Biodiversitas 21: 5811-5817. Determining the diversity of freshwater fish species in Central Java, Indonesia has been confounded by temporal and spatial limitations in past studies. The Serayu Basin is a large watershed in Central Java that is believed to have high freshwater fish diversity. We aimed to determine the diversity and community structure of freshwater fish species in three tributaries of the Serayu, elucidate the factors influencing this diversity, and determine the extent to which these tributaries contribute to the total freshwater fish species richness for southwest Central Java. We conducted gill net sampling from May to August 2018 on the Tulis, Mrawu, and Kali Sapi Rivers located at Banjanegara district Sampling followed a random group technique in upstream, midstream, and downstream river sections of each tributary. In total, we observed 21 freshwater fish species. Broadly, fish species diversity was relatively low in the study area, and water temperature and dissolved oxygen were important in maintaining fish diversity. These three tributaries house 27–46% of the freshwater fish species reported for southern Central Java.

Abstract. Retnowati A, Kusumawaty A, Apandi I, Haryadi A. 2020. Short Communication: Two newly recorded species of Agaricales (Basidiomycota) from Banggai Kepulauan, Sulawesi, Indonesia. Biodiversitas 21: 5615-5621. Two species of Agaricales are reported here as new records for Indonesia, Mycena picta (Fr.) Harmaja (Mycenaceae) and Inocybe stellata E. Horak, Matheny & Desjardin (Inocybaceae). These new records will contribute to the information on mushroom diversity in Indonesia. Mycena picta is characterized by having cylindrical and olive pileus; adnate to slightly decurrent and distant lamellae; filiform, greyish-yellow to dull yellow stipe. Inocybe. stellata is distinguished by having brown conical pileus with squamules surface; adnexed or sinuate and close lamellae with 2 series of lamellulae; terete, glabrous with finely pruinose, base slightly bulbous, light brown stipe; echinulate, globose or subglobose basidiospores; metuloid hymenial cystidia and caulocystidia present. Photographs, descriptions, and line drawings of both macroscopic and microscopic features of the new records are provided herein and give a brief discussion on their taxonomy.

Abstract. Widiarso, Nurcahyo W, Kurniasih, Prastowo J. 2021. The ultrastructure changes of Haemonchus contortus exposed to bamboo leaves (Gigantochloa apus) aqueous extract under in vitro condition. Biodiversitas 22: 1-5. The ultrastructural changes induced in adult Haemonchus contortus in vitro using the aqueous extract of bamboo leaves were assessed using scanning electron microscopy (SEM). The H. contortus adult females were obtained from three groups and treatment was repeated thrice. The first group (T0) was not treated with bamboo leaves; 100% of the worms lived. The second group (T1), treated with 0.1% bamboo leaf-extract, had 50% mortality 4 h after examination. The third group (T2), treated with 1% bamboo leaf-extract, had 100% mortality 4 h after examination. Five worms used per treatment were submerged in ethanol and incubated for 24 hours. The ultrastructural changes observed by SEM revealed structural alteration of the worm surface after in vitro contact with the bamboo leaf aqueous extract and compared to the control worms. The main changes concerned the anterior end or cephalic region, cuticle surface, and vulva flap area. The structural modification of the external part of the female reproductive system was found only in vitro. The structural changes found in the worms exposed to the bamboo leaves might affect their motility and nutrition with possible consequences on their reproduction. Transmission electron microscopy may help to understand the external changes observed in H. contortus.

Dietary Bacillus NP5 supplement impacts on growth, nutrient digestibility, immune response, and resistance to Aeromonas hydrophila infection of African Catfish, Clarias gariepinus. Biodiversitas 22: 253-261. This study aims to investigate the effects of Bacillus NP5 supplementation as a probiotic on growth, immune response, and resistance of African catfish to Aeromonas hydrophila infection. Catfish with an initial weight of 6.8 ± 0.1 g were fed with different doses of Bacillus NP5 (0%, 1.1%, 1.2% B, 1.3%, and 1.4%) with three replications and reared for 60 days. The first 45 days were used to observed growth performance, and the last 15 days were used to perform challenge tests against A. hydrophila infection. The results showed that the treatment of 1.2% Bacillus NP5 results in the highest specific growth rate (2.55±0.28 day-1) and increased protein and lipid digestibility significantly (P<0.05). Treatment of 1.1-1.3% Bacillus NP5 increase amylase and lipase activity that significantly higher than 0% Bacillus NP5 treatment. Supplementation of Bacillus NP5 significantly increased the leukocyte, phagocytic index, and survival rate in African catfish after A. hydrophila infection. Therefore, the supplementation of 1.2% Bacillus NP5 in the feed increased the growth, immune response, and African catfish resistance to the infection.

Brown seaweeds have the potential to produce bioactive compounds. Bacteria associated with seaweeds are involved in the production of metabolites. Microbes may be present as a living symbiotic in association with other algae as epiphytes or endophytes. In this study, bacteria isolated from brown seaweed (Turbinaria conoides) were tested for antibacterial activity. A total of 14 bacteria were isolated, of which 6 were isolated from external tissue, while 8 from internal tissue. Results of an antagonistic test revealed that 7 isolates showed inhibitory activity against Staphylococcus aureus and only 1 isolate showed the inhibition against both S. aureus and Escherichia coli. Phenotypic and genotypic analysis showed that the symbiont bacteria was Lactobacillus plantarum.

Abstract. Wakhid, Rauf A, Krisanti M, Sumertajaya IM, Maryana N. 2021. Aquatic insect communities in headwater streams of Ciliwung River watershed, West Java, Indonesia. Biodiversitas 22: 30-41. Ciliwung is one of the major rivers flowing from Cisarua (Bogor) to Jakarta. This study was conducted to assess and compare aquatic insect biodiversity and community structure in various headwater streams with respect to different land-use types. Ten stream reaches of the first and second orders representing three different land-use types in the surrounding area (forest, tea plantation, village) were selected. At each stream reach, the aquatic insects were collected using a D-net (0.4 mm mesh). Physico-chemical properties of water were directly measured in the field. Samples were collected every two months from April until December 2018. A total of 26531 aquatic insect specimens belonging to 75 species/morphospecies, 46 families, and 10 orders were recorded. The order Trichoptera was the most abundant comprising 51.28% of the total collected insects, followed by Diptera (30.5%), Ephemeroptera (7.52%), and Plecoptera (6.25%). The highest proportion of functional feeding group was filtering-collectors (61.34%), followed by shredders (16.29%), gathering-collectors (15.32%), and predators (6.94%). Abundance of Diptera and Plecoptera was significantly higher in forest streams, while Ephemeroptera in plantation streams, and Trichoptera in village streams. The peak population of Diptera, Ephemeroptera, and Trichoptera occurred in June (dry period), while Plecoptera in December (wet period). The forest streams exhibited the highest species richness and diversity. Analysis of similarity (ANOSIM) showed a low dissimilarity among streams of the three land uses (Global R=0.217; P<0.001). The non-metric multidimensional scaling (NMDS) analysis showed that forest streams somewhat separated from plantation and village streams. The study provided important information on aquatic insect biodiversity, and indicated the need for forest area preservation in the headwater streams of Ciliwung River watershed.

Abstract. Ardiyani F, Utami ESW, Purnobasuki H, Paramita SA. 2020. Development and regeneration of somatic embryos from leaves-derived calli of Coffea liberica. Biodiversitas 21: 5829-5834. Coffea liberica is an important and potentially commercial plant with a high economic value from the Coffea genus. Therefore, the availability of planting material is needed to increase productivity and ensure the sustainability of its farming. Somatic embryogenesis is a powerful propagation method used to produce clonal plants from limited genetic material. In the present research, we have shown that C. liberica could be successfully regenerated in vitro via somatic embryogenesis from leaves derived embryogenic callus. These calli were cultured on Murashige Skoog (MS) medium added with 1 mgL-1 BAP or in combination with 2.4 D (0.5, 1.0, 1.5 and 2 mgL-1) for embryo development induction. Furthermore, the medium containing only BAP was best for embryo development induction after culturing for 12 weeks, with the highest number of cotyledonary stage embryos (17.8%) and producing a total of embryo (20.2). Following cotyledonary stage embryo were cultured on new MS medium containing 0.5 mgL-1 BAP, 0.5 mgL-1 IAA, 0.5 mgL-1 NAA only, and 0.5 mgL-1 BAP in combination with 0.5 mgL-1 IAA or 0.5 mgL-1 NAA. Interestingly, the results showed that cotyledonary stage embryos were converted into complete plants at all treatment, but the MS medium containing 0.5 mgL-1 BAP was found to be the most effective in promoting regeneration with 2.6 leaves per-plantlet and height of 5.2 mm. Based morphological analysis confirm that the development of somatic embryo from leaves-derived calli of Coffea liberica started with the formation of embryo globular, heart, torpedo, cotyledonary stages, and finally conversion of cotyledonary embryo into complete plant.

Genetic variability in the A microsatellite at SLC11A1 gene and possible implications with innate resistance against brucellosis in Algerian native goats. Biodiversitas 21: 5630-5636. Goat rearing is among the major agricultural activities practiced in Algeria. However, brucellosis represents an important threat to the goat sector and to public health. The aim of our work was to characterize the genetic variability of the A microsatellite at the 3' untranslated region (3' UTR) of SLC11A1 gene in Algerian native goat breeds as it was associated with Brucellosis genetic resistance in goat. Genomic DNA samples of 90 goats belonging to the four Algerian native breeds; Mekatia (n = 32), Arbia (n = 30), Dwarf of Kabylia (n = 14) and Mozabit (n = 14) have been analyzed. Polymorphism of region A microsatellite was determined by capillary electrophoresis identifying 8 alleles and 20 genotypes. Genetic parameters were also estimated. Polymorphic information content was PIC = 0.60, the observed and the expected heterozygosity were Ho = 0.569 and He = 0.595, respectively, and the values of FIS, FIT and FST were 0.044, 0.064, and 0.021, respectively. The A microsatellite was highly polymorphic and alleles associated genetic resistance against brucellosis in other breeds or species have been identified. These results open a promising opportunity to implement a genetic improvement program to reduce goat brucellosis spread in Algeria.

Updating of Makiling Biodiversity Information System (MakiBIS) and Analysis of Biodiversity Data. Biodiversitas 22: 211-226. Mt. Makiling is a Philippine biodiversity hotspot listed as one of the 170 conservation priority areas established by the Philippine government. With the vast amount of ecological and biological information about the flora and fauna of Mt. Makiling, the Makiling Biodiversity Information System (MakiBIS) was developed to serve as a repository for these data. MakiBIS may serve as a prototype in the development of a national BIS that will be useful in addressing the need to set a national baseline, measurable targets, and indicators to facilitate the assessment of biodiversity status and as a basis for decision-making.The MakiBIS project started in 2011 and was last updated in 2014. In 2019, a new version of the information system was updated to take advantage of current web technologies in software development. The new MakiBIS’ (MakiBIS 2.0) frontend was written using ReactJS, while its backend functions were coded using NodeJS, and the database tables were compressed and optimized for faster queries. MakiBIS 2.0 was published online and can be accessed at www.makibis.com.The MakiBIS 2.0 database was populated with biodiversity information from published botanical and faunal survey studies conducted in Mt. Makiling. Biodiversity information includes taxonomic classification, botanical description, habitat, mode of reproduction, economic use, endemism, and conservation status. The MakiBIS database was analyzed for patterns, trends, distribution, and potential threats that will be useful towards the conservation and management of these biodiversity resources.

The association of follicle stimulating hormone receptor (FSHR) gene polymorphism of on egg productivity in hybrid chicken (Gallus gallus gallus, Linnaeus 1758). Biodiversitas 22: 1221-1226. Pelung chicken genetics Improvement by selective breeding to Layer Lohmann Brown was successfully developed F1 which yielded 140 eggs for 300 days of production. BC1 chicken derived from ? Layer and ? F1 (? Layer vs ? Pelung chicken). Polymorphism on cFSHR gene promoter was potential genetic marker candidate to assist the selection. This research aims to study the BC1 chicken egg-related traits performance for 16 weeks and study the correlation of cFSHR polymorphism to egg productivity. FSHR gene promoter was showed using sanger sequencing. Chicken were grouped based on their haplotype. Chicken were maintained in battery cage for observation of egg production. The results show that there is a difference of BC1 chicken DOC weight from different egg weight. The six SNP polymorphisms exist on cFSHR gene promoter fragments on 10, 51, 59, 121, 233, 331 nucleotides and conducted 7 haplotype group. The highest egg production in BC1 chicken on TTGCYA and lowest egg production on TTGYYG haplotype. Based on the correlation test there was a positive correlation at p> 0.05 between BC1 chicken TTGCYA haplotype with egg production and positive correlation at p <0.05 between BC1 TAGTTA haplotype with egg length.
