeric plants of diploid and tetraploid genomes. Immersing germinating seeds for 1 day in 15 or 60 µM oryzalin concentration effectively induced autopolyploid in Moringa. Tetraploid plants exhibit typical morphological characteristics of a tetraploid, such as low stomata density, larger stomata size, and larger leaflet size compared to the diploid counterpart. The induced tetraploid plants have significantly increased protein, fat, and calcium content compared to the diploid counterpart emphasizing their values as functional food. This polyploidy induction method can be used to produce Moringa tetraploid lines from various accessions in the attempts of developing and selecting high yielding tetraploid Moringa cultivars.

Multivariate analysis unravels genetic diversity and relationship between agronomic traits, protein, and dietary fiber in yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.). Biodiversitas 21: 5662-5671. Yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.) is a vegetable legume that possesses the highest amount of protein and total dietary fiber (TDF) content among other vegetables. However, protein and TDF content diversity are found among yardlong bean genotypes. Genetic improvement to produce cultivars with high yield and improved nutritional content is required. This study aimed to analyze genetic diversity and identify the relationship between important agronomic traits from 21 yardlong bean genotypes assessed by multivariate statistical analysis. The experiment was arranged in alpha design with three replicates, each consist of six blocks. Six pod-related important agronomic traits, protein, and TDF content were observed in this study. Total pod weight, pod weight, and pod length were found to be negatively correlated with protein and TDF content based on correlation and principal component analysis (PCA). Although negatively correlated, several variables possess a large and positive direct effect on protein and TDF content, such as the number of pod, pod weight, and days to flower. D2 analysis and Tocher’s method classify genotypes used in this study into five clusters with diverse traits value. Information from this study laid a foundation for the improvement of important agronomic traits as well as nutritional content in yardlong bean.

Abstract. Novianti V, Indradewa D, Maryani, Rachmawati D. 2020. Selection of local swamp rice cultivars from Kalimantan (Indonesia) tolerant to iron stress during vegetative stage. Biodiversitas 21: 5595-5606. Kalimantan with the large swamp area is a potential region in Indonesia to develop local rice cultivars with high tolerance to iron toxicity in tidal lowlands. This research was conducted to select iron toxicity-tolerant in local Kalimantan-rice cultivars, namely: Amas (AMS), Pandan Ungu (PDU), Kambang (KMB), Suatek Merah (SM), Siam Unus Mayang (SUM), Siam Unus Kuning (SUK), and Siam 11 Panjang (S11P). As control, two rice cultivars: Ciherang (iron toxicity-susceptible) and Inpara 5 (iron toxicity-tolerant) were also used. The seeds were grown hydroponically for 35 DAP (days after planting) in Yoshida nutrient solution containing 0 ppm (control), 100 ppm (low), and 400 ppm (high) FeS2 as pyrite treatments. Morphological analysis (plant height, leaf length and width, and leaves number) showed that PDU and KMB cultivars have better characters for tolerating iron toxicity. The highest level of chlorophyll and carotenoid contents during iron toxicity treatment was observed in KMB cultivar. AMS cultivar had the highest proline level under high iron stress. Antioxidant enzyme analysis showed that AMS, PDU, and KMB cultivars had a high percentage increase in the activity of superoxide dismutase (SOD), hydrogen peroxide (H2O2), and ascorbate peroxidase (APX) enzymes when exposed to high pyrite conditions. Based on morphophysiological characteristics, biochemical component, and enzymatic antioxidant activity, it can be concluded that AMS, PDU, and KMB cultivars are potential candidates as iron toxicity-tolerant rice.

First chromosome analysis of Thai pufferfish Pao cochinchinensis (Steindachner, 1866). Biodiversitas 21: 4309-4316. Here first analysis of chromosomes and nucleolar organizer region (NOR) pattern in pufferfish Pao cochinchinensis (Steindachner, 1866) was undertaken. Chromosomal preparations were obtained from kidney of P. cochinchinensis from Chi River basin in Thailand. Chromosomal characteristics were analyzed by Giemsa staining, Ag-NOR banding as well as fluorescence in situ hybridization (FISH) using microsatellites d(CA)15 and d(CGG)10 probes. P. cochinchinensis had 2n = 40 with the fundamental number (NF) 74, both in male and female. The karyotype exhibited 12 metacentric (m), 10 submetacentric (sm), 12 acrocentric (a) and 6 telocentric (t) chromosomes. No differentiated heteromorphic sex chromosomes were observed. NORs were located on short arms adjacent to telomere of the metacentric chromosome pair 4, which coincide with signals of d(CGG)10 probe. FISH with d(CGG)10 sequences were also displayed at the telomeres of most other chromosomes, whereas d(CA)15 repeats highly accumulated throughout almost all entire chromosomes except for centromeric regions. The results of conventional Giemsa staining presented the differentiation even the same genus. The localization of NORs on one pair of chromosomes only is a common characteristic found in many fish groups as well as other vertebrates. Mapping of two distinct microsatellites demonstrated the remarkable chromosomal diversification that characterizes evolution in the genus Pao. Both, conventional and molecular cytogenetics are excellent tools to study, and better understand chromosomal evolution, as well as to uncover biodiversity among fishes.

Abstract. Gafur A. 2020. Short Communication: Updated tabular key and improved browser-based interactive key to species of Pratylenchus Filipjev (Nematoda: Pratylenchidae). Biodiversitas 21: 3780-3785.  Pratylenchus is the genus of plant-parasitic nematodes that attack many crop plants and cause significant economic losses worldwide. Attempts have been made to control the parasitic nematodes. However, incorrect identification of species has hindered biological studies on the nematodes and has led to improper and inefficient control measures. To facilitate identification of Pratylenchus species, a prototype of a cross-platform, browser-based computer-assisted identification key for species of Pratylenchus has been developed, based on the pre-existing tabular key. This paper described updates and improvements that have been made to enhance the performance of the prototype key. Updates on the tabular key, which the browser-based key was based on, were made primarily to incorporate all-new species that have been subsequently published and new data from further reports of existing species. Illustrations of characters and character states were added to the key to help users decide which character states best correspond to their specimens. Drawings of each species were also given to provide general ideas on how the species look like.  The user interface has been revised in which species name(s) relevant to selected character state(s) was listed, instead of highlighted from a lengthy full list.

Abstract. Prihastanti E, Hastuti ED, Suedy SWA. 2020. Short Communication: Comparing the growth of stem explants between Citrus reticulata var. Tawangmangu and C. reticulata var. Garut using in vitro culture methods. Biodiversitas 21: 5845-5849. Several efforts have been made to preserve Citrus reticulata var. Tawangmangu and Citrus reticulata var. Garut as indigenous Indonesian mandarin cultivars, including in vitro tissue culture methods. This study aimed to determine growth differences of the stem explants of C. reticulata var. Tawangmangu and C. reticulata var. Garut,which planted on the same Murashige and Skoog (MS) media. The treatment groups were derived from different explants, grown in 4 separate culture bottles for 35 days at 25°C. The observed parameters included the percentage of explants indicating callus development browned-colored explants, and the contaminated explants. Among C. reticulata var. Tawangmangu explants, 23.53% indicated callus development, 29.42% were browned-colored explants, and 0% indicated contamination. In contrast, among the C. reticulata var. Garut explants, 0% indicated callus development, 7.14% brown-colored, and 7.14% indicated contamination. The stems explants from C. reticulata var. Tawangmangu showed a tendency to develop calluses, but the explants of C. reticulata var. Garut was able to support the growth of shoots. C. reticulata var. Tawangmangu and Garut mandarin stem explants showed differences of shoot growth because physiological conditions varied according to the variety.
