Abstract. Darniati, Setiyaningsih S, Agungpriyono DR, Handharyani E. 2021. Development of monospecific polyclonal antibodies against hypervirulent Klebsiella pneumoniae. Biodiversitas 22: 99-105. The aim of the research was to produce monospecific antibodies against Klebsiella pneumoniae serotype K1 and K2. Klebsiella pneumonia isolates were recovered from pneumonic lungs of Aceh cattle. The bacteria were identified by rpoB (RNA polymerase ? subunit) PCR amplification specific for K. pneumoniae. The presence of  capsule cluster gene magA  and k2A  was used to characterize capsular serotype K1 and K2, respectively. Antisera were produced using serial immunization of New Zealand White rabbits with K. pneumoniae serotypes K1 and K2. The presence of antibodies was determined by using immunodiffusion test and purification was performed by immunoaffinity purification method. K. pneumoniae antibodies began to appear on the 14th day after the first injection and progressively intensified after the 1st and 2nd booster. The cross-reactivity between the antigen was eliminated by absorbing the antisera with the opposite antigen and several serotypes of non-K1/K2. After purification, serum protein concentrations were substantially decreased from 58.48 µg/µL and 53.99 µg/µL to 2.38 µg / µL and 3.72 µg / µL for K1 and K2, respectively). The SDS-PAGE analysis showed two bands with molecular weight at 54 kDa and 25 kDa representing the heavy and light chains of immunoglobulin G, respectively. Purified polyclonal antiseras against K. pneumoniae serotypes K1 and K2 showed high affinity and specificity for homologous antigens in immunodiffusion and direct agglutination tests, and immunohistochemical staining.

Mitochondrial DNA suggests the existence of two distinct species in Moluccas and New Guinea within Nyctimystes infrafrenatus (Günther, 1867). Biodiversitas 22: 3287-3297. Nyctimystes infrafrenatus is a widespread frog species distributed across northern Australia, New Guinea, Bismarck archipelago and in the eastern part of Indonesian archipelago. The species thus encompasses two biogeographic regions, Papua and Wallacea. We evaluated the phylogenetic relationships among the populations from Papua and Moluccas based on two mitochondrial loci (16S rRNA and Cytochrome Oxidase Subunit I). Two major subclades are recovered within Nyctimystes infrafrenatus with subclade A being represented by populations from New Guinea and northern Australia and subclade B by Moluccan populations (Halmahera and Tidore). Genetic distances (2.3-4.2% in 16S rRNA) between these subclades suggest they could belong to distinct species. Since New Guinea populations correspond to the nominal species and that Nyctimystes tennuigranulatus (Boettger 1895), currently considered as a junior synonym of Nyctimystes infrafrenatus, is available for the northern Moluccan populations, we proposed to remove this taxon from synonymy. However, samples used in this study come from northern Moluccas, further studies including samples from Ambon (where another synonym was described i.e. Calamita dolichopsis) is needed to determine the taxonomic status of the southern Moluccan population whether they are conspecific with northern Moluccan population or not.

Abstract. Simangunsong BCH, Manurung EGT, Elias, Hutagaol MP, Tarigan J, Prabawa SB. 2020. Tangible economic value of non-timber forest products from peat swamp forest in Kampar, Indonesia. Biodiversitas 21: 5954-5960. Development of alternative economy based on non-timber forest products usage is an important means to prevent forest conversion and preserve the quality of a forest ecosystem. The main objectives of this study were to identify and calculate the economic value from tangible/marketable perspective of the non-timber forest products (NTFPs) provided by peat swamp forest in Kampar, Sumatra, Indonesia. This is one of the largest remaining peat swamp forest ecosystems in the country. Seven villages located around that forest ecosystem with total forest cover area of 201,224 ha were chosen as samples. A conversion return approach was used to estimate the economic value of non-timber forest products. The results showed 18 kinds of non-timber forest products were identified, but only seven were used commercially by community. They are jungle rubber (Hevea brasiliensis), sago (Metroxylon sp.), swiftlet nest (Aerodramus fuciphagus), honey, betel nut (Areca catechu L), Agathis resin (Agathis dammara), and pandan (Pandanus amaryllifolius Roxb.) The obtained economic value of NTFPs was very low, which was about Rp 9,367.4 (or US$0.68 ha-1 yr-1). Market development of non-timber forest products is then crucial, and the government should play an important role in promoting it in collaboration with private companies. Non-marketed non-timber forest products and various environmental services should also be taken into account to measure that peat swamp forest ecosystem value. Otherwise, concerns about the sustainability of that forest ecosystem in Indonesia due to undervaluation of forest resources will continue.

Antifungal activities of the rhizome extract of five member Zingiberaceae against Candida albicans and Trichophyton rubrum. Biodiversitas 22: 1509-1513. Fungal infections have now become serious health issues. One of the strategies to avoid the problems of fungal infections is by using natural product from plants that are effective against many human pathogenic fungi. The study portrayed the use of the extracts of plant rhizomes as the alternatives to fight against number of human pathogenic fungi. This research aimed to investigate the antifungal activities of crude ethanol extract of five member of the family Zingiberaceae (Curcuma longa, Alpinia galanga Zingiber officinale. var. rubrum, Zingiber officinale var. officinarum and Zingiber officinale var. amarum), which are widely used as folk medicines against Candida albicans and Trichophyton rubrum. Crude ethanol extracts of five members of Zingiberaceae were evaluated for their antifungal activities and the results were calculated based on the zones of inhibition using the diffusion method. The extract showed antifungal activity against Candida. albicans in the agar well diffusion assay (10.2-27.1 mm inhibition diameter) and against T. rubrum (27.3-44.3 mm inhibition diameter). The data have revealed that all rhizomes have the potential to be developed as antifungal agents, particularly against C. albicans and T. rubrum. Studies on the antifungal activity against yeast-like (C. albicans) and filamentous (T. rubrum) can provide new information about the benefits of members Zingiberaceae as a source of natural antifungal. Researchers can select the type of rhizome that has more potential for further extraction to obtain pure compounds that can be used as antifungals.

Several species of mistletoe are parasitizing and infesting cultivated plants in Purwodadi Botanic Garden, Pasuruan, East Java i.e. Dendrophthoe pentandra (L.) Miq., Macrosolen tetragonus (Blume) Miq., Scurrula atropurpurea (Blume) Danser, Viscum articulatum Burm.f., and Viscum ovalifolium Wall. ex DC. The population of the mistletoes is dynamics affected by genetic and environment. The study aimed to investigate the population dynamics of mistletoe species on Cassia fistula L. in Purwodadi Botanic Garden, Indonesia. Data collection was conducted by direct observation in the garden on the mistletoe species, parasite density, and host prevalence in four block locations and 28 trees of C. fistula. Data were analyzed by two-way ANOVA and tested by Least Significantly Difference (LSD) if there was a significant difference in blocks and year. The results showed two species of mistletoe found on C. fistula, i.e. D. pentandra and V. articulatum. Parasitic density was significant different and infested host prevalence also different in blocks and years. The highest parasite density per host plant was reached in block III.D in 2015,i.e. 7.00±2.95 and 2.28±0.85 individuals per plant for D. pentandra and V. articulatum, respectively. The highest infested host prevalence of 100.00 was reached in block III.D in 2015 and XVI.A since 2012.

Abstract. Imron M, Tawaqal MI, Yusfiandayani R. 2021. Fishing ground and tuna productivity by tuna longline based on Benoa Bay, Bali, Indonesia. Biodiversitas 22: 961-968. The success of a longline fishing operation depends on several things such as the skill of the crew, bait used, fishing season, fishing operation, the total of fish caught, the price of the fish, productivity, and fishing grounds. Information about productivity and fishing ground becomes important to increase effectiveness and optimal profit. The methodology is carried out by conducting a survey to collect data to be processed and analyzed using productivity analysis based on Landing per Unit Effort (LPUE) and fishing activity analysis from the Vessel Monitoring System (VMS) to determine the Fishing Ground. We calculate tuna productivity from catch production (landing) per effort, meanwhile fishing ground use VMS data. Production tuna was landed at Benoa Bali from 2016-2018 fluctuated. In 2016 production of tuna albacore took a portion of 34.60%, yellowfin tuna 43.56%, bigeye tuna 15.44%, and southern bluefin tuna 6.26%. Production of tuna albacore took a portion of 35.61%, yellowfin tuna 42.64%, bigeye tuna 12.49%, and southern bluefin tuna 9.26% in 2017. Production albacore took a portion of 36.41%, yellowfin tuna 41.56%, bigeye tuna 11.79%, and southern bluefin tuna 10.24% in 2018. The highest productivity of albacore was in August 2018 with LPUE value 1.0099, yellowfin tuna was in July 2018 with LPUE value 1.2431, big eye tuna was in November 2018 with LPUE value 0.5538, and bluefin was at December 2017 with LPUE value 0.3864. The result of VMS data processing showed that tuna longline vessel based at Benoa has several locations of fishing grounds based on fishing activity for example Hindia High Seas, WPP NRI 714 (Telo gulf and Banda Sea), WPP NRI 718 (Aru Sea, Arafura Sea, and Timor Sea), ZEEI WPP NRI 718.

Lingula anatina is one of brachiopods found in tropical regions, however, the reports on this species from South East Asia is currently limited. This study, therefore, aims to identify the Cytochrome Oxidase subunit I (COI) sequence and phylogenetic of lamp shells from Probolinggo, Indonesia. A total of five samples of lamp shells were collected from Probolinggo, Indonesia, then identified based on morphological characters, and the identification was confirmed using molecular data. Subsequently, molecular characterization and identification were conducted based on 657 bp of COI gene similarity, sequence variation, genetic distance, phylogenetic topology, and BOLD System. According to the results, the Lingula species found in Probolinggo Indonesia was Lingula anatina. The median-joining network and Automatic Barcode Gap Discovery (ABGD) analysis showed this Lingula anatina population from Probolinggo had relatives of 22 haplotypes and divided into six haplogroups. Furthermore, the population belonged to the same haplogroup but had different haplotypes. Thus, this study provides more information for Lingula anatina phylogenetic and haplotype studies.

Short Communication: Characterization and nutrient analysis of seed of local cowpea (Vigna unguiculata) varieties from Southwest Maluku, Indonesia. Biodiversitas 21: 85-91. Cowpea (Vigna unguiculata (L.) Walp.) is a legume species that have many local varieties across regions in Indonesia, including in Southwest Maluku District. It has been utilized by the community as raw materials, but mainly for food sources of self-consumption. While there are rich local varieties of cowpea in Southwest Maluku, the data about its morphological characters and nutritional compositions are not available. The objectives of the research were to analyze morphological characters and nutritional compositions of local seven cowpea varieties from Southwest Maluku and classify these varieties according to such characters and compositions. Morphological characters observed consisted of shape, color, texture, eye pattern, eye color, size (length, width, thickness), and 100-grain weight. Nutritional composition consisted of proximate analysis tested using AOAC (Association of Official Analytical Chemist) method and folic acid content using HPLC (High-Performance Liquid Chromatography) method. The results showed that the varieties have significant variation in morphological characters and nutrition composition. There are differences in shape, color, texture, eye pattern, and color. Seed size in terms of length, width and thickness have range values of 5-9 mm, 4-6 mm, 3-4 mm, respectively while seed weight ranges 11-19 g. Furthermore, nutrition composition is significantly affected by varieties. Moisture content ranges between 11-17%, ash content of 3.13-3.97%, fat content of 0.58-1.42%, protein of 15.5-20.76%, carbohydrate of 58.46-63.48% and folic acid content of 100.13-131.57 µg/mL. There is significant correlation between morphology characters and nutrition composition. Principal Component Analysis (PCA) showed that local varieties can be classified into three clusters. Therefore, KM1, KM3, KM4 and KM7 varieties that have the highest size, weight, and nutrition composition can be selected in plant breeding.

Chrysanthemum is one ornamental plant with high potential to be developed commercially. It has many varieties such as colors, types, and shapes of flowers. Paclobutrazol (PBZ) is one of plant growth regulators used to suppress plant growth. The PBZ action mechanism is to inhibit gibberellin synthesis. The application of PBZ with a certain concentration on Chrysanthemum plants has the potential to form potted Chrysanthemum. The aim of this study was to analyze the anatomical variation of potted mums varieties, i.e. Jaguar Red, Snow White and Fiji White, and also the effect of PBZ with various concentrations on the anatomy of the stems and leaves of three Chrysanthemum varieties. The research was conducted by applying PBZ concentrations of 0, 50, 100, 150 ppm to the three Chrysanthemum varieties and grown for 8 weeks to form potted Mums. Then, the anatomy of stems and leaves was studied using the paraffin and freehand section methods. Data analysis was performed using ANOVA and DMRT (Duncan Multiple Range Test) for anatomical characters in stems and leaves of three Chrysanthemum varieties. The results showed that the length of the epidermal cells of Fiji white variety was longer than those of Snow White and Jaguar Red. Meanwhile, Jaguar Red produced greater stem thickness, stomata, and leaf thickness than those of the other two varieties. PBZ could increase the size of the anatomical parameters of Chrysanthemum stems and leaves for all varieties. The PBZ concentration of 150 ppm increased leaf thickness and stem diameter, as well as tracheal diameter. The PBZ concentration of 100 ppm increased the size of the guard cells, palisade and spongy tissue, also peripheral tissues. Meanwhile, the PBZ concentration of 50 ppm had an effect on the increase in the number of stomata and pith diameter. The optimum PBZ concentration to form potted Chrysanthemum was 150 ppm.

Abstract. Suciyati A, Suryadarma IGP, Paidi Abrori FM. 2021. Ethnobotanical study based on the five dimensions of basic life needs in Tidung Tribe of North Kalimantan, Indonesia. Biodiversitas 22: 3199-3208. The Tidung tribe is one of the original inhabitants of North Kalimantan, Indonesia. Research results related to the Tidung tribe, especially those related to ethnobotany, are still rare. This research aimed to make an inventory of plant species used by Tidung Tribe based on the five dimensions of basic life needs of Tidung Tribe. The five dimensions of basic life needs of Tidung Tribe is that plants are useful as food (ngakan), shelter (baloy), health/medicine (sihat), traditional ceremony (adat), and clothing (memana). Informants were obtained by conducting snowball sampling consisting of 22 family heads and as many as 65 people. There were three instruments in the data collection: questionnaires of list of plant species, quantitative plant assessment instruments, and semi-structured interviews. According to the inventory result, Tidung Tribe utilize 60 plant species in their daily life. Most of these plants are used as food (ngakan) and traditional medicine (sihat). The Availability Index and Frequency Use Index data showed that Arecaceae, Pandanaceae, Solanaceae, Zingiberaceae, and Poaceae families were the most widely available plants in the research location and most often used by the community.
