Biodiversitas 22: 555-562. Antibiotic is the first option treatment for infectious diseases both in human and animal. However, the excessive usage and misuse of antibiotics have driven antibacterial resistances worldwide and the increasing case of antibiotic resistance leads to limited options for treatment. This study aimed to observe antimicrobial susceptibility and molecular identification of carbapenem-resistant human clinical bacteria. A total of nine isolates in this study were collected in 2020 from a teaching hospital in Indonesia. All isolates were originated from various human clinical specimens, including urine, blood, pus, and sputum. Identification using 16s rRNA-based showed that these isolates were closely related to Klebsiella pneumoniae (1/9), A. baumannii (5/9), Escherichia coli (2/9), and Lysinibacillus fusiformis (1/9). According to minimum inhibitory concentration using Vitek Automated Machine, four isolates of multi-drug resistant (MDR) bacteria were found. In contrast, five of them were categorized as extensively-drug resistant (XDR). Interestingly, all of the XDR isolates belonged to A. baumannii. These isolates were resistant to at least seven different antimicrobial classes. A comparison of partial 16s rRNA showed two E. coli had similar variance. While in A. baumannii isolates, we found one of five isolates had a different variance sequence, which suggests different clonality among this species. This study gives an insight into the prevalence of carbapenem-resistant bacteria with XDR criteria in Indonesia.

Morphological, molecular and resistance responses to soft-rot disease variability among plantlets of Phalaenopsis amabilis regenerated from irradiated protocorms. Biodiversitas 22: 1077-1090. Phalaenopsis amabilis (L.) Blume is a prominent donor for the white petal and sepal trait in Phalaenopsis breeding. However, it has an undesirable character, such as susceptible to soft-rot disease. Therefore, developing soft-rot resistance mutants through gamma irradiation could be explored. This study aimed to evaluate the variability of plantlets regenerated from irradiated and non-irradiated protocorms using morphology, stomatal size and molecular markers and to test responses of the plantlets against soft-rot disease. The plantlets were regenerated from irradiated (5, 10, 15 or 20 Gy) and non-irradiated protocorms. The results showed that P. amabilis plantlet variants were successfully identified based on their leaf morphology and stomatal size variations. A few plantlets have low stomatal densities, large stomatal size, and high chloroplast numbers, which indicated they were polyploids. Leaf disc assay for soft-rot disease response grouped most of the plantlets into very susceptible or susceptible. Moreover, four soft-rot resistant plantlets regenerated from irradiated and non-irradiated protocorms were successfully identified. The resistant plantlets were identified after three consecutive periods of inoculations with pathogens causing soft-rot disease. The evaluation also confirmed nucleotide variation in the Pto gene isolated from different levels of plantlet variant resistance responses.

Characteristics of vegetation as determinant of Timor Friarbird (Philemon inornatus) distribution in Bipolo Landscape of West Timor Island, Indonesia. Biodiversitas 22: 2617-2635. This study aims to identify vegetation factors as determinants of the distribution of Timor Friarbird (Philemon inornatus) in Timor Island. Timor Friarbird is one of the seven endemics bird species widely distributed in Timor Island, from the eastern region of Timor Leste to western region of Indonesia. This research was conducted at Bipolo Landscape, located in Timor Island, and has diversity of tropical flora diversity surrounding its Natural Tourism Park, which provides Timor Friarbird habitat. The characteristics of vegetation ecology were obtained by sampling design through Biogeography Branch's Sampling Design Tool for ArcGIS, while land use cover type was obtained by maximum likelihood method. The structure and composition of vegetation were observed through nested plot sampling, while Timor Friarbird species were assessed using the combination of point count and line transect method. The result obtained was formulated by discriminant linear regression. The vegetation parameters influencing the presence of Timor Friarbird were tree land use cover, number of trees and their species, pole land use cover, number of poles and their species, sapling land use cover, number of saplings and their species. Timor Friarbird is more likely to be present in the land cover area of poles with a minimum number of individual trees, poles, and saplings (press value Q = 6,63 ? 7). The results of discriminant analysis obtained a new group (nine variables), used collectively as a determinant factor for the presence of Timor Friarbird in their natural habitat on Timor Island

Agronomic characteristics of 30 promising lines of aromatic, red, and black rice and their antioxidant and cytotoxic effects in some cancer cells. Biodiversitas 22: 1695-1700. In our effort to increase rice production, some activities were done such as increased land production, rice planting patterns, and crossbreeding some rice with the special characteristic. We have cross-bred aromatic rice with red rice and black rice. These activities resulted in thirty lines of rice varieties. The thirty lines of rice variation were extracted with ethanol and evaluated the extracts for their phytochemicals, antioxidant activity, and cytotoxicity effect on cancer cell lines (MCF-7, HeLa, and OVK-18). The phytochemicals of thirty lines variation of rice showed variation in their contents. The line of F2BA201 extracts showed the strongest in antioxidant with IC50 2.43 ?g/mL while Vitamin C was 32.93 ?g/mL. The line of F2LD20 extracts displayed potent inhibition with value of 40.2% against OVK-18 cancer cells while 5FU as positive control revealed percentage inhibition with value of 21.2%. Based on the results, the thirty line rice variations exhibited antioxidant and anti-cancer potential.

Reproductive aspects of naleh fish (Barbonymus sp.): A native species from Nagan river, Aceh Province, Indonesia. Biodiversitas 22: 2682-2690. Naleh fish (Barbonymus sp.) is a native species in Indonesia and reproductive biology is key information to planning a better conservation strategy for this species. Hence, the present study aimed to examine the spawning season, sex ratio, and fecundity of the naleh fish harvested from Nagan Raya District, Aceh Province, Indonesia. Sampling was conducted from January to December 2016 at three sampling locations that were determined purposively based on the recommendations of local fishermen. The fish samples were caught using casting nets and gillnets at two-week intervals. A total of 761 fish samples, comprising 135 females and 626 males, with males predominating at a ratio of 1:4.6 (female: male). The gonadosomatic index (GSI) average of the females ranged from 0.58% to 4.28%, while 1.24% to 4.18% for males. The male fish first matured at 73 mm, whereas the female reached maturity at a size of 85 mm. The total fecundity ranged from 656-5725 eggs with an average of 2663.2 eggs, whereas the relative fecundity from 35.69-254.65 eggs gram-1 body weight with an average of 136.88 eggs gram-1 body weight. Our finding able to demonstrate that reproductive properties of Barbonymus sp. allow us to better estimate their spawning season either females and males, which can be useful for conservation planning and also their fecundity data was important for aquaculture development in the future.

Abstract. Moualki N, Boukrouma N. 2021. The influence of environmental factors on the distribution and composition of plant species in Oued Charef dam, North East of Algeria. Biodiversitas 22: 346-353. Identification of the primary factors that influence the ecological distribution of species groups is important to managers of Oued Charef dam in northern Algeria. This study aimed to identify main ecological species groups, describe the site conditions associated with these species groups, and the relationships between environmental factors and the distribution of ecological species groups using Ward’s cluster analysis for classification and principal component analysis (PCA). For this purpose, 50 plots (200 m2 each) were sampled using the Braun- Blanquet method. Soil samples were collected and analyzed to study soil properties. Multivariate analysis methods were used to classify and determine the relationship between species composition and environmental factors and to recognize ecological species groups. The R i386 (version 4.0.3) software was used for data analyzing. Ward's cluster analysis when applied on terrestrial species data gives three groups distinctly distributed on ordination plan. In cluster groups of terrestrial species Group (1) is dominated by Daisies chrysanthemum, Group (2) by Cynodon dactylon L, and Group (3) dominated by Fumana thymifolia. The groups of terrestrial species are readily superimposed on PCA ordination plane. The most important environmental factors associated with terrestrial species composition in Oued Charef dam communities were conductivity (EC), FSA, FSI, clay, salinity, phosphorus (PO4), TN (nitrogen), nitrates (NO3), and nitrites (NO2). While among the edaphic factors only pH showed a negative correlation to plant species this may due to the anthropogenic disturbances however further studies are needed to explore the rest of parts of the said regions. This study gives important insights on ecological relationships between plant biodiversity and soil chemical in a primary wetland ecosystem in northeast of Algeria.

Abstract. Ramona Y, Darmayasa IBG, Kusuma AANN, Line MA. 2021. Diversity of biocontrol agents, isolated from several sources, inhibitory to several fungal plant pathogens. Biodiversitas 22: 298-303. This study investigated the inhibitory potential of diversity of antagonist bacteria residing in the rhizosphere zone and mature compost to counter fungal plant pathogens. Soils collected from rhizosphere of lettuce farms in Bali-Indonesia and Tasmania-Australia, mature compost, commercial biocontrol (Dipel®), and laboratory contaminants with significant inhibition against tested fungal pathogens were used as sources of antagonist bacteria. These antagonists were isolated by applying dilution and spread method on trypticase soya agar (TSA) or potato dextrose agar (PDA), and their ability to inhibit Sclerotinia minor, Sclerotinia sclerotiorum, Fusarium spp., and Rhizoctonia solani was assessed in dual culture assays. The results showed that 67 out of more than 100 isolates had antagonistic activity in vitro against at least one of tested fungal pathogens. In the preliminary identification, Bacillus spp. or Pseudomonas spp. were found to be pre-dominant isolates. Following screening studies in a non-replicated glasshouse experiment against S. minor and S. sclerotiorum, 8 of the most promising isolates were further identified using molecular methods based on their 16s rDNA sequences aligned with those deposited at the GeneBank. These 8 isolates were identified as Pseudomonas corrugata, Bacillus megaterium, Bacillus polymyxa, Bacillus mojavensis, Bacillus pumilus, Bacillus thuringiensis, Exiguobacterium acetylicum, and Chryseobacterium indologenes.

Abstract. Widiarti R, Zamani NP, Bengen DG, Madduppa H. 2021. Morphotype diversity of Prorocentrum lima in the western part of Indonesian waters. Biodiversitas 22: 609-616. Prorocentrum lima is one of the toxic benthic dinoflagellates, known to produce various toxins, including okadaic acid and dinophysis toxins. The species have a wide range of morphological variability, and possess morphotype diversity, which makes it essential to have detailed morphology observation for identification and other purposes. However, such comprehensive observation has never been reported from Indonesian waters. This study aims to determine the morphological characteristics (including pore size and number of pores) of P. lima morphotypes, in the western part of Indonesian waters (Bintan Island, Belitung Island, Seribu Islands, and Karimunjawa Islands). The results showed three different and unique morphotypes of P. lima, namely morphotype 1, morphotype 2, and morphotype 3. Three clusters were presented by Cluster Analysis, corresponded to the three morphotypes, which were Belitung Island clusters, Seribu Islands and Karimunjawa Islands cluster, and Bintan Island cluster, respectively. These findings support distribution of P. lima and its potential risk of toxicity in Indonesian waters, which prompts the necessity of conducting future research, to avoid the negative impact.

Abstract. Ismail AS, Rizal Y, Kasim A. 2021. Identification of bioactive compounds in gambier (Uncaria gambir) liquid by-product in West Sumatra, Indonesia. Biodiversitas 22: 1474-1480. Gambier liquid by-product (GLB) is a by-product of gambier production, alleged to contain similar flavonoid compounds. Currently, there is no available information on the active components of GLB. This study, therefore, aims to identify the active compounds in GLB using Fourier transform infrared spectroscopy (FTIR), and screen for active compounds using liquid chromatography coupled to quadrupled the mass spectrometry time of flight (LC-MS/MS QTOF) positive (ESI+) and negative (ESI-) ionization modes. The results of the functional-group identification spectra showed 5 peaks, and the GLB was characterized by a hydroxyl group (OH) at 3423.22 cm-1 wavelength, indicating a phenolic compound. Meanwhile, CO2 was identified at 2360.92 and 2344.84, C=O at 1654.43 and C-H stretching at 1400.26 cm-1 wavelength. LC-MS/MS screening in ESI+ mode identified that the GLB contained 22 active compounds, while the ESI- mode confirmed that the GLB had 14 active compounds. Based on these two ionization modes, nine compounds, quercetin 3-glucoside, gallocatechin-4beta-OI, (-)-epi-afzelechin, quercetin-3-O-(2G-?-Lrhamnosyl)-rutinoside, kaempferol-3-O-?-rutinoside, quercetin-3-O-?-L-rhamnoside, epigallocatechin 3-O-P-coumarate, quercetin 3-O-(6''-acetyl-galactoside) 7-O-rhamnoside and quercetin 3-(4''-acetylrhamnoside) 7-rhamnoside were discovered. Therefore, GLB was discovered to contain 27 active compounds that all of these compounds are flavonoid group, and some of these substances are also present in the gambier product.

Abstract. Soesanto L, Mugiastuti E, Manan A. 2021. The use of alternative liquid media for propagation of pathogenic fungi and their effect on weeds. Biodiversitas 22: 719-725. Weeds cause high yield losses and are controlled by chemical, physical and mechanical control. Weed pathogenic fungi could be used as a biological control agent. However, the propagation media of them are still a higher cost, therefore, the new alternative and inexpensive media for weed pathogenic fungi propagation are required. The propagation of weed pathogenic fungi in liquid media in-vitro used randomize completely design consisted of three pathogenic weed fungi, i.e., Chaetomium sp., Fusarium sp., and Curvularia lunata and three liquid media, i.e., PDB, rice washing water, and tofu liquid waste. The in-planta tests used split-plot design used weed pathogenic fungi multiplied in the best formula from in-vitro results (rice washing water) and control as the main plot and four weeds, i.e., Cyperus kyllingia, Cynodon dactylon, Portulaca oleracea, and Ageratum conyzoides as subplot. The variables observed were conidia density, incubation period, disease intensity, number of leaves, and weed height. The results showed that rice washing water and tofu liquid waste were suitable media for the propagation of the weed pathogenic fungi. Chaetomium sp. was virulent fungi against broad leaves weeds, especially A. conyzoides. This fungus can be promoted to manufacture organic herbicides against the weed.
